Iron promotes cadmium binding to citrate.

Iron-cadmium interactions are important in cadmium toxicity. Dietary iron supplements may decrease cadmium retention after oral cadmium exposure but the underlying mechanism is not known. Using a CdS/AgS ion selective electrode to measure [Cd2+] in physiological saline solution at pH 7.4, we show that Fe2+ promotes Cd2+ binding to citrate thereby decreasing the availability of free Cd2+. This suggests the formation of high molecular weight Cd2+-Fe2+-citrate complexes. We confirm this suggestion by showing that 109Cd2+ is retained by 1 kDa cut off filters when present with total 50 microM Fe2+ plus 1 mM citrate but not when present with citrate alone. The formation of high molecular weight complexes may prevent Cd2+ absorption. As citrate is part of the diet, we suggest that these iron-cadmium interactions may contribute to the protective effect of iron against cadmium toxicity.

Prognosis in bladder cancer. A study of cytometric, morphometric and immunohistochemical techniques.

Bladder cancer affects about 2,000 people in Sweden every year. For adequate treatment, prognostic information is essential. Most of the prognostic factors applied to date still lack satisfactory predictive value. This investigation was conducted on 230 bladder tumours with a known prognosis in order to improve prognostication methods. A new modified technique for the measurement of the DNA content in formalin-fixed paraffin-embedded bladder tumour tissue was developed, by introducing a new enzyme and density gradient centrifugation before performing flow cytometry. The results were comparable with those obtained by analysing fresh or frozen tissue. DNA ploidy was found to correlate positively to stage, histological grade and progression. A methenamine-silver staining technique, which facilitates the identification of mitoses, was developed and applied for determining mitotic frequency and density. The results proved to yield prognostic markers superior to subjective grading. Intermediate grade tumours could be separated into two prognostically highly different groups. Reduced PCNA antigenicity in archival, formalin-fixed, paraffin-embedded bladder tumour tissue was observed. Antigen expression could be retrieved by heating the slides in distilled water in a microwave oven. The application of a citrate buffer could further improve the staining, which correlated to prognosis but had no independent prognostic value. Image analysis was applied in an attempt to develop computer-based grading systems. Analysis of Feulgen-stained tissue sections was performed by an object-based method, digitizing, segmenting and analyzing the images automatically. Furthermore, a texture analysis method, which estimated grey-scale co-occurrence probability matrices on digitized images, was developed; both types of grading were found to correlate significantly to subjective grading.

Effect of inorganic additives on solutions of nonionic surfactants VI: Further cloud point relations.

Disperse dosage forms stabilized with nonionic surfactants frequently contain electrolytes as active ingredients or adjuvants. Salting out of the surfactants by these electrolytes may cause breakdown of the dosage forms. The cloud point of an aqueous solution of octoxynol 9 was used to measure the salt effects. Electrolytes which salt octoxynol 9 out lower its cloud point, while salting-in electrolytes raise it. The observed cloud point effects are discussed according to the mechanisms involved. Salting out by dehydration in competition with octoxynol 9 for the available water was observed with sulfate and phosphate anions, sodium, potassium, and ammonium tribasications, and the nonelectrolyte sorbitol. The extensive self-association of water by hydrogen bonds at and below room temperature weakens its solvent power. Ions which reduce this self-association, breaking the structure of water, increased the cloud point of octoxynol 9. Among them were the iodide, thiocyanate, and nitroprusside anions. Ions which tighten the structure of water and enhance its self-association salted the surfactant out, lowering its cloud point. Among these were the fluoride and hydroxide anions. Complex formation between the ether linkages of octoxynol 9 and the following cations increased its cloud point: hydrogen (from strong acids), silver, magnesium, and zinc. Including published data, the only cations which do not form complexes with polyoxyethylated surfactants (and are, therefore, unable to salt them in) were the alkali metal ions sodium, potassium, and cesium and the ammonium ion.(ABSTRACT TRUNCATED AT 250 WORDS)

Early development of the adrenal glands in the grass snake Natrix natrix L. (Lepidosauria, Serpentes).

The aim of the study was to investigate the development and differentiation of the adrenal glands in the grass snake (Natrix natrix L.) during the early stages of ontogenesis, i.e., from egg-laying to hatching of the first specimens. The material used for the studies consisted of a collection of embryos of the grass snake. The Natrix eggs were incubated in the laboratory at a constant temperature of 30 degrees C and 100% relative humidity. Embryos were isolated in a regular sequence of time from egg-laying to hatching. The age of the embryos was qualified according to normal tables for this species. For histological and histochemical investigations, the smallest embryos were fixed in toto. From the oldest embryos, the medial region with the mesonephros and adrenal primordium were resected. Depending on the requirements of histochemical methods, the material was fixed in various fixatives, namely, 10% formalin solution, Bouin, Wood and Millonig fluid, embedded in paraffin and sectioned into serial transversal, sagittal and longitudinal sections. The sections for review were stained with H&E and azan. For detection of adrenaline and noradrenaline in chromaffin tissue, the Wood and Honoré methods were used. SGC cells were detected with the silver stain method after Bodian. For electron microscopic studies, the adrenal gland was fixed in 2.5% glutaraldehyde and 2.0% paraformaldehyde 1:1 in 0.1 M phosphate buffer at pH 7.4 and post-fixed in 1.5% osmic acid in the same buffer. The fixed sections of the adrenal glands were embedded in Epon 812. Semithin and ultrathin sections were cut on ultramicrotome ultratome IV. Semithin sections were stained with methylene blue and ultrathin sections were routinely contrasted with uranyl acetate and lead citrate, then examined and photographed with the JEM JEOL 1220 electron microscope. According to morphological and metrical observation in the course of the grass snake embryo development, one can distinguish 12 stages of development. The primordia of the adrenal cortex appear at the first trimester of egg incubation as two asymmetrical strands between the mesonephros and aorta dorsalis. They are made of dense mesenchymal cells. At the second trimester of development, primordia are penetrated by chromaffinoblasts and capillaries. The mesenchymal cells differentiate into interrenal cells, while chromaffinoblasts are arranged dorsally of the gland. The glands are enclosed by the capsule which separates them from the mesonephros. At the third trimester of the eggs incubation, only noradrenaline appears in a chromaffin tissue. At the moment of snake hatching, the adrenal glands are completely differentiated, both in their structure and their function. The primordia of the interrenal tissue differentiate from mesenchymal cells similarly to mammals. During the development of the snake interrenal tissue, several types of cells can be recognized, varying in the degree of differentiation and in ultrastructural features: 1. Undifferentiated cells with features of mesenchymal cells 2. Differentiating mesenchymal cells 3. Transitional cells with features of mesenchymal and steroidogenic cells 4. Differentiating interrenal cells with pleomorphic mitochondria and numerous lipid droplets 5. Embryonic interrenal cells containing circular lipid droplets and underdeveloped smooth endoplasmic reticulum 6. Transitional interrenal cells containing mitochondria with tubular and vesicular cristae, large lipid droplets, numerous myelin structures, and well-developed smooth endoplasmic reticulum 7. Degenerating cells of embryonic interrenal tissue 8. Differentiating mesenchymal cells with features of fibroblasts The above classification is very schematic and presumptive. In developing adrenal glands at the first trimester of egg incubation type 1 and 2 cells predominate. Type 3 and 4 cells were observed at the second trimester of the adrenal primordia development. At the third trimester of egg incubation, embryonic adrenal glands were composed of the type 5 cells. At the moment of snake hatching, interrenal tissue contained type 5 and 6 cells. In the next days of the adrenal gland development, at the border between the cortex and in medulla as under the capsule, numerous cells were degenerated. During the entire development period the adrenal capsule was built from type 7 cells. The chromaffin tissue of the adrenal glands is derived from the neural crest. These findings agree with the findings of all embryologists. The first chromaffinoblasts infiltrated the adrenal cortex primordium around stage IV of development. They were mixed with interrenal cells and just at hatching they were localized dorsally of the gland. The chromaffinoblasts differentiated gradually from neuron-like cells to typical chromaffinocytes. All the chromaffinoblasts contained the chromaffin granules. The size and numerical density of the chromaffin granules increased with development. Just before hatching, the cells of the chromaffin tissue contained only noradrenaline. Differentiation chromaffinoblasts into chromaffin cells are probably stimulated and controlled by the influence of hormones, which are produced by the cells of the interrenal tissue. According to morphological, histochemical and ultrastructural observation in the course of adrenal differentiation and development in the grass snake, six morphological phases can be distinguished.

[Accumulation and localization of 67Ga citrate in cancerous lung tissue during diagnostic use of the preparation].

Altogether 89 patients were examined using scintigraphy with 67Ga-citrate. Radiometric, scintigraphic, radioautographic and histological investigation of the operation material of 15 patients was conducted. The diagnostic accuracy was 92.1%, sensitivity 98.6%, specificity 75%. The investigation of the operation material showed that RP accumulation in the peripheral part of tumor was 4-4.5 times higher and in its central part 2-3 times higher than in the unaffected pulmonary tissue. Elevated RP accumulation was found in the capsular wall in inflammatory processes as well as in lymph nodes with anthracosis. Recovered silver grains on histoautoradiograms of tumor tissue zones were mainly localised around isolated cells and their groups in the intercellular space.

Bismuth tracing in organotypic cultures of rat hippocampus.

Bismuth is known to have neurotoxic side effects in humans and animals. In the 1970s France experienced about a thousand cases of patients suffering from bismuth-induced encephalopathy. Studies suggest that bismuth may provoke a selective degeneration of CA1 pyramidal cells in the organotypic cultures of rat hippocampus. A currently established technique for the histochemical visualization of bismuth was applied on hippocampal tissue cultures allowing the tracing of bismuth in concentrations hitherto not possible. The accumulation and subcellular localization of bismuth is demonstrated in the tissue cultures of rat hippocampus. CA1 pyramidal cells in the rat hippocampus exhibit the highest uptake of bismuth. High bismuth citrate concentrations (10 microM) are able to totally destroy the cytoarchitecture of the hippocampus. At ultrastructural levels bismuth was found to be located exclusively in lysosome-like organelles.

Self assemblies of nanocrystals: preparation, collective properties and uses.

Self-organization of nanocrystals depends on the type of nanomaterial and the coating. With silver nanocrystals, the self-organization is partially perturbed by the latter. With cobalt nanocrystals, the size distribution and thus the self-organization is controlled by the amount of reducing agent added during the chemical reaction and not by the micellar solution. It is possible to make "supra" crystals of cobalt nanoparticles in a face centered cubic (fcc) structure. By applying, during the evaporation process, an external magnetic field to ferrite nanocrystals dispersed in solution, nanocrystal organizations markedly change with their coating. Hence tubes are obtained with nanocrystals coated with citrate ions whereas thick films are produced when the coating is replaced by dodecanoic acid. Collective magnetic properties due to the organization in tubes are observed with a behavior similar to that observed with nanowires. The use of nanocrystals as a mask to produce various patterns on silicon is described.

Macrophage accumulation of inhaled gallium-67 citrate in normal lungs.

Injected 67Ga has been used extensively to monitor inflammatory processes in the peripheral lung. We hypothesized that inhaled 67Ga may be useful in marking early airway inflammation in smokers. Eight nonsmokers and eight smokers breathed a 67Ga aerosol and imaging was performed immediately and 24 and 96 hr later. Approximately two-thirds of the initial dose remained in the lungs at 24 hr in both groups and no difference was seen between the groups. Only a very slight decrease was seen in both groups at 96 hr suggesting the gallium becomes bound to lung tissue or to cells not rapidly removed from the lungs. Autoradiography was performed on tissue from two smoke-exposed guinea pigs and two human patients undergoing resection surgery who breathed the gallium aerosol 24 hr prior to tissue removal. Silver grain accumulations were seen only over macrophages. We conclude that macrophage associated accumulation of 67Ga occurs in healthy lungs, and that it is not feasible to use aerosolized gallium to assess airway inflammation in smokers.

Synthesis and characterization of deoxyribonucleic acid-conjugated gold nanoparticles.

Gold nanoparticles functionalized with thiol-modified single-stranded oligonucleotides are highly useful reagents for a variety of applications, ranging from materials science to bioanalytics. In this chapter, the preparation of citrate stabilized 15-nm Au nanoparticles is described. The nanoparticles are conjugated with 3'-thiol-modified deoxyribonucleic acid oligomers and the resulting conjugates are characterized by determining their shape, size, and surface coverage. The hybridization capabilities are quantified in a microplate assay.

Effects of any epoxymethano stable analogue of prostaglandin endoperoxides (U-46619) on human platelets.

U-46619 is a stable epoxymethano analogue of cyclic endoperoxide PGH2. We studied platelet aggregation, 14C-5HT release, LDH extrusion and prostaglandin and thromboxane production induced by this compound in platelet-rich plasma samples from 15 healthy volunteers. Each subject was tested both before and 90 min after aspirin (500 mg) ingestion. The threshold aggregating concentration (TAC) of U-44619 ranged between 0.18 and 0.90 micro M. Aggregation was maximal between 40 and 60 min after venipuncture and was concentration-dependent. At concentrations below the TAC, U-44619 induced primary reversible aggregation with minimal 14C-5HT release. At TAC or higher concentrations aggregation and release proceeded as parallel events. Neither prostaglandin or thromboxane production nor LDH loss could be detected in any of the situations tested. Aspirin ingestion did not modify the pattern of platelet responses. In unstirred, not aggregated platelet samples 14C-5HT release by U-46619 occurred to a similar extent as in stirred, aggregated platelet samples. Addition to citrated PRP of 0.3 mM Na2 EDTA blocked both aggregation and release induced by U-46619. This compound, however, aggregated washed platelets resuspended in Ca++-free-tyrode-albumin containing fibrinogen. The mechanism by which U-46619 activates platelets differs from that of all other common aggregating agents.

A rabbit lung model for testing reaction to inhaled dental restorative particles.

A study was designed to test the outcome of the deposition of particles of plastic composite dental restorative material in rabbit lungs. Grinding and polishing these restorations in situ produces some particles in the 0.5- to 10-microns size range that easily enter and remain in human lungs and are associated with industrial lung disease. Dental restorative plastic material was ground in the laboratory, suspended in saline solution, and injected transtracheally into four New Zealand white rabbits. Two control rabbits were similarly injected with saline solution transtracheally. Twenty-four hours later, the rabbits were injected with 1 mCi of 67Ga citrate intravenously and subsequently reanesthetized for scanning. Baseline scans were obtained in the six animals prior to the injection of the test particles. Positive gallium scans were obtained 72 h after the administration of particulate material in the four test rabbits. The gallium scans of the control rabbits remained no different from baseline. The study was repeated one month later. The animals were killed seven days after the last gallium scan. Light microscopy and transmission electron microscopy of the lungs of the test animals showed foci of chronic inflammation around particles of the restorative material. Particles were in vacuoles within alveolar macrophages and also free in interstitium. Control animals had normal histologic conditions. Silver amalgam and gold dental restorations have years of clinical use but the new plastic composite restorative materials are rapidly being introduced into human clinical dental practice. Normal use involves polymerization, grinding, and polishing of the material within the mouth. The chronic inflammation in the lungs of rabbits indicates a need to test dental restorative material for lung biocompatibility before further, extensive clinical use.

Thiobacillus acidophilus sp. nov.; isolation and some physiological characteristics.

After a brief exposition to glucose, Thiobacillus acidophilus was isolated from a culture of iron-grown T. ferrooxidans. Physicochemical analysis of its DNA showed a G+C content of 62.9-63.2%. The new isolate grows best at 25-30 degrees C and at pH 3.0. Growth is possible between pH 1.5 and 6.0. Thiobacillus acidophilus is apparently strictly aerobic. Ammonium salts are the only suitable source of nitrogen. The bacterium is a facultative autotroph. In addition to elemental sulfur, it obtains energy from organic compounds such as D-glucose, D-galactose, D-fructose, D-mannitol, D-xylose, D-ribose, D-arabinose, L-arabinose, sucrose, sodium citrate, malic acid,dl-aspartic acid, and dl-glutamic acid. Thiobacillus acidophilus possesses the key enzymes of the tricarboxylic acid (TCA) cycle including NAD-and NADP-linked isocitric dehydrogenase and alpha-ketoglutarate dehydrogenase, and the key enzymes of the hexose monophosphate pathway (glucose-6-phosphate and 6-phosphogluconate dehydrogenase, and fructose 1,6-diphosphate aldolase). NADH oxidase has been found in particulate fraction of extracts. Rhodanese and thiosulfate oxidase have also been detected.

Isolation and reconstitution of the n-butylmalonate-sensitive dicarboxylate transporter from rat liver mitochondria.

The mitochondrial dicarboxylate carrier has been substantially purified from rat liver mitoplasts by extraction with Triton X-114 in the presence of cardiolipin followed by chromatography on hydroxylapatite. Upon incorporation of the hydroxylapatite eluate into phospholipid vesicles, an n-butylmalonate-sensitive malonate/malate exchange has been demonstrated. This exchange activity is enhanced 226-fold relative to the starting material (i.e. detergent-extracted mitoplasts). Silver-stained sodium dodecyl sulfate-polyacrylamide gradient gels verify the high purity of this fraction relative to the starting material. Nonetheless, the banding pattern indicates that several protein species are still present. As isolated, the dicarboxylate transporter is rather unstable but can be stabilized either by the addition of 10% ethylene glycol and subsequent storage at -20 degrees C or by incorporation into phospholipid vesicles in the presence of malate followed by freezing in liquid nitrogen. Such proteoliposomes catalyze a [14C]malonate uptake which is characterized by a first order rate constant of 1.02 min-1 and a t 1/2 of 41 s. This uptake can be inhibited by dicarboxylates (e.g. succinate, malate, unlabeled malonate) but not by either alpha-ketoglutarate or by tricarboxylates (e.g. citrate, threo-Ds-isocitrate). Furthermore, the reconstituted malonate transport is dependent on internal malate and can be inhibited by n-butylmalonate, mersalyl, p-chloromercuribenzoate, and Pi, but not by N-ethylmaleimide. It is concluded that this highly purified fraction contains a reconstitutively active dicarboxylate transporter which, based on its substrate specificity and inhibitor sensitivity, appears to be identical to the native dicarboxylate transport system found in intact rat liver mitochondria.

Design and development of a highly stable hydrogen peroxide biosensor on screen printed carbon electrode based on horseradish peroxidase bound with gold nanoparticles in the matrix of chitosan.

The design and development of a screen printed carbon electrode (SPCE) on a polyvinyl chloride substrate as a disposable sensor is described. Six configurations were designed on silk screen frames. The SPCEs were printed with four inks: silver ink as the conducting track, carbon ink as the working and counter electrodes, silver/silver chloride ink as the reference electrode and insulating ink as the insulator layer. Selection of the best configuration was done by comparing slopes from the calibration plots generated by the cyclic voltammograms at 10, 20 and 30 mM K(3)Fe(CN)(6) for each configuration. The electrodes with similar configurations gave similar slopes. The 5th configuration was the best electrode that gave the highest slope. Modifying the best SPCE configuration for use as a biosensor, horseradish peroxidase (HRP) was selected as a biomaterial bound with gold nanoparticles (AuNP) in the matrix of chitosan (HRP/AuNP/CHIT). Biosensors of HRP/SPCE, HRP/CHIT/SPCE and HRP/AuNP/CHIT/SPCE were used in the amperometric detection of H(2)O(2) in a solution of 0.1M citrate buffer, pH 6.5, by applying a potential of -0.4V at the working electrode. All the biosensors showed an immediate response to H(2)O(2). The effect of HRP/AuNP incorporated with CHIT (HRP/AuNP/CHIT/SPCE) yielded the highest performance. The amperometric response of HRP/AuNP/CHIT/SPCE retained over 95% of the initial current of the 1st day up to 30 days of storage at 4 degrees C. The biosensor showed a linear range of 0.01-11.3mM H(2)O(2), with a detection limit of 0.65 microM H(2)O(2) (S/N=3). The low detection limit, long storage life and wide linear range of this biosensor make it advantageous in many applications, including bioreactors and biosensors.

Arachidonic acid induces human platelet-fibrin retraction: the role of platelet cyclic endoperoxides.

Arachidonic acid (0.2-0.8 mM) retracts clots formed in human citrated platelet-rich plasma by batroxobin. Extracellular calcium ions, but not the secretion of ADP by platelets, are required. AA-induced clot-retraction requires cyclo-oxygenase but not thromboxane synthetase activity since the retraction is inhibited by aspirin but not by selective inhibitors of thromboxane synthesis. The data indicate that endogenous cyclic endoperoxides mediate the retraction. Moreover, intact endoperoxide/thromboxane receptors also seem to be necessary because clot retraction is inhibited by thromboxane receptor antagonists.

[Preliminary proteome analysis for Saccharomyces cerevisiae under different culturing conditions].

For the investigation of the metabolic regulation of Saccharomyces cerevisiae under different culture conditions, the proteins of cell utilizing various carbon sources were separated by two-dimensional electrophoresis with immobilized pH gradients as the first dimension and SDS-PAGE as the second. Samples were taken in the log phase of batch culture using glucose or lactic acid as carbon source, while another sample was taken from the broth when glucose was consumed up and ethanol accumulated in the previous phase was further metabolized. After electrophoresis, the protein spots were detected by silver-stain in a Hoefer Automated Gel Stainer with a protein silver staining kit. Silver-stained gels were scanned and digitized to create computer images. About 500 protein spots were detected by employing the 2D proteome image analysis system Image Master 2D Elite and SWISS-2DPAGE proteome database. Most of the protein expressed and involved in the glycolysis, pentose phosphate (PP) pathway, anaplerotic pathway, as well as TCA cycle were analyzed. The metabolism regulation of protein level for Saccharomyces cerevisiae under various carbon sources, as well as during different phase of growth, was studied. The expression of several glycolytic enzymes (glk, pgi, pgk, eno, pyk) was up-regulated while the expression of enzymes in oxidative pentose phosphate pathway (zwf, gnd) was down-regulated when ethanol and lactic acid were taken as carbon source. Simultaneously, frucotose 1,6-biphosphatase was found to be up-regulated due to the gluconeogenic requirement. Citrate synthase and Malate dehydrogenase do not exhibit significant difference, indicating TCA cycle is necessary when utilizing glucose, ethanol or lactic acid as carbon source. Thus, the NADPH loss due to the repressed pentose phosphate pathway could be compensated by TCA cycle in cases of ethanol and lactic acid. The expression of malic enzyme and isocitrate lyase are activated to a large extent when metabolizing ethanol, indicating glyoxylate shunt is essential in transferring ethanol to generate four carbon precursors for the biosynthesis and the NADP-dependent malic enzyme could also serve as compensation mechanism for NADPH loss in this case.

Diagnosis of chronic and postoperative osteomyelitis with gallium 67 citrate scans.

The effectiveness of gallium 67 citrate in diagnosing the exacerbations of chronic osteomyelitis or postoperative osteomyelitis secondary to open reduction of fractures was tested in thirteen patients. This isotope demonstrated an area of infection in every case in which such an area existed. The control subjects consisted of patients with noninfected wounds after open reduction. In this group, gallium was not incorporated into the post-operative hematoma. No false-negative results were observed in our series, demonstrating that this element is useful in the early diagnosis of bone infections.
