Tracking genes from seed to supermarket: techniques and trends.

Analytical techniques to track plant genes in the environment and the food chain are essential for environmental risk assessment, government regulation and production and trade of genetically modified (GM) crops. Here, I review laboratory techniques to track plant genes during pre-commercialization research on gene flow and post-commercialization detection, identification and quantification of GM crops from seed to supermarket. At present, DNA- and protein-based assays support both activities but the demand for fast, inexpensive, sensitive methods is increasing. Part of the demand has been generated by stringent food labeling and traceability regulations for GM crops. The increase in GM crops, changes in GM crop design, evolution of government regulations and adoption of risk-assessment frameworks will continue to drive development of analytical techniques.

How multiagency partnerships can successfully address large-scale pollution problems: a Hawaii case study.

Oceanic circulation patterns deposit significant amounts of marine pollution, including derelict fishing gear from North Pacific Ocean fisheries, in the Hawaiian Archipelago [Mar. Pollut. Bull. 42(12) (2001) 1301]. Management responsibility for these islands and their associated natural resources is shared by several government authorities. Non-governmental organizations (NGOs) and private industry also have interests in the archipelago. Since the marine debris problem in this region is too large for any single agency to manage, a multiagency marine debris working group (group) was established in 1998 to improve marine debris mitigation in Hawaii. To date, 16 federal, state, and local agencies, working with industry and NGOs, have removed 195 tons of derelict fishing gear from the Northwestern Hawaiian Islands. This review details the evolution of the partnership, notes its challenges and rewards, and advocates its continued use as an effective resource management tool.

The use of erythropoietin in neonates.

Although much information has been accumulated about the clinical use of Epo in preterm infants, many questions remain unanswered. The evolution of clinical practice in the care of extremely ill, preterm infants continues to affect the number of transfusions required during hospitalization. Decreasing phlebotomy losses and instituting standardized transfusion guidelines have both been shown significantly to decrease the transfusion requirements of preterm infants. The administration of Epo likely decreases transfusion need even further; however, the direct impact of each of these actions has not been studied prospectively. It is likely that the combination of instituting rigorous and standardized transfusion guidelines, decreasing phlebotomy losses, and the appropriate use of Epo will have the greatest impact in decreasing transfusion requirements in all preterm and term neonates, regardless of the cause of their anemia.

Rotavirus VP7, VP4 and VP6 genotypes co-circulating in Tehran, Iran, between 2003 and 2004.

Rotaviruses were detected by enzyme-linked immunosorbent assay (ELISA) in 92 out of 374 faecal samples collected between November 2003 and October 2004 at the Markaz Tebbi Koudakan Hospital, Tehran, Iran, from children aged 6 months to 5 years. Analysis of clinical and disease severity data showed a significant association between rotavirus infection and diarrhoea, vomiting and severe dehydration. Ninety-two samples (64 rotavirus ELISA-positive and 28 ELISA-negative samples) were sent to the Enteric Virus Unit, Virus Reference Department, Centre for Infection, Health Protection Agency, UK for rotavirus characterization by G-typing, P-typing and subgrouping (SG) using reverse transcriptase (RT)-PCR, semi-nested PCR and sequencing methods. In this study, both common and uncommon rotavirus genotypes were detected. The most prevalent types were G1P[8], SGII (59.2%) followed by G9P[8] SGII (15.5%) which has not been previously reported from Iran. Unusual genotypes G1P[10] SGI (2.8%) and G12P[8] SGII (1.4%) and strains derived from reassortment between common co-circulating genotypes such as G1P[4] SGII represented 5.6% of strains. Mixed infections with combinations of G1+G4P[8] SGII and G1+G9P[8] SGII were also found. This contrasts with previous reports from Iran in which a small number of common rotavirus strains (G1 and G4) were found. This study highlights the need for continued surveillance and characterization of rotaviruses to take account of the rapid evolution and introduction of novel rotaviruses into the human population.

Brief-time-series analysis of depressive symptomatology in older people.

This research shows the utility of systematic data-gathering from older people and of a statistical analysis procedure for interpreting the data. Four cases of institutionalized older people are presented, and their scores of depressive symptomatology over a period of one to two years is analyzed. Time-series analysis showed a significant positive trend of depression symptomatology in two of the cases, perhaps too subtle for detection in routine clinical check-up, but statistically verifiable. In one of these cases, none of the factors of the scale stands out, despite the fact that the general index shows a statistically significant change over the 36 observations made, so that the results obtained are related to the syndrome of depletion or exhaustion. In the second of the cases, we detect a significant change in the depressive mood factor, which may indicate a subclinical depressive form in its initial stages. Continuous registers can reveal valuable information about situations and progress in the evolution of an older person's mood, with regard to natural development, the prelude to a mood disorder, or follow-up in clinical cases.

Computerized quantitative radionuclide assessment of heterotopic ossification in spinal cord injury patients.

We evaluated the progression of heterotopic ossification (HO) in 17 spinal cord injury patients by comparing radiographs, quantitative radionuclide bone scans, and serum alkaline phosphatase levels. Evidence of maturation of HO appeared earlier (3 months to 6 years post injury) in radiographs, whereas, during the same time frame, radioactive nuclide assessment showed continued progression of HO in 10 out of the 17 patients. The evolution of HO appeared to take place over a period ranging between 3 and 80 months. We believe that stabilization of HO may be reasonably defined in terms of uptake ratios of 2.0 or less in patients with initial uptake ratios over 3.0 but below 5.0, and of ratios of 3.0 or less when the initial values are over 5.0.

[Early amniocentesis].

Preliminary clinical experience with early amniocentesis is reported. Fifty-two amniocenteses were performed before the end of the 14th week following the last menstrual period. Cytogenetic and biochemical analyses (AFP, AChE) were performed. Increasing experience with amniotic fluid samples containing small-cell populations and the use of combined culture media improved the poor initial results to a 100% level, thus enabling the use of this technique for diagnostic purposes. The sampling technique and the post-procedural evolution of twelve amniocenteses in pregnant women, whose pregnancy is to be continued, are presented. The possibility of performing amniocentesis in early pregnancy is discussed with reference to anatomical aspects (amniotic and chorionic cavity).

Dynamical correlations in Brownian hard rods.

We analyze the time decay of small amplitude density perturbations in systems of highly packed Brownian hard rods, relaxing towards a uniform density distribution. The results of Brownian dynamics simulations and those of the deterministic dynamic density functional (DDF) theory, are contrasted with a new theoretical approach beyond the DDF assumptions. We characterize dynamical correlation modes which, having the lowest relaxation time, determine the late time evolution of the system. The spectrum of possible time decays has a continuous band structure, with pockets of discrete values, near the minima of the DDF results, where the validity of that theory appears to be well established.

Immunophenotypic analysis of hematogones (B-lymphocyte precursors) and neoplastic lymphoblasts by 4-color flow cytometry.

Hematogones are identified by 4-color flow cytometry in most bone marrow specimens. They are more commonly found and are generally present in higher numbers in children. There is a general decline in hematogones with increasing age but a broad range exists at all ages and marrow from some adults contains relatively high numbers. They are often increased (> 5%) in regenerating marrow and in some clinical conditions, particularly various types of cytopenias and neoplastic diseases. Hematogones may morphologically resemble the neoplastic lymphoblasts of precursor B ALL and their immunophenotype also has features in common with neoplastic lymphoblasts. Distinguishing hematogones from neoplastic lymphoblasts may be problematic in post-chemotherapy and post-bone marrow transplant regenerating marrow. With 4-color flow cytometry using optimal antibody combinations the distinction can nearly always be made. Hematogone populations always exhibit a continuous and complete maturation spectrum of antigen expression typical of the normal evolution of B-lineage precursors; they lack aberrant or asynchronous antigen expression. The neoplastic lymphoblasts in precursor B ALL deviate from the normal B-lineage maturation spectrum and exhibit maturation arrest and over-, under-, and asynchronous expression of antigens observed on normal B-cell precursors and they often aberrantly express myeloid-associated antigens.

Continuous exchange of sequence information between dispersed Tc1 transposons in the Caenorhabditis elegans genome.

In a genome-wide analysis of the active transposons in Caenorhabditis elegans we determined the localization and sequence of all copies of each of the six active transposon families. Most copies of the most active transposons, Tc1 and Tc3, are intact but individually have a unique sequence, because of unique patterns of single-nucleotide polymorphisms. The sequence of each of the 32 Tc1 elements is invariant in the C. elegans strain N2, which has no germline transposition. However, at the same 32 Tc1 loci in strains with germline transposition, Tc1 elements can acquire the sequence of Tc1 elements elsewhere in the N2 genome or a chimeric sequence derived from two dispersed Tc1 elements. We hypothesize that during double-strand-break repair after Tc1 excision, the template for repair can switch from the Tc1 element on the sister chromatid or homologous chromosome to a Tc1 copy elsewhere in the genome. Thus, the population of active transposable elements in C. elegans is highly dynamic because of a continuous exchange of sequence information between individual copies, potentially allowing a higher evolution rate than that found in endogenous genes.

Ambulatory antimicrobial use: the value of an outcomes registry.

The opportunity for treating many serious infections in the community or ambulatory setting is growing. Outpatient parenteral antimicrobial therapy (OPAT) provides many potential advantages to the patient, hospital and clinician, including quality of life, cost savings and reduced risk of hospital-acquired infections due to antibiotic-resistant organisms. However, despite the evolution of this type of care in many countries, there have been continued questions and concerns about its safety and effectiveness. As with many new forms of therapy in medicine, the value of OPAT is in doubt because of the lack of published information concerning outcomes and its impact on patient care. In order to examine the quality of such programmes, an outcome-based registry of patients has been developed for OPAT. The core outcomes measures include clinical effectiveness, eradication of bacteria and adverse antibiotic events. The registry may also be adapted for benchmarking for quality assurance, surveying performance of new antimicrobials, cost effectiveness studies and comparisons of different antibiotics and their side effects.

Increased body iron stores in elite road cyclists.

BACKGROUND: One third of French elite road cyclists were found to have hyperferritinemia on antidoping control tests performed during the Tour de France in 1998. PURPOSE: This study was undertaken to determine whether hyperferritinemia corresponded to elevated body iron stores or not and, affirmatively, what were its mechanism, its clinical consequences, and its spontaneous course. METHODS: 83 elite road male cyclists presenting with hyperferritinemia, defined as serum ferritin level greater than 300 microg.L-1, were studied with respect to consumption of iron and other drugs, serum iron tests, HFE mutations, and hepatic iron concentration (HIC; N < 35 micromol.g-1 dry weight). RESULTS: All cyclists were asymptomatic and had normal physical and cardiac examination. Their median (range) serum ferritin, serum iron, and transferrin saturation levels were 504 microg.L-1 (306-1671), 20 micromol.L-1 (8.5-36.3), and 39% (20-76), respectively. HIC was increased in 24/27 up to 187 micromol.g-1. Allelic frequency of the H63D mutation was increased in cyclists when compared to controls (P = 0.04). However, iron tests did not differ according to HFE genotypes. Most cyclists (89%) had been supplemented with iron. The median iron supplementation was 25.5 g (range: 1.4-336) and correlated well (P = 0.002) with serum ferritin. Evolution of serum ferritin levels did not differ whether cyclists had been continuing iron supplementation or not. CONCLUSION: Hyperferritinemia in elite road cyclists accounted for increased body iron stores caused by and persisting after cessation of excessive iron supplementation. Even when mild, iron excess may expose to long-term complications and should be removed, at least at the time when professional cyclists retire. To prevent iatrogenic iron overload, supplementation with iron must be done according to serum ferritin follow-up and not either blindly or on the basis of serum iron determination only.

The prebiotic molecules observed in the interstellar gas.

Over 130 molecules have been identified in the interstellar gas and circumstellar shells, the largest among them is a carbon chain with 13 atoms and molecular weight of 147 (twice that of the simplest amino acid glycine). The high reliability of astronomical identifications, as well as the fairly accurate quantitative analysis which can often be achieved, is emphasized. Glycine itself has been claimed, but a recent analysis indicates that few, if any, of the astronomical radio lines attributed to glycine are actually from that molecule. Polycyclic aromatic hydrocarbons (PAHs) have long been proposed as the source of the unidentified infrared bands between 3 and 16 microm, but no single PAH has been identified in space, partly because PAHs generally have weak or non-existent radio spectra. A remarkable exception is the non-planar corannulene molecule (C20H10) that has a strong radio spectrum; in the rich molecular cloud TMC-1, it is found that less than 10-5 of the carbon is contained in this molecule, suggesting that PAHs are not the dominant large molecules in the interstellar gas, as has been claimed. Owing to inherent spectroscopic limitations, determining the structures of the large molecules in space may require capture of the dust grains, which are continually entering the outer Solar System.

Classical dynamics on graphs.

We consider the classical evolution of a particle on a graph by using a time-continuous Frobenius-Perron operator that generalizes previous propositions. In this way, the relaxation rates as well as the chaotic properties can be defined for the time-continuous classical dynamics on graphs. These properties are given as the zeros of some periodic-orbit zeta functions. We consider in detail the case of infinite periodic graphs where the particle undergoes a diffusion process. The infinite spatial extension is taken into account by Fourier transforms that decompose the observables and probability densities into sectors corresponding to different values of the wave number. The hydrodynamic modes of diffusion are studied by an eigenvalue problem of a Frobenius-Perron operator corresponding to a given sector. The diffusion coefficient is obtained from the hydrodynamic modes of diffusion and has the Green-Kubo form. Moreover, we study finite but large open graphs that converge to the infinite periodic graph when their size goes to infinity. The lifetime of the particle on the open graph is shown to correspond to the lifetime of a system that undergoes a diffusion process before it escapes.

Octupolar perturbation of a single ion in a Penning trap.

We investigate the classical dynamics of a single ion trapped in a Penning trap perturbed by a octupolar electrostatic perturbation that introduces nonlinearities in the motion. We show that the dynamics is controlled by a single external parameter that combines the influence of the electric and magnetic fields. Through the variation of this parameter, we explore the evolution of the phase space structure of the system by the numerical continuation of the families of periodic orbits.

Propagating waves in one-dimensional discrete networks of coupled units.

We investigate the behavior of discrete systems on a one-dimensional lattice composed of localized units interacting with each other through nonlocal, nonlinear reactive dynamics. In the presence of second-order and third-order steps coupling two or three neighboring sites, respectively, we observe, for appropriate initial conditions, the propagation of waves which subsist in the absence of mass transfer by diffusion. For the case of the third-order (bistable) model, a counterintuitive effect is also observed, whereby the homogeneously less stable state invades the more stable one under certain conditions. In the limit of a continuous space the dynamics of these networks is described by a generic evolution equation, from which some analytical predictions can be extracted. The relevance of this mode of information transmission in spatially extended systems of interest in physical chemistry and biology is discussed.

Dynamics of stock prices.

We show that the dynamics of stock prices can be accurately described as a continuous time random walk with a time dependent diffusion coefficient. The time evolution of the diffusion coefficient can be derived from tick by tick databases provided the stock price is characterized in terms of a couple of values describing the best ask and the best bid. We are then led to a finding and, namely, that the transition rate of the random walk process is different from the frequency of transactions. Our results allow us to obtain a fast and reliable determination of the diffusion coefficient and precisely confirm that fat tails in the distribution of price variations are due to volatility fluctuations.

Generalized eruptive histiocytoma evolving into xanthoma disseminatum in a 4-year-old boy.

We report a 4-year-old boy who developed a generalized and symmetrical eruption of brownish papules over a period of 8 months, with spontaneous regression of some lesions. Clinical, histopathological and ultrastructural studies were suggestive of the diagnosis of generalized eruptive histiocytoma (GEH). The clinical features subsequently changed to a generalized eruption of confluent, yellowish papules, and diabetes insipidus developed. The clinical, histopathological and ultrastructural features of the new lesions were those of xanthoma disseminatum with cerebral involvement. This evolution suggests that GEH and xanthoma disseminatum may be variants of a continuous spectrum of histiocytic diseases.
