A computer program that simulates cloning of DNA.

Easy Cloner is a computer program that manipulates DNA sequences as in cloning experiments and produces maps of the resulting plasmids. The program runs in the graphics mode of an IBM PC or compatible computer and is operated by using a mouse to point to the required actions. The program is available in the public domain.

Computer programs that teach the interpretation of image-based laboratory tests.

OBJECTIVE: To review the effort of the University of Washington (UW) Department of Laboratory Medicine to develop and use personal computer programs to teach the interpretation of image-based clinical laboratory tests to medical technologists and other health care workers. DATA SOURCES: Professional journals and books; Software owned by and licensed by the University of Washington. STUDY SELECTION: Not applicable. DATA EXTRACTION: Not applicable. DATA SYNTHESIS: We have been developing interactive personal computer (PC) programs for teaching image-based laboratory tests to medical technologists and other health care workers. The programs, called "Laboratory Tutors," are useful for teaching microscope-based tests and tests based on electrophoresis. Our programs include ANA-Tutor, which teaches the immunofluorescence assay for anti-nuclear antibodies; Gram Stain-Tutor, which teaches the direct Gram stain; Electrophoresis-Tutor, which teaches the interpretation of agarose gel protein electrophoretic patterns; Urinalysis-Tutor, which teaches the microscopic examination of urine sediment; in addition to other programs. The tutorials are all based on high-quality digital images that were acquired and processed using digital imaging systems. They require minimal computer literacy and have a number of advantages over standard approaches to teaching image-based laboratory tests. The computer tutorials are used in UW's medical technology and medical school curriculum, where they are used as supplements to traditional instruction. CONCLUSION: Laboratory tutors are computer programs that use high resolution digital images to teach the interpretation of image-based laboratory tests. We plan to continue to develop these programs, study their educational effectiveness, and update them periodically.

Using 'off the shelf', computer programs to mine additional insights from published data: diurnal variation in potency of ACTH stimulation of cortisol secretion revealed.

We describe the use of available computer programs to mine additional insights from published data. Graphs from four studies of ACTH and cortisol plasma levels collected throughout the 24 h day in humans were scanned into digital form and the data points extracted. To investigate the magnitude of ACTH stimulation of cortisol secretion across the 24 h, Monte Carlo methods were used to fit the parameters of a computer model of the ACTH-adrenal axis to the extracted data. ACTH was found to have a greater effect on cortisol secretion during the peaks of the cycle than at the nadir. This finding could not be explained by previously published dose response curves of ACTH effect. This implies that other modulators influence the effect of ACTH on the adrenal. This study also demonstrates how available computer programs can be used to examine models of physiologic regulation using data already available in the literature.

Developing a dose-volume histogram computation program for brachytherapy.

A dose-volume histogram (DVH) computation program was developed for brachytherapy treatment planning in an attempt to benefit from the DVH's ability to present graphically information on 3D dose distributions. The program is incorporated into a planning system that utilizes a pair of orthogonal radiographs to localize the radiation sources. DVHs are calculated for the volume of tissue enclosed by an isodose surface (e.g. half the value of the reference isodose). The calculation algorithm is based on a non-uniform random sampling that gives a denser point distribution at the centre of the implants. Our program was tested and proved to be fast enough for clinical use and sufficiently accurate (i.e. computation time of 20 s and less than 2% relative error for one point source, for 100,000 calculation points). The accuracy improves when a larger calculation point number is used, but the computation time also increases proportionally. The DVH is presented in the form of a simple graph or table, or as Anderson's 'natural' DVH graph. The cumulative DVH tables can be used to extract a series of indexes characterizing the homogeneity and the dose levels of the distribution in the treatment volume and the surrounding tissues. If a reference plan is available, the DVH results can be assessed relative to the reference plan's DVH.

The reliability and internal consistency of an Internet-capable computer program for measuring utilities.

OBJECTIVE: The purpose of this study was to assess the reliability and internal consistency of measurements of utilities performed with a computer program (iMPACT2) designed for Internet surveys and Internet patient decision-support systems. METHODS: We implemented the Internet Multimedia Preference Assessor Construction Tool, version 2 (iMPACT2) program using the combination of a web server, HTML files, and a web-accessible database. The program randomized subjects, screened their responses for missing data and failures of internal consistency, assisted patients with resolving certain inconsistencies, and, upon a subject's completion of the protocol, provided a report of results to the research assistant administering the program. To validate the iMPACT2 program, we recruited 60 healthy community volunteers and elicited preferences in a research-lab setting using a visual analog scale (VAS) and the standard gamble (SG) for subject's current health and three hypothetical states. For purposes of comparison, we also administered a Short Form-12 (SF-12) health-assessment questionnaire. Subjects used the computer software on two occasions separated by 2-4 weeks of time. RESULTS: Visual analog scale and standard gamble ratings for subjects' current health were reliable (intraclass correlation coefficient (ICC) of 0.82 and 0.84 (two outliers excluded -0.60 without exclusions), respectively) were comparable with the reliability of the Physical and Mental Component scales of the SF-12 (ICCs of 0.84 and 0.75, respectively). Subjects could easily discriminate between hypothetical states (D scores 0.74 for SG and 0.90 for VAS), and 94% had a completely internally consistent ordering of preference ratings for states. CONCLUSIONS: iMPACT2 produces measurements of standard gamble utilities that are reliable and have a high degree of internal consistency. Procedures for assessment of utilities developed for desktop computer programs can be translated to software designed for the Internet, facilitating the use of utilities and endpoints in clinical trials and development of web-based decision-support applications for patients. However, further testing, including direct comparisons with traditional interviewer administered utility elicitation protocols, is needed.

Are calculated log P values for some guanine derivatives by different computer programs reliable?

The log P values of n-octanol/water for some guanine derivatives, acyclovir, deoxyaciclovir and their acetyl congeners, were calculated by some commercially available computer programs for log P calculation. These values were compared with those obtained by the conventional shake-flask method. It was established that the calculations of log P values for examined guanine derivatives by these computation programs do not give reliable results.

A computer program for constructing multivariate reference models.

From a statistical point of view the simultaneous interpretation of multiple variables should be performed with a multivariate reference model rather than with multiple univariate reference intervals. A computer program for constructing and testing multivariate reference models is described. The use of the computer program is illustrated with a data set of total serum calcium concentrations and serum albumin concentrations from 222 2nd year medical students. Using a single univariate reference interval for total serum calcium, 17 students were classified as having an abnormal calcemic status while using a bivariate reference model for total serum calcium and serum albumin, 13 of these 17 students had in fact normal total serum calcium concentrations, taking into account their serum albumin concentrations.

Computer program for quick work scheduling of nursing staff.

A new computer program is reported that provides work schedules for nursing staff. Designed to run on the NEC PC-9801 personal computer using the N88-BASIC(86) interpreter, this program is intended for use by the senior nurses of each ward and removes most of the manual burden of staff work scheduling. The word work schedule can be created in only 90 seconds by the program and allows for individual nursing staff requests for particular work assignments.

[Computer program of genetic map construction and database of gene localization].

Computer data base containing information about gene localization in the chromosomes of various organisms is composed. Computer program of genetic map constructing is created. The program represents graphically the information conserving in the data base. The data base and the program are intended for realization on the IBM-compatible computers.

A computer program for the on-line computation of cardiac output from thermodilution curves.

An accurate, rapid calculation of Cardiac Output is of great importance during the treatment of critically ill patients. This paper describes a complete, self-contained mini-computer system, based on a Data General NOVA 1220 which can accurately sample data and compute the results in about 30 sec. The system uses the thermodilution method with a Swan-Ganz catheter to collect data and a weighted least-squares technique to compute the results.

A computer program in BASIC for estimation of ED50 and LD50.

A BASIC computer program for calculating ED50 and LD50 values and their confidence limits by probit analysis and weighted linear regression is presented. It also includes the Chi square procedure for testing the mathematical model and allows on-screen high resolution graphic display. The portable program can easily be modified for use in different models of microcomputers.

Hippin: a semiautomatic computer program for selecting hip prosthesis femoral components.

This work illustrates a computer program designed to aid surgeons in selecting the hip prosthesis femoral component during the preoperation planning stage of hip replacement surgery. Starting from the processing of the patient's coxo-femoral region X-ray image, the program, called Hippin, interacts with the user to outline the femoral region, including the head and the inner contour of the proximal femur. It automatically examines all possible couplings with the patient's femur outlines from a database containing the outlines of the available prostheses created by digitizing the templates normally used in preoperation planning. The resulting images enable the surgeon to visually compare all the alternatives. In addition, the program provides numerical values for the distances between the physiological rotation and prosthesis centers, helping the surgeon in selecting from among the possibilities. The program has been validated by comparing the computer results with actual surgeon selections.

The ability of a computer program based on the Marquette Matrix-12 short measurement matrix to replicate coding by the Minnesota ECG coding laboratory.

The study was undertaken to determine whether a computer program that uses "short measurement matrix" data from the Marquette Matrix-12 system can replicate Minnesota electrocardiogram (ECG) coding laboratory interpretations. An agreement was found between coding of median complex ECGs at the Minnesota ECG coding laboratory and coding based on Marquette Matrix-12 short measurement matrix. The comparison was based on 763 ECGs plus chest pain history and serum enzyme values for a stratified random sample of 141 patients hospitalized in 1990 or 1991 for an event coded as HICDA 410.x (acute myocardial infarction), 411 (other acute and subacute forms of ischemic heart disease), 413 (angina pectoris), or 796.9 (other ill defined and unknown causes of morbidity and mortality). The population was reconstructed from the stratified random sample to enable population-based inferences. Exact agreement between Matrix-12 and Minnesota coding laboratory interpretation on 4 ECG patterns (evolving diagnostic, diagnostic, equivocal, or other ECG pattern) was 74.5% (Kappa = 0.63 +/- 0.05) for the stratified random sample and 78.8% (Kappa = 0.66 +/- 0.05) for the reconstructed population. For coding myocardial infarction based on the ECG, serum enzyme levels, and ischemic chest pain, agreement was 91.5% (Kappa = 0.85 +/- 0.04) for the stratified random sample and 90% (Kappa = 0.83 +/- 0.04) for the reconstructed population. Although ECG interpretation by a computer program based on the short measurement matrix of the Matrix 12 system results in better agreement than prior attempts to replicate the Minnesota coding laboratory, interpretation remains unacceptably discordant.

SOLPRO: theory and computer program for the prediction of SOLution PROperties of rigid macromolecules and bioparticles.

Single-valued hydrodynamic coefficients of a rigid particle can be calculated from existing theories and computer programs for either bead models or ellipsoids. Starting from these coefficients, we review the procedures for the calculation of complex solution properties depending on rotational diffusion, such as the decays of electric birefringence and fluorescence anisotropy. We also describe the calculation of the scattering from factor of bead models. The hydrodynamic coefficients and solution properties can be combined to give universal, shape-dependent functions, which were initially intended for ellipsoidal particles, and are extended here for the most general case. We have implemented all three developments in a new computer program. SOLPRO, for calculation of SOLution PROperties, which can be linked to existing software for bead models or ellipsoids.

A review of small canned computer programs for survey research and demographic analysis.

A variety of small canned computer programs for survey research and demographic analysis appropriate for use in developing countries are reviewed in this article. The programs discussed are SPSS (Statistical Package for the Social Sciences); CENTS, CO-CENTS, CENTS-AID, CENTS-AIE II; MINI-TAB EDIT, FREQUENCIES, TABLES, REGRESSION, CLIENT RECORD, DATES, MULT, LIFE, and PREGNANCY HISTORY; FIVFIV and SINSIN; DCL (Demographic Computer Library); MINI-TAB Population Projection, Functional Population Projection, and Family Planning Target Projection. A description and evaluation for each program of uses, instruction manuals, computer requirements, and procedures for obtaining manuals and programs are provided. Such information is intended to facilitate and encourage the use of the computer by data processors in developing countries.

A computer program for Wiener filtering of evoked potential data.

We have developed a computer program for Wiener filtering of evoked potential data. The basic algorithm involves computation of the difference berween the power spectrum of the sweep sum and the sum of power spectra of individual sweeps. Power spectra are computed by means of the discrete Fourier transform. The program is now being run on a LSI-11 computer in a neurophysiology research laboratory to analyze somatic evoked potential data from monkeys.
