Systematic studies of structure and physiological activity of alapyridaine. A novel food-born taste enhancer.

By application of taste dilution analysis (+)-(S)-1-(1-carboxyethyl)-5-hydroxy-2-(hydroxymethyl)-pyridinium inner salt was recently successfully identified as a multimodal taste enhancer in beef bouillon. While being taste-less on its own, this so-called alapyridaine was found to intensify the human perception of sweet, salty, and umami taste. To gain information on the molecular requirements of this novel class of taste enhancer, a range of structurally related pyridinium betaines were synthesized, purified, and their physiological activities sensorially evaluated. Removal or modification of the hydroxyl and the hydroxymethyl group, respectively, induced a loss in bioactivity, thus indicating the 2-(hydroxymethyl)-5-hydroxypyridinium moiety as an essential structural element for taste enhancement. Regarding the amino substituent, neither the prolongation or removal of the alkyl chain or the carboxy function in the 1-(1-carboxy-2-ethyl)-moiety, nor the incorporation of an additional carboxy function led to any active derivative, thus demonstrating that also the structure of the nitrogen substituent is rather conserved for taste enhancement. But substitution of the methyl group by a benzyl group yielded a compound showing similar taste enhancing activities as found for alapyridaine. Interestingly, additional insertion of glycine between the 1-(1-carboxy-2-phenylethyl)-moiety and the pyridinium ring resulted in a compound eliciting comparable taste enhancing effects as shown for the compound lacking the glycine spacer. In contrast to these multimodal taste enhancers, substitution of the alanine moiety in alapyridaine by an arginine moiety revealed an one-dimensional taste enhancer exclusively increasing the human sensitivity for salty taste.

In vivo single-voxel proton MR spectroscopy in brain lesions with ring-like enhancement.

It is often difficult to make a correct diagnosis of ring-like enhanced lesions on Gd-enhanced MR brain images. To differentiate these lesions using proton MR spectroscopy (1H-MRS), we retrospectively evaluated the correlation between the 1H-MR spectra and histopathological findings. We evaluated proton MR spectra obtained from the lesions in 45 patients, including metastasis (n = 19), glioblastoma (n = 10), radiation necrosis (n = 7), brain abscess (n = 5), and cerebral infarction (n = 4). The rate of misdiagnosis was found to be lowest at the threshold level of 2.48 for the (choline containing compounds)/(creatine and phosphocreatine) ratio (Cho/Cr) obtained from the whole lesions, which include the enhanced rim and the non-enhanced inner region. That is, the positively predictive values of a Cho/Cr greater than 2.48 for diagnosing metastasis or glioblastoma was 88.9 and 60.0%, respectively, and the positively predictive value of a Cho/Cr less than 2.48 for diagnosing radiation necrosis or cerebral infarction was 71.4 and 100%, respectively. For further differentiating between metastasis and glioblastoma, information about the presence and absence of an N-acetyl-aspartate (NAA) peak and lipid- or lactate-dominant peak was found to be useful. In 73.7% of metastasis cases a lipid-dominant peak was observed in the whole lesion without an NAA peak in the inner region, whereas the same pattern was observed in only 10% of the glioblastoma cases. Correlation with the histopathological findings showed that a high Cho signal is suggestive of neoplasm. Lipid signal in the non-enhanced central region was correlated to necrosis. Lactate signals were often observed in glioblastoma, abscess and sometimes metastasis, presumably reflecting the anaerobic glycolysis by the living cells in the ring-like enhanced rim. Single-voxel proton MR spectroscopy may serve as a potential tool to provide useful information of differentiation of ring-like enhanced lesions that cannot be diagnosed correctly using enhanced MR images alone.

Enhanced resolution triple-quadrupole mass spectrometry for fast quantitative bioanalysis using liquid chromatography/tandem mass spectrometry: investigations of parameters that affect ruggedness.

In order to increase sample analysis throughput, the use of fast liquid chromatography in quantitative bioanalysis based on liquid chromatography coupled with tandem mass spectrometry (LC/MS/MS) has become prevalent. Therefore, it is important to increase the specificity of such bioanalytical methods. This can be done by enhancing both the chromatographic and mass resolving power. Increasing the mass spectrometric resolving power to minimize interference from endogenous compounds in the biological matrix is the subject of this paper. We present the results of our experience with developing and validating SRM-based, enhanced resolution bioanalytical methods using a new triple-quadrupole mass spectrometer with enhanced resolution capability. We have shown that SRM bioanalytical methods using better than unit-mass resolution (Q1 FWHM = 0.2 Th, Q3 FWHM = 0.7 Th) can be developed which are as rugged as unit resolution methods (Q1 FWHM = 0.7 Th, Q3 FWHM = 0.7 Th). The enhanced resolution methods require more attention to detail than unit resolution methods. For instance, the mass setting for precursor ion selection is more critical because the mass peak is narrower. Because of this, enhanced resolution methods may be more easily influenced by temperature changes in the laboratory. We have shown that there is good correlation between the shift in the precursor ion mass and the ambient temperature. Other studies carried out to investigate the effects on mass peak shape and response (both in the SIM and SRM mode) as the result of varying the FWHM revealed some interesting results. For instance, the decrease in response with the decrease in the FWHM was larger using SRM compared to that using SIM. However, the decrease in both SRM and SIM response with decreasing FWHM was significantly smaller compared with the decrease obtained using an older generation instrument. We demonstrate that, at concentrations near the limit of detection, the signal specificity can be improved by using an enhanced resolution method. To compare the performance of an enhanced resolution method against a unit resolution method under optimized mass spectrometric conditions, we analyzed calibration standards and quality control samples using a lower limit of quantitation that could be easily achieved by either method. Under these conditions, the two methods were essentially the same, demonstrating that the enhanced resolution method is as accurate, precise and rugged as the unit resolution method. We propose system suitability procedures, based on precursor ion scan, product ion scan, SRM with fractional mass changes, or SIM with a narrow scan width, for the updating of the SRM set masses before the start of analysis. We also recommend that Q1 SRM masses be determined during and at the end of analysis in order to ascertain whether or not the precursor masses have shifted during the course of the analysis.

Antibody-mediated immune enhancement in coxsackievirus B3 myocarditis.

The aim of the present study was to explore the contribution of antibody-mediated immune enhancement in coxsackievirus B3(CB3) infection. Murine macrophage-like P388D1 cells were exposed to various concentrations of anti-CB3 immunoglobulin G (anti-CB3 IgG) or the Fab fragment of anti-CB3 IgG, and were infected with CB3 in Experiment I. High concentrations of anti-CB3 IgG showed a virus-neutralizing activity; however, a subneutralizing antibody concentration of IgG significantly enhanced virus replication. This infectious enhancement was blocked not only by the pretreatment of heat-aggregated gamma-globulin but by a specific Fc receptor (Fc gamma III/II receptor) antibody treatment. In contrast, the Fab fragment of anti-CB3 IgG did not enhance CB3 infection, but showed a rational neutralizing activity to CB3. These findings suggest the presence of Fc receptor mediated enhancement of CB3 infection in vitro. In Experiment II, C(3)H/He mice were inoculated with various amounts of an amyocarditic variant of CB3 followed 15 days later by myocarditic CB3. By this rechallenge, myocarditis was not induced in the mice with high neutralizing antibody titers. There was an inverse relationship between preexisting neutralizing antibody titers and the severity of myocarditis. The severity of myocarditis and myocardial CB3 titers, however, were markedly enhanced in the mice with a subneutralizing level of immunity compared to those with no immunity. The distribution of myocardial Fc receptor-bearing cells and serum macrophage inflammatory protein-2 levels paralleled the severity of myocarditis. By another virus rechallenge in Experiment III, enhanced infection of CB3 was not observed in vivo. These findings suggest that antibody-mediated immune enhancement might be involved in the pathogenesis of CB3 myocarditis.

Acute stress rapidly and persistently enhances memory formation in the male rat.

Previous studies, as well as the present one, report that acute exposure to intermittent tailshocks enhances classical eyeblink conditioning in male rats when trained 24 h after stressor cessation. In Experiment 1, it was determined that the facilitating effect of stress on conditioning could also be obtained in response to a stressor of acute inescapable swim stress but not inescapable noise or the unconditioned stimulus of periorbital eyelid stimulation. These selective responses arose despite comparable enhancements of the stress-related hormone corticosterone in response to tailshocks, periorbital eyelid stimulation, noise stress, and supraelevation in response to swim stress. Although corticosterone is necessary for the enhanced learning in response to stress (Beylin & Shors, 1999), these results suggest that it is not sufficient. In addition, the results suggest that the enhancement is not dependent on common characteristics between the stressor and the conditioning stimuli (stimulus generalization). In Experiment 2, it was determined that the facilitating effect of the stressor on conditioning occurs within 30 min of stressor cessation. Thus, the mechanism responsible for facilitating memory formation is rapidly induced as well as persistently expressed. In Experiment 3, it was determined that exposure to the stressor does not enhance performance of the conditioned response after the response has been acquired. Thus, exposure to the stressor enhances the formation of new associations rather than affecting retention or performance of the motor response. These studies extend the circumstances under which stress is known to enhance associative learning and implicate neural mechanisms of memory enhancement that are rapidly induced and persistently expressed.

Multiple copies of virG enhance the transient transformation of celery, carrot and rice tissues by Agrobacterium tumefaciens.

In an effort to improve the T-DNA-mediated transformation frequency of economically important crops, we investigated the possible enhancement effect of multiple copies of virG genes contained in Agrobacterium tumefaciens strains upon the transient transformation of celery, carrot and rice tissues. Four days after A. tumefaciens infection, we performed histochemical beta-glucuronidase (GUS) assays to determine the frequency of transient transformation of calli from celery and carrot, and explants from rice and celery. Additional copies of octopine- and agropine-type virG genes in A. tumefaciens strains containing an agropine-type Ti-plasmid enhanced the frequency of transient transformation of celery and rice. This enhancement ranged from 25% to five-fold, depending upon the source of the virG gene and the plant tissues inoculated. For both rice and celery, we observed a greater enhancement of transformation using A. tumefaciens strains containing additional copies of an octopine-type virG gene than with strains harboring additional copies of an agropine-type virG gene. Multiple copies of virG genes contained in A. tumefaciens strains harboring a nopaline-type Ti-plasmid had a smaller enhancing effect upon the transformation of celery tissues, and no enhancing effect upon the transformation of rice. In contrast, we obtained a three-fold increase in the transient transformation frequency of carrot calli using an A. tumefaciens strain harboring a nopaline-type Ti-plasmid and additional copies of an octopine-type virG gene. Our results show that multiple copies of virG in A. tumefaciens can greatly enhance the transient transformation frequency of celery, carrot and rice tissues, and that this enhancement is influenced by both the type of Ti-plasmid harbored by A. tumefaciens and by the infected plant species.

Evaluation of IBMX-enhanced ocular hypotension after adrenergic agonists in the rabbit eye.

BACKGROUND: 3-Isobutyl-1-methylxanthine (IBMX), a phosphodiesterase inhibitor, enhanced the reduction of intraocular pressure more after administration of norepinephrine and epinephrine than after isoproterenol. The question arises of whether or not the IBMX-induced enhancement of ocular hypotension is exclusively due to beta 2-adrenoceptor/cAMP stimulation. METHODS: In groups of eight rabbits the ocular hypotensive responses after selective adrenergic agonists were studied in the presence and absence of phosphodiesterase inhibition with IB-MX. RESULTS: Pretreatment with IBMX 1%, applied topically, did not enhance the ocular hypotensive responses after phenylephrine (alpha 1), B-HT920 (alpha 2) and dobutamine (beta 1). The ocular hypotensive responses induced by salbutamol (0.001-0.5%) and higher concentrations of terbutaline were significantly enhanced by IBMX. Combined treatments of terbutaline 0.01% and B-HT920 0.2%, dobutamine 3% and phenylephrine 2%, and dobutamine 3% and B-HT920 0.2% were not associated with enhanced ocular hypotensive responses in the presence of IBMX. The only combination that was associated with a significant enhancement of ocular hypotension when combined with 1% IBMX was phenylephrine 2% and terbutaline 0.01%. A subthreshold dose of phenylephrine 0.1% further increased the enhanced ocular hypotensive responses induced by salbutamol 0.025, 0.2 and 0.5% in combination with IBMX. CONCLUSIONS: Phosphodiesterase inhibition with IB-MX enhances the ocular hypotensive effect induced by catecholamines not only by beta 2-adrenoceptor/cAMP stimulation, but also by simultaneous alpha 1-adrenoceptor stimulation.

Enhancement of Ca2+ channel currents in human neuroblastoma (SH-SY5Y) cells by phorbol esters with and without activation of protein kinase C.

The effects of phorbol esters on Ca2+ channel currents in human neuroblastoma SH-SY5Y cells were studied using whole-cell patch-clamp recordings. Bath application of 12-O-tetradecanoylphorbol-13-acetate (TPA) or phorbol 12,13-dibutyrate (PDBu; 100 nM to 1 microM), known activators of protein kinase C (PKC), enhanced Ca2+ channel currents in a voltage-dependent manner similar to that of Bay K 8644. TPA also enhanced Ca2+ channel currents during cell dialysis with the PKC pseudosubstrate, PKC(19-36), and in cells which had been pre-incubated with 500 nM staurosporine, and which were exposed to staurosporine during recordings. Application of 4 alpha-phorbol-12,13-didecanoate (4 alpha-PDD; 100 nM), which does not activate PKC, caused current enhancement similar to the effects of TPA. However, intracellular dialysis of TPA was without effect on Ca2+ channel currents. Residual Ca2+ channel currents recorded after exposure to 1 microM omega-conotoxin GVIA were still enhanced by TPA, but in the presence of either Bay K 8644 (5 microM) or nifedipine (5 microM), TPA was without effect. When cells were pre-incubated for 10 min at 37 degrees C with 100 nM TPA, currents subsequently recorded in its absence were enhanced as compared to untreated cells; 5 microM nifedipine still inhibited currents to the same degree. This enhancement was not mimicked by 4 alpha-PDD, and was inhibited by staurosporine. Our results indicate that acute applications of phorbol esters (at concentrations commonly used to activate PKC) enhance L-type Ca2+ channel currents in SH-SY5Y cells via a PKC-independent mechanism which appears similar to that induced by Bay K 8644. By contrast, pre-incubation with TPA enhances both L- and N-type currents via activation of PKC.

Sequential gadolinium-DTPA enhanced MRI studies in neuro-Behçet's disease.

Sequential gadolinium-DTPA (Gd-DTPA) enhanced MR images were obtained before and after steroid therapy in a case of neuro-Behçet's disease. Multiple scattered lesions, which could not be detected on pre- and post-contrast CT, were demonstrated mainly in the white matter of the pons and/or the cerebrum with both T1- and T2-weighted images. Some of these lesions, however, were not enhanced at all by infusion of Gd-DTPA. The Gd-DTPA infusion study demonstrated marked enhancement in the white matter of the pons and cerebrum. Some lesions not seen with T2-weighted images were also strongly enhanced by Gd-DTPA infusion at the acute stage. After steroid therapy, the symptoms and abnormal laboratory findings were resolved. The pontine and cerebral lesions on plain MR images remained unchanged even after resolution of the symptoms, suggesting that they were inactive old foci. On the other hand, the lesions detected in the enhancement study before steroid therapy disappeared with the repeat Gd-DTPA enhanced MR images hich were performed after resolution of the symptoms. Some active inflammatory lesions in neuro-Behçet's disease may be demonstrated only on Gd-DTPA enhanced MR images. Gd-DTPA enhanced MR imaging appears to be potentially useful for detecting active inflammatory lesions in neuro-Behçet's disease and for evaluating the efficacy of treatment.

Intracranial meningiomas. Comparison of plain and contrast-enhanced examinations in CT and MRI.

Fifty patients with intracranial meningiomas underwent plain and contrast-enhanced examinations with CT and MRI. Each of the MR studies consisted of three plain (T1, proton density and T2-weighted) and a post-contrast series (0.1 mmol Gd-DTPA/kg body weight). All techniques (plain CT, plain MRI, contrast-enhanced CT, contrast-enhanced MRI) proved to be highly efficient as regards tumour detection: depending on the technique, an intracranial lesion was demonstrated in 47-50 cases. The image contrast was assessed as good or excellent in 21 cases having plain CT and in 33 cases having plain MRI, but in 46 and 50 of the contrast-enhanced CT and MRI studies respectively. Adequate tumour delineation was achieved in 18 cases with plain CT, in 35 cases with plain MRI and in 46 and 50 cases of the contrast-enhanced CT and MRI examinations. The contrast-enhanced studies proved to be superior to the plain CT and MRI studies as regards image contrast and tumor delineation. Because of the methodological advantages of the MRI technique, contrast-enhanced MRI was judged to be slightly superior to contrast-enhanced CT.

Membrane permeability transition promoted by phosphate enhances 1-anilino-8-naphthalene sulfonate fluorescence in calcium-loaded liver mitochondria.

Phosphate and a number of other compounds induce membrane permeability transition (MBT) in Ca(2+)-loaded mitochondria. 1-Anilino-8-naphthalene sulfonate (ANS) was used as a fluorescent probe to investigate perturbations on the inner membrane during MBT. Induction of MBT caused ANS fluorescence enhancement with a biphasic rate that reached a plateau. The enhancement is analogous to that reported for de-energization of mitochondria. The fluorescence level was independent of whether ANS was added before or at different times after phosphate. In the absence of ANS, fluorescence was low and remained unchanged. The initial time course of MBT, as followed by large-amplitude swelling, was similar to that of fluorescence enhancement. Ruthenium red, EGTA, ADP, and cyclosporin A inhibited the enhancement. Only EGTA + ADP (or ATP) reversed the enhancement when added after phosphate. Efflux of matrix Ca2+ by sodium acetate or A23187 did not alter ANS fluorescence. The binding parameters (Kd and number of binding sites) were not significantly different, but the fluorescence maximum was more than doubled after MBT. Although the fluorescence of bound ANS showed a nonlinear relationship, it was always higher (73.0 +/- 19.0%) after reaching the plateau. Since ANS binding to membranes is nonspecific, the exact mechanism of the enhanced fluorescence is not apparent. The dependence of the initial rate of fluorescence enhancement on Ca2+ concentration was nonlinear, with 45 microM at half-maximal rate. The dependence on phosphate was hyperbolic with 0.7 mM at half-maximal rate, which is close to the Km value of phosphate carrier. The kinetics is compatible with Ca2+ binding to some membrane component(s) during MBT and cause ANS fluorescence enhancement.(ABSTRACT TRUNCATED AT 250 WORDS)

Hyperthermia enhances the cytotoxicity and platinum-DNA adduct formation of lobaplatin and oxaliplatin in cultured SW 1573 cells.

The cytotoxicity of cisplatin and cisplatin-DNA adduct formation in vitro and in vivo is clearly enhanced by hyperthermia. We investigated whether cytotoxicity and platinum-DNA adduct formation of two promising new third-generation platinum derivatives, lobaplatin [1,2-diamminomethylcyclobutane platinum(II) lactate] and oxaliplatin [oxalato-1,2-diaminocyclohexane platinum(II)], are also enhanced by hyperthermia. Cisplatin was used for comparison. SW 1573 cells were incubated with cisplatin, lobaplatin or oxaliplatin at different concentrations for 1 h at 37 degrees, 41 degrees and 43 degrees C. The reproductive capacity of cells was determined by cloning experiments. Immunocytochemical detection of platinum-DNA adducts was performed with the rabbit antiserum NKI-A59. At 37 degrees C, cisplatin was the most cytotoxic, followed by oxaliplatin and lobaplatin. Hyperthermia clearly enhanced the cytotoxicity of cisplatin, lobaplatin and oxaliplatin. There was no further increase in cytotoxicity at 43 degrees C compared to 41 degrees C for cisplatin and oxaliplatin. A further increase in cytotoxicity at 43 degrees C was observed for lobaplatin. At 43 degrees C thermal enhancement was higher for lobaplatin than for oxaliplatin, with the reverse pattern at 41 degrees C. For both drugs, thermal enhancement of cytotoxicity was lower than observed for cisplatin. Immunocytochemical detection of platinum-DNA adducts was feasible for all the drugs. Adduct formation was enhanced at 43 degrees C for cisplatin, lobaplatin and oxaliplatin with a relative increase of 410%, 170% and 180%. These results seem to confirm that an increase in platinum-DNA adduct formation is involved in the in vitro thermal enhancement of cytotoxicity. The observed thermal enhancement of cytotoxicity of lobaplatin and oxaliplatin in vitro warrants further in vivo investigations.

A study on the mechanism of antibody-dependent enhancement of feline infectious peritonitis virus infection in feline macrophages by monoclonal antibodies.

Enhancement of feline infectious peritonitis virus (FIPV) infection of feline macrophages was studied using monoclonal antibodies (MAbs) to the FIPV strain 79-1146. Adherent cells recovered from the feline lung and peritoneal cavity phagocytosed fixed red blood cells, and formed Fc-mediated rosettes. Enhancement of virus infection by MAb was investigated by inoculating alveolar macrophages with a mixtures of viral suspension and MAb, and examining the cells for intracellular viral antigen by the immunofluorescence assay and the amount of infectious virus in the supernatant fluid after incubation. The replication of FIPV in macrophages was enhanced by non-neutralizing MAbs recognizing peplomer protein (S) and transmembrane protein (M) of the virus. Even among the MAbs having the ability to neutralize FIPV strain 79-1146, some reversely enhanced virus infection when they were diluted. The enhancement was suppressed by pretreatment of the MAb with protein A. The enhancement was reduced by the use of F(ab')2 fragment of MAb. These results demonstrated antibody-dependent enhancement (ADE) of FIPV infection in macrophage. The replication of FIPV 79-1146 strain in macrophages from FIPV antibody-positive cats was more enhanced than in those from antibody-negative cats.

Enhancement characteristics of hepatic focal nodular hyperplasia and its scar by dynamic magnetic resonance imaging.

RATIONALE AND OBJECTIVES: To determine the relationship between the lesion and the scar enhancement characteristic in a series of hepatic Focal Nodular Hyperplasia (FNH) lesions studied with dynamic MR imaging. METHODS: Nine patients with FNH were studied. The slice showing the largest scar was selected for the dynamic single slice T1-weighted Gradient-echo sequence before and after contrast administration (15 images, one every 20 s). Analysis was performed with ROI measurements in the lesion and the scar. Signal-intensity and enhancement curves were obtained from both structures. RESULTS: Dynamic MRI showed the typical homogeneous early enhancement of the lesion with delayed enhancement of the scar. The scar enhanced early and vigorously in all cases. Two patterns of enhancement curves were defined. In the parallel pattern, both curves started early, quickly reaching a plateau maintained over time (77.8%). In the divergent pattern the curve of the scar was above the curve of the FNH (22.2%), after the maximum slope was reached, with progressive separation of the curves. CONCLUSION: There is a hypervascular scar enhancement within FNH lesions with either a parallel or divergent course after the maximum early enhancement.

Subdural enhancement on postoperative spinal MRI after resection of posterior cranial fossa tumours.

In malignant brain tumours which may disseminate staging, usually by cranial and spinal MRI is necessary. If MRI is performed in the postoperative period pitfalls should be considered. Nonspecific subdural contrast enhancement on spinal staging MRI is rarely reported after resection of posterior fossa tumours, which may be mistaken for dissemination of malignancy. We investigated the frequency of spinal subdural enhancement after posterior cranial fossa neurosurgery in children. We reviewed 53 postoperative spinal MRI studies performed for staging of paediatric malignant brain tumours, mainly infratentorial primitive neuroectodermal tumours 2-40 days after surgery. There was contrast enhancement in the spinal subdural space in seven cases. This was not seen in any of eight patients who had been operated upon for a supratentorial tumour. After resection of 45 posterior cranial fossa tumours the frequency of subdural enhancement was 15.5%. MRI showing subdural enhancement was obtained up to 25 days postoperatively. No patient with subdural enhancement had cerebrospinal fluid (CSF) examinations positive for tumour cells or developed dissemination of disease in the CSF. Because the characteristic appearances of subdural contrast enhancement, appropriate interpretation is possible; diagnosis of neoplastic meningitis should rarely be impeded. Because of the striking similarity to that in patients with a low CSF-pressure syndrome and in view of the fact that only resection of tumours of the posterior cranial fossa, usually associated with obstructive hydrocephalus, was followed by this type of enhancement one might suggest that rapid changes in CSF pressure are implicated, rather the effects of blood introduced into the spinal canal at surgery.

Post-contrast FLAIR MR imaging of the brain in children: normal and abnormal intracranial enhancement.

OBJECTIVE: To describe the normally enhancing intracranial structures on fluid-attenuated inversion recovery (FLAIR) MRI and evaluate the usefulness of postcontrast FLAIR images of the brain in the assessment of enhancing lesions by comparing postcontrast FLAIR imaging with postcontrast T1-weighted (T1-W) imaging in children. MATERIALS AND METHODS: In 218 children, 249 pre- and postcontrast FLAIR MRI examinations of the brain were obtained consecutively between August 2001 and April 2002. The normally enhancing intracranial structures on FLAIR imaging were assessed in 77 MRI studies of 74 children who showed normal intracranial imaging findings. In 86 MRI studies in 68 children who showed enhancing intracranial lesions, lesion conspicuity on postcontrast FLAIR imaging was compared with that on postcontrast T1-W imaging for all lesions ( n=107), intra-axial lesions ( n=40), or extra-axial lesions ( n=67). RESULTS: The normally enhancing intracranial structures on FLAIR MRI were the choroid plexus (99%, 76/77), pituitary stalk (84%, 65/77), pineal gland (71%, 55/77), dural sinuses (26%, 20/77), and cortical veins (9%, 7/77). Of all the enhancing lesions, lesion conspicuousness on postcontrast FLAIR imaging was better than postcontrast T1-weighted imaging in 42, equal in 28, and worse in 37. Of 40 intra-axial lesions, lesion conspicuousness on postcontrast FLAIR imaging was better in 6, equal in 10, and worse in 24. Of 67 extra-axial lesions, lesion conspicuity on postcontrast FLAIR imaging was better in 36, equal in 18, and worse in 13. Conspicuousness of extra-axial lesions was significantly better than that of intra-axial lesions on postcontrast FLAIR imaging ( P<0.001). CONCLUSIONS: The choroid plexus, pituitary stalk, pineal gland, dural sinuses, and cortical veins show normal enhancement on postcontrast FLAIR MRI in children, and postcontrast FLAIR imaging appears better than postcontrast T1-W imaging in the assessment of extra-axial enhancing lesions in children.

Chondroblastic osteosarcoma: characterisation by gadolinium-enhanced MR imaging correlated with histopathology.

OBJECTIVE: To identify specific features of chondroblastic osteosarcoma on gadopentetate dimeglumine (Gd)-enhanced magnetic resonance (MR) imaging. DESIGN AND PATIENTS: Nine patients with chondroblastic osteosarcoma and a control group of 20 patients with conventional central osteosarcoma were included in this study. The histopathological findings of the surgical specimens were compared with enhancement patterns on static Gd-enhanced MR images. RESULTS: In chondroblastic osteosarcoma septonodular and peripheral rim enhancement represented tumour with a pure chondroid matrix. Non-enhancing and heterogeneous enhancing areas represented tumour with both chondroid and osteoid matrix. In the tumours in the control group enhancement was predominantly heterogeneous but in one it was homogeneous. All these areas corresponded to necrotic or viable osteoid tumour tissue or fibrovascular tissue in areas of necrosis. CONCLUSION: Gd-enhanced MR imaging can assist in obtaining diagnostic biopsy material of chondroblastic osteosarcoma by identifying both osteoid- and chondroid-forming areas.

The enhancement of riboflavin-mediated photo-oxidation of doxorubicin by histidine and urocanic acid.

PURPOSE: Previously we have shown that doxorubicin (Adriamycin, ADR) can be inactivated by light-excited riboflavin. The inactivation of the drug results from its direct oxidation by the excited triplet riboflavin in a type I photosensitization reaction, and 3-methoxysalicyclic acid is an ADR breakdown product. In the present study, we investigated the enhancement of this process by histidine and some other imidazole analogs. METHODS: ADR solutions containing various concentrations of riboflavin and other agents were exposed to 365 nm light for various time periods and then the absorbance spectrum of ADR was measured by a double beam spectrophotometer. These measurement were used to calculate the half-time of the ADR degradation process. The degraded ADR solutions were analyzed by HPLC. RESULTS: The rate of bleaching of ADR by light-excited riboflavin was enhanced in the presence of histidine in a concentration-dependent manner. This enhancement was more pronounced at higher riboflavin concentrations. Histidine also enhanced the riboflavin-mediated photobleaching of N,N-dimethyl-4-nitrosoaniline (RNO), a compound known to be resistant to oxidation by singlet oxygen but sensitive to oxidation by the trans-annular peroxide of histidine. RNO was found to block the histidine enhancement of the riboflavin-mediated photobleaching of ADR in a competitive manner. Among the imidazole analogs of histidine tested, urocanic acid was found to be the most efficient enhancer of the riboflavin-mediated photobleaching of ADR. Superoxide anion radicals which retard the oxidation of ADR were quenched by urocanic acid but not by histidine. It was shown that the oxidation of ADR by the trans-annular peroxide of histidine resulted in the formation of 3-methoxysalicylic acid. CONCLUSIONS: In contrast to singlet oxygen, the trans-annular peroxide, formed by the interaction of histidine and the singlet oxygen produced by photoexcited riboflavin, is an efficient oxidizer of ADR. The enhancement of the riboflavin-mediated photobleaching of ADR by histidine analogs depends on the rate of their conversion to a trans-annular peroxide and on the efficiency of these products in oxidizing ADR. However, for some analogs of histidine, as shown for urocanic acid, other mechanisms could also be involved. The presence of urocanic acid in the skin suggests that significant degradation of ADR could occur in the presence of biologically relevant concentrations of riboflavin if patients treated with ADR are exposed to sunlight. The finding that histidine also enhanced the degradation of ADR to 3-methoxysalicylic acid, suggests that the process of ADR oxidation by the trans-annular peroxides is similar to the direct oxidation of ADR by excited triplet riboflavin.
