Rapid separation of DNA molecules by agarose gel electrophoresis: use of a new agarose matrix and a survey of running buffer effects.

This report describes the use of a new type of agarose (FastLane agarose) for faster separation of DNA by agarose gel electrophoresis. DNA molecules separated in this agarose exhibited electrophoretic mobilities up to 30% higher than similar separations in standard analytical grade agarose. DNA molecules of all sizes examined showed higher mobilities in FastLane agarose. The mobility increase was predominantly due to the low electroendosmosis of FastLane agarose and was most pronounced in pulsed field gel electrophoresis separations. The magnitude of mobility increase varied depending on the conditions used for electrophoresis.

Recurrent vulvovaginitis candidiasis; an overview of traditional and alternative therapies.

Vulvovaginitis is a common inflammatory condition that can significantly disrupt a woman's life. The term vulvovaginitis actually encompasses a variety of inflammatory lower gential tract disorders that may be secondary to infection, irritation, allergy or systemic disease (Table 1). This article focuses on candida-caused vulvovaginitis, a condition known as vulvovaginitis candidiasis. Approximately 75% of U.S. women expreience vulvovaginitis candidiasis during thier reproductive years. Between 40% and 50% of these women have recurrent episodes, and 5% to 8% experience chronic candida infections. Approximately 3 million women have recurrent candidial infections. The prevalence of vulvovaginitis candiasis is expected to rise due to the growing number of non-C albicans species (which are immune to most antifungal medcationa) and as a result of more widespread antifungal resistance.

[Vulvovaginal candidiasis: a therapeutic approach].

The vulvovaginal candidiasis represents, after the bacterial vaginosis, the most frequent cause of vaginal affection. It is esteemed that around the 75% of the women of reproductive age suffered from an episode of vulvovaginitis from candida and 40-45% have had more episodes, of which 10-20% in complicated form. The kind of candida more frequently isolated in the vagina of symptomatic women is the Candida albicans: in the 10-20% of the cases the agent is present in absence of symptomatology, and we can almost consider it a saprophytic. On the other hand, always with greater frequency fetterses can be isolated of not albicans Candida, particularly the tropicalis and the glabrata kind, usually resistant to the common therapies. The classification of the vulvovaginal candidiasis proposed by Sobel, and by now universally approved, foresees 2 clinical forms of vulvovaginal candidiasis, the vulvovaginitis from not complicated candida (VVC) and the vulvovaginitis from complicated candida (VVCC): different for pathogenesis, elapsed clinical, symptomatology and frequency. They have to be considered in the substance 2 different nosological entities, and they request a diagnostic approach and a well different therapeutic appointment. In this study we will shortly reassume the principal characteristics of it, detaining us on the most recent acquisitions in theme of therapy. The base medicines of ac. boric, to parity of effectiveness, seem to introduce the most contained cost and the best compliance, and they offer him to a complementary use or, in some cases, alternative to the more you consolidate therapies with azoli.

[Ion solvation and ion association in high-concentration of NaBF4/DMF solutions].

Ion solvation and ion association in solutions of NaBF4/DMF were studied by infrared and Raman spectroscopy and ab initio quantum chemistry. It was found that the sodium cation interacts on oxygen atom of C=O group in DMF molecules. The solvation number of Na+, four, was obtained according to the change analysis of CH3 rocking band. The splitting of the nu1 mode of BF4(-) anion indicates the presence of contact ion pairs. Ionic association degree increases with increasing molality of the salt. The solvated structure of Na+ and the geometry of Li+ BF4(-) ion pair were also suggested based on the ab initio calculations.

[Quantitative analysis of cantide in plasma by capillary gel electrophoresis].

Cantide is a 20-mer antisense phosphorothioate oligonucleotide that inhibits telomerase catalytic subunit hTERT, pharmacologic results showed that it had promising antitumor activity. In order to study the pharmacokinetic properties of Cantide, a capillary gel electrophoretic (CGE) method with internal standard was used for the determination of Cantide in rat plasma. Cantide and the internal standard had approximately equal percentage of base composition. Extraction of the phosphorothioate oligonucleotides from plasma was accomplished using two solid-phase extraction columns, a strong anion-exchange column to remove plasma proteins and lipids, followed by a reversed-phase column to remove plasma salts. A second desalting step, achieved by dialysis utilizing a membrane, was required to remove residual ionic material from the extracted sample. The size of the capillary column was 31 cm x 100 microm i.d. with an effective length of 20 cm. The running buffer was a mixture of Tris-boric acid-urea (pH 8.5). The calibration curve was linear in the range of 12.5 - 400 mg/L, with correlation coefficient (r) of 0. 999 8. Intra-day and inter-day relative standard deviations (RSDs) for the extracted samples were 0.398% - 2.46% and 2.75% - 6.07%, respectively. The range of recoveries was 99.53% - 102.1%. The results demonstrate the high accuracy, stability and reproducibility of the procedure.

[Speciation analysis of mercury by capillary electrophoresis on-line coupled with inductively coupled plasma mass spectrometry based on a micro mist nebulizer and a removable interface].

A method for mercury speciation analysis was developed by CE-ICP-MS with a Micro Mist nebulizer and a removable interface. Contamination of ICP-MS detector by the rising solution was avoided with the use of a removable interface during capillary rinsing. Baseline separation of methylmercury (MeHg) and inorganic mercury (Hg2+) was obtained by CE in a 45 cm x 75 microm i.d. fused-silica capillary at 22.5 kV, while a mixture of 30 mmol(-1) H3BO3 + 10% CH3OH (pH 8.7) acted as running electrolyte. The precisions (RSD, n=5) of migration time and peak area for the mercury species were in the ranges of 3.5%-3.8% and 2.3%-5.1%, respectively. The limits of detection (3sigma) of MeHg and Hg2+ were 47 and 48 microg x L(-1), respectively.

[Determination of osteopontin at trace levels by non-gel sieving capillary electrophoresis].

A method of non-gel sieving capillary electrophoresis (NGSCE) was established to determine osteopontin at trace levels. The capillary used was uncoated fused silica with a size of 57 cm x 75 microm i. d. and an effective length of 50 cm. The electrode buffer was a 150 mmol/L boric acid-borate buffer containing 30 g/L polyethylene glycol 20000 (pH 10.0). Other conditions were as follows: separation voltage 23 kV; detection wavelength 214 nm; pressure of injecting sample 3.4 kPa (0.5 psi) x 5 s; and column temperature 25 degrees C. The NGSCE method had excellent linearity with correlation coefficient of 0. 996, and reproducibility with the relative standard deviation of migration time of osteopontin less than 5% . The recovery was 95% and better, the sensitivity was 0. 079 g/L. Osteopontin secreted by vascular smooth muscle cells was determined by the NGSCE method at different times after serum withdrawal, and the results were in agreement with those of Western blot method. The results indicate that NGSCE is a simple and rapid method of determining osteopontin at trace levels. This method only needs a micro-amount of sample and is easily automated.

Biological efficacy of boronated low-density lipoprotein for boron neutron capture therapy as measured in cell culture.

Low-density lipoproteins (LDLs) are known to be internalized by the cell through receptor-mediated mechanisms. There is evidence that LDLs may be taken up avidly by tumor cells to provide cholesterol for the synthesis of cell membranes. Thus, the possibility exists that LDLs may provide an ideal vehicle for the transport of boron to tumor cells for boron neutron capture therapy. A boronated analogue of LDL has recently been synthesized for possible application in boron neutron capture therapy. The analogue was tested in cell culture for uptake and biological efficacy in the thermal neutron beam at the Brookhaven Medical Research Reactor. It was found that boron concentrations 10 times higher than that required in tumors for boron neutron capture therapy were easily obtained and that the amount of uptake was consistent with a receptor-mediated binding mechanism. The measured intracellular concentration of approximately 240 micrograms 10B/g cells is significantly higher than that obtained with any other boron compound previously evaluated for possible clinical application.

Diagnostic test for mucopolysaccharidosis. I. Direct method for quantifying excessive urinary glycosaminoglycan excretion.

This direct method for quantifying excessive urinary glycosaminoglycan excretion exploits the specific binding of 1,9-dimethylmethylene blue (DMB). The procedure obviates cumbersome and labor-intensive procedures for separating glycosaminoglycans from other constituents of urine. Pediatric pharmaceutical formulations (except heparin), in concentrations expected in urine, do not interfere with spectrophotometry, nor does protein. Results can be expressed in terms of urinary creatinine; thus the test is applicable to very small urine specimens (0.1 mL), such as those obtainable from neonates. In a pilot study, results of the direct DMB test for 48 urine specimens agreed with the clinical diagnosis, and quantitative measurements correlated moderately (r = 0.76) with results of a commonly used procedure (carbazole-borate reactivity after precipitation with cetylpyridinium chloride). The present method was also used to assess metabolic correction in a patient with Hurler's syndrome after treatment by bone-marrow transplantation. This quantitative method surmounts the major technical problems of developing mass screening programs for infants, thus offering the potential for earlier diagnosis and treatment of mucopolysaccharidosis diseases.

An open multiple dose study of Optrex Eye Lotion in eye irritation due to hayfever.

Twenty-four patients consulting their general practitioner with eye irritation due to hayfever entered a seven-day open, multiple dose study of a newly formulated Optrex Eye Lotion. Patients self-administered Optrex by irrigation into their left eye three times daily for seven days, with an option to use the same preparation in their right eye if they thought this to be of benefit. Assessment was by means of daily diary cards completed by the patient each evening for the seven-day period. Following the first instillation, the treated eye felt significantly better at 20 seconds and at four minutes when compared with the untreated eye. Differences between the eyes for degree of redness, comfort and clearness of vision were not significant, but 15 patients (63 per cent) optionally used Optrex in their right eye. Seventeen patients (71 per cent) reported that they derived overall benefit from the use of Optrex Eye Lotion during the study period. Two patients reported side effects during the study but, in each case, the investigator did not consider the event to be therapy related. One patient withdrew on Day 7 of the trial due to worsening of their allergic conjunctivitis. It can be concluded that some subjective benefit was gained by the majority of patients in that a considerable number of them chose to treat both eyes for the duration of the study.
