[Reproductibility of the morphological identification of schisocytes and evaluation of non observer-dependent methods].

Schistocytes are red blood cell fragments observed on a blood smear. They are a mark of mechanical haemolytic anaemias whose group of the thrombotic micro-angiopathies requires an urgent treatment. The detection of the schistocytes and sometimes their quantification are thus of primary importance. To evaluate this search for schistocytes, several surveys of practice were carried out (1999-2003) including pictures of blood fields (identification of schistocytes among abnormal red blood cells) near biologists of variable level of specialization. The aim was to try to lead to a consensus, in particular for the morphological criterias of identification. Our results indicated that: 1) the biologists are badly sensitized with the importance of this research and the consequences of their response for the diagnosis; 2) the morphological identification of the schistocytes is difficult with an important variability of the criteria according to the observers. An investigation overviewed by the French Group of Cellular Hematology (Delphi method) allowed the development of a morphological consensus (fragments of triangular/crescent/helmet forms with rectilinear zone testifying to the zone of break). In order to cancel the observer-dependent identification of the schistocytes, a software of morphometric analysis (Q-WIN, Leica) was developed for sorting, starting from digitalized microscopic fields, the fragmented - among the normal - red blood cells. The results appeared encouraging, but not yet optimized. An automated analyzer (Bayer ADVIA 120) was also evaluated for the measurement of the schizocytes ("fragmented red blood cells" parameter). The moderate over-estimation of the real schizocytes (+ 0,4%) encouraged to observe the clinical value of the fragmented red blood cells detection in a group of patients that undergone a bone-marrow transplantation. The predictive value of the test (98%) was satisfactory.

[Strategy for the protein identification of human proteome expression profile: selection of searching database].

Widely used method of protein identification for high-throughout proteome expression profile studies was database-dependent, so the selection of databases for the protein identification was very important. Despite the deficiency of available human protein databases, the complementarity of human proteins could be got mainly from human genome but not from the protein databases of other organisms. According to the comparison of the current protein databases from different aspects, IPI was recommended for the basic identification for the studies of human proteome expression profile, and other human protein or nucleic acid databases were needed for the complementary identification and novel protein mining.

From tissue phenotype to proteotype: sensitive protein identification in microdissected tumor tissue.

Correlation of disease phenotype with protein profile (proteotype) is a significant challenge for biomedical research. The main obstacles have been the need to insure sufficient quantities of pure protein sample, the reproducibility of protein display, and rapid and accurate protein identification. We present a modified approach that combines enhanced detection sensitivity with tissue microdissection from frozen primary renal cancer tissues of different histological subtypes, followed by 2D gel analysis and protein identification with MALDI mass spectrometry. We obtained reliable and highly consistent results in phenotypically similar tumors of each individual subtype by performing strict morphological control of the analyzed tumor cells without physical or chemical alteration of the frozen tissue samples. By application of non-oxidizing silver staining, proteins were resolved and identified with high levels of specificity and sensitivity. This new combination of techniques allows not only for sensitive identification of specific protein patterns that correspond to a histological tumor phenotype, but also for identification of specific disease-associated protein targets.

[Molecular identification of species within the Mycobacterium tuberculosis complex using regions of difference].

The differentiation within the Mycobacterium tuberculosis complex (MTBC) based on phenotypic methods is long and does not give an unambiguous result in every case whereas the advance in genetic knowledge leads to new views. Thus, regions of difference (RD), that seem to characterize the species of the MTBC, have been identified. Amplification methods, targeted on these zones, have been developed then. The study of four regions (RD1, RD5, RD9, RD10) has been done on 64 isolates formerly identified thanks to phenotypic methods. Genotypic results confirm phenotypic identifications except in one case. This strain initially identified as M. tuberculosis and isolated from a Gabonese patient, corresponds, according to genotypic identification, to M. africanum. Since phenotypic characterization of M. africanum is difficult, this method would allow to accurately determine the true prevalence of this specie. Moreover, the study of RD10 doesn't seem to be informative. The amplification of only two RD, RD1 and RD9 carries out the identification of all M. tuberculosis and M. bovis BCG isolates. M. bovis and M. africanum will be then identified thanks to RD5. Thus, this easy and rapid method of identification of the major species of MTBC seems to be appropriated for a routine use.

The killing field of Khao Lak: forensic odontology in Thailand tsunami victim identification.

Forensic odontology is the science of dental identification. This paper describes the contribution of forensic odontology to tsunami victim identification in Thailand, with particular reference to the Singaporean victims. Thirteen Singaporeans were reported missing in Phuket following the Indian ocean tsunami on 26 December 2004. To date, 10 victims have been found and identified, eight of whom were identified by dental records. The author travelled twice to southern Thailand and spent 5 weeks there. First, in December 2004 as part of a Singapore Police Force Disaster Victim Identification team deployed in Khao Lak, and later in July 2005 at the Thai Tsunami Victim Identification Information Management Centre in Phuket.

[Study on sex-identification of ruminant using SRY gene and microsatellite markers].

UNLABELLED: Multiplex PCR amplification of four microsatellite markers on chromosomes X and Y and the SRY gene was used to identify the sex and genotype the ruminant simultaneously in one step. Result indicated that the d SRY primers was highly specific and could be used as the main basis for sex-identification. But primer specificity was not good for Y chromosome markers MCM158 and MAF45, and therefore these markers were not suited for sex identification. The primers of two X chromosome markers MILVET09 and AE25 are amplified, but the SRY gene is not, then the individual being examined is female. Furthermore the genotype of MILVET09 and AE25 dose not affect sex determination of the female. The genotype of markers MCM158 and MAF45 dose not affect the result of sex-identification, only were able to further confirm the male identity. IN CONCLUSION: when SRY, MCM158 and MAF45 on chromosome Y and MILVET09 and AE25 on chrosomome X can all be amplified simultaneously, and when the genotype of MILVET09 and AE25 is homozygous, the animal is determined to be normal male; When only MCM158, MAF45 and MILVET09 and AE25 are amplified, but the SRY gene is not, then the individual being examined is female. Furthermore the genotype of MILVET09 and AE25 does not affect sex determination of the female. The genotype of markers MCM158 and MAF45 dose not affect the result of sex-identification.

Computer-aided identification of renal corpuscle elements in RGB and HLS color images.

UNLABELLED: The qualitative and quantitative changes in the structure of the renal corpuscle allow the assessment of the glomerular involvement degree, defining each type of glomerulonephritis. Recently, several works dedicated to glomerulonephritis focus on the quantitative investigation, achieved by digital (numerical) image processing, where the identification of the interest elements is a key stage. OBJECTIVES: The aim of the study was the elaboration of a technique for constructing reliable color models in the RGB and HLS spaces for each element of the structure of the renal corpuscle. MATERIAL AND METHODS: We analyzed images captured from trichrome stained fragments of renal biopsy. Each image contained a single renal corpuscle. The identification procedure used the software tools specialized in color image analysis provided by the Zeiss KS400 environment. RESULTS: There were defined specific chromatic domains in the RGB and HLS space for the identification of the following elements: the Bowman space and the glomerular capillaries (white colored regions), the nuclei (brown colored regions), the mesangium (green colored regions). DISCUSSIONS: In order to get a relevant picture of the role played by the decisions made in the definition of the chromatic domains, we performed two tests for the modification of the chromatic domain allocated to the mesangium in the RGB and HLS space, respectively. CONCLUSION: The procedures proposed for the identification of the RGB and HLS color models can successfully replace the manual delineation of the renal corpuscle elements. Such models represent an important step in our research visualizing the future development of computer programs for automated measurements of renal corpuscle elements.

[The application of several types of large-scale exactitude equipments in the identification of jadeite].

Infrared spectrometer, Raman spectrometer, EPMA, and X-ray powder diffractometer are very effective equipments for the identification of jadeite. Some kinds of raw jadeite fakes occurred in the identification and jewelry market recently. With the methods of infrared spectrometer, Raman spectrophotometer, EPMA, and X-ray powder diffractometer etc., these typical fakes have been systematically studied. These fakes have superficial resemblances such as texture, outer characters and color of minerals in rocks. The relevant information of these fakes has been obtained. Meanwhile, several kinds of rough materials with fake weathering crust and cutting groove were analyzed. These identification methods were compared with each other. Type-B can be tested by infrared spectrometer, Raman spectrophotometer, and X-ray powder diffractometer. The present paper means a lot for the identification, evaluation and business of jadeite.

Verification of single-peptide protein identifications by the application of complementary database search algorithms.

Data produced from the MudPIT analysis of yeast (S. cerevisiae) and rice (O. sativa) were used to develop a technique to validate single-peptide protein identifications using complementary database search algorithms. This results in a considerable reduction of overall false-positive rates for protein identifications; the overall false discovery rates in yeast are reduced from near 25% to less than 1%, and the false discovery rate of yeast single-peptide protein identifications becomes negligible. This technique can be employed by laboratories utilizing a SEQUEST-based proteomic analysis platform, incorporating the XTandem algorithm as a complementary tool for verification of single-peptide protein identifications. We have achieved this using open-source software, including several data-manipulation software tools developed in our laboratory, which are freely available to download.

Classifications used by Australian forensic odontologists in identification reports.

Forensic odontologists are repeatedly called upon to assist in the identification of deceased persons. A great deal of information is available in the literature as to how and why comparative dental investigation of identification is performed but there is little information on the descriptive terms used in reporting these identifications. A forensic odontology report sets out the findings of a comparison between antemortem and postmortem evidence and indicates the odontologist's opinion on the identification. This opinion needs to be defendable in a court of law. This paper investigates the classifications utilised in the six states and two territories of Australia and reflects on the differences. Three states of Australia use American Board of Forensic Odontology classifications, whilst the remaining regions use a modified format. Since there are no significant legal, cultural or religious differences, and similar practitioners and clients, variation between regions within Australia would seem hard to justify. National standard terminology should be encouraged.

[Positive identification of dental porcelain in a case of murder].

Dental prostheses can provide important evidence for use in personal identification. In recent years, the variety of materials used in prostheses has increased with the diversification of dental treatment. The detailed analysis of these materials can provide significant information to assist in personal identification. This paper reports a case when identification was achieved by the comparative analysis of fragment of fused dental porcelain. In April, 1990, a woman aged 24 years was found murdered in her apartment in Hamamatsu City, Shizuoka Prefecture. Near her body a small mass of fused porcelain was recovered. Subsequently, a 48 year old male suspect was arrested. An examination of his teeth revealed a small fragment of dental porcelain adhering to an abutment tooth. This was removed for testing. The suspect denied the charge of murder. A comparative analysis of the porcelain found in the apartment and the porcelain that was removed from the suspect's abutment tooth, was carried out by scanning electron microscopy (SEM) and electron probe X-ray microanalysis (EPMA). The SEM examination demonstrated remarkable similarity in respect of the structure of the porcelain and the nature and distribution of the air bubbles produced during the baking process in both samples. The EPMA showed the elemental composition of each sample to be identical. This case demonstrated that detailed analysis of dental materials may affords a useful aid for personal identification.

Positive identification of victims by comparison of ante-mortem and post-mortem dental radiographs.

Radiography of jaws and teeth can provide one of the most reliable sources of information for comparison between ante-mortem and post-mortem conditions leading to definitive evidence in cases of identification. Teeth and dental restorations are resistant to destruction by fire and are therefore very important in identification. The forensic odontologist utilizes dental radiographs taken of the victim before death and compares them to dental data from the remains to assist the identification. Three cases described in this paper illustrate the procedures and techniques of identification using ante-mortem and post-mortem radiographs.

Identification of peripheral visual images in a laterally restricted gaze field.

When the peripheral visual field is restricted or distorted, as occurs with certain spectacle lenses, the identification of objects in the periphery requires a coordinated head and eye movement. Initial experiments on the identification of peripheral images under such restrictions show that the degradation in performance is defined by a consistent additional delay in the time required to identify the image correctly. An analysis of the motor movements shows that performance is solely determined by movements of the head; eye movements are sufficiently precise and fast so they do not limit performance. A quantitative model of the identification task was developed and model simulations confirmed the experimental findings that head movement variables, specifically response latency and movement duration, uniquely determine identification performance. Hence, improved performance under these conditions must come from modifications in head-movement control either through training or adaptive processes.

[Evaluation of Sen-SIR, a new microsystem for the identification and determination of bacterial sensitivity].

The use of semiautomatic methods for identification and study of bacterial sensitivity to antimicrobials is increasingly widespread in clinical Microbiology. In the present study, the evaluation of a new micromethod (Sen-SIR, BBL) for the identification and determination of bacterial sensitivity is reported. 115 clinical isolates of enterobacteria, 10 strains of Acinetobacter calcoaceticus and 57 gram positive cocci, previously identified as belonging to the geni Staphylococcus and Streptococcus were evaluated. In gram negative organisms, the concordance with classical identification methods was 98.4%. In gram positive organisms, the concordance was lower only for the differentiation between group D streptococci and enterococci. The observed sensitivities were similar to those seen with the disk method, except basically for Morganella morganii and Proteus vulgaris. This method is quick, simple and reliable; thus, it can represent an alternative to classical identification methods.

[100 years of identification (from man to his genome)].

It is a set of descriptive features or traits characterising an individual that serves as the basic for the identification of his person. Some of these traits are of interest mainly in police investigations; others, of which the interest is more particularly scientific, are rapidly gaining recognition. Anthropometric methods enable the identification of recidivist criminals. Anthropometric description is based on bone measurements taken in conjunction with the spoken portrait and recorded distinguishing marks--the features concretely represented in the descriptive photograph. This method was first supplemented and then supplanted, both for description and identification, by dactyloscopy. Investigations involving corpses are often extremely difficult. Several facts concerning bone and/or teeth could be relevant in determining the race, sex, height and age of the person whose remains are examined. Traces and stains as "clues" of hereditary and congenital properties of blood (erythrocytic, enzymatic, plasmic and leucocytic systems, factors and groups) and of sperm (with, in this case, secretor and non secretor properties) do provide and increasingly precise description of an individual but only a certain degree of probability for his identification; at best, they can only exclude an individual, but cannot prove his involvement. A knowledge of the patterns of the nucleotides in the Deoxyribo Nucleic Acids, which are the same in all the cells of an individual but differ from individual to individual, provides an ultimate means of arriving at certainty in the comparison: samples-suspect.(ABSTRACT TRUNCATED AT 250 WORDS)

[A study on tooth marks--morphological differences between dentitions and tooth marks thereof and selection of measurement items effective in identification].

Tooth marks in human skin assume so various aspects depending upon such factors as the forms and properties of the bitten regions that they do not necessarily represent the dentition of a suspect as it is. In the present study a cylindrical dummy with skin-like elasticity was used as a recording medium on which to make experimental tooth marks. Morphological comparisons were made of the tooth marks with the oral models by the use of measurements of 28 items. Furthermore, the selection of the effective items for identification was also considered by applying the discriminant analysis to the rate of morphological change on the base of the measurements. The results obtained were as follows: 1) In respect of the tooth marks and the oral models derived from the same subject, the values of their difference (i.e., the measured value of the former minus that of the latter) were calculated for each item so that the directional changes of the tooth marks should be investigated. It was found that the values for the lengths of the dental arch showed a tendency to be positive and that those for the widths negative, and that the values for the angles of lateral incisors and canines tended to be positive. 2) Among the mean values of the rates of morphological change for each item on the same subject, those for the lengths of the dental arch were larger, whereas those for the widths were smaller. Regarding the widths of tooth edge, large were those values for the mandibular canines in particular. 3) The identifying indices were calculated from the rate of morphological change on the basis of the measured values for 28 items in 400 combinations of the oral models with the tooth marks. They indicated that, out of the combinations derived from different subjects, 11.3% for maxillary overlapped those from the same subject and so did 8.7% for mandible in the distribution of their indices. 4) A discriminant analysis was carried out for identification. The range of coefficients thereby obtained rendered it possible to consider how much each item contributed to identification. As a result, the range of the inter-canine distance was largest. It could be considered that these items are effective in identification for tooth marks by using measurements.

Hearing disabled children--epidemiology and identification.

An investigation of hearing disabled children was performed in the Copenhagen City and County in order to: evaluate the prevalence and incidence of congenital/early acquired hearing disability and delineate any change in these parameters compared to the 1970'ies; evaluate the time of identification of the hearing impaired child; indicate who first suspects the child's hearing disability. It was demonstrated that the prevalence of hearing aid treated children had been reduced with 40-42%, however, the incidence was unchanged 0.13%. An annual variance of 0.06-0.20% was present. No significant difference in the incidence was found between the two geografical areas. The median age at identification was 15-18 months, and only 32-48% of the hearing disabled children had been identified at the age of 1 year, showing an improvement in comparison to the 15-16% identified at the same age in the 1970'ies. Children, who could be registered as auditory at-risk were not identified earlier than the non-at-risk children. In 57-67% of the children the parents were the first to suspect the hearing disability. On the basis of the investigation it is concluded that the incidence of congenital/early acquired hearing disability in children is unchanged over a 10 year period, and that a delayed identification of these children is present although an improvement in the early identification has taken place.
