Reversible intracellular displacement of the cytoskeletal protein and organelles by ultracentrifugation of the sympathetic ganglion.

Superior cervical ganglia of rats were centrifuged at 40,000 rpm (160,500 g) for 1 h at 4 degrees C. Most neuronal somata exhibit a minor centripetal domain free of organelles and a major centrifugal domain rich in organelles. The former is occupied by numerous fine granules having low electron density unlike ribosomes in epoxy sections stained with uranium and lead, and is occupied by a meshwork of microtrabecular or filamentous elements similar to that of the centrifugal domain as well as that of the normal cells in PEG (polyethylene glycol)-processed embedment-free sections without staining. The latter centrifugal domain contains regular cell organelles except for neurofilaments without stratification. All the organelles are suspended in the meshwork of microtrabecular or filamentous elements. In immunolight microscopy, NFPs (neurofilament proteins) are confined to the centripetal domain. In immunoelectron microscopy using ultrathin cryosections and the protein A-gold labeling, numerous gold-particles for NFPs were deposited randomly in the centripetal cytoplasmic domain without long linear alignment. In the PEG sections the gold-labelings for NFPs are randomly deposited on portions of the microtrabecular strands in the centripetal domain. After incubation of the centrifuged ganglia in the anterior eye chamber overnight, NFPs-immunoreactivity appears again diffusely throughout the entire cytoplasm of all neuronal somata in immunolight microscopy. The organelle-free domain of the cytoplasm is no longer visible in electron microscopy. The present findings are discussed in relation to the state of the cytoplasmic soluble proteins and the reality of the microtrabecular or filamentous elements in the cytoplasm.

Cataract-related changes in protein aggregates of human lens studied by ultracentrifugation.

In human lens, cataract development causes an increase in the amount of protein aggregates. Their specific density of 1.25 g/ml is much higher than that of 1.14 g/ml and 1.06 g/ml obtained for water-soluble proteins. During the formation of water-insoluble aggregates their size varies up to some micrometers. Infrared spectra of water-soluble protein and in the aggregates confirm that the content of bound water in aggregates decreases.

The use of zonal ultracentrifugation in the investigation of the binding of delta9-tetrahydrocannabinol by plasma lipoproteins.

The major classes of lipoprotein, very low density, low density, and high density, as well as the lipoprotein-free protein were isolated from plasma by rate-zonal centrifugation. Fractionation of plasma after the addition of delta9-tetrahydrocannabinol (THC) showed that over 60% of the drug is associated with the lipoprotein; the remainder of the drug appears to be bound by albumin. In human plasma the low density lipoprotein is the major lipoprotein; in rat plasma the very low density and high density lipoproteins predominate. The distribution of THC in the lipoprotein reflected this species difference. In both species the distribution of THC among the lipoproteins appears to be related to their content of neutral lipid or total lipid rather than that of phospholipid or protein. Fractionation of plasma after the addition of estradiol, imipramine, prostaglandin E2, digitoxin, or dicumarol demonstrated that the lipoproteins contribute little to their binding. Thus, even among lipid-soluble compounds, the binding of THC in plasma is unusual.

Characterization of androgen receptor by high performance liquid chromatography and sucrose density gradient ultracentrifugation in normal and malignant human prostatic tissues.

In the present study we have characterized androgen-receptor complexes of normal and malignant human prostate cytosols using sucrose density gradient centrifugation, gel permeation and anion exchange high performance liquid chromatography (HPLC). Our results indicated that: 1) malignant tissue cytosols differed from normal by the presence of a 4-5S androgen receptor form which accounted for 30% of total specific-binding of malignant tissue cytosols, 2) 8-9S androgen-receptor complexes in normal and malignant prostate cytosols were estimated as 270kDa by gel permeation HPLC, 3) 8-9S complexes were retained and could be eluted by 0.22M KCl on a linear gradient anion exchange HPLC, 4) 4-5S androgen-receptor complexes were estimated as 90kDa by gel permeation HPLC and were not retained on anion exchange HPLC in our experimental conditions, and 5) either 10X dilution of the 4-5S complexes and subsequent anion exchange HPLC, or anion exchange chromatography of 8-9S complexes at 22 degrees C were causing fragmentation of the androgen receptor molecule from normal and malignant tissues. These fragments had enhanced affinity for anion exchange columns. These results are discussed in relation to the composition of the nontransformed androgen receptor macromolecule.

[Effectiveness of isolation ultracentrifugation in the combined treatment of heart failure refractory to therapy developing in the presence of severe arterial hypertension with stable and malignant courses].

The feasibility and practicability of isolated ultrafiltration, or extracorporeal evacuation of excessive body fluid, was assessed in patients with severe stable and malignant arterial hypertension, complicated by refractory heart failure. This procedure, added to combined treatment, is a means to overcome resistance to medication, achieve a stable compensation of circulatory insufficiency symptoms, bring down arterial blood pressure and use smaller doses of hypotensive and diuretic drugs. Isolated ultrafiltration proves less effective in patients with cachectic circulatory insufficiency.
