Diastereoselective multicomponent cyclizations of Fischer carbene complexes, lithium enolates, and allylmagnesium bromide leading to highly substituted five- and six-membered carbocycles.

The one-pot sequential reaction of a chromium alkoxycarbene complex, a ketone or ester lithium enolate, and allylmagnesium bromide enabled the selective synthesis of novel diastereomerically pure pentasubstituted cyclopentanols or tetrasubstituted 1,4-cyclohexanediols, depending on the degree of substitution at the Cbeta position of the enolate anion. A few exceptions have been encountered in which tetrasubstituted cyclopentanols or pentasubstituted 1,4-cyclohexanediols were selectively formed. The use of 2-iodoethoxycarbene complexes gave access to 1,2,4-cyclohexanetriols. These multicomponent-coupling reactions involved the formation of lithium alkylpentacarbonylchromates as key intermediates, which further evolved through intramolecular processes, such as insertion of an alkene, CO insertion or addition to a carbonyl group, and, moreover, could be trapped in intermolecular reactions with different electrophiles and styrene. The substitution pattern of the alkylchromate carbon chain has been proposed to control the nature of the annulation process.

Benzylation of aromatic compounds with different crystallites of MgO.

Magnesium oxide nanocrystals are shown to be selective catalysts for the benzylation of aromatics with benzyl chloride, in the order xylene > toluene >> benzene. Normal polycrystalline MgO is not as effective as the nanomaterials. However, at the highest surface area, normally most reactive MgO samples (aerogel prepared AP-MgO) were poorer catalysts than a hexagonal platelet form (CP-MgO) of lower surface area. During the catalysis, the MgO is partially converted to MgCl(2), but crystal shape differences are not washed out. Rate data, differential thermal analysis, and consideration of crystal shapes and faces suggest that the CP-MgO form is most catalytically active because of optimal adsorption, molecular trafficking, and desorption energies. The AP-MgO apparently adsorbs the reactants too strongly, and thereby this hinders the catalytic process. These results serve as another example of where nanocrystalline shape plays an important role.

The effects of magnesium and ammonium additions on phosphate recovery from greenhouse wastewater.

Phosphorus recovery from greenhouse wastewater, using precipitation-crystallization, was conducted under three levels of calcium concentration, 304 mg/L (7.6 mmol/L), 384 mg/L (9.6 mmol/L), and 480 mg/L (12 mmol/L), and also with additions of ammonium and magnesium into the wastewater. Jar test results confirmed high phosphate removal, with more than 90% of the removal achieved with a pH as low as 7.7. Under the low calcium concentration, ammonium addition affected the chemical reactions at pH lower than 8.0, where struvite was produced; when the pH was raised to 8.8, other calcium compounds dominated the precipitation. Under the medium calcium concentration, ammonium and magnesium addition helped struvite precipitation in the low pH range. Hydroxyapatite (HAP) was the main product. Under the high calcium concentration, ammonium addition showed no effects on the precipitation.

DESCRIPTIVE ANALYSIS OF DIVALENT SALTS.

Many divalent salts (e.g., calcium, iron, zinc), have important nutritional value and are used to fortify food or as dietary supplements. Sensory characterization of some divalent salts in aqueous solutions by untrained judges has been reported in the psychophysical literature, but formal sensory evaluation by trained panels is lacking. To provide this information, a trained descriptive panel evaluated the sensory characteristics of 10 divalent salts including ferrous sulfate, chloride and gluconate; calcium chloride, lactate and glycerophosphate; zinc sulfate and chloride; and magnesium sulfate and chloride. Among the compounds tested, iron compounds were highest in metallic taste; zinc compounds had higher astringency and a glutamate-like sensation; and bitterness was pronounced for magnesium and calcium salts. Bitterness was affected by the anion in ferrous and calcium salts. Results from the trained panelists were largely consistent with the psychophysical literature using untrained judges, but provided a more comprehensive set of oral sensory attributes.

[Efficacy and tolerance of a calcium-magnesium-aspartate solution in the treatment of hypocalcemic parturient paresis in cows].

The systemic tolerance of a solution of calcium aspartate and magnesium aspartate was studied in 7 cows. Intravenously administered dosages of 500 ml per cow were well tolerated. A twofold increase of the serum calcium concentration was measured. In 2 cows which were treated with 1000 ml of the solution a threefold increased calcium concentration and heart arrhythmia were found. The clinical efficacy of the solution was demonstrated in a study with 44 hypocalcemic cows. A long lasting increase of the serum calcium as well as an enhanced phosphorus concentration were measurable. In conclusion, the calcium-magnesium-aspartate solution seems to be an efficacious and well tolerated alternative for the treatment of hypocalcemia in cows.

Sensitivity of FRDA lymphoblasts to salts of transition metal ions.

Friedreich's ataxia (FRDA) is an autosomal recessive neurodegenerative disease resulting from decreased expression of the nuclear-encoded mitochondrial protein, frataxin. FRDA patients have characteristic iron deposits and dysfunction of mitochondrial enzymes in the heart. Inactivation of the frataxin homologue in yeast causes dysregulation of both mitochondrial iron levels and iron export. Previously, we have observed sensitivity of FRDA fibroblasts to FeCl3 and hydrogen peroxide, results consistent with the hypothesis that FRDA cells may experience increased Fenton chemistry. To determine whether the sensitivity of FRDA cells to transition metal ions is a general or specific property, we have compared the sensitivity of lymphoblasts from FRDA patients and healthy controls to the transition metal salts CoCl2, CuSO4 FeCl3 FeSO4, MnCl2, and ZnCl2. FRDA lymphoblasts were significantly more sensitive to FeCl3 and MnCl2 than control cells. However, there were no significant differences observed in sensitivity to CoCl2, CuSO4, FeSO4 and ZnCl2 in the concentration ranges studied. Thus, the sensitivity of FRDA lymphoblasts exposed to transition metals appears to be specific, and could be relevant to the pathophysiological mechanism, which is discussed.

Formation of physically stable amorphous drugs by milling with Neusilin.

Each of four drugs (ketoprofen, indomethacin, naproxen, and progesterone) was milled with Neusilin (amorphous magnesium aluminosilicate) to effect conversion from crystalline to amorphous states, and the physical stability of the resultant drugs was investigated. Ball milling the drugs alone for 48 h resulted in no amorphization. X-ray powder diffractometry (XPD), birefringence, and differential scanning calorimetry (DSC) data indicated amorphization of all the four drugs on ball milling with Neusilin. Fourier transform infrared spectroscopy (FTIR) data showed a reduction in the absorbance of the free and the hydrogen-bonded acid carbonyl peaks accompanied by a corresponding increase in the absorbance of the carboxylate peak, indicating an acid-base reaction between the carboxylic acid-containing drugs and Neusilin on milling. On storage of milled powders (at 40 degrees C and 75% RH for 4 weeks), XPD, birefringence, and DSC data showed the absence of reversion to the crystalline state, and FTIR data revealed continued absence of the carbonyl peaks. Whereas the carboxylic acid-containing drugs convert from their crystalline acid form to amorphous salt form on milling with Neusilin, progesterone seems to interact with Neusilin via hydrogen bonding. The amorphous Neusilin-bound states of all four drugs were physically stable during storage. The water of adsorption seems to mediate the conversion of the crystalline state to an amorphous Neusilin-bound state.

Synthesis, characterization and DNA binding of magnesium-ciprofloxacin (cfH) complex [Mg(cf)2] * 2.5H2O.

Interactions of the tested systems (title compound [Mg(cf)(2)] * 2.5H(2)O (1), ciprofloxacin (cfH) and ciprofloxacin in the mixture with MgCl(2)), with single and double stranded calf thymus DNA, poly[d(AT)] * poly[d(AT)] and poly[d(GC)] * poly[d(GC)] were studied by UV-spectrophotometric (melting curves) and fluorescence emission measurements. Pronounced quenching of ciprofloxacin's fluorescence intensity has been observed for all the tested compounds after titration with various GC containing DNA molecules. It seems probable that quenching originates in the electron transfer from guanine to the photo-excited fluoroquinolone. The UV-spectrophotometric results obtained for 1 are substantially different from the other solutions and the biggest differences were observed for GC containing DNAs. Solution of 1 provokes a large thermal destabilization of poly[d(GC)] * poly[d(GC)]. This process is irreversible which suggests that the species present in solution of 1 alone inhibit re-annealing by associating irreversibly with the single strands. We have realized that aqueous solutions of 1 are colloidal and we propose that colloidal particles are involved in specific binding to GC containing sequences, most probably in the major groove of DNA.

[Solubility and absorption of magnesium fluoride in the intestinal tract].

The solubility rate of various substances was determined via solubility experiments using magnesium fluoride on various specific surfaces. This rate controls the resorption of fluorine ions from MgF2 during its passage through the intestinal tract. The solubility rate for MgF2, as for other hard-to-dissolve substances, is proportional to the specific surface and the mass of the substance. In light of this information, resorption tests with magnesium fluoride showed that this compound is "biodisposable". The amount of fluorine which will be resorbed from MgF2 can be estimated.

The magnesium-ene cyclization stereochemically directed by an allylic oxyanionic group and its application to a highly stereoselective synthesis of (+/-)-matatabiether. Allylmagnesium compounds by reductive magnesiation of allyl phenyl sulfides.

The first example of a magnesium-ene cyclization stereochemically directed by an allylic oxyanionic group is demonstrated by a highly stereoselective synthesis of the bicyclic terpene matatabiether 10. The synthetic method is particularly valuable, not only because of the stereochemical control and the utility of the versatile hydroxyl group introduced into the product, but also because the precursor of the allylmagnesium is an allyl phenyl sulfide, which is more stable and more easily prepared in a connective fashion than the usual allyl halide precursor. Since the presence of lithium ions encourages undesirable proton transfer to the cyclized organometallic and is detrimental to the stereochemical control, the conversion of the allylic thioether to the allylmagnesium utilizes a lithium-free method involving direct reductive magnesiation in the presence of the magnesium-anthracene complex.
