Evaluation of commercial and newly-synthesized amine accelerators for dental composites.

The characteristics of newly synthesized tertiary aromatic amines as accelerators for restorative resins have been evaluated. Comparison of the composites prepared with these and presently used accelerators indicate that resins formulated with 4-N,N-dimethylaminophenylacetic acid, its methyl ester or N,N-dimethylaminoglutethimide have properties generally better than comparable resins prepared with commercially used amines.

Effects of lithium on the beta-adrenergic receptor-adenylate cyclase system in rat cerebral cortical membranes.

The effects of lithium on the beta-adrenoceptor-adenylate cyclase system in cerebral cortical membranes of rats were investigated. Lithium chloride inhibited adenylate cyclase activity in a concentration-dependent manner in vitro. However, relatively high concentrations of lithium were needed for this inhibition; and at 1 mM, no significant reduction in adenylate cyclase activity was seen under any condition. Administration of lithium carbonate for 21 days decreased the maximum number of [3H]dihydroalprenolol binding sites without changing the apparent dissociation constant. Activation of adenylate cyclase by (-)-isoproterenol in the presence of 1 microM guanyl-5'-ylimidodiphosphate (Gpp(NH)p) was significantly attenuated in lithium-treated rats compared with the controls. Lithium treatment reduced the Gpp(NH)p-stimulated adenylate cyclase activity in the presence of 10 microM (-)-isoproterenol, but not in the absence of this beta-adrenergic receptor agonist. Basal activity or adenylate cyclase activity stimulated by forskolin or manganese was not affected, whereas the activity stimulated by sodium fluoride was significantly attenuated by long-term lithium treatment. These results indicate that chronic lithium treatment induces subsensitivity in the beta-adrenoceptor-adenylate cyclase system, for which down-regulation of beta-adrenergic receptors is chiefly responsible.

[The protective action on the myocardium of antihypoxants in acute occlusion and subsequent reperfusion of the coronary artery].

In experiments on anesthetized cats with acute occlusion of the coronary artery followed by its reperfusion antihypoxants piracetam, lithium hydroxybutyrate and pyrid oxynyl-glyoxylate (glyo-6) were shown to stabilize the contractile and pumping functions of the heart. GABA derivatives piracetam and lithium hydroxybutyrate in contrast to glyo-6 were found not only to stimulate the compensatory processes in the conventional intact area of the ischemized left ventricle but also improve the energy metabolism in the ischemic focus itself.

[Contents of serotonin and 5-hydroxyindoleacetic acid in the rabbit brain after long-term administration of lithium oxybutyrate].

Serotonin (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) levels in rat brain were analysed 24 hours after 7-, 15-, 29- days lithium hydroxybutyrate (LH) injections (10 mg/kg daily). After 7 days the drug reduced 5-HT in hypothalamus and 5-HIAA in the mid brain by 35%. After 15 days LH decreased 5-HT in striatum, hypothalamus by 32 and 17% and 5-HIAA in thalamus, hypothalamus by 28 and 44% respectively. After 29 days LH diminished 5-HT in striatum, hippocampus, amygdala by 24, 29 and 32% and 5-HIAA--in hypothalamus by 42%. The role of adaptative changes and stabilization processes in the central serotoninergic system in mechanism of LH psychotropic effects is discussed.

Lithium increases turnover of phospholipids in flight muscles of Periplaneta americana L.

The effect of prolonged lithium administration on the phospholipid metabolism of flight muscles of the cockroach Periplaneta americana has been studied. Following daily injections of LiCl in a dose of 19.25 mumol LiCl per gram of wet weight [32P]- orthophosphate were injected and its incorporation into the phospholipids was measured 2, 12 and 24 h later. Lithium administration did not change the content of phospholipids but increased the 32P incorporation into phosphatidylinositol, phosphatidylcholine, phosphatidylethanolamine and sphingomyeline 1.87, 2.13, 2.02 and 1.87 times, respectively, as compared with the control values. These increases were neither due to an increased permeability of the tissue for inorganic phosphate nor to an increased turnover of gamma-P-ATP. It is concluded that prolonged lithium treatment increases the turnover of all phospholipids in insect flight muscle tissue.

[Effect of lithium preparations on cardiac arrhythmias due to strophanthin in waking rats].

It was shown in experiments on conscious rats that intravenous injection of strophanthine in toxic doses provokes heart arrhythmias and death of the animals. Lithium drugs (lithium chloride and lithium hydroxybutyrate) injected during arrhythmias led to a short-lived effect of heart rhythm normalization. Lithium hydroxybutyrate was more effective if administered shortly after strophanthine injection, reducing the latter's cardiotoxic effect and preventing the death of the majority of the animals.

[Randomized double-blind placebo-controlled study of drugs with anti-ulcer action and their use in associated pathology].

The effect of lithium preparations (lithium carbonate, lithium oxybutyrate), calcium channel blocking agents (verapamil, nifedipine), beta 2-adrenoagonist (salbutamol), cromolyn sodium and cimetidine on clinical efficacy and frequency of ulcer cicatrization was assessed in 201 patients (33 women, 168 men) with endoscopically verified stomach ulcer (52 patients) and duodenal ulcer (166 patients) during treatment for 4 weeks as compared to placebo and standard antiulcerative therapy (cholinolytics, antacids and reparents). All the tested drugs (excluding cromolyn sodium) significantly increased as compared to placebo the frequency of ulcer cicatrization (p less than 0.001 in the use of lithium preparations and cimetidine; p less than 0.01 in the use of salbutamol and verapamil; p less than 0.05 in the use of nifedipine). The effect of lithium preparations and cimetidine exceeded that of standard antiulcerative multimodality therapy.

[Effect of lithium oxybutyrate on the viability of preserved donor kidney].

The experiments on 10 dogs showed that replacement of sodium oxybutyrate for lithium oxybutyrate in the solution for preservation increases the period of preserving the structural and functional integrity of the donor kidney, contributes to its survival and delays the development of the transplant rejection.

[Development of progeny after antenatal exposure to lithium oxybutyrate].

Experiments on rats were made to study the effect of lithium hydroxybutyrate and to compare it with lithium carbonate effect on the development of the progeny after antenatal exposure to the drugs. It was established that the lithium salts under study did not produce any conclusive effect on the postnatal development of young rats. The activating component was discovered to be predominant in the action of lithium hydroxybutyrate, whereas the action of lithium carbonate was marked by the inhibitory component. As regards the effect on the development of the progeny, lithium hydroxybutyrate was less hazardous than lithium carbonate.

[Effect of lithium oxybutyrate on excitability of the cortex and some subcortical structures in rabbit brain].

Lithium oxybutyrate microinjections (10 mg/ml) produce a depressant action on spontaneous bioelectrical activity of the cortex and subcortical structure. The drug brings down excitability of the motor cortex, hippocampus, caudate nucleus, thalamus, posterior hypothalamus and mesencephalic reticular formation; it also raises excitability of the tonsils. The depressant effect of lithium oxybutyrate is superior to that of lithium chloride.

[Effect of lithium hydroxybutyrate and lithium chloride on heart rhythm disorders of various origins].

It has been shown in experiments on anesthetized cats and dogs that lithium hydroxybutyrate produces antiarrhythmic action in central aconitic and strophanthine arrhythmias, prevents the development of ventricular fibrillation, and shows antiarrhythmic activity in strong occlusion on the coronary artery. The drug does not remove the atrial and aconitic patterns of arrhythmia. Lithium chloride also exhibits antiarrhythmic action in central aconitic arrhythmia, being inferior to lithium hydroxybutyrate. The drug does not influence central strophanthine arrhythmia and does not show antifibrillation activity.
