A CAG repeat polymorphism in the androgen receptor gene is associated with reduced bone mass and increased risk of osteoporotic fractures.

Osteoporosis is a common disease with a strong genetic component. Hypogonadism results in low bone mass and it increases significantly the risk of osteoporosis in both sexes. The estrogen and androgen receptor genes are therefore strong candidates for mediating the genetic influence on bone mass and risk of osteoporosis. A CAG repeat in the first exon of the androgen receptor (AR) is associated with reduced transcriptional activity of the AR. We therefore examined whether this CAG repeat polymorphism is associated with changes in bone mass and risk of osteoporotic fractures in 284 osteoporotic patients with vertebral fractures and 327 normal individuals. The number of CAG repeats varied between 13 and 30 in men and between 7 and 34 in women. The short and long alleles comprised 19.2 +/- 2.5 and 19.0 +/- 2.3 repeats (ns) and 22.7 +/- 2.4 and 21.9 +/- 2.4 (P < 0.01) in women with vertebral fractures and normal women, respectively. This difference was also reflected in the average number of CAG repeats: 21.0 +/- 2.0 in osteoporotic women vs. 20.5 +/- 2.0 in normal women (P < 0.05). 54.8% of women with osteoporotic fractures vs. 45.9% of normal women had average number of CAG repeats of 21 and more (chi2 = 3.11, P = 0.08). Logistic regression analyses revealed that both the average number of CAG repeats and the length of the long allele were significant predictors of osteoporotic fractures in women (P < 0.05 and P < 0.01, respectively). Men with vertebral fractures had 20.0 +/- 2.8 CAG repeats compared with 20.7 +/- 2.5 CAG repeats in normal men (ns). Linear regression analysis revealed that the length of the long allele was negatively correlated with BMD of the lumbar spine (P < 0.05) and femoral neck (P < 0.05) in women. In men, linear regression analyses demonstrated that BMD of the lumbar spine (P < 0.05), femoral neck (P < 0.05) and total hip (P < 0.05) was positively correlated with length of the CAG repeat polymorphism. In conclusion, we have demonstrated that the CAG repeat polymorphism in the first exon of the AR gene is associated with reduced bone mass and increased risk of osteoporotic fractures in women.

Body pain area and pain-related negative affect predict clinical pain intensity in patients with fibromyalgia.

UNLABELLED: Patients with fibromyalgia (FM) report widespread chronic musculoskeletal pain. Palpation of 9 paired tender points (TPs) is commonly used for the diagnosis of FM according to criteria specified by the American College of Rheumatology. Although TP palpation can be used to assess deep tissue hypersensitivity, it has failed as a reliable indicator of clinical pain intensity in FM. The sum of local areas of pain (SLAP) obtained from a body pain diagram represents a relevant measure of the spatial extent of clinical pain, a feature most likely important for FM pain. Because spatial summation of pain can be an important determinant of clinical pain intensity, we hypothesized that this measure would predict clinical pain intensity in FM patients. Because pain is strongly associated with negative emotions, we evaluated the relationship of pain-related negative affect (PRNA) with clinical pain intensity in FM. The independent contributions of SLAP, PRNA, and TP count to the variance of clinical pain intensity were assessed in 280 FM patients. Clinical pain intensity of 280 FM patients was measured by using a visual analogue scale. FM patients shaded all painful body areas on body pain diagrams. Dolorimetry was used for TP evaluations. PRNA was assessed with the Medical College of Virginia Pain Questionnaire. Hierarchical linear regression was used to test the association of SLAP, TPs, and PRNA with clinical pain intensity. FM patients' mean visual analogue scale rating (0 to 100) of usual clinical pain was 50.1. Mean SLAP, TP count, and PRNA were 11.4, 16.0, and 44.3, respectively. Hierarchical linear regression analysis identified SLAP, TP count, and PRNA as independent predictors of clinical pain that accounted for 45% of the variance in clinical pain intensity ratings in FM patients. Consistent with the literature, TP count predicted only a small part (4%) of this variance. Our statistical model of body pain areas and negative affect predicts a large portion of the variance of pain intensity in FM. This result suggests that the extent of pain areas and negative emotions are uniquely associated with clinical pain intensity in FM. PERSPECTIVE: The number of painful body areas obtained by body pain diagrams is a better predictor of clinical pain intensity than TPs in FM patients. The combination of painful body areas, TP counts, and PRNA predicts 45% of the clinical pain intensity of FM patients. This finding might be useful for clinical evaluations of FM patients.

The diuretic effects of alcohol and caffeine and total water intake misclassification.

Data from the 1994 USDA nationwide survey (CSFII) on 190 non-smoking males (aged 20-29) were used to propose a method for adjusting total water intake for the diuretic effects of caffeine and alcohol, and evaluate the potential for related misclassification bias. The data were processed on a per meal basis. Under the assumption that subjects were in water balance at the start of the survey day, water losses due to caffeine (1.17 ml/mg caffeine) and alcohol (10 ml/g alcohol) were subtracted from crude intake estimates. If water intake for one meal was inadequate for excretion of the associated osmotic load at 750 mosmol/l, water losses for the subsequent meal were reduced by 32%. Unadjusted and adjusted mean total water intakes differed by 321.5 g. Misclassification appeared worst at higher water intakes. Linear regression models, each with a water intake variable as an independent variable and body mass index as the outcome, were fit to evaluate the potential for alcohol- and caffeine-related misclassification bias. Misclassification resulted in large changes (all >10%) in linear regression estimates of effect. Future studies of water-disease relationships, especially those intending to compare extremes of total water intake, should consider caffeine- and alcohol-related misclassification bias.

Anal sphincter electromyography after vaginal delivery: neuropathic insufficiency or normal wear and tear?

The study was performed to evaluate the potential role of vaginal delivery on innervation of the external anal sphincter (EAS) muscle. Forty-four women, 18 nulliparous and 26 of varying parity (1-4), without genitourinary prolapse, major urogynecological, anorectal, or neurological dysfunction were included. Participants' histories were evaluated by a questionnaire. Quantitative concentric needle electromyography (EMG) using multi-MUP analysis for sampling motor unit potentials (MUPs) in all volunteers, and "turn/amplitude" analysis for interference pattern (IP) analysis in 13 nulliparous and 23 parous women were applied. Pools of MUPs and IPs of parous and nulliparous women were formed and compared using the Mann-Whitney U test. Multiple linear regression analysis was used for evaluation of parity and obstetric variables. No difference between the groups was found in any MUP parameter, while a significant difference was found in two of five IP parameters. On multiple linear regression analysis, the number of deliveries was related to several MUP and IP parameters: the time elapsed since last delivery to MUP, and slight stress urinary incontinence to IP parameters. A group of parous women with (slight) stress incontinence had less "pathologic" MUP parameters, compared to those without. Vaginal delivery is indeed related to EAS muscle EMG abnormalities. However, these are minor and seem not to indicate loss of sphincter function. Our study casts some doubt on the commonly accepted preconception that significant damage to peripheral innervation of the EAS occurs even during uncomplicated deliveries.

MIC-III--an integrated software package to support experiments using the radioactive microsphere technique.

MIC-III is a versatile program system to support gamma spectrometry data management and data collection for experiments using the radioactive microsphere technique. It is mainly written in PASCAL and running on a minicomputer. Hierarchical organ dissection schemes are used for unique classification and identification of samples. For each series of experiments, up to 12 different nuclides and up to 20 different organ dissection schemes can be handled simultaneously. Sample weights are collected on-line and sample radioactivities are measured automatically by gamma spectrometry under process control. The spectra with a resolution of 1022 channels are analyzed using a modified linear regression technique and a strategy to compensate for global spectrum shifts. The spectrum deconvolution algorithm was evaluated by calculating the recovery indices for known nuclide mixtures and compared to the stripping method, matrix method and a linear regression technique using window counts. Recovery values produced by MIC-III are better balanced than those of the other methods. MIC-III showed the smallest dispersion of the recovery index and exhibited least error accumulation in case of spectrum shift. MIC-III computes regional blood flow and arterio-venous shunt, and provides interfaces to customized programs or statistical software packages for further analysis. It has successfully been employed in more than 140 experiments.

Use of the mitral valve-tricuspid valve distance as a marker of fetal endocardial cushion defects.

OBJECTIVE: The purpose of this study was to compare the mitral valve-tricuspid valve distance in second-trimester fetuses with normal cardiac anatomy versus those fetuses with endocardial cushion defects. STUDY DESIGN: We identified fetuses between 16 and 24 weeks of gestation. The distance between the insertions of the medial leaflets of the mitral and tricuspid valves were obtained. Linear regression curves were generated. RESULTS: The mean mitral valve-tricuspid valve distance for 86 fetuses with normal cardiac anatomy was 2.02 mm, compared with 0.37 mm in 13 fetuses with endocardial cushion defects ( P = .0001). Linear regression curve correlating mitral valve-tricuspid valve distance with gestational age showed a gradual slope (R 2 = 0.28; P < .0001). With a mitral valve-tricuspid valve distance < 5th percentile as a marker for the diagnosis of endocardial cushion defect gave a sensitivity of 69.2%, a specificity of 100%, a positive predictive value of 100%, a negative predictive value of 95.6%, and a false-positive rate of 0% ( P = .0001). CONCLUSION: The mitral valve-tricuspid valve distance is useful clinically in the detection of endocardial cushion defects in second-trimester fetuses.

Medtronic Freestyle valve for right ventricular reconstruction in pediatric Ross operations.

BACKGROUND: The use of homograft conduits to reconstruct right ventricle (RV) to pulmonary artery (PA) connections is an essential component of the Ross operation. Homograft availability and cost may be problematic when considering the Ross operation. We elected in January 1998 to utilize commercially available xenografts as an alternative to homografts for RV/PA reconstruction in the pediatric Ross operation. Our early results using the Medtronic Freestyle valve (Medtronic, Minneapolis, MN) for RV/PA reconstruction are presented. METHODS: We reviewed our database for all Ross operations performed on children since January 1998. A total of 16 patients were identified. Eleven children received a Medtronic Freestyle valve, 2 children received a homograft, and 3 children received another type of xenograft. Echocardiographic evaluation of all children who received the Medtronic Freestyle valve was performed at hospital discharge and at two subsequent outpatient evaluations. RESULTS: The median peak instantaneous pressure gradient across the xenograft was 16 +/- 9 mm Hg (immediately after surgery before hospital discharge); 22 +/- 20 mm Hg at 23 +/- 11 months (first postdischarge follow-up); and 27 +/- 20 mm Hg at 35 +/- 9 months (second postdischarge follow-up). Linear regression analysis revealed an increasing pressure gradient with time (R(2)-adjusted = 0.44, p < 0.0001). At the same three observation points, the xenograft annulus diameter decreased: 25 +/- 1.2 mm; 19 +/- 4.3 mm; and 20 +/- 1.8 mm. Linear regression analysis revealed a decreasing annulus diameter with time (R(2)-adjusted = 0.41, p < 0.0001). CONCLUSIONS: The Medtronic Freestyle valve provides a possible alternative to homografts for the reconstruction of the RV/PA connection in the pediatric Ross operation. Long-term follow-up is necessary to evaluate this xenograft as an alternative to the homograft.

The sensitivity and specificity of nerve fiber layer measurements in glaucoma as determined with scanning laser polarimetry.

PURPOSE: To determine the sensitivity and specificity for detecting glaucoma by scanning laser polarimetry and to assess the relation between nerve fiber layer (NFL) measurements and visual field indices. METHODS: The peripapillary NFL was divided into four segments: superior, inferior, temporal, and nasal. The mean polarimetric NFL for each segment was calculated out of six selected areas of 256 pixels each. Ratios relative to the nasal segment were determined for the superior and inferior segments. With the use of previously obtained normograms for polarimetric NFL readings, the sensitivity of scanning laser polarimetry was assessed in 200 glaucomatous eyes (155 subjects). The specificity was assessed in a normal population of 150 eyes (150 subjects). The relation between hemifield polarimetric NFL and visual field indices was assessed by linear regression analysis. RESULTS: The sensitivity of scanning laser polarimetry was 96% and the specificity was 93%. The correlation between NFL parameters and visual field indices ranged from -0.18 to +0.26. The amount of variation by the linear regression model ranged from 3% to 6%. CONCLUSIONS: Although quantitative measurements of the NFL with scanning laser polarimetry relate poorly to visual field indices, the technique seems to be promising for screening populations for glaucoma. Whether measurements of the NFL with scanning laser polarimetry are also sensitive enough to detect change over time requires further study.

Type I skin reactivity to native and recombinant phospholipase A2 from honeybee venom is similar.

Phospholipase A2 is the major allergen in honeybee venom. Recombinant phospholipase A2 was produced in prokaryotes and tested for its biologic activity by intracutaneous skin testing with serial 10-fold dilutions in comparison with native and deglycosylated phospholipase A2 in patients allergic to bee venom. Linear regressions of the log of the wheal area versus the log of the allergen concentration were calculated for all allergens in each patient. The relative allergenic potency of the various preparations was analyzed by comparing the linear regressions. Native phospholipase A2 was about 10 times more potent than whole bee venom. None of 58 patients allergic to bee venom was missed by testing with native phospholipase A2 alone. This allergen and deglycosylated native phospholipase A2 resulted in similar skin reactions, indicating that the sugar residues were of little relevance for IgE-binding in the patients tested. Native phospholipase A2 also had relative potency similar to that of recombinant refolded phospholipase A2, whereas recombinant nonrefolded phospholipase A2 had almost no biologic activity in skin testing. These results demonstrate in vivo activity of the recombinant bee venom allergen phospholipase A2. Although correct refolding is a prerequisite for type I skin reactivity, glycosylation seems to be less important.

Quantitative analysis of chemical warfare agent degradation products in reaction masses using capillary electrophoresis.

Quantitative methods have been developed for the analysis of chemical warfare agent degradation products in reaction masses using capillary electrophoresis (CE). This is the first report of a systematic validation of a CE-based method for the analysis of chemical warfare agent degradation products in agent neutralization matrixes (reaction masses). After neutralization with monoethanolamine/water, the nerve agent GB (isopropyl methylphosphonofluoridate, Sarin) gives isopropyl methylphosphonic acid (IMPA) and O-isopropyl O'-(2-amino)ethyl methylphosphonate (GB-MEA adduct). The nerve agent GD (pinacolyl methylphosphonofluoridate, Soman), [pinacolyl = 2-(3,3-dimethyl)butyl] produces pinacolyl methylphosphonic acid (PMPA) and O-pinacolyl O'-(2-amino)ethyl methylphosphonate (GD-MEA adduct). The samples were prepared by dilution of the reaction masses with deionized water before analysis by CE/indirect UV detection or CE/conductivity detection. Migration time precision was less than 4.0% RSD for IMPA and 5.0 RSD for PMPA on a day-to-day basis. The detection limit for both IMPA and PMPA is 100 micrograms/L; the quantitation limit for both is 500 micrograms/L. For calibration standards, IMPA and PMPA gave a linear response (R2 = 0.9999) over the range 0.5-100 micrograms/mL. The interday precision RSDs were 1.9, 1.0, and 0.7% for IMPA at 7.5, 37.5 and 75.0 micrograms/mL, respectively. Corresponding values for PMPA (again, RSD) were 2.9, 1.1, and 1.0% at 7.5, 37.5 and 87.5 micrograms/mL, respectively, as before. Analysis accuracy was assessed by spiking actual neutralization samples with IMPA or PMPA. For IMPA, the seven spike levels used ranged from 20 to 220% of the IMPA background level, and the incremental change in the found IMPA level ranged from 86 to 99 % of the true spiking increment (R2 = 0.9987 for the linear regression). For PMPA, the five spike levels ranged from 10 to 150% of the matrix background level, and similarly, the accuracy obtained ranged from 95 to 97% of the true incremental value (R2 = 0.9999 for the linear regression).

A significant correlation between clonogenic radiosensitivity and the simultaneous assessment of micronucleus and apoptotic cell frequencies.

PURPOSE: To study the relationship between clonogenic radiosensitivity and a simultaneous assessment of the percentages of radiation-induced micronuclei (MN) and apoptotic cells. MATERIALS AND METHODS: Five mammalian cell lines were studied. Following irradiation, measurements were made of the percentages of MN, apoptotic cells and floating cells. Each factor was compared, both alone and as the simple sum of the factors, with cell survival. The factors were also evaluated using a multiple linear regression analysis. RESULTS: The simple sum of the MN and apoptotic cell percentages, the sum of the MN and floating cell percentages, and the sum of all three factors correlated with cell survival. However, the multiple linear regression analysis was the best method of predicting cell survival. CONCLUSIONS: These results suggest the usefulness of the simultaneous assessment of the MN frequency and the detection of floating cells in culture medium or apoptotic cells among attached cells to predict cellular intrinsic radiosensitivity. In order to predict cell survival after irradiation from these factors, the use of multivariate analyses is more useful than simple summation.

Measuring left ventricular dimensions by conductance catheter in the rabbit.

A miniaturized (3.5 F), six-electrode conductance catheter was tested in 18 anaesthetized adult rabbits (weight 3.8-4.6 kg, ethylurethane 2.5 g kg-1). In eight animals, the reference stroke volume (ref-SV) was obtained by an electromagnetic flow probe, while reference end-diastolic volume (ref-LVEDV) was computed by dividing ref-SV by undamped thermal dilution ejection fraction (ref-EF) estimates. Comparisons with conductance indexes (z-SV, z-LVEDV and z-EF) were made at baseline, subsequent levels of graded haemorrhage and reinfusion state. In 10 animals intraventricular segmental conductance was compared with echocardiographic left ventricular cross-section (5 MHz short-focus probe), in the basal state and during acute left ventricular volume changes generated by inferior vena cava balloon occlusion. In each experiment, parallel conductance due to the tissues surrounding the left ventricle (Gp) was determined by infusing a 5M NaCl solution bolus into the right ventricle. Linear regression analysis showed fairly good correlations between z-SV, z-LVEDV and z-EF and reference indexes (r = 0.84, r = 0.83, and r = 0.72, respectively; P less than 0.001 in all cases). A linear regression analysis from 17 interventions (inferior vena cava balloon occlusion) showed a good correlation between left ventricular echocardiographic cross-sectional area and conductance, and higher correlation coefficients, r ranging from 0.870 to 0.986 were obtained from continuously sampled conductance and echographic measurements. Parallel conductance Gp was correlated (r = 0.807, P less than 0.01) with the intercept of the regression line of echographic vs conductance data. The determination of Gp thus improved the accuracy of the left ventricular dimension estimate. These results add further evidence for the possibility of continuous monitoring of left ventricular dimension by means of a conductance catheter, and demonstrate the feasibility of such studies on small experimental animals.

Cytomorphometric parameters and the metastatic potential of cutaneous and uveal melanoma: a comparison with prognostic factors.

The Mean of the Ten Largest Nuclei (MTLN1) is one of the most important prognostic factors in uveal malignant melanoma. This study was performed to investigate the applicability of nuclear and nucleolar measurements as a prognostic factor for cutaneous melanoma. A routine light microscope (Carl Zeiss: Axiophot EL-Einnsatz; #451888) at 640 times magnification with a Micrometer Eye Piece (Carl Zeiss: #444034) was used to evaluate the correlation of MTLN1 and Mean of Ten Largest Nuclei (MTLN) with the occurrence of metastasis in 58 primary cutaneous melanoma. For uveal melanoma, cytologic classification was used for comparison. Prognostic value was determined by univariate and multivariate linear regression analysis. MTLN1 was the only significant factor for uveal melanoma (p = 0.05). For cutaneous melanoma, all factors were significantly associated with disease progression. MTLN1 was the only one to remain significant (p + 0.027) in multivariate linear regression analysis. Nuclear and nucleolar morphometry are significant prognostic factors for uveal and cutaneous melanoma.

Prediction of thoracic dimensions and spine length based on individual pelvic dimensions in children and adolescents: an age-independent, individualized standard for evaluation of outcome in early onset spinal deformity.

STUDY DESIGN: A retrospective review of spine, chest, and pelvic dimensions of normal CT scans from 1999 to 2004 was performed. OBJECTIVE: To determine age-independent predictors of spine and chest dimensions. SUMMARY OF BACKGROUND DATA: Progressive thoracic deformity is associated with early-onset spinal deformities. Patients with severe early-onset scoliosis are commonly smaller and slower to grow than normal. Newer treatment approaches emphasize spine length and chest growth. Assessing treatment outcomes is hampered by the lack of normal values appropriate for diseased children. This study establishes normal range standards of chest and spine dimensions to aid in assessment of treatment outcomes. METHODS: A total of 198 CT scans of previously healthy patients 0 to 21 years of age were measured. Measurements taken from CT scans include maximum inner chest width, anteroposterior chest depth, thoracic spine height, lumbar spine height, total spine height, and pelvic inlet width. Linear regression was applied to establish gender-specific reference ranges for spine and chest dimensions based on pelvic width. RESULTS: Chest and spine dimensions were significantly correlated with pelvic inlet width and gender (P < 0.001). Multiple linear regression indicated that pelvic inlet width provided excellent fit in establishing gender-specific normal ranges independent of age. CONCLUSION: Pelvic inlet width is an age-independent predictor of pediatric chest width and thoracic dimensions that may be used to assess thoracic and spine growth in early onset spine deformity patients.

Implication of maternal nitrogen balance in the regulation of circulating levels of insulin-like growth factor-I in human pregnancy.

Nutrition plays an important role in regulating the circulating levels of insulin-like growth factor-I (IGF-I). We have demonstrated that reduced nitrogen balance is related to changes in serum IGF-I levels but not serum growth hormone levels in pregnant rats. In the present study, we investigated the effects of changes in nitrogen balance on serum IGF-I levels in normal and malnourished pregnant women (defined as having negative nitrogen balance). Forty-eight pregnant women (threatened miscarriage and premature labor, 39; pre-eclampsia, 3; hyperemesis, 3; ileus, 2; bleeding from the colon, 1) and 19 non-pregnant women admitted in Kobe University Hospital were enrolled in this study. Blood samples of normal pregnant controls were also obtained from 172 healthy pregnant women attending the outpatient clinic. Serum IGF-I levels and nitrogen balance were measured serially in 9 pregnant women with threatened miscarriage and premature labor and 9 malnourished pregnant women out of 48 pregnant women. Serum IGF-I and urinary nitrogen levels were measured by radioimmunoassay with acid-ethanol extraction and the Dumas method, respectively. Nitrogen balance was expressed as the difference between daily nitrogen intake and nitrogen excretion assessed by urinary nitrogen levels. Serum IGF-I levels in normal pregnant controls significantly increased in the third trimester of pregnancy compared with non-pregnant controls. No difference in serum IGF-I levels in any trimester of pregnancy was observed between normal pregnant controls and pregnant women with threatened miscarriage and premature labor. There was no significant difference in nitrogen balance between the pregnant women with threatened miscarriage and premature labor and non-pregnant controls. In the longitudinal study, no correlation was found between the changes in serum IGF-I levels and those in nitrogen balance in the 9 pregnant women with threatened premature labor (daily nitrogen balance > 0 g/day) on the basis of linear regression analysis. On the other hand, the changes in serum IGF-I levels in the 9 malnourished pregnant women were significantly correlated with those in nitrogen balance (y = 1.72x + 17.5; r = 0.60; P < 0.05: linear regression analysis). These results indicate that maternal nutritional states have a major effect on serum IGF-I levels in malnourished pregnant women, but not in pregnant women with daily nitrogen balance > 0 g/day. Serum IGF-I levels can be a potent index of nutritional states under malnutrition during human pregnancy.

Risk factors for hospitalization and infection in Canadian Inuit infants over the first year of life--a pilot study.

OBJECTIVES: Inuit infants experience higher mortality and poorer health than other Canadian infants, and suffer disproportionately from bacterial and viral infections. A wide range of inter-related factors affect their health and susceptibility to infection. The objective of the study was to describe hospitalization and morbidity patterns in a cohort of 46 healthy Inuit infants from Iqaluit, Nunavut, over their first year of life. STUDY DESIGN: Risk factors for hospitalization and infections were assessed using multiple linear regression. RESULTS: Infants experienced an average of four respiratory tract infections (RTIs) annually, which accounted for half of the hospitalizations in the cohort. Some interesting trends were evident from the assessment of risk factors using multiple linear regression. Adoption was associated with adverse health effects in addition to those that would be expected due to lack of breast-freeding alone; among infants who were not breast-fed, adopted infants had three more RTIs per year than non-adopted infants. CONCLUSION: The results of this pilot study provide support for undertaking larger epidemiological studies in order to clarify the role of these risk factors, so that future preventive efforts can be informed and effective.

Analyte comparisons between 2 clinical chemistry analyzers.

The purpose of this study was to assess agreement between a wet reagent and a dry reagent analyzer. Thirteen analytes (albumin, globulin, alkaline phosphatase, alanine aminotransferase, amylase, urea nitrogen, calcium, cholesterol, creatinine, glucose, potassium, total bilirubin, and total protein) for both canine and feline serum were evaluated. Concordance correlations, linear regression, and plots of difference against mean were used to analyze the data. Concordance correlations were excellent for 8 of 13 analytes (r > or = 0.90); the correlations for albumin, potassium, and calcium were clinically unreliable. The linear regression analysis revealed that several analytes had slopes significantly different from unity, which was likely related to methodological differences. Compared to the wet reagent analyzer, the dry reagent analyzer showed excellent agreement for alkaline phosphatase, alanine aminotransferase, amylase (feline), urea nitrogen, cholesterol, creatinine, glucose, total bilirubin (canine), and total protein. However, it showed only slight to substantial agreement for amylase (canine), calcium, albumin, potassium, and total bilirubin (feline).

Application of geographic information technology in determining risk of eastern equine encephalomyelitis virus transmission.

Geographic information system (GIS) technology and remote sensing were used to identify landscape features determining risk of eastern equine encephalomyelitis virus (EEE) transmission as defined by the abundance of Culiseta melanura (the enzootic vector) and 6 putative epidemic-epizootic vectors in Massachusetts. Landsat Thematic Mapper data combined with aerial videography data were used to generate a map of landscape elements at epidemic-epizootic foci in southeastern Massachusetts. Geographic information system technology was used to determine the proportion of landscape elements surrounding 15 human and horse case sites where abundance data were collected for Culiseta melanura, Aedes canadensis, Aedes vexans, Culex salinarius, Coquillettidia perturbans, Anopheles quadrimaculatus, and Anopheles punctipennis. The relationships between vector abundance and landscape proportions were analyzed using stepwise linear regression. Stepwise regression indicated wetlands as the most important major class element, which accounted for up to 72.5% of the observed variation in the host-seeking populations of Ae. canadensis, Ae. vexans, and Cs. melanura. Moreover, stepwise linear regression demonstrated deciduous wetlands to be the specific wetland category contributing to the major class models. This approach of utilizing GIS technology and remote sensing in combination with street mapping can be employed to identify deciduous wetlands in neighborhoods at risk for EEE transmission and to plan more efficient schedules of pesticide applications targeting adults.
