Effect of ruminal ammonia-nitrogen concentration on protein degradation in situ.

Two experiments were conducted to determine the effect of ruminal ammonia-nitrogen concentration on rate of ruminal protein degradation. In Experiment 1, four Holstein steers were fed a basal diet of corn grain and corn silage at hourly intervals. Continuous intraruminal infusions of solutions containing sodium bicarbonate and either sodium chloride or ammonium chloride resulted in ruminal ammonia-nitrogen concentrations that averaged 4.8 and 17.3 mg/dl. Ruminal fluid pH, fluid volume, and turnover rate of fluid and molar percentage of acetate, propionate, and butyrate were similar across treatments, reflecting steady state conditions. Rates of nitrogen and dry matter disappearance from polyester bags containing soybean protein supplements with 10.2 or 50.1% soluble nitrogen were not affected by increase of ruminal ammonia-nitrogen concentrations from 4.8 to 17.3 mg/dl. In Experiment 2, Holstein steers were fed twice daily a basal diet of urea-supplemented corn grain and corn silage. Polyester bags containing soybean protein supplements were placed in the rumen at -4, 0, or 4 h with reference to feeding and incubated from 1 to 12 h. Peak ruminal ammonia-nitrogen concentrations occurred during different periods of incubation for each treatment. Ruminal ammonia-nitrogen concentrations ranged from 3 mg/dl at 6 h postfeeding to 46 mg/dl at 1 h postfeeding. Nitrogen and dry matter disappearance rates during 0 to 1 and 1 to 12 h of incubation did not differ among treatments.

Repair of spinal dural defects. An experimental study.

The search for an ideal substance for duraplasty has stimulated clinical and experimental investigations. To date a large number of materials have been employed for dural repair, although there is as yet no unanimity regarding the ideal material. Most of these studies have been concerned with cranial dura, and spinal duraplasty has received less attention. This study was designed to examine the repair of spinal dural defects in the dog. The materials chosen for this experiment were autologous fat, a polyester fiber mesh (Mersilene) and silicone-coated Dacron (Dura Film). Nineteen dogs were used in this study. Following lumbar laminectomy and the excision of elliptical pieces of dura (1.0 X 0.5 cm) at three noncontiguous levels, each of the defects was repaired using one of the three materials. Groups of animals were sacrificed at each of 3, 6, 12, and 24 weeks after dural repair. The lumbar region was removed en bloc and prepared for histological examination. Repair of the dural opening was achieved in all cases. The polyester fiber mesh was quite effective for dural repair, serving as a scaffold through which a neomembrane grew and united the dural edges. The results with autologous fat were similarly favorable. On the other hand, results with silicone-coated Dacron showed encapsulation by connective tissue, with the ventral aspect of the graft frequently compressing the underlying cord.

The contribution of forensic geology and other trace evidence analysis to the investigation of the killing of Italian Prime Minister Aldo Moro.

In May 1978 the body of the kidnapped Italian Prime Minister, murdered by the Red Brigades, was found in a car parked in the center of Rome. This paper discusses the findings from the investigations conducted on the evidence found on Mr. Moro's clothes, shoes (beach sand, bitumen, vegetals and polyester fragments), and on the car. To get a comprehensive picture of the characteristics of the various pieces of evidence, use was made of a multiple-technique approach. The sand was identified as coming from the seashore close to Rome. A tract of shore with a limited number of roads leading to the beach was defined as compatible with the textural and compositional characteristics of the sand. The study of the vegetal fragmenta suggested that they had been picked up in a period of time close to the killing. Thermosetting polyester, of the type used in boat manufacturing was found under the fenders, in the tires and inside the car, as well as under Mr. Moro's shoes, supporting proximity of a beach. Pollen analysis showed that adhesion of volcanic soil to the car fenders antedated adhesion of the sand.

Monofilament absorbable sutures in median sternotomy.

BACKGROUND: The most common material used for closure of median sternotomy incision is steel suture in open heart surgery. Some complications and disadvantages have been investigated recently. These complications are the breaking down of steel suture, erosion of sternum tabulae especially in osteoporotic patients, erosion of the dermis especially in patients with thin subdermic layer and cause of infection. Another disadventage of steel suture material is cosmetic problems or discomfort. For these reasons some suture materials such as silk, polyfilament polyester, monofilament material, polypropylene have been used recently. Silk and polyester have a risk of high infection, and polypropylene causes granulation tissue according to the number of knots. These facts encouraged the usage of an absorbable suture material. The available polyfilament absorbable sutures in the market a few years ago had a short absorption time, causing sternal infection and dehiscence. Polydiaxone, a monofilament suture material introduced recently has a considerably longer absorption time. METHODS: 153 sternal closures were performed with monofilament absorbable suture material in a period of seven months at the Koşuyolu Heart and Research Hospital. The mean age of the patients was 32.55, ranging from 8/12 to 71 years. The mean body weight is 48.37, ranging between 7 kg and 75 kg. RESULTS: Only two patients had sternal dehiscence. CONCLUSIONS: We conclude that monofilament absorbable suture is a safe alternative for all kinds of steel suture material for closure of sternotomy.

Material property determination of sub-surface objects in a viscoelastic environment.

Modeling human organs and soft tissue or anatomic regions for the purpose of medical training and simulation is a relatively new area. The data presented here is the groundwork for our ongoing development of a real-time haptic virtual environment for abdominal soft tissue palpation. The purpose of modeling the human abdomen is twofold. First, to provide a mathematical description of soft tissue and organs and second, to simulate the behavior of realistic interactions in real-time within a virtual environment. We have developed a, non-invasive, system that will allow us to determine mathematical functions that model the deformation of individual layers of soft tissue within the human abdomen. This system has been tested, experimentally, with a viscoelastic polyester foam model. We have been able to determine the stiffness and force/displacement function of an object beneath two layers of foam having different material characteristics. These experimental results correlate well with known polyester foam material characteristics. In general, the calculated stiffness constants were within 5% of the actual value. The data presented in this study shows that this system may be a viable tool for accurate measurement of human soft tissue properties and behavioral response to palpation.

[In vitro experimentation of a new model of radial flow bioreactor containing isolated hepatocytes].

Hepatocyte based artificial liver support systems are under investigation to support acute liver failure patients. The main purpose of such systems is to serve as a bridge to liver transplantation, or to promote spontaneous liver recovery. Limitation in mass-transfer capacity makes hollow-fiber bioreactors unsuited for long-term functioning of hybrid devices. We developed a novel radial-flow bioreactor in which the fluid perfuses the module from the center to the periphery, after having diffused through a space occupied by a three-dimensional structure filled with the hepatocytes. Five grams of freshly isolated porcine hepatocytes were seeded into uncoated, woven-non woven, hydrophilic polyester fabric, overlaid by two polyethersulfone membranes. Liver cells were perfused with 37 degrees C-warm, oxygenated, serum-free tissue culture medium, in which NH4Cl and Lidocaine were added at the final concentration of 1 mM and 60 micrograms/ml, respectively. Ammonium chloride removal, urea synthesis, monoethylglycinexylide (MEGX), pO2, pCO2, and pH were measured throughout the 14 day duration of the study. In a separate set of experiments, a scaled-up version of the radial flow bioreactor containing 150 grams of cells was perfused for 7 h with recirculating human plasma and MEGX production was monitored. During the 2 weeks of the study, an increasing production of urea was paralleled by constant ammonium removal. MEGX concentration after Lidocaine addition increased throughout the 14 days of perfusion with tissue culture medium, as well as after 7 hour perfusion with human plasma. Under transmission and scanning electron microscopy cells appeared attached to the polyester and one to each other, displaying ultrastructural features typical of functioning hepatocytes. Our study showed that liver cells were metabolically active when perfused into the radial-flow bioreactor. This configuration allowed close contact between media, or plasma, and cells at a physiological flow rate, by equalizing the concentration of the perfusing components, including O2, throughout the module. Our results suggest a potential use of this system for temporary extracorporeal liver support in acute hepatic failure patients.

[Endovascular treatment of aorto-iliac aneurysms. Made-to-measure endoprotheses increase the feasibility].

The authors describe their experience of tailoring endoprostheses for endovascular treatment of aorto-iliac aneurysms with components available on the market. Between January 1996 and December 1999, 188 aorto-iliac aneurysms were treated by tailor-made endoprostheses using self-expanding Z stents made of stainless steel compiled with polyester ligatures and covered with standard commercially available polyester prostheses. These endoprostheses were implanted with an 18 to 24 Fr (usually 20 Fr) introducer and positioned by a surgical approach. This method allows construction of tubular, bifurcated, digressive or occlusive endoprostheses associated with an extra-anatomical bypass graft. It increased the number of endovascular procedures for aorto-iliac aneurysms in the authors' department. This number has been further increased by using endoprostheses with an uncovered proximal or distal stent for cases with particularly short or angled necks and by using hybrid endoprostheses with one or more extremities without a stent, allowing surgical suture of the anastomosis. The authors' results show that tailoring endoprostheses considerably increased the feasibility of endovascular treatment of aorto-iliac aneurysms, even in unselected patients whilst providing an effectiveness and safety to justify the continuation of this experience.

[The content of double bonds in blood serum lipids from patients with myocardial infarction].

Hypocholesterolemia and hypotriglyceridemia during the first days of myocardial infarction (MI) is a result of enhanced absorption of saturated fatty acids in the form of triglycerides within very low density lipoproteins (VLDL) through apoE/B-100 receptors. This is followed by inhibition of this endocytosis, accumulation of saturated fatty acids VLDL in the blood, and hypertriglyceridemia. Permanent level of double bonds in MI (ozone titration) results not from MI but from blocking of cell absorption of polyenic fatty acids in the form of cholesterol polyesters in low-density lipoproteins (LDL) through apoB-100 receptors, which is characteristic of atherosclerosis. High molar ratio of double bonds/cholesterol in comparison with the double bonds/glycerol ratio indicates that the majority of polyenic fatty acids in the blood lipoproteins present as cholesterol polyesters. Lipoprotein transfer and cell absorption of saturated fatty acids alone are selectively impaired in MI. Selective impairment of cell absorption of saturated or polyenic fatty acids alone or a combination of both is observed in some diseases. Ozone titration of serum lipid double bonds reflects impaired absorption of essential polyenic fatty acids by cells.

[Surgical treatment of great vessel injuries of thoracic outlet].

OBJECTIVE: To summarize the experience in surgical treatment of great vessel injury of the thoracic outlet. METHODS: Thirty-two cases (24 men and 8 women aged 2-48 years) treated by surgery were collected during the period of 1970-1998. Sharp injury, blunt injury or decelerated injury was the causes. The vessels of injury included the common carotid artery, subclavian artery and accompanying veins. Emergency suture or repair of the artery with polyester patch was performed in 15 patients, aneurysmectomy in 6, resection of vessels with end-to-end anastomosis in 5, Gore-Tex vessel reconstruction in 2, and transplantation of the great saphenous vein in 3. RESULTS: Twenty-four patients (75%) were cared and 7 (21.8%) died. CONCLUSIONS: In the early period, patients with great vessel injury of the thoracic outlet died from acute bleeding, cerebral ischemia or combined multiple injuries, in the later period, however, false aneurysm develop. The operation is appropriate to the recondition of damaged cut, the effect of evolution in false aneurysm is approving. For large damaged cut or for bigger vessel injuries, the repair with polyester patch, Gore-Tex of blood vessel reconstruction or the transplantation of the great saphenous vein must be used.

Production of 1,3-propanediol by Klebsiella pneumoniae.

1,3-Propanediol (1,3-PD) has numerous applications from polymers to cosmetics, foods, lubricants, and medicines. Recently, there are strong industrial interests in a new kind of polyester, polytrimethylene terephthalate, with 1,3-PD as a monomer. This new polyester shows significant promise for use in carpeting and textiles. In this article we introduce a mild aerobic fermentation process using a strain screened from Klebsiella pneumoniae ATCC 25955, which is insensitive to oxygen, to produce 1,3-PD. We also describe a two-step fermentation process starting with glucose that was converted into glycerol with a glycerol-producing yeast, followed by K. pneumoniae that converts glycerol into 1,3-PD without intermediate isolation and purification of glycerol.

[Tailored endoprostheses for the endovascular treatment of aorto-iliac aneurysms: current features and perspectives].

We describe here our preliminary experience with the almost exclusive use of a range of made to measure stent-grafts home-made from commercially available components. From January 1996 to December 1999, 188 aortoiliac aneurysms (AIA) were treated with stent-grafts that were home-made to measure using Z autoexpandable stainless steel stents connected with polyester sutures and covered with commercially available polyester vascular prostheses. These stent-grafts were implanted through 18 to 24 (typically 20) Fr. commercially available introducers via a surgical remote access. Made to measure tubular, bifurcated, tapered, and/or blind stents combined with extra-anatomic bypass designs increased the rate of endovascular treatment (ET) of AIA in this series. This rate was further increased through the use of uncovered proximal or distal stents when dealing with short or tortuous necks near major side branches and through use of hybrid, partly surgical designs, one with stented and the other with stentless ends, the latter allowing for a surgically made anastomosis. The results of our experience with these techniques show that use of home-made to measure stent-grafts greatly increases the feasibility of the ET of AIA among unselected patients while offering enough efficiency and safety to deserve further investigation. Future perspectives are discussed.

Adsorption of metals to membrane filters in view of their speciation in nutrient solution.

Filtration of a solution may lower metal concentrations through adsorption of metal species to the filter. Processes such as filter-sterilizing nutrient solution and filtration of field water are sensitive to these sorption artifacts, yet basic data on the affinity of different filters for metals are lacking. This article describes the adsorption of five metals to eight types of 0.2-microm membrane filters used for sterilizing a plant (Lemna minor L.) culture medium. Filters of cellulose acetate, cellulose nitrate, mixed cellulose ester, nylon, polyamide, polycarbonate, polyester, and polyvinylidene fluoride were tested for their affinity toward mono- (K), di-(Mn, Cu, Zn), and trivalent (lanthanum [La]) metals. Metal concentrations were quantified using radioisotopes and speciation was calculated. Results showed that metals had the lowest affinity for polycarbonate and nylon filters and the highest affinity for cellulose- and polyester-type filters. Furthermore, it was shown that the metal load on cellulose filters correlated best with free ion concentrations (indicating electrostatic attraction), while loads on other filters correlated better with total metal concentrations. Filtering a 5-ml solution of pH 5 did not affect its metal concentrations, ranging from 10 nM (La) to 49 microM (K). To minimize filtration artifacts, we propose using polycarbonate or nylon filters, especially when dealing with low volumes of high pH and low metal species concentrations.

[Combined operation for left ventricular aneurysm and ventricular septal perforation with normal coronary arteries; report of a case].

We experienced a rare case of left ventricular aneurysm (LVA) and ventricular septal perforation (VSP) with normal coronary arteries, and had a good outcome by undergoing Dor procedure and direct closure of VSP individually. A 68-year-old female was printed out cardiac systolic murmur. Echocardiogram revealed LVA and VSP, but coronary arteriogram showed no significant stenosis. The apex was incised longitudinally, lateral and parallel to the interventricular groove. VSP was directly closed by 2 pieces of 3-0 polypropylene suture with felt strip. Purse-string stitch with 3-0 polyester suture around the transitional zone between fibrotic tissue and viable tissue was secured to firm fibrous tissue. The residual orifice was closed with an elliptical composite patch of equine pericardial patch and polyester velour with 3-0 polypropylene suture. We considered based on the operative finding that myocardial infarction (MI) led to LVA and VSP. Coronary thrombosis rather than coronary spasm may have provoked MI because spasm provocative test was negative.

[Biodegradable plastic production by activated sludge with different carbon sources].

Polyhydroxyalkanoates(PHAs) are polyesters synthesized by numerous bacteria in sludge as intracellular carbon and energy storage compounds under the condition of unbalance growth. Another important aspect of PHAs is thermoplastic polyester with the remarkable characteristics of biodegradability. In this paper, using sodium acetate, sodium propionate and sodium butyrate as the unique carbon source, 5.58%, 3.90% and 4.98% PHAs of dry sludge weight were obtained by assimilated sludge as PHAs synthesis strains in the anaerobic condition, and 12.32%, 9.55% and 11.35% PHAs of dry sludge weight were obtained in the anaerobic condition. It was shown that there were large quantities strains which could accumulate PHAs as granules in the cytoplasm of cells, and the yield of PHAs in the anaerobic process were higher than that in the aerobic process. The best result was that 12.32% PHAs of dry sludge weight was obtained in the best experiment condition, and the yield of the PHAs to substrate was 30.65%. The structures of PHAs were analyzed by 1HNMR spectrum, and the relationship of the structure of PHAs and the carbon source was explained.

Complications of Mersilene sutures in penetrating keratoplasty.

BACKGROUND: After penetrating keratoplasty, many corneal surgeons adjust sutures to reduce astigmatism and then leave the sutures in place indefinitely. Nylon sutures can hydrolyze and break years after surgery. In a series of human penetrating keratoplasties, we studied the use of polyester (Mersilene) sutures that do not hydrolyze. METHODS: We performed two prospective studies of polyester (Mersilene) sutures in human penetrating keratoplasty done by one surgeon. Study I was a randomized comparison of combined running and interrupted Mersilene and nylon sutures in 45 consecutive eyes. Study II was a case series of single running Mersilene suture with postoperative adjustment of suture tension to manage astigmatism in 23 consecutive eyes. We evaluated the performance of the suture and the control of astigmatism. RESULTS: The interrupted running suture study demonstrated that interrupted Mersilene sutures were 5.5 times more likely to have handling-related complications than nylon interrupted sutures (p = .01); in addition, they were three times as likely to have tissue-related complications as nylon (p = .16). The running suture study demonstrated an unacceptable complication rate of 69% when Mersilene was used as a single adjustable running suture. At 6 months postoperatively, the median refractive astigmatism for the adjustable cases was 3.37 diopters (mean, 4.03 +/- 2.37 D). Eyes with significant suture-related complications were 2.85 times more likely to have greater than 4.00 D of refractive astigmatism than were eyes without suture-related complications. CONCLUSIONS: Mersilene is an undesirable suture for use in penetrating keratoplasty.

[Experimental study on the reinforced effect of light curing composite resins used for crowns and bridges].

OBJECTIVE: To evaluate the reinforced effect of the light curing composite resins used for crowns and bridges. METHODS: Three light curing composite resins which were used for crowns and bridges were chosen, and three polyester fiber sieves and three stainless steel sieves in different mesh were used as the additional reinforced materials. Compressive strength and three point flexural strength of test bars made of those materials were evaluated. The reinforced bridges with special fibers were used as control groups. RESULTS: (1)There was significant increase in the stainless steel sieves groups. Nevertheless, there was some decrease after use of the polyester fibers as the additional reinforced material. (2)The increase of the reinforced crowns was especially obvious. (3)Among the three resins, the property of Targis was better than that of Arglass and Solidex. CONCLUSION: The properties of the whole composite material were closely correlated with the additional reinforced materials, the resistance to compression of the sieves are better than its resistance to bend.

A new bioactive molecule for improving vascular graft patency: exploratory trials in dogs.

Myxalin is a new bioactive molecule that we have isolated from the culture medium of Myxococcus xanthus, a non-pathogenic Gram negative bacterium. This glycopeptide possesses an antithrombotic effect in vivo and has been shown to promote human endothelial cell growth in vitro. With the object of exploring its ability to improve vascular graft healing and patency, myxalin was immobilized on 6 mm diameter knitted polyester prostheses using gelatin as a carrier, and the prosthesis was then implanted as an infrarenal abdominal arterial substitute in dogs for a period of 2 weeks. Two additional series of implantations were conducted for control purposes: one with gelatin-coated prostheses without myxalin, the other following normal preclotting of the polyester grafts. In order to select adequate sterilization conditions which can preserve the biological activity of myxalin, the prostheses were sterilized according to 3 different sterilization processes (gamma radiation and ethylene oxide either at 63 degrees C or 37 degrees C). At the sacrifice, all grafts were patent. The myxalin treated prostheses exhibited improved blood compatibility in terms of fewer thrombotic deposits and significant inhibition of platelet and fibrinogen uptake on their luminal surfaces. In addition, the development of a thin collagenous internal capsule with endothelial cells secreting high levels of prostacyclin was observed at both anastomoses of the myxalin-treated grafts sterilized by gamma radiation.

[Late intestinal perforations after alloplastic repair of postoperative eventrations].

Late intestinal perforation is the most rare and the most serious complication following the repair of abdominal parietal defects with alloplastic material. The authors have gathered in 7 years 6 cases of perforations which occurred between 1 and 10 years after using nonabsorbable synthetic mesh prostheses made of polyester (4 cases) and polypropylene (2 cases) as parietal substitution materials. The perforations were discovered either by the appearance of complete enteric fistulas after exploration of circumscribed parietal suppurations (3 cases) or during surgery of suppurations without external fistulas. All cases involved perforations of the small intestine and in only one case there was also a perforation of the transverse colon. In 4 cases, where eventration relapse co-existed, the polyester prostheses were found partially or totally detached from the abdominal wall. The treatment of enteric perforation consisted of 5 segmental enterectomies (one of those a double one) followed by anastomoses and one enteroraphy. The colic perforation was excised and sutured. The mesh was totally removed in 5 cases and partially removed in 1 case (around the perforation). The postoperative evolution was simple in only 3 cases. In the other 3 cases anastomotic fistulas occurred and one of them (with reduced leakage) was treated by nonoperative management. It did not close spontaneously even after 12 months. The other 2 cases required multiple reinterventions (enterectomies, ileostomies, ileocoloanastomosis). One of these patients has been lost away after 200 days of hospitalization and multiple reinterventions. In all cases, the closure of the abdominal wall was simplificad by cutaneous suture only whereas consecutive relapsed eventrations remain to be treated by subsequent reinterventions.
