[Contact allergy to olive oil].

Sensitization to olive oils has not yet been reported in the German literature. A 43 year old female developed increasing eczematous reactions after treatment of her leg with a boric acid/zinc oxide preparation in an oil vehicle. When different components of this zinc oxide/oil compound were tested, olive oil proved positive. A patch testing with other vegetable oils gave negative results.

Simple spectrophotometric quantification of urinary excretion of glycosaminoglycan sulfates.

We describe a simple, rapid, precise, and sensitive spectrophotometric method for measuring urinary glycosaminoglycan (GAG) sulfate excretion. The GAG sulfates are precipitated with cetylpyridinium chloride, resuspended in water, and mixed with the basic dye 1,9-dimethylmethylene blue to produce a complex with the polyanionic molecule of sulfated GAGs. Absorbance is read at 535 nm. The standard curve for reaction was linear up to 12 micrograms of the different GAGs: dermatan sulfate, heparan sulfate, keratan sulfate, chondroitin 4-sulfate, and chondroitin 6-sulfate. Within- and between-run precision (CV), measured at three different GAG concentrations (normal and pathological), varied from 1.6% to 2.5% and from 1.8% to 4.5%, respectively. Analytical recovery ranged from 71% to 107%. Urinary GAG excretion, measured by this procedure, correlates (r = 0.837; p less than 0.001) with the values obtained with the borate-carbazole reaction (Anal Biochem 1962;4:330-4).

Resistotype heterogeneity of Escherichia coli strains isolated from humans, animals and sewage waters.

The tests commonly used for bacterial identification, especially in the field of microbial environmental analyses frequently do not provide a sufficient strain differentiation. Considering the importance that accurate characterization of bacterial pollution indicators could have as epidemiological tools, this study used a resistogram subtyping method for Escherichia coli as a tentative method of effecting a good monitoring of the environmental spread of this microorganism. The resistance of 313 E. coli strains of different origin (human, animal, sewage), previously identified by standard biochemical reactions, to 8 chemical compounds (inorganic, organic and dyes) and to 7 antibiotics was tested. The results indicated this method has a higher discriminatory power for chemicals than for drugs. Some typical resistotype patterns for E. coli from various sources are described.

Microbiologic characteristics of unpreserved saline.

This study was designed to assess the microbiological risk associated with using unpreserved saline buffered with either borate or phosphate salts. The results indicate that borate buffer has measurable antimicrobial activity and can control some microbiological contamination. However, phosphate-buffered saline can become easily contaminated and usually promotes growth or maintains viability of adventitious contaminants.

Microbiologic evaluation of canine urine: direct microscopic examination and preservation of specimen quality for culture.

Urine specimens were obtained from 115 dogs. Each specimen was divided into 2 aliquots; one aliquot was placed into a sterile container (non-preserved), and the other was preserved in a boric acid-glycerol-sodium formate at 4 C. Both aliquots were then transported to a laboratory, and specimens of each aliquot were bacteriologically cultured immediately upon arrival at the laboratory. Specimens of the preserved aliquot also were cultured after 24, 48, and 72 hours of preservation. In 47 specimens containing greater than or equal to 1 X 10(4) colony-forming units (CFU)/ml, the results of quantitative bacteriologic culturing of preserved urine at 0, 24, 48, and 72 hours were the same as the result of the immediate quantitative bacteriologic culture of non-preserved urine. In 2 of 5 specimens containing greater than or equal to 1 X 10(3) CFU/ml but less than 1 X 10(4) CFU/ml, the results of quantitative bacteriologic culture of preserved urine differed from the corresponding immediate culture of nonpreserved urine. Bacteria were not isolated from 63 specimens (less than 1 X 10(3) CFU/ml). Bacteriologic cultures of preserved urine were also negative for bacterial growth at 0, 24, 48, and 72 hours. Bacteriuria was detected by direct microscopic examination of gram-stained smears of uncentrifuged urine in 50 of 52 urine specimens from dogs with urinary tract infection. Bacteria were observed in 1 of 63 specimens that did not have bacterial growth when cultured.

Toleration of artificial tears - the effect of pH.

We studied the effect of altering the pH of an artificial tear preparation on the tolerance of 20 patients with keratoconjunctivitis sicca. Of those who expressed a preference, 11/17 preferred a tear substitute more alkaline than any preparation now available in North America. We recommend an alkaline tear substitute as a valuable alternative in patients with keratoconjunctivitis sicca.

Metabolism of prostaglandin D2 in isolated rat lung: the stereospecific formation of 9 alpha,11 beta-prostaglandin F2 from prostaglandin D2.

The metabolic transformation of exogenous prostaglandin D2 was investigated in isolated perfused rat lung. Dose-dependent formation (2-150 ng) of 9 alpha,11 beta-prostaglandin F2, corresponding to about 0.1% of the perfused dose of prostaglandin D2, was observed by specific radioimmunoassay both in the perfusate and in lung tissue after a 5-min perfusion. To investigate the reason for this low conversion ratio, we analyzed the metabolites of tritium-labeled 9 alpha,11 beta-prostaglandin F2 and prostaglandin D2 by boric acid-impregnated TLC and HPLC. By 5 min after the start of perfusion, 9 alpha,11 beta-prostaglandin F2 disappeared completely from the perfusate and the major product formed remained unchanged during the remainder of the 30-min perfusion. The major product was separated by TLC and identified as 13,14-dihydro-15-keto-9 alpha,11 beta-prostaglandin F2 by GC/MS. In contrast, pulmonary breakdown of prostaglandin D2 was slow and two major metabolites in the perfusate increased with time, each representing 56% and 11% of the total radioactivity at the end of the perfusion. The major product (56%) was identified as 13,14-dihydro-15-ketoprostaglandin D2 and the minor one (11%) was tentatively identified as 13,14-dihydro-15-keto-9 alpha,11 beta-prostaglandin F2 based on the results from radioimmunoassays, TLC, HPLC, and the time course of pulmonary breakdown. These results demonstrate that the metabolism of prostaglandin D2 in rat lung involves at least two pathways, one by 15-hydroxyprostaglandin dehydrogenase and the other by 11-ketoreductase, and that the 9 alpha,11 beta-prostaglandin F2 formed is rapidly metabolized to 13,14-dihydro-15-keto-9 alpha,11 beta-prostaglandin F2.

[Effect of biologically active dressings of wounds on the course of the wound process].

A comparative analysis of the effectiveness and expediency of the local application of collocyl (128 patients), trypsin immobilized on textile cellulose and kapron matrix (186 patients) and gauze bandage moistened with a solution of enzymes and 10% sodium chloride was made. It was shown that collocyl as well as trypsin modified gauze and kapron accelerated cleansing the wounds of nonviable tissues, decreased their infectivity, reduced intoxication of the organism and improved the course of the wound process.
