A computerized technique for vertebral morphometry.

A new technique for morphometric measurement of vertebral bodies has been described, in which a computerized image analysis system was used to digitize and measure standardized lateral radiographs of the thoracic and lumbar spine. The sources of error in the radiographic system and the image analysis system were assessed using vertebral phantoms and test images. Oblique projection in the radiographic image of a vertebra did not produce a significant error in the measured area. The radiographic magnification was approximately 35%. Optical non-uniformity was detected in the image analysis system, and was ascribed to the lens optics. The maximum intra-observer and inter-observer variations in vertebral area were 3.1% and 5.3% respectively for experienced observers. The technique is applicable to clinical studies of large populations, and has value in investigating vertebral deformity in the management of metabolic bone disease.

The geometric configuration and morphometry of the rabbit oesophagus during luminal pressure loading.

The aim of the present study was to determine the geometric configuration and morphometric dimensions in the rabbit oesophagus during luminal pressure loading. The geometric configuration is of fundamental importance because it is related to stress and strain and determines the resistance to flow. The oesophagus was excised from rabbits, transferred to an organ bath and stretched to the in vivo length. Cannulas were inserted into both ends and each oesophagus was subjected to a luminal pressure up to 0, 1, 2, 5 and 10 cm H(2)O. After equilibrium was reached, the oesophagus was snap-frozen in liquid nitrogen. Sections were cut from five locations in the oesophagus and videotaped in the frozen state. Geometric and morphometric data including perimeter, circularities, area, thickness, buckles and strains in different layers were obtained from the video images. These variables did not show axial variation in the oesophagus. All variables changed as a function of the pressure. The whole wall thickness decreased as an inverse function of the pressure. The thickness decrease was mainly due to thinning of the submucosa and muscle. The number of buckles decreased from 4.05 +/- 0.34 at 0 cm H(2)O pressure to 0.07 +/- 0.07 at 10 cm H(2)O. Circumferential Green strain-pressure curves for the various layers all showed an exponential pattern. The curve obtained at the submucosa-muscle layer interface was located to the right of the other curves, indicating that the muscle layer was softest. The data obtained in this study will be useful for further modelling of the mechanics of the oesophagus.

Simple morphometry of axonal swellings cannot be used in isolation for dating lesions after traumatic brain injury.

Disruption of fast axonal transport as a result of traumatic brain injury is characterized by the accumulation of beta-amyloid precursor protein (APP) in axonal swellings. A recent report has suggested a correlation between the size of axonal swellings and survival time up to about 85 h after blunt head injury. The authors of the report concluded that this correlation, in conjunction with other evidence, might be useful in forensic science for timing injuries. To test this hypothesis we have used image analysis software to measure a number of different morphological parameters of axonal swellings. Paraffin sections from 63 cases of fatal head injury were stained with an antibody raised against the N-terminus of APP and counterstained with haematoxylin. Three different measurements were made of the APP-immunoreactive axonal swellings from the corpus callosum: (i) minimum and (ii) maximum Feret diameters, and (iii) area. Linear regression revealed a significant correlation between survival time and the minimum Feret diameter (p < 0.0001) and the area (p < 0.001) of axonal swellings. Our findings are in agreement with the previous study in that there is a significant correlation between axonal swelling size and survival time. However, we would suggest that the large variability in swelling size within individual cases and the heterogeneity of the original trauma seriously compromise the utility of such information in the timing of lesions.

Computer-assisted cell morphometry and ploidy analysis in the assessment of thyroid follicular neoplasms.

Conventional cytology cannot discriminate between benign and malignant follicular neoplasms. Our study evaluated the diagnostic role of computer-assisted image analysis in the presurgical assessment of thyroid follicular neoplasms. Fifty-eight patients (14 males, 44 females, age range, 45-75 years) who underwent surgery for cytologic diagnosis of thyroid follicular neoplasm were studied. All patients were first evaluated on clinical grounds and assigned a high/low suspicion of malignancy on the basis of gender, age, and nodule size. Cell image analysis was subsequently performed using a Cytometrica BYK Gulden microscope image processor on Feulgen-stained thyroid cytologic smears. A different population of 50 benign and 50 malignant, histologically evaluated nodules was studied in order to establish image analysis criteria suggestive of thyroid malignancy. Ploidy histogram, proliferation index (PI), nuclear area coefficient of variation (NACV), and anisocariosis ratio (AR) were studied. Thyroid cancer was diagnosed in 16 of 58 follicular neoplasms. Only 7 of these lesions were clinically suspicious (43.7%), whereas 14 of 16 (87.5%) malignant tumors were identified by image analysis. Positive and negative predictive values of image analysis versus clinical evaluation were 46.6% versus 30.4% and 92.8% versus 74.3%, respectively. The distribution of ploidy pattern was different in benign versus malignant follicular neoplasms (chi2 8.25, p = 0.016), malignant lesions showing an increased frequency of heteroclonal aneuploid DNA content (37.5% vs. 7.1%). Increased PI (mean +/- standard deviation (SD) = 11.3 +/- 5.7 vs. 7.1 +/- 6.1; p < 0.01) and NACV (mean +/- SD = 25.28 +/- 1.89 vs. 20.14 0.91; p < 0.01) levels were also observed in malignant follicular neoplasms. In conclusion, computer-assisted image analysis may profitably support clinical evaluation in the assessment of thyroid follicular neoplasms.

Sural nerve morphometry in diabetic autonomic and painful sensory neuropathy. A clinicopathological study.

Observations have been made on a selected series of insulin-dependent patients with neuropathy, subdivided into three groups: (1) severe autonomic neuropathy with an accompanying painless sensory neuropathy; (2) severe autonomic neuropathy with a chronic painful sensory neuropathy; and (3) chronic or acute painful sensory neuropathy with no autonomic neuropathy. All three groups showed a loss of large and small myelinated nerve fibres in sural nerve biopsy specimens which was greater in Groups 1 and 2. Regenerative activity was prominent in all three groups, but least in Group 3. Teased fibre studies showed evidence both of axonal regeneration and remyelination. Active fibre degeneration was rare. Measurements of g ratio (axon diameter:total fibre diameter) gave no indication of axonal atrophy. The density of unmyelinated axons was reduced in all three groups, as was their median diameter. Vibration sense threshold was positively correlated with the total number of myelinated fibres and thermal sensory threshold with median unmyelinated axon diameter but not with total unmyelinated axon numbers. No correlation between the occurrence of pain and active degeneration of myelinated fibres or with regenerative activity either in myelinated or unmyelinated axons was detectable. Assessment of differential loss of large or small myelinated nerve fibres was difficult because of the presence of large numbers of small regenerating myelinated axons. The results are discussed in relation to the concept of 'diabetic small fibre neuropathy' and the causation of pain in diabetic neuropathy.

Morphometry of the Sylvian fissure and the corpus callosum, with emphasis on sex differences.

The relationship between anatomical asymmetries in the perisylvian region and the sizes of different regions of the corpus callosum was investigated post-mortem in 40 brains of right-handed hospital admissions (20 males, 20 females) with no cortical involvement. There were no sex differences either in anatomical asymmetries or in regional size of the callosum. There was a negative correlation between the absolute value of Sylvian fissure (planum temporale) asymmetries and the size of the isthmus in males but not in females. Further, there was a significant negative correlation between the size of the Sylvian fissure (or planum temporale) and the size of the callosal mid-body in males but not in females. There was no correlation between the asymmetry of the planum temporale magnitude of left-right and total size of the planum (left+right). These findings constrain theories about the ontogenesis of hemispheric specialization through changes in callosal connectivity and about sex differences in interhemispheric organization.

Assessment of lesion pathology in multiple sclerosis using quantitative MRI morphometry and magnetic resonance spectroscopy.

Quantitative measurement of MRI-defined brain lesions can provide an index of the extent and activity of disease in multiple sclerosis patients. However, the relationships between these indices and clinical features are not well-understood. Heterogeneity of the pathological changes underlying MRI lesions may be an important factor determining the correlation between MRI lesion volumes and clinical measures. Recent studies have suggested that with magnetic resonance spectroscopy (MRS), it may be possible to define chemical changes that better reflect the pathological changes in multiple sclerosis. Here we report results of combined quantitative brain T2-weighted MRI lesion volume and proton MRS examinations that demonstrate heterogeneity of the chemical pathology underlying brain lesions in patients selected on the basis of similar clinical disability but differing with respect to the presence or absence of clinical relapses. We examined 29 patients with disease characterized by either clear relapses with at least partial remissions (RR) or secondary, chronic progression after an earlier history of a more relapsing and remitting course (SP). Total hemispheric lesion volume was greater (P < 0.04) in the RR (32.5 +/- 20.9 cm3) than in the SP (16.2 +/- 9.0 cm3) patients, despite the longer duration of disease in the latter group. Central brain N-acetyl aspartate: creatine (NAA:Cr) ratios were reduced relative to normal controls (4.0 +/- 0.3, n = 19) by similar amounts in the two patients groups (RR, 3.1 +/- 0.5; SP, 3.2 +/- 0.4; P < 0.0001). The ratio lesion volume:(NAA:Cr) was greater for the RR group (11.7 +/- 9.3 cm3) than for the SP group (5.4 +/- 3.3 cm3, P < 0.05), implying a greater average degree of axonal loss per unit lesion volume defined by MRI for subjects in the SP group or, alternatively, a greater proportion of lesions without axonal damage or loss in the RR group. Our results emphasize a limitation of using T2-weighted MRI lesion volume alone and suggest that combined analysis of MR-based chemical and imaging data might allow improved non-invasive assessment of lesion pathology in order to better understand its relationship to clinical features of multiple sclerosis.

Morphometry of human superficial dorsal and dorsolateral column fibres: significance to spinal cord stimulation.

In spinal cord stimulation (SCS) large diameter cutaneous (Abeta) fibres in the dorsal columns (DCs) are activated and have an inhibiting effect on the transmission of pain signals by Adelta and C fibres from the corresponding dermatome(s). The largest Abeta fibres can be activated up to a maximum depth of about 0.25 mm in the DCs. No data are available on the distribution of the large fibres in this superficial human DC layer at the common SCS levels Th(10-11). Such data are indispensable to improve the predictive capability of a computer model of SCS. The whole myelinated fibre population in the superficial 300 microm of the dorsal column (DC(0-300)) at Th(10-11 )of two human subjects was morphometrically analysed. Some data was obtained from a third subject. The superficial dorsolateral column (DLC(0-300)) was included in this analysis because it was hypothesized that large dorsal spinocerebellar tract fibres could also be activated by SCS. Only very few fibres larger than 10.7 microm were found: a mean of 68 (0.5%) in DC(0-300) and 114 (2%) in DLC(0-300). Considering that the effect of SCS is primarily attributed to activation of these largest fibres, it is concluded that a surprisingly small average amount of 2.4 fibres per running 0.1 mm width and 6 fibres per segmental division of the DC is involved. Distinct mediolateral heterogeneity in fibre composition was found in both DC(0-300) and DLC(0-300). In the DC(0-300), the mean diameter of fibres > or =7.1 microm increases significantly by 5% from medial to lateral. Density (i.e. number of fibres per 1000 microm(2)) and frequency (i.e. percentage of a fibre size group compared to its parent population) of the large fibres increase significantly from medial to lateral in the DC(0-300). For fibres > or =10.7 microm, these parameters increase by 200 and 269%, respectively. It is concluded that the difference in stimulation threshold of large Abeta fibres in the median and lateral DC can be mainly attributed to the absence and presence, respectively, of collaterals at the stimulation site. Marked differences were found between DC(0-300) and DLC(0-300). The largest DLC(0-300) fibres (> or =10.7 microm) have a 320% higher frequency and a 473% higher density. Their mean diameter is, however, only 2% larger. The largest DLC(0-300) fibres are not likely to be recruited by SCS, since they are not larger than their DC(0-300) counterparts, they lack collaterals (which would reduce the threshold stimulus substantially) and they are more remote from the stimulation electrode.

Functional and anatomical cortical underconnectivity in autism: evidence from an FMRI study of an executive function task and corpus callosum morphometry.

The brain activation of a group of high-functioning autistic participants was measured using functional magnetic resonance imaging during the performance of a Tower of London task, in comparison with a control group matched with respect to intelligent quotient, age, and gender. The 2 groups generally activated the same cortical areas to similar degrees. However, there were 3 indications of underconnectivity in the group with autism. First, the degree of synchronization (i.e., the functional connectivity or the correlation of the time series of the activation) between the frontal and parietal areas of activation was lower for the autistic than the control participants. Second, relevant parts of the corpus callosum, through which many of the bilaterally activated cortical areas communicate, were smaller in cross-sectional area in the autistic participants. Third, within the autism group but not within the control group, the size of the genu of the corpus callosum was correlated with frontal-parietal functional connectivity. These findings suggest that the neural basis of altered cognition in autism entails a lower degree of integration of information across certain cortical areas resulting from reduced intracortical connectivity. The results add support to a new theory of cortical underconnectivity in autism, which posits a deficit in integration of information at the neural and cognitive levels.

Morphometry of the small arteries and arterioles in the rat heart: effects of chronic hypertension and exercise.

STUDY OBJECTIVE: The aim of the study was to characterise small coronary arteries and arterioles quantitatively in rat left ventricle and septum under various experimental conditions. DESIGN: Morphometric analysis of resistance vessels was carried out in rats divided into four experimental groups: (1) control animals sedentary; (2) control animals exercised (moderate swimming); (3) renal hypertensive animals sedentary; (4) renal hypertensive rats exercised. Hypertensive animals received the two kidney/one clip Goldblatt procedure; control animals were sham operated. SUBJECTS: Experimental animals were 28 female Sprague-Dawley rats. MEASUREMENTS AND RESULTS: In hypertensive rats, there was a significant increase in left ventricular mass and a reduced coronary reserve. Morphometric analysis of 10,000 resistance vessels in the coronary bed showed that cardiac hypertrophy in hypertensive rats was characterised by increased wall thickness in arterioles and small arteries and an increased lumen to wall ratio, particularly in vessels of 30-100 microns size. The number of capillary profiles per arteriole in cross section was also decreased in the hypertensive rat. Chronic exercise increased the number of capillaries per arteriole in controls but not in hypertensive rats. Swimming did not affect the architecture of the resistance vessels. CONCLUSIONS: Chronic renal hypertension, but not swimming, produces subtle but significant changes in structure of coronary resistance vessels.

Morphometry of arterioles and capillaries in hearts of senescent mice.

OBJECTIVE: The aim was to characterise quantitative changes in the coronary arterioles and capillaries with aging in mice. METHODS: Morphometric analysis of the coronary resistance vessels was carried out in 12 "young" mice (63 days) and 11 "old" mice (650 days). RESULTS: Compared to hearts from young mice, those from senescent mice were heavier, contained less water, and differed in several variables characterising terminal vessels. The main changes were a decreased capillary and arteriolar density, increased heterogeneity of capillary spacing, and larger arterioles with thicker walls. CONCLUSIONS: Aging of the murine myocardium is characterised by important changes at the level of the terminal vascular bed, the most prominent being a decrease in capillary and arteriolar density. The decrease in arteriolar density is most prominent in the classes of smallest vessels. All these changes represent an impairment of geometrical conditions for the oxygen supply to the myocardial tissue.

Products of cells cultured from gliomas. V. Cytology and morphometry of two cell types cultured from glioma.

Explants of cells of a human glioma were evaluated with the nuclear fluorochrome 4',6-diamidino-2-phenylindole, by phase-contrast illumination, and by Giemsa staining correlated with double immunofluorescence for glial fibrillary acidic protein (GFAP) and fibronectin (FN). FN-positive (FN+) cells lacked GFAP detectable by immunofluorescence. Their mean nuclear-to-cytoplasmic ratio was large (0.192). Actual mean areas of nuclei (1,252 microns2) and cytoplasm (8,376 microns2) of FN+ cells compared with mean areas of fibroblasts suggested that the high nuclear-to-cytoplasmic ratio of FN+ cells was due to their microscopically evident reduced cytoplasmic spreading rather than to larger nuclei. Some FN+ cells showed marked variation in nuclear and nucleolar size and shape. Others had abnormal mitoses or hyperchromatic nuclei. GFAP-positive (GFAP+) cells lacked FN detectable by immunofluorescence. GFAP+ cells were smaller and less round than FN+ cells. Their usual location was growing on a layer of FN+ cells. The mean nuclear-to-cytoplasmic ratio (0.245) of GFAP+ cells was the highest in the study, surpassing the ratio of the continuous glioma line LM (0.176). Mean areas of nuclei (289 microns2) and of cytoplasm (1,350 microns2) of GFAP+ cells suggested that their high nuclear-to-cytoplasmic ratio was due to their microscopically evident reduced cytoplasmic spreading. Reduced spreading was associated with extension of long, thin cytoplasmic processes. The majority of GFAP+ cells showed marked cytoplasmic basophilia, nuclear hyperchromasia, and clumped chromatin. Features observed in both FN+ and GFAP+ cells from this high-grade astrocytoma are features associated with malignant transformation in more thoroughly studied tumor systems.

Morphometry of interstitial fibrosis.

Several clinical studies have confirmed that histomorphometric changes in the tubulointerstitial compartment contain the best correlating parameters to predict the development of progressive renal insufficiency. The process of interstitial fibrosis is accompanied by an influx of inflammatory cells, up-regulation of fibrogenic cytokines such as transforming growth factor-beta and basic fibroblast growth factor, transient down-modulation of their antagonists, generation and proliferation of myofibroblasts, and, finally, by accumulation of interstitial collagens and proteoglycans. A careful morphometric analysis of interstitial fibrosis requires sensitive parameters through which the severity can be quantified and by which the progression into renal insufficiency can be predicted. We have addressed these issues by morphometric analysis of both human biopsies and by refining existing experimental models in the rat. Morphometric analysis was performed using a Zeiss microscope equipped with a full colour 3CCD camera and KS-400 image analysis software from Zeiss-Kontron. For studies with human material, biopsies were examined from patients with various renal diseases including patients with chronic allotransplant dysfunction. The development of interstitial fibrosis was correlated with clinical parameters. In experimental models, we analysed the interstitial composition and eventual glomerular alterations in rats with bovine serum albumin (BSA)-induced protein overload nephropathy and with human IgG-induced chronic serum sickness nephritis. Finally, we adapted and refined the model of ureter obstruction-induced interstitial fibrosis in the rat. For this purpose, custom-made titanium clips (S&T, Neuhaus, Switzerland) were implanted around the ureter in the abdomen of rats to obstruct the ureter without causing necrosis. The clips were removed at various time points after obstruction of the ureter (1-14 days). The subsequent remodelling of the interstitium was studied thereafter, in order to establish whether uraemia-induced interstitial fibrosis remains reversible at all times. In rat models, we have found that both protein overload-induced and serum sickness-induced interstitial fibrosis are accompanied by the development of focal and segmental glomerulosclerosis. Only in the ureter obstruction model did selective interstitial fibrosis develop, and remained reversible at all times studied. For the reliable assessment of interstitial fibrosis we have found that the best correlating parameters of interstitial fibrosis with renal function were: (i) the ratio of protein accumulation of TGF-beta-1 and its antagonist decorin; (ii) interstitial expression of smooth muscle alpha-actin; and (iii) accumulation of interstitial collagens (as determined by immunoperoxidase and by Sirius red staining).

Comparison of mandibular morphology in Korean and European-American children with Class III malocclusions using finite-element morphometry.

The purpose of this study was to determine whether the morphology of the mandible differed in subjects of diverse ethnic origin exhibiting Class III malocclusions. Lateral cephalographs of 147 children of either Korean or European-American descent aged between 5 and 11 years were compared. The cephalographs were subdivided into seven age- and sex-matched groups, traced, and eight mandibular homologous landmarks digitized. Average mandibular geometries, scaled to an equivalent size, were computed using Procrustes superimposition and subjected to ANOVA. Graphical analysis using a colour-coded finite element (FEM) programme was used to localize differences in morphology. Results indicated that the overall mean Korean and European-American mandibular configurations differed statistically (P < 0.001) and statistical difference was maintained at all age-wise comparisons. Comparing Korean and European-American Class III mandibular configurations for local size-change, FEM analysis revealed that the Korean condylar and mental regions generally were smaller (approximately 15-20 per cent decrease in size, respectively). However, an antero-posterior increase in the size of the mandibular corpus was most apparent in the incisor alveolus region (approximately 35 per cent increase in size). For shape-change, the Korean and European-American mandibular configurations were fairly isotropic except in the symphyseal and incisor alveolus regions. Dissimilarities in mandibular morphology are identifiable particularly in the dento-alveolar regions in subjects of diverse ethnic origin exhibiting Class III malocclusions. These differences may reflect genetic and/or environmental influences that might determine the severity and prevalence of the condition, and its subsequent clinical management.

Relationship between endometrial histology, morphometry and ultrasound texture in the follicular phase of infertile women with natural menstrual cycles.

The relationship between endometrial histology and ultrasonographic texture in the follicular phase was investigated. The endometrial sonographic texture of 32 infertile women with normal menstrual cycles was classified into three patterns (L, H and I) and histological and morphometrical analyses were performed. Endometrial specimens from pattern L, which showed multi-layered endometrium characterized by three hyperechogenic lines with inner hypo-echogenic regions, had smaller, similar-sized endometrial glands and few stromal cells. Those from pattern H which showed entirely homogeneous hyperechogenic endometrium, had larger, various-sized glands and more stromal cells. Those from pattern I which showed heterogeneous hyperechogenic and partially hypo-echogenic endometrium, had the largest, most variable-sized glands and many stromal cells. The differences in sonographic texture may be related to the histological and morphometrical findings.

Variations in leaf morphometry and nitrogen concentration in Betula pendula Roth., Corylus avellana L. and Lonicera xylosteum L.

Relations between leaf dry weight to leaf area (LWA), leaf nitrogen concentration and irradiance inside a natural canopy were studied in Betula pendula Roth., Corylus avellana L. and Lonicera xylosteum L. In all species, LWA increased with increasing irradiance. Relative variability in LWA was smaller in Betula pendula than in the other two species. In Corylus avellana, LWA also depended on total plant height. Foliar nitrogen concentration (on a dry weight basis) increased with increasing irradiance and LWA in Betula pendula, but decreased in the other two species. The interspecific variation in response to light availability and in nitrogen partitioning may be caused by different light demands or different life forms (trees versus shrubs), or both, of the species examined, and must be considered in contemporary canopy models.

Testis morphometry, seminiferous epithelium cycle length, and daily sperm production in domestic cats (Felis catus).

There is very little information regarding the testis structure and function in domestic cats, mainly data related to the cycle of seminiferous epithelium and sperm production. The testis weight in cats investigated in the present study was 1.2 g. Compared with most mammalian species investigated, the value of 0.08% found for testes mass related to the body mass (gonadosomatic index) in cats is very low. The tunica albuginea volume density (%) in these animals was relatively high and comprised about 19% of the testis. Seminiferous tubule and Leydig cell volume density (%) in cats were approximately 90% and 6%, respectively. The mean tubular diameter was 220 microm, and 23 m of seminiferous tubule were found per testis and per gram of testis. The frequencies of the eight stages of the cycle, characterized according to the tubular morphology system, were as follows: stage 1, 24.9%; stage 2, 12.9%; stage 3, 7.7%; stage 4, 17.6%; stage 5, 7.2%; stage 6, 11.9%; stage 7, 6.8%; and stage 8, 11 %. The premeiotic and postmeiotic stage frequency was 46% and 37%, respectively. The duration of each cycle of seminiferous epithelium was 10.4 days and the total duration of spermatogenesis based on 4.5 cycles was 46.8 days. The number of round spermatids for each pachytene primary spermatocytes (meiotic index) was 2.8, meaning that significant cell loss (30%) occurred during the two meiotic divisions. The total number of germ cells and the number of round spermatids per each Sertoli cell nucleolus at stage 1 of the cycle were 9.8 and 5.1, respectively. The Leydig cell volume was approximately 2000 microm3 and the nucleus volume 260 microm3. Both Leydig and Sertoli cell numbers per gram of testis in cats were approximately 30 million. The daily sperm production per gram of testis in cats (efficiency of spermatogenesis) was approximately 16 million. To our knowledge, this is the first investigation to perform a more detailed and comprehensive study of the testis structure and function in domestic cats. Also, this is the first report in the literature showing Sertoli and Leydig cell number per gram of testis and the daily sperm production in any kind of feline species. In this regard, besides providing a background for comparative studies with other fields, the data obtained in the present work might be useful in future studies in which the domestic cat could be utilized as an appropriate receptor model for preservation of genetic stock from rare or endangered wild felines using the germ cell transplantation technique.
