[Morpholgical, histological and histochemical observations on the effect of albendazole on encysted larvae of Trichinella spiralis in mice].

AIM: To study the mechanism of action of albendazole on encysted larvae of Trichinella spiralis in mice. METHODS: Twelve Kunming strain mice each infected with 200 T. spiralis larvae were equally divided into two groups. Six mice of the treatment group were treated with 30 mg of albendazole/kg daily for 5 days. Morphological, histological and histochemical methods were used. RESULTS: The encysted larvae of albendazole-treated group became significantly damaged, most worms were shrunken and surrounded by inflammatory cells. Histochemical study demonstrated that the glycogen and RNA content of the larvae in the treated group was decreased, the activities of SDH, ATPase, ACP were lower than those of the control group. CONCLUSION: Albendazole can affect the physiological function of Trichinella spiralis.

Histochemical demonstration of neuraminidase effects in pneumococcal meningitis.

In three cases of pneumococcal meningitis the in vivo action of pneumococcal neuraminidase could be demonstrated. The removal of sialic acid was demonstrated in necrospy material by the use of labeled peanut agglutinin, which has a high specific affinity for the subterminal disaccharide beta-D-galactopyranosyl-(1-3)N-acetyl-D-galactosamine, thereby exposed. Furthermore, this lectin was used for a rapid in vitro histochemical assay of neuraminidase activity in cerebrospinal fluid and culture medium taken from these cases. From the clinical point of view the exposure of the disaccharide which represents the immunodominant group of the Thomsen-Friedenreich antigen may induce immunologic reactions, because all human sera contain antibodies to this cryptic antigen. Thereby, neuraminidase can contribute to the poor prognosis of pneumococcal meningitis.

Effect of parasympathectomy on the histochemical maturation of myoepithelial cells of the rat sublingual salivary gland.

This effect of parasympathectomy was assessed by fluorescent microscopy using nitrobenzoxadiazole-phallacidin, which is known for its specific binding to actin filaments. Resection of the chorda tympani within 48 h after birth inhibited the normal formation of actin in the myoepithelial cells and the maturation of myoepithelial cells. The effect of denervation decreased progressively if the operation was delayed. Denervation 30 days after birth had no effect on development and differentiation of myoepithelial cells. Moreover, the procedure produced no histochemical changes in the myoepithelial cells in mature rats, even two months later. These findings suggest that the parasympathetic innervation is closely involved in the maturation of myoepithelial cells. Further, its involvement is limited to the first 48 h after birth, during which it exerts its neurotrophic effect on the myoepithelial cells present in all the acinar buds.

Effects of prenatal undernutrition on prevertebral sympathetic neurons in the rat: a morphological and fluorescence histochemical study.

The effects of maternal undernutrition during the last seven and last fourteen days of pregnancy on neurons of the rat coeliac-superior mesenteric ganglion has been investigated in neonates and in adult animals. The parameters studied were neuron diameter and the level of neuronal noradrenaline fluorescence. The morphology was more permanently affected by fourteen than by seven days undernutrition as shown by the smaller neuron diameters persisting at six months. Both periods of undernutrition affected noradrenaline levels permanently: seven days undernutrition producing a rise and fourteen days producing a depression. These changes demonstrate the permanent effects of maternal undernutrition on the development and maturation of prevertebral sympathetic neurons.

[The effect of steroids on organ-cultured porcine trabecular meshwork--an ultrastructural, biochemical, and lectin histochemical study].

The effect of corticosteroid administration on porcine trabecular meshwork (PTM) was studied by ultrastructural and biochemical analysis. Electron microscopic observations showed an increase of elastic fibers in PTM treated with steroids. Two-dimensional polyacrylamide gel electrophoresis with silver staining showed an increase of proteins with molecular weights of 52 kDa, 66 kDa and 70 kDa. The 66 kDa protein was assumed to be a glycoprotein containing N-acetyl-D-glucosamine, as it was stained positively in the lectin blot analysis with Triticum vulgaris (WGA) and Datura stramonium (DSA). The elastic fibers reacted positively to WGA and DSA, and the degree of this reaction was strengthened by steroid treatment. These results demonstrated that steroid treatment induces ultrastructural and biochemical changes in PTM, and an increase of elastic fibers containing glycoprotein with N-acetyl-D-glucosamine.

[Histochemical observation of the effect of electroacupuncture on the livers of rats with endotoxic shock].

The experimental rats were randomly divided into three groups: i.e. control group; endotoxic shock group (the model of endotoxic shock was induced by intravenous administration of E. Coli endotoxin, 16 mg/kg); and electroacupuncture (EA) group ("Renzhong" or "Zusanli" acupoint was stimulated for 15 minutes at 1 hr after injection of endotoxin). The experimental rats were decapitated at 75 minutes after injecting endotoxin. Their livers were taken out for cryostat section, and histochemical observation. The results were as follows: 1) The glycogen in the hepatic cells of endotoxic shocked rats were almost completely depleted. The activities of SDH Mg(++)-ATPase and G-6-Pase and 5'-Nase were decreased; especially the activities of 5'-Nase in the biliary canaliculi and sinusoids were apparently reduced. 2) The content of hepatic glycogen in EA group was increased, but some of them was still depleted. The activities of SDH, Mg(++)-ATPase and G-6-Pase were slightly increased as compared with that of the endotoxic shock group. The activity of 5'-Nase was obviously increased after EA. The preliminary results indicated that EA at "Renzhong" or "Zusanli" acupoint of rats with endotoxic shock might play certain role on improving the hepatic metabolism and promoting the membrane transport action.

A histochemical study of the effects of high doses of sodium fluoride on dipeptidyl peptidase II activity in the rat incisor ameloblast.

Female Wistar rats, 3 weeks old, were given sodium fluoride in saline solution (isotonic) by intraperitoneal injection at a dose of either 0, 10 or 20 mg per kg body weight. This treatment was given 9 times over 4.5 days. After fixation by perfusion and demineralization in neutral EDTA, hemi-mandibles were sectioned in a cryostat. Sections were stained for dipeptidyl peptidase II activity, using the specific substrate Lys-Ala-MNA and the coupler Fast Blue B for histochemical localization. Staining indicative of dipeptidyl peptidase II was found in the enamel organ of the incisor, particularly in cells of the stratum intermedium and in both secretory and maturation ameloblasts. This staining was markedly reduced in ameloblasts of rats given either 10 or 20 mg sodium fluoride per kg body weight.

Morpho-histochemical study of the biological effects of sodium dodecyl sulphate on the digestive gland of the Portuguese oyster.

In the present work we have studied the effects on survival as well as histopathological and histochemical alterations on the digestive gland of the Portuguese oyster (Crassostrea angulata, Lmk.) induced by acute action of the anionic tensioactive Sodium Dodecyl Sulphate (SDS) at different concentrations (50, 100, 200 and 400 mg/L). Firstly, the LC 50 at 96 h was found to be 136 mg/L SDS. Secondly we calculated the exposure time required for 50% mortality of the specimens exposed to different concentrations, and thirdly we examined histopathological alterations (degenerative processes ranging from inflammatory responses to extreme vacuolation) and histochemical changes in the distribution of carbohydrates and proteins that appeared at each employed concentration. The degree of these alterations and the effects on survival were dependent upon the SDS concentration. The above established modifications indicate that exposure to SDS has a detrimental effect on oyster digestive gland, perturbing metabolic and nutritional functions, what may have some influence on oyster survival.

Effects of Streptomyces hyaluronidase digestion on the histochemical reactions of proteoglycans in cartilage compared with its effects on certain non-cartilaginous tissues.

To test the value of Streptomyces hyaluronidase in carbohydrate histochemistry, the effects of digestion with the enzyme on the staining of cartilage and non-cartilaginous tissues by Alcian Blue (AB) pH 1.0, AB pH 2.5, high iron diamine, low iron diamine, aldehyde fuchsin, dialysed iron-ferrocyanide and AB pH 2.5-periods acid-Schiff were studied by light microscopy. The results obtained lead to the conclusion that the Streptomyces enzyme releases not only hyaluronic acid but also chondroitin sulphates and keratan sulphates in cartilage. Since hyaluronic acid is known to be linked to chondroitin sulphate proteoglycans, the enzyme is of limited value in localizing hyaluronic acid in cartilage. However, it is useful in localizing hyaluronic acid in most non-cartilaginous tissues.

Effects of alpha-latrotoxin on the early developmental events of the sea urchin Paracentrotus lividus: a histochemical study.

The effects of the spider toxin alpha-latrotoxin (alpha-LTX) on gametes, zygotes, and early embryos of the sea urchin Paracentrotus lividus have been investigated by in vivo experiments and by histochemical studies of acetylcholinesterase (AChE) activity. Treatment of unfertilized eggs with nanomolar amounts (1 to 0.3 nmol/l) of alpha-LTX neither triggered cortical granule exocytosis, nor prevented the elevation of the fertilization layer by sperms. Instead, fertilized eggs exposed to alpha-LTX showed noticeable alterations in cell surface topography, including the appearance of prominent membrane-limited blebs. Moreover, the zygotes treated with 1 nmol/l alpha-LTX failed to cleave. The histochemical staining of treated zygotes revealed a very strong AChE activity in the cortical region, including blebs. An enzyme reaction was also found in the perivitellin space. Our results suggest the hypothesis that some alpha-LTX receptors may appear after fertilization, supporting the awareness that fertilized eggs display excitable cell features.

Quantitative histochemical studies of the effect of levamisole on splenic T-cells in thymectomized Bufo bufo larvae.

The ability of levamisole (LVS) to correct the effect of thymectomy on splenic T-cell population in Bufo bufo larvae was tested. Using the nonspecific acid alpha-naphthyl acetate esterase (ANAE) technique, the T-lymphocytes displayed a characteristic reddish brown spot (T-pattern). The total number of white cells per spleen and the number of T-pattern cells per 100 splenic white cells were determined. In addition, differential counts of the T-patterns were made on the basis of the size of their spots. Thymectomized larvae in comparison to sham-operated larvae possessed a lower total number of splenic white cells with a similar proportion of T-pattern cells, but a lower proportion of cells with a large T-pattern. After seven days of LVS treatment, the thymectomized larvae showed an increased total number of splenic white cells and an increased proportion of cells with a large T-pattern, both data falling within normal limits. The effect of LVS appeared marked in thymectomized larvae, but was slight or absent in sham-operated larvae. Differential counts of the large T-pattern cells, based on the size of ANAE reaction product, showed the capacity of LVS to stimulate T-cell maturation. It was concluded that LVS induced proliferation and differentiation of splenic T-cells and that the different sizes of T-pattern reaction represent different maturational stages of T-cells.
