Osteogenesis in a rat model: use of bone marrow cells and biodegradable gelatin matrix carrier.

We investigated the use of a biodegradable porcine gelatin matrix (Gelfoam) as a carrier for marrow cells that induce osteogenesis at ectopic sites in rats. Bone marrow cells obtained from the long bones of 6-week-old Sprague-Dawley rats were dissociated enzymatically and the cells reconstituted in rat serum. Twenty million cells in 0.1 mL of serum were then adsorbed into 1-cm3 pieces of gelatin matrix substrate and implanted into ectopic sites in live Sprague-Dawley rats. The implants were retrieved and analyzed histologically for bone and cartilage formation 3, 4, 6, and 8 weeks after implantation. Woven bone was seen as early as 3 weeks and persisted through 8 weeks. No cartilage was observed. Osteoclasts first appeared at 3 weeks, peaking in number at 4 weeks. By 6 and 8 weeks, only small islets of substrate remained, surrounded by dense, woven bone. Control implants, consisting only of serum adsorbed into the carrier, showed no bone formation. We conclude that biodegradable gelatin matrix can serve as a carrier for the osteogenic cells of bone marrow in rats in ectopic sites. Such a system may be further developed to augment bone healing.

[Effect of shale kerogen oxidation products on biodegradation of petroleum and petroleum products in soil and water].

Effect of oxidation products of shale kerogen (high-molecular-weight acids with 6-22 carbon atoms) on biodegradation of oil and oil products in soil and water was studied. High-molecular-weight acids (HMWA) not only affected the layer of oil and/or oil products and dispersed it into small particles, but also stimulated growth of Rhodococcus erythropolis VKM AS-1339D, degraders of oil and oil products. Addition of 0.001-0.003% HMWA to a medium to be purified from oil products increased the extent of bacterial biodegradation by a factor of 1.1-5.0.

Studies on bioremediation of polycyclic aromatic hydrocarbon-contaminated sediments: bioavailability, biodegradability, and toxicity issues.

The widespread contamination by polycyclic aromatic hydrocarbons (PAHs) has created a need for cost-effective bioremediation processes. This research studied a chronically PAH-contaminated estuarine sediment from the East River (ER; NY, USA) characterized by high concentrations of PAHs (approximately 4-190 ppm), sulfide, and metals and a marine sediment from New York/ New Jersey Harbor (NY/NJH; USA) with only trace quantities of PAHs (0.1-0.6 ppm). The focus was to examine the relationship between bioavailability of PAHs and their biological removal in a slurry system. Freshwater and marine sediment toxicity tests were conducted to measure baseline toxicity of both sediments to amphipods, aquatic worms, fathead and sheepshead minnow larvae, and a vascular plant; to determine the cause of toxicity; and to evaluate the effectiveness of the biotreatment strategies in reducing toxicity. Results showed the ER sediment was acutely toxic to all freshwater and marine organisms tested and that the toxicity was mainly caused by sulfide, PAHs, and metals present in the sediment. In spite of the high toxicity, most of the PAH compounds showed significant degradation in the aerobic sediment/water slurry system if the initial high oxygen demand due to the high sulfide content of the sediment was overcome. The removal of PAHs by biodegradation was closely related to their desorbed amount in 90% isopropanol solution during 24 h of contact, while the desorption of model PAH compounds from freshly spiked NY/NJH sediment did not describe the bioavailability of PAHs in the East River sediment well. The research improves our understanding of bioavailability as a controlling factor in bioremediation of PAHs and the potential of aerobic biodegradation for PAH removal and ecotoxicity reduction.

Effect of dissolved organic matter of various origins and biodegradabilities on the bioaccumulation of polycyclic aromatic hydrocarbons in Daphnia magna.

As a preliminary study of the influence of urban organic matter on the bioavailability of polycyclic aromatic hydrocarbons (PAHs), the effect of different types of dissolved organic matter (DOM) on the bioaccumulation of fluoranthene, pyrene, or benzo[a]pyrene in Daphnia magna was studied. Commercial humic substances, DOM from the aeration basin of a wastewater treatment plant, and highly biodegradable DOM (algae or animal extracts) were tested. The bioaccumulation of benzo[a]pyrene was reduced by each DOM (up to 80% reduction with humic substances). Pyrene bioaccumulation was also decreased by each DOM to a lesser extent. Fluoranthene bioaccumulation was affected by the presence of humic acids only. In each experiment, the solution containing humic DOM led to the lowest bioaccumulation. Supposing that only dissolved PAHs were bioavailable, the reduction of bioaccumulation allowed a biological estimate of the partition coefficients of DOM and PAH, K(DOC). The estimated coefficients were positively related to the aromaticity of DOM and negatively related to its biodegradability.

[Study on biodegraded characteristics of endocrine disruptor di-n-butyl phthalate].

The biodegraded Characteristics of di-n-butyl Phthalate (DBP), endocrine disruptor were studied. The activated sludge was acclimated using DBP as the only carbon in the culture medium. DBP concentration was increased progressively from 0.05 g/L to 0.5 g/L. The degradation tests were carried out on the constant temperature table using the acclimated sludge at different PH, temperature, DBP concentration and hydraulic residence time. The results showed that DBP could be rapidly degraded-nearly 90% in 48 hours. The appropriate condition for DBP degradation was pH 6.0-8.0, temperature 25 degrees C-35 degrees C, DBP concentration no more than 300 mg/L, and hydraulic residence time 12 h-24 h. The form of biodegradation of DBP can be described as the first-order reaction model.

Comparison of di-n-methyl phthalate biodegradation by free and immobilized microbial cells.

OBJECTIVE: To compare the biodegradation of di-n-methyl pathalate by free and immobilized microbial cells. METHODS: The enrichment and isolation technique was used to isolate the microorganism. The PAV-entrapment method was utilized to immobilize the microorganisms. The scanning electron microscophy (SEM) was used to observe the growth and distribution of microbial cells immobilized inside the PVA bead gels. The GC/MS method was used to identify the main intermediates of DMP degradation. RESULTS: The microbial cells could grow quite well in PVA gel. The metabolic pathway did not change before and after immobilization of the microbial cells. The degradation rate of immobilized cells was higher than that of free cells. CONCLUSION: The immobilized microbial cells possess advantages than free cells when applied to the biodegradation of toxic organic pollutants.

[Research advances in biodegradation of chlorophenols in environment].

This paper examines the biodegradation of chlorophenols by microbes and deals with the chlorophenols-degrading microbes and the usage of biotechnology with special emphasis on degradation mechanisms. Dechlorination is the first critical step in the bacterial degradation of many chloroniated pollutans. Under aerobic condition, the degradation of mono- and dichlorophenols is shown to be initiated by oxygenation into chlorocatechols, and dechlorination occurs only after ring cleavage of the chlorocatechols. The degradation of polychlorinated phenols starts by hydrolytic para-hydroxylation, yielding chlorinated para-hydroquinone. The anaerobic biodegradation of chlorophenols occurs by reductive dechlorination, a process by which chlorines are replaced with hydrogen.

Evaluation of biodegradable microspheres as vaccine adjuvant for hepatitis B surface antigen.

Biodegradable microspheres were evaluated as vaccine adjuvants based on their ability to provide prolonged release of incorporated agents. Hepatitis B surface antigen (HBSA) prepared by recombinant DNA technology was chosen as a model antigen and encapsulated into polyglycolic acid (PGA) by solvent extraction and solvent evaporation techniques. Five microsphere formulations were prepared to evaluate effect of microsphere size and the presence of immunostimulants such as muramyl dipeptide (MDP) or aluminum hydroxide. The microspheres were characterized for size distribution, surface morphology and antigenicity. Guinea pigs were chosen as the animal model for evaluation of antigenicity of the formulations. The animals were divided into seven groups of four animals each and the microsphere formulations were injected intraperitoneally, using alum adsorbed HBSA as positive control and placebo microspheres as negative control. Blood samples were withdrawn from the animals by toe clipping at two, four, six and sixteen weeks and plasma was analyzed for antibodies against hepatitis B by an enzyme linked immunoassay. At sixteen weeks, the animals were reinjected and evaluated for antibody response at two, four and six weeks post second injection. Antibody response to the microspheres was higher than control. Smaller size microspheres elicited earlier antibody response while the larger size microspheres provided delayed and longer duration of antibody production. Microspheres with MDP potentiated the antibody response. The results demonstrate the applicability of biodegradable microspheres for immunization against hepatitis B.

Experimental investigation and modelling of the effect of sulfate on anaerobic biodegradation processes in municipal solid waste.

The experimental investigations have been carried out in two parts. First, the biodegradation of the organic compounds in the municipal solid waste has been investigated with focus on different sulfate concentrations influencing the methane formation. Second, the inhibition effect of hydrogen sulfide in solution (H2S(aq)) on the acetate, and methane formation, respectively, has been studied at different pH and temperature values. In solution the equilibrium of hydrogen sulfide (H2S(aq)) and the hydrogen sulfide anion (HS-(aq)) mainly depends on pH. At pH 6.3 the favoured species is H2S(aq) whereas at pH 7.7 the HS-(aq) species is favoured. Additionally, the experiments have been carried out at two different temperatures (35 degrees C and 55 degrees C). According to the acetate and methane formation the H2S(aq) species is observed to have an inhibitory effect. All simulations based on a biodegradation model represented are in good agreement with the experimental data obtained.

Simultaneous biodegradation of a phenol and 3,4-dimethylphenol mixture under denitrifying conditions.

Denitrification is a feasible alternative for the treatment of phenolic bearing-wastewaters. The aim of this study was to evaluate the biodegradability of phenolic compounds, as the only carbon and energy source in batch and continuous experiments, using nitrate as a final electron acceptor. Experiments in a continuous upward anaerobic sludge bed reactor demonstrated the possibility of biodegrading a mixture of phenol and 3,4-dimethylphenol at organic loads of 251.6 and 39.5 mg/L-d, respectively, at a COD/NO3(-)-N ratio of 2.57. A nitrogen production efficiency of 86% was obtained according to the nitrate consumption. GC-MS analyses demonstrated that m-cresol was an intermediate of 3,4-dimethylphenol degradation in batch conditions, and had an inhibitory effect on phenol degradation.

[Effect of the biodegradable chitosan external stent on the early changes in the rabbit vein grafts].

OBJECTIVES: To investigate the effect of chitosan biodegradable external stent (CES) on the early changes of rabbit vein graft (VG). METHODS: Rabbit vein grafting models were divided into S group (with perivenous CES) and NS group (without perivenous CES). The VG were harvested in 1 week, 2 weeks, 4 weeks after operation, respectively. The expression of proliferating cell nuclear antigen (PCNA) was used for evaluating the proliferation of the smooth muscle cell (SMC). The thickness, area of neointima and media of the VG were calculated by computer imaging analysis system. RESULTS: CES began to degrade in 2 weeks after operation. The thickness, area of both neointima and media of the VG in S group, increased mildly in 1 week after operation, and kept steady in 1 or 2 weeks after grafting, which was significantly less than NS group (both P < 0.01), then increased mildly in 4 weeks after grafting but still less than NS group (P < 0.05). The expression of PCNA of SMC decreased significantly in comparison with NS group though increasing mildly in four weeks after operation. Both neointimal formation and cell proliferation in the graft wall were significantly reduced by external stenting as compared to the results with unstented grafts. CONCLUSIONS: CES may reduce early intimal and medial hyperplasia, and may be beneficial in improving the long term patency of the VG. The biodegradable characteristics of the CES may influence its effect.

Effect of oxygen on the stability and inducibility of the biodegradative capability of benzoate.

Anoxic zones in biological nitrogen removal systems are typically open to the atmosphere and receive oxygen from the atmosphere and the recirculation flow from the aerobic zone. This raises the question of how such oxygen input might influence the stability and inducibility of the enzyme systems involved in biodegradation of aromatic compounds. To investigate this, various amounts of oxygen were added to mixed culture denitrifying chemostats receiving benzoate at 667 mg/h as chemical oxygen demand (COD), and the stability and inducibility of the culture's benzoate biodegradative capability (BBC) were tested in aerobic and anoxic fed-batch reactors (FBRs). Cultures from chemostats receiving oxygen at 0, 33, 133, 266, and 466 mg O2/h lost almost all of their anoxic BBC within one hour after being transferred to an aerobic FBR and the first three cultures did not recover it upon being returned to anoxic conditions. The last two cultures recovered their anoxic BBC between 9 and 16 h during the 16 h aerobic exposure period that preceded their return to anoxic conditions and continued to increase their anoxic BBC as they were retained under anoxic conditions. In contrast, the culture from a chemostat receiving oxygen at 67 mg O2/h retained its anoxic BBC longer, recovered it within 3 h after its return to anoxic conditions, and increased it linearly thereafter. None of the cultures developed any aerobic BBC during the 16 h aerobic exposure period in FBRs. The results suggest that higher oxygen inputs into anoxic reactors helped the mixed microbial cultures recover and/or induced anoxic BBC more easily when they were exposed to alternating aerobic/anoxic environments. The exceptional behavior of the culture from the chemostat receiving oxygen at a rate of 67 mg O2/h may have been caused by the presence of a protective mechanism against the toxic forms of oxygen.

An off-line respirometric procedure to determine inhibition and toxicity of biodegradable compounds in biomass from an industrial WWTP.

Determining whether a certain compound is toxic (or inhibitor) or not to a biomass of a WWTP is crucial in order to avoid WWTP malfunction. Respirometric techniques have been commonly used to assess the toxicity of a certain compound by evaluating the oxygen uptake rate (OUR) profile obtained when a pulse of substrate is added to endogenous biomass and comparing it with the OUR profile obtained when the pulse is a mixture of substrate and the possible toxic or inhibitor (PTI). However, when using this method with biodegradable compounds some difficulties arise because the PTI consumption implies oxygen consumption as well. In this study, a modified procedure for toxicity assessment using respirometry is developed which overcomes the difficulties caused by the toxic biodegradability. This procedure is based on the comparison of different respirograms obtained with pulses of control substrate before and after adding the PTI and with a pulse of PTI and control substrate together. Moreover, some examples are shown with p-phenylenediamine and phenol as an example of toxic and inhibitor for nitrifying biomass.

Biodegradation of mixture of VOC's in a biofilter.

Volatile organic compounds(VOC' s) in air have become major concern in recent years. Biodegradation of a mixture of ethanol and methanol vapor was evaluated in a laboratory biofilter with a bed of compost and polystyrene particles using an acclimated mixed culture. The continuous performance of the biofilter was studied with different proportion of ethanol and methanol at different initial concentration and flow rates. The result showed significant removal for both ethanol and methanol, which were composition dependent. The presence of either compound in the mixture inhibited the biodegradation of the other.
