[Determination of adenosine and thymidine in Fritillaria bulbs by dual wavelength ultraviolet spectrophotometry].

A convenient method for the quantitative analysis of adenosine and thymidine in Fritillaria bulbs was developed by means of dual wavelength ultraviolet spectrophotometry. This method has good linear relationship and the interrelated coefficient of standard curve for adenosine and thymidine were all found to be 0.9999. The methanol extracts of four species of Fritillaria have been analyzed with this method. The results show that the the bulbs of four species contain different quantities of adenosine and thymidine, which indicates that the nucleosides may be responsible for the anti-coagulation activity of Fritillaria.

[Determination of adenosine and thymidine in Fritillaria bulbs by ultraviolet spectrophotometry].

A convenient method for quantitative analysis of the nucleosides in Fritillaria bulbs has been developed by means of thin layer chromatography and ultraviolet spectrophotometry. The MeOH extracts from four species of Fritillaria were analyzed with this method. The results show that both adenosine and thymidine in the four species are quite disparate, suggesting that the nucleosides may be responsible for the anti-coagulative activity of Fritillaria.

Simultaneous determination of enalapril maleate and hydrochlorothiazide by first-derivative ultraviolet spectrophotometry and high-performance liquid chromatography.

Two methods are described for the simultaneous determination of enalapril maleate and hydrochlorothiazide in combined pharmaceutical tablets. The first method depends on first-derivative ultraviolet spectrophotometry, with zero-crossing and peak-to-base measurement methods. The first-derivative amplitudes at 224 and 260 nm were selected for the assay of enalapril maleate and hydrochlorothiazide, respectively. The second method is based on high-performance liquid chromatography on a reversed-phase column using a mobile phase of acetonitrile-water (20:80, v/v) (pH 3.8) with programmable detection at 215 and 275 nm. Both methods showed good linearity, precision and reproducibility. The proposed methods were successfully applied to the determination of these drugs in laboratory-prepared mixtures and in commercial tablets.

Deuteration kinetics of deoxyguanosine, deoxycytidine, and their polynucleotides by means of ultraviolet spectrophotometry.

The kinetics of the hydrogen-deuterium exchange reactions of deoxyguanosine (dG), deoxycytidine (dC), double-helical poly[d(G-C)] X poly[d(G-C], and double-helical poly(dG) X poly(dC) have been examined at 20 degrees C, pH 7.0, and in low-salt (0.15 M NaCl) medium by stopped-flow ultraviolet spectrophotometry, in the spectral region of 260 to 320 nm. The rate constant was found to be 78.9 s-1 for dG-NH, 2.2 s-1 for dG-NH2, 39.3 s-1 for dC-NH2, 2.4 s-1 (fast) and 0.94 s-1 (slow) for poly[d(G-C)] X poly[d(G-C)], and 2.2 s-1 (fast) and 0.92 s-1 (slow) for poly(dG) X poly(dC). From these values, the probability of base-pair opening of the G X C containing B-form double helix is estimated to be (3 +/- 1) X 10(-3). This is much greater than what is expected from an extrapolation of the van't Hoff plot at the helix-coil transition region, i.e. at about 110 degrees C. The mechanism of these base-pair openings at 20 degrees C (as well as the mechanism of base-pair reformation) is suggested to be totally different from those in the melting temperature range.

UVA/UVB sunscreen determination by second-order derivative ultraviolet spectrophotometry.

UVA/UVB protective agents are important constituents in many cosmetics and sunscreen preparations. Analytical methodologies which enable the determination of combined substances with different spectrophotometric characteristics are therefore useful and cost-effective. To this end, the following substances in a sunscreen gel were analyzed: the UVB and UVA protective substances, 2-hydroxy-4-methoxy-benzophenone-5-sulphonic acid (BZP) and 2-phenylbenzymidazole-5-sulphonic acid (PBS), respectively, were evaluated by second-order derivative ultraviolet spectrophotometry in distinct solvents and wavelengths. The method used for determination of BZP in 95% ethanol was the zero-peak (ZP) at 356.0 nm. For PBS, the solvent used was 0.1 mol l-1 triethanolamine, and the methods were the ZP at 317.5 nm and the peak-peak (PP) at 317.5-323.0 nm. Statistical results for the methods showed linearity of 0.9999 and an average precision of 1.12% (ZP) for BZP, and 0.55% (ZP) and 0.63% (PP) for PBS. The average recovery for BZP and PBS were 97.28% (ZP), 103.13% (ZP) and 103.71% (PP), respectively.

[Preparation of liposome podophyllotoxin chitosan film and determination the podophyllotoxin content in the film by ultraviolet spectrophotometry].

OBJECTIVE: To optimize the formula of liposome podophyllotoxin chitosan film and accurately determine the podophyllotoxin content in the film. METHODS: Liposome podophyllotoxin chitosan films were prepared according to different formula derived from the results of orthogonal design by selecting chitosan, acetic acid and gelatin as the 3 factors for preparation, each factor including 3 levels with liposome podophyllotoxin as the blank factor. Comprehensive evaluation of the formulas was performed on the basis of the conglutination and solubility of the prepared film to select the optimal formula. The content of podophyllotoxin was determined by ultraviolet spectrophotometry at 292 nm. RESULTS: The conglutination and solubility were optimal of the film prepared from 2% chitosan, 1% acetic acid, 2% gelatin and 2.8% liposome podophyllotoxin chitosan, showing good linearity within podophyllotoxin concentration range of 5-25 microg/ml (r=0.999 89) with an average recovery rate of 99.7% (n=5) and RSD of 1.11%. CONCLUSION: The film prepared according to the orthogonal design results conforms to the requirements in terms of conglutination and solubility, and the method we adopted is fast and accurate, yielding qualified film with censored podophyllotoxin content that distributes homogeneously.

Diagnosis of Smith-Lemli-Opitz syndrome by ultraviolet spectrophotometry.

Smith-Lemli-Opitz syndrome (SLOS) is an autosomal recessive disorder due to an inborn error of cholesterol metabolism, characterized by congenital malformations, dysmorphism of multiple organs, mental retardation and delayed neuropsychomotor development resulting from cholesterol biosynthesis deficiency. A defect in 3 -hydroxysteroid-delta7-reductase (delta7-sterol-reductase), responsible for the conversion of 7-dehydrocholesterol (7-DHC) to cholesterol, causes an increase in 7-DHC and frequently reduces plasma cholesterol levels. The clinical diagnosis of SLOS cannot always be conclusive because of the remarkable variability of clinical expression of the disorder. Thus, confirmation by the measurement of plasma 7-DHC levels is needed. In the present study, we used a simple, fast, and selective method based on ultraviolet spectrophotometry to measure 7-DHC in order to diagnose SLOS. 7-DHC was extracted serially from 200 l plasma with ethanol and n-hexane and the absorbance at 234 and 282 nm was determined. The method was applied to negative control plasma samples from 23 normal individuals and from 6 cases of suspected SLOS. The method was adequate and reliable and 2 SLOS cases were diagnosed.

Analysis of trifluralin and other dinitroaniline herbicide residues by zero-order and derivative ultraviolet spectrophotometry.

The utility of zero-order and first- and second-derivative ultraviolet (UV) spectrophotometry for the identification of benfluralin, trifluralin, isopropalin and oryzalin is discussed. These four herbicides were determined by zero-order and first-derivative UV spectrophotometry, with linear calibration graphs established between 50 and 100 concentration units and limits of detection ranging from 1 to 7 micrograms ml-1. The application of these techniques to the residue analysis of fortified soils and niebe and peanut leaves is described. Trifluralin residues were found to be 6.7, 8 and 1.7 micrograms ml-1 in samples of fortified soils, niebe leaves and peanut leaves, respectively. Isopropalin residues were found to range from 62 to 154 micrograms ml-1 in samples of fortified niebe leaves.

Determination of amlodipine in pharmaceutical dosage forms by liquid chromatography and ultraviolet spectrophotometry.

A liquid chromatography (LC) method and an ultraviolet (UV) spectrophotometric method were developed and validated for quantitative determination of amlodipine in tablets and compounded capsules. The isocratic LC analyses were performed on an RP18 column using a mobile phase composed of 0.1% (v/v) ortho-phosphoric acid (pH 3.0) -acetonitrile (60 + 40, v/v) at a flow rate of 1.0 mL/min. The UV spectrophotometric method was performed at 238 nm. The analytical methods were validated according to International Conference on Harmonization Guidelines. The calibration graphs were linear [correlation coefficient (r) > 0.999] in the studied concentration range of 10-30 microg/mL for LC and 10-35 microg/mL for UV spectrophotometry. The relative standard deviation values for intraday and interday precision studies were less than 2%, and the accuracy was greater than 98% for both methods. The specificity of the LC method was proved using forced degradation. Statistical analyses showed no significant difference between the results obtained by the 2 methods. The proposed methods are precise and accurate and can be applied directly and easily to the oral pharmaceutical preparations of amlodipine.

Determination of absorption coefficients of purified proteins by conventional ultraviolet spectrophotometry and chromatography combined with multiwavelength detection.

The direct, ultraviolet spectrophotometric determination of protein absorption coefficients was found to be more reproducible and accurate when diluting was replaced by chromatography and multiwavelength detection. Four different ultraviolet spectrophotometric methods, described in the literature, were compared by calculating A0.1%280 values from the spectra of 25 proteins, obtained by chromatography. Only two methods, i.e., one based on the absorbance at 210 nm and the other on the absorbance at 205 nm, corrected for the absorbance of aromatic amino acids at that wavelength, were sufficiently accurate to be of potential use for the determination of unknown proteins. It was found, however that with uncorrected A203 values even better results could be achieved. Using 7 well-defined proteins the equation A0.1%280 = 38.69 X A280/A203 - 0.01 was established by linear regression. A0.1%280 values for 14 pure proteins calculated with this equation showed a mean deviation of only 4% from literature values. Since similar deviations were seen with 5 chromophoric and 7 glycoproteins, 3 and 7% respectively, the method may have universal applicability. In the configuration used, only 40 micrograms of a protein is required for the chromatographic determination of its absorption coefficient.

Ultraviolet spectrophotometry as an index parameter for estimating the biochemical oxygen demand of domestic wastewater.

The relationship between ultraviolet absorbance at 280 nm (UV280) and the 5-day Biochemical Oxygen Demand (BOD5) test was evaluated using wastewater samples collected during March - December 1998 from the Fort Meade wastewater treatment plant (Maryland, U.S.A.). Three types of samples were collected: raw influent wastewater, primary effluent, and the effluent from the nitrification settling basin. A regression of BOD5 on UV280 was obtained using half of the data, with the other half of the data used to test application of the equation. The presence of NO3 and NO2, did not interfere with the BOD5/UV relationship. However, the relative fraction of organic compounds that absorb at UV280 and are biodegradable did appear to decrease across the treatment plant, thereby reducing the strength of the association between BOD5 and UV280 further along the treatment train. Interestingly, the exclusion of solids > 1 microm from the BOD5 test did not strengthen the association between BOD5 and UV280. These results suggest that simple UV absorbance measurements may be a useful analytical tool for wastewater treatment personnel, allowing them to quickly monitor for changes in the BOD5 during the treatment process and to quickly estimate the BOD5 when determining what dilutions to use in the standard BOD5 test. However, such relationships are likely to be wastewater and treatment plant specific and variable with time and treatment.

[Determination of plasma ethambutol with liquid chromatography and ultraviolet spectrophotometry].

The method presented here is based on the ability of ethambutol to give chelates with divalent cationic metals. With copper salts, the chelate presents a characteristic ultraviolet absorbance at 270 nm, making it possible to titrate ethambutol extracted from plasma with chloroform in alkaline medium. The column which was used contained silica (LiChrosorb Si 60, 5 microns) and the mobile phase was a mixture of equal parts of water and acetonitrile, containing copper sulphate and ammonia. The internal standard was a lower homologue of ethambutol with the same chelating ability. The detection limit was 0.15 mg/l. No interference with other antitubercular agents was observed.

The photoadaptive response to ultraviolet exposure in human skin using ultraviolet spectrophotometry.

BACKGROUND: Both pigmentation and non-pigmentary processes contribute to the development of photoadaptation yet the exact contribution of either in the resting state and in response to ultraviolet (UV) radiation is unclear. The purpose of this study was to estimate independently these changes occurring in the epidermis following repeated exposure to UV in two groups with differing degrees of constitutive pigmentation. METHODS: We describe a mathematical model for explaining the spectral absorbance of excised human epidermis based on the absorbance of constituent chromophores. The model was applied to spectral absorbance data measured on samples of epidermis excised from pre-irradiated skin and from skin obtained following UV irradiation on 3 successive days. RESULTS: We found that in Asian skin there was only a mild photoadaptive response, principally by a small increase in pigmentation. On the other hand, the significant adaptive response in Caucasian skin was through hyperplasia of the epidermis, with tanning contributing only to a much smaller degree. CONCLUSION: This study has enabled us to study independently the pigmentary and non-pigmentary pathways and has shown that in those people with a lower degree of constitutive pigment, the primary mechanism of photoadaptation is via the non-pigmentary route.
