Kinetic and donor stabilization of organotellurenyl iodides and azides.

The first tellurium compounds containing the extremely bulky tris(phenyldimethylsilyl)methyl (Tpsi) and 2,6-bis(2,4,6-triisopropylphenyl)phenyl (2,6-Trip(2)C(6)H(3)) moieties have been synthesized and isolated. Careful oxidation of the tellurolate TpsiTeLi (1) resulted in the formation of the crowded ditellane (TpsiTe)(2) (2), and iodination of 2 gave the alkanetellurenyl iodide TpsiTeI (3). In a similar fashion, the terphenyl-substituted ditellane (2,6-Trip(2)C(6)H(3)Te)(2) (9) and the arenetellurenyl iodide 2,6-Trip(2)C(6)H(3)TeI (10) were prepared. Reaction of the iodides TpsiTeI (3) and 2,6-Trip(2)C(6)H(3)TeI (10), as well as TripTeI, MesTeI (Trip = 2,4,6-triisopropylphenyl, Mes = 2,4,6-tri-tert-butylphenyl), and the donor-stabilized 2-Me(2)NCH(2)C(6)H(4)TeI, with AgN(3) resulted in the formation and isolation of the corresponding tellurenyl azides TpsiTeN(3) (4), TripTeN(3) (7), MesTeN(3) (8), 2,6-Trip(2)C(6)H(3)TeN(3) (11), and 2-Me(2)NCH(2)C(6)H(4)TeN(3) (12). Furthermore, the corresponding tris(ethyldimethylsilyl)methyl-containing (Tesi) tellurium compounds (TesiTe)(2), TesiTeI (5), and TesiTeN(3) (6) have been prepared but could not be isolated in pure form. The crystal structures of TpsiTeLi (1), (TpsiTe)(2) (2), TpsiTeN(3) (4), 2,6-Trip(2)C(6)H(3)TeI (10), 2,6-Trip(2)C(6)H(3)TeN(3) (11), and 2-Me(2)NCH(2)C(6)H(4)TeN(3) (12) have been determined by X-ray diffraction. Additionally, computational studies of the molecules for which experimental structural data were available were performed.

Toward a protecting-group-free halogen-metal exchange reaction: practical, chemoselective metalation of functionalized aromatic halides using dianion-type zincate, tBu4ZnLi2.

A versatile preparation method for aromatic zincate compounds through a halogen-zinc exchange reaction using dilithium tetra-tert-butylzincate (tBu4ZnLi2) has been developed. This reagent permits efficient preparation of highly functionalized aromatic zincates, particularly, those with electrophilic functional groups, such as ester, amide, alcohol, and phenol. Halogen-zinc exchange reactions followed by electrophilic trapping (with allyl bromide or benzaldehyde) proved to be a powerful tool for C-C bond formation on functionalized aromatic rings. The functionalized aromatic zincate intermediate was also found to undergo copper- and palladium-catalyzed C-C bond-forming reactions with good yields and high chemoselectivity.

Lithium potassium borate and lithium rubidium borate: new non-linear optical crystals

The title compounds, LiKB(4)O(7) and LiRbB(4)O(7), are newly developed non-linear optical crystals containing two kinds of anionic groups, namely (B(3)O(8))(7-) and (B(5)O(10))(5-). The (B(3)O(8))(7-) groups form infinite spiral chains parallel to the [100] direction, which are interconnected by sharing O atoms with (B(5)O(10))(5-) groups.

Synthesis of 2',3'-Didehydro-2',3'-dideoxynucleosides by Reaction of 5'-Protected Nucleoside 2',3'-Dimesylates with Telluride Dianion: A General Route from Cis Vicinal Diols to Olefins.

2',3'-Dimesylates of 5'-protected nucleosides are converted into the corresponding 2',3'-didehydro-2',3'-dideoxy compounds by treatment with telluride dianion in the form of the sodium or lithium salt. The method is well-suited to the preparation of unsaturated nucleosides that can be converted into compounds that are believed to be useful in the treatment of AIDS. The deoxygenation is general for vicinal dimesylates that have, or may adopt, a synperiplanar conformation. With straight chain compounds the reaction is stereospecific. In some cases, similar, but slower, deoxygenations can be performed with selenide dianion.

The immunostimulating and antimicrobial properties of lithium and antidepressants.

Eicosanoids are products of arachidonic acid (AA), an essential fatty acid. They include prostaglandins (PGs), prostacyclin (PGI2), thromboxanes (TXs), leukotrienes (LTs) and hydroxy fatty acids. AA is derived enzymatically from membrane phospholipids and to a lesser extent the diet. Eicosanoids self-regulate every cell, including those synthesizing serotonin, norepinephrine and dopamine and those subserving immune function, such as T-cells, B-cells, natural killer cells, macrophages, monocytes and dendritic cells. There is objective evidence that prostaglandins regulate the physiology of the hypothalamic-pituitary-adrenal axis (HPA). Elucidation of the structure and metabolic pathways of eicosanoids galvanized researchers into illuminating their role in physiology, pathology and pharmacology. Striking contradictions arose: eicosanoids were shown to activate and suppress microorganisms, potentiate and suppress immunity and possess pro- and anticancer properties. As prostaglandins are the most heavily studied eicosanoids in the context of mood and immunity I will focus on them in this article. I will present evidence of the immunostimulating and antimicrobial properties of lithium and antidepressants and propose that these properties are linked to the antiprostaglandin actions of these compounds.

New anilinotropone-based titanium complexes: synthesis, characterization and application as catalysts for olefin polymerization.

New titanium(IV) dichloride complexes containing 2-anilinotropone ligands have been synthesized and characterized. Bis(ligand)titanium dichloride complexes 1 and 2 were synthesized from reaction of TiCl4(THF)2 with 2 equivalents of the corresponding sodium salts of 2-(2,6-diisopropylanilino)tropone (L1) and 2-(2,3,4,5,6-pentafluoroanilino)tropone (L2), respectively. The mixed cyclopentadienyl-anilinotropone compound 3 was synthesized by reaction of the lithium salt of with CpTiCl3. The Cp-mixed micro-O bimetallic complex 4 was also isolated as a by-product owing to the adventitious presence of moisture. The molecular structures 1-4 of have been determined by single-crystal X-ray diffraction studies. Complexes 1 and 2 are isostructural and exhibit a C2-symmetric octahedral geometry, with a trans(N,N), cis(O,O) arrangement in complex 1, but with a trans(O,O), cis(N,N) arrangement in complex 2. The Cp-mixed complex 3 has a distorted square-pyramidal structure with the Cp ligand in the apical position. Bimetallic complex 4 shows a similar coordination geometry for the five-coordinate titanium atom and a pseudo-tetrahedral coordination for the second metallic centre. All new complexes, when activated with methylaluminoxane, are active in the polymerization of ethylene and propene.

Palladium-catalyzed alpha-arylation of esters.

A new and simple one-pot procedure for the palladium-catalyzed intermolecular alpha-arylation of esters is described. A number of esters can be functionalized with a wide range of aryl bromides using Pd(OAc)(2) or Pd(2)(dba)(3) and bulky electron-rich o-biphenyl phosphines 1-3. Under the reaction conditions, using LiHMDS as base, alpha-arylation proceeds at room temperature or at 80 degrees C with very good yields and high selectivities for monoarylation. Important nonsteroidal antiinflammatory drug derivatives such as (+/-)-naproxen tert-butyl ester and (+/-)-flurbiprofen tert-butyl ester can be prepared in 79% and 86% yield, respectively. The catalyst system based on the di-tert-butylphosphine (2) is also active for the alpha-arylation of esters using aryl chlorides. Furthermore, using (3) the alpha-arylation of trisubstituted ester enolates can be accomplished to provide compounds that have quaternary centers.

Effect of potentiated solutions on mercury(II) signal in inversion voltammetry.

We studied the effects of homeopathically potentiated substances in ultralow doses on physicochemical properties of solutions. Inverse voltammetric signals during Hg(II) oxidation-reduction in the presence of potentiated water, lithium chloride, and mercury nitrate were studied by alternating-current inversion voltammetry. Potentiated substances modified oxidation-reduction processes in the electrochemical system. Potentiated solutions of Hg(II) increased the effective concentration (activity) of mercury ions. The effect was most significant in the presence of mercury nitrate in homeopathic dilutions C6+C12+C24. Our results indicate that potentiated substances specifically modulate kinetic and thermodynamic characteristics of solutions containing the same compounds.

Differential up-regulation of Na+-Ca2+ exchange by phosphoarginine and ATP in dialysed squid axons.

1. The aim of this study was to characterize further the two main metabolic pathways of regulation of the Na+-Ca2+ exchanger in squid axons induced by its two naturally ocurring high-energy compounds: ATP and phosphoarginine (Pa). [Na+]o-dependent Ca2+ efflux (forward Na+o-Ca2+i exchange) and [Ca2+]o-dependent Ca2+ efflux (Ca2+o-Ca2+i exchange) were measured in internally dialysed squid axons at 16-17 C. 2. Measurements of changes in the apparent affinity of the Na+-Ca2+ exchanger for transporting (Na+o, Na+i, Ca2+o, Ca2+i) and regulatory (Ca2+i) ions induced by ATP and Pa show marked differences for the two substrates: (i) ATP strongly alters the affinity for Na+o and Na+i, while Pa does not, and (ii) in the absence of Na+i, ATP has no stimulatiory effect; on the other hand, Pa causes a dramatic increase in Na+o-Ca2+i exchange with little activation of Ca2+o-Ca2+i exchange. 3. The MgATP analogue chromium-ATP (CrATP) completely inhibits MgATP stimulation of the Na+-Ca2+ exchanger. Nevertheless, even with the effects of the nucleotide blocked, Pa exhibits its usual activation of the [Na+]o-dependent Ca2+ efflux. 4. None of the classical serine-threonine-tyrosine kinase inhibitors, nor the PP1 and PP2 phosphatase inhibitors, affects either the ATP or the Pa effect. However, intracellular microinjections of an exogenous phosphatase (alkaline phosphatase) completely reverses the stimulation of the Na+-Ca2+ exchange induced by ATP and Pa. 5. Prolonged intracellular dialysis with highly permeable porous capillaries (18 kDa molecular weight cut-off), which normally induces a complete run-down of the MgATP effect, does not alter the Pa stimulation of the exchanger, even after 6 h of continuous dialysis. 6. We conclude that the ATP and Pa modulation of Na+-Ca2+ exchange in an invertebrate nerve fibre are two genuinely different mechanisms, which affect the carrier properties in very different ways. An interesting similarity between ATP and Pa is that a phosphorylation-dephosphorylation process seems to be a common feature of these two regulation modes.

Synthesis of multi-1,10-phenanthroline ligands with 1,3-phenylene linkers and their lithium complexes.

The synthesis of two multisite ligands containing four and five 1,10-phenanthroline (phen) chelates in line, respectively, is presented. The connectors are 1,3-phenylene linkers. The two ligands were prepared following multistep procedures, the two key reactions being the Suzuki coupling reaction between aromatic nuclei and the nucleophilic addition of aryllithium derivatives onto a phen fragment. The coordination chemistry of both ligands with Li+ ions was very clean and selective, whereas their reaction with copper(I) led to intractable mixtures of insoluble complexes. The tetraphen and the pentaphen compounds afforded almost quantitatively the four- and five-lithium double-stranded helical complexes, respectively. The helical systems are probably highly wound, as indicated by NMR measurements. The pronounced strain of the 5-Li+ complex is reflected by the easy loss of a lithium cation, as shown by electrospray mass spectrometry.

Synergism of organic zinc salts and sulfhydryl compounds (thiols) in the protection of mice against acute ethanol toxicity, and protective effects of various metal salts.

Organic zinc salts such as zinc aspartate, zinc orotate, zinc histidine and zinc acetate protected mice against the lethality of an acute intraperitoneal challenge with ethanol. A similar activity was also provided by salts of cobalt, zirconium, lithium and magnesium. Organic zinc salts acted synergistically with sulfhydryl compounds in protecting the mice and potentiation between the two categories of agents was seen. The results are in analogy to radioprotective effects by zinc and thiols and imply that organic zinc salts may, alone or in conjunction with thiols, reduce in a wider context tissue injury caused by free radical-mediated mechanisms.

Complex biohopanoids synthesis: efficient anchoring of ribosyl subunits onto a C(30) hopane.

Bacteriohopanoids represent a particularly important series of triterpenoids, widely distributed in bacteria. One of the common features of these pentacyclic hopanepolyols is the presence of an extended non-terpenoid and polyhydroxylated side chain attached to the triterpenic moiety through a C--C bond. The biological function of biohopanoids also has to be addressed when one considers the broad diversity in both structures and functionalities found in the side chain. Moreover, the stereochemistries of some biohopanoids are still unconfirmed, due to the lack of synthetic methods to prepare them. In this study we describe an efficient methodology for the formation of the C--C bond between the C(30)-hopane component and C(5)-polyhydroxylated carbohydrates through the use of a hopanyllithium intermediate, which has enabled us to synthesize several biohopanoid derivatives. We also report the first synthesis of hopanepentol bearing an additional hydroxy group at position C31.
