Influence of radioprotective agents on the activities of isoenzymes of blood serum alkaline phosphatase in irradiated dogs.

Changes of the total serum activity of alkaline phosphatase (AP) and its bone and intestinal isoenzymes were studied in dogs gamma-irradiated by 3.0 Gy. The activities of AP isoenzymes were determined by means of a heat inactivation-inhibition method. After irradiation the serum AP activities were lower in general. In case of the bone isoenzyme the decrease was most pronounced. The changes of the total AP activity and of the intestinal isoenzyme were less distinct. The i.m. administration of the radioprotective mixture cystamine with mexamine (24 mg/kg + 4 mg/kg) before irradiation led only to a moderate reduction of the radiation-altered AP values in the dogs. The present results indicate that the radioprotective way used here renders only an unsatisfactory protection for large laboratory animals against ionizing radiation.

[The alkaline phosphatase activity of the blood serum as a factor reflecting the chronic inflammatory process in the skin in generalized forms of psoriasis, neurodermatitis, eczema and trophic ulcers].

Blood serum alkaline phosphatase (AP) level was found elevated in the patients suffering from eczema, neurodermatitis, eczema with large skin sites involved, and in those with trophic ulcers of various origins, as against normal subjects and reference patients with sarcoidosis and Kaposi's sarcoma. Elevated level of the enzyme activity is directly related to exacerbation stage and size of the skin site involved; this level reduced after therapy and this reduction was associated with clinical improvement. No concomitant visceral abnormalities (calculous cholecystitis, liver cirrhosis, or malignant tumors) were detected in the examinees, that might influence the blood serum AP activity.

Labelling of T cell subsets under field conditions in tropical countries. Adaptation of the immuno-alkaline phosphatase staining method for blood smears.

Immuno-alkaline phosphatase (AP) staining for T cell subsets (CD4 and CD8) of smears from fingerprick blood functioned well under tropical climatic conditions when smears were stored frozen with silica gel before being labelled. Unlabelled smears were stored for up to 12 months and could be transferred abroad without antigenic damage. Identical total CD4 and CD8 counts were obtained on venous and capillary blood, when compared using a FACS analyser. Although the AP method gave somewhat higher total CD4 and CD8 counts, the ratio remained the same. The major advantages of the method are: (i) no expensive equipment is required, (ii) only minute amounts of blood are needed, and (iii) slides can be stored for long periods before labelling and can be preserved for later reading. The method is suitable for community studies where there is a need for assessing the immune status of the population.
