The response of recombinant inbred strains of mice to bacterial lipopolysaccharides.

Fourteen recombinant inbred strains of mice have been produced by the inbreeding of the F2 generation of a cross between C57BL/6J and C3H/HeJ progenitor mice. The responses of these BXH strains to bacterial lipopolysaccharides (LPS) have been characterized. Four BXH strains are high LPS responders and nine strains are low LPS responders. One BXH strain shows intermediate responsiveness which may reflect residual heterozygosity. F1 hybrid mice from low x high responder strains were intermediate in their response to LPS suggesting additive genetic control. The LPS responses in backcross mice from the F1 x low LPS responders showed segregation consistent with LPS responsiveness being determined by a single gene. In 13/14 BXH strains, there was concordant inheritance of LPS responsiveness and the major urinary protein locus Mup-1b. The association of the expression of the Mup-1 alleles with LPS responsiveness in the BXH strains suggests that the defective LPS response gene in C3H/HeJ mice is located on chromosome 4.

Coumarin 7-hydroxylase in inbred strains of mice: comparison with other microsomal monooxygenase activities and induction by pyrazole.

This study was carried out in order to find out the effects of pyrazole on liver drug metabolism in several inbred mice: D2, B6, BALB, and AKR and in the outbred mouse NMRI. Compared to control pyrazole treatment decreased the microsomal cytochrome P-450 content of liver to 60-70% and benzo(a)pyrene hydroxylase and ethylmorphine N-demethylase activities to 40-55% and increased 7-ethoxycoumarin O-deethylase activity to 100-200% in AKR, BALB, B6, and NMRI mice and 300% in D2 mouse. Coumarin 7-hydroxylase was increased only 200-300% in B6, AKR, and BALB mice and as much as 700% in D2 and 900% in NMRI mice compared to control. Coumarin 7-hydroxylase is under different genetic control from other monooxygenase activities and differently expressed in various strains of mice.

Susceptibility differences of inbred strains of mice to blastomycosis.

The susceptibility of inbred strains of mice to pulmonary blastomycosis was studied to derive information relevant to host resistance and genetic background. Initial studies with eight strains with various H-2 backgrounds revealed the C3H/HeJ strain to be highly susceptible and DBA/1J mice to be resistant. These observations were confirmed with various challenge inocula. These differences were not dependent on the size of the challenge, the strain of Blastomyces dermatitidis, host age, or ability of the challenge to penetrate to the lower airways. Differences between the susceptible and resistant strains in lymphocyte proliferation in vitro and delayed-type hypersensitivity in vivo after nonlethal subcutaneous infection were not demonstrated; the susceptible strain made a significantly greater antibody response to blastomyces antigens as determined by an enzyme-linked immunosorbent assay. The resistance of the C3H/HeN strain of mice, which differs from the C3H/HeJ in sensitivity to lipopolysaccharide and lacks the macrophage cytotoxicity defect of the latter, suggests that the susceptibility of C3H/HeJ mice is not related to their C3H background or the H-2 locus. As the A/HeJ strain, which also has a macrophage cytotoxicity defect, was found in this study to be the second most susceptible strain, this also suggests macrophages as the subject for further study with respect to the mechanism of genetic resistance to this infection.

[Sex ratio of inbred strains of mice].

A study of the sex ratio in mice of the inbred strains (CBA and C3H) and connection of the postimplantation embryonic mortality in mice of these strains with the sex distribution of embryos demonstrated that the sex ratio in these mice was 1:1. Literature and the author's personal data suggested that genetic features of mice of the inbred strains failed to influence significantly the sex ratio of the offspring. The postimplantation embryonic death rate in the C3H mice exceeded that in the CBA mice (14.4 and 9.3%, respectively). However, the balanced sex ratio in mice of these strains points to the absence of selective mortality of the embryos of any one sex during embryogenesis.

Genetically determined susceptibility and resistance to diet-induced atherosclerosis in inbred strains of mice.

To determine whether recombinant inbred strains derived from C57BL/6J and A/J mice would provide a good model in which to study the genetics of diet-induced atherosclerosis, male mice of the parent strains were compared in a number of experiments designed to correlate various biochemical changes with susceptibility or resistance to the disease. In both strains fed an atherogenic diet containing 27% coconut oil and 4.5% cholesterol, there was a significant rise in serum very low-density plus low-density lipoprotein cholesterol levels, but only C57BL/6J mice developed discernible fatty lesions in the aortic wall. In A/J mice a significant rise in high-density lipoprotein cholesterol was also observed, which corresponded to the appearance of a second species of high-density lipoprotein in the serum, but in C57BL/6J mice there was a fall. In susceptible C57BL/6J mice, free cholesterol is secreted into bile, which becomes supersaturated, leading to the formation of gallstones. In the resistant strain, however, dietary cholesterol accumulates in the liver. A difference in hepatic cholesterol metabolism between the two strains may thus be a factor in determining their different susceptibilities to diet-induced atherosclerosis.

Identification of sublines of inbred strains of mice and assessment of genetic relationships between substrains or sublines by DNA fingerprinting.

Identification of substrains or sublines of inbred mice and assessment of genetic relationships among them were performed on the basis of DNA fingerprinting using M13 phage DNA as a probe. We used eight C57BL/6 sublines (J parallel Jcl, J parallel Jms, J parallel Slc, J parallel Nrs, J parallel Yok, Jah, N parallel Crj, N parallel Jcl) and eleven C3H/He sublines, (J, J parallel Jcl, J parallel Yok, J parallel Nrs, J parallel Jms, N, N parallel Jcl, N parallel Crj, Slc, Jah, Nrs). Two kinds of restriction endonucleases (HinfI and PstI) were used. It was found that: 1) DNA fingerprint within each subline showed identical patterns. 2) Most sublines of C57BL/6 and C3H/He could be identified using DNA fingerprinting with HinfI except between N parallel Crj and Slc, and among J, J parallel Nrs and J parallel Yok in C3H/He. DNA fingerprints with PstI endonucleases showed low polymorphic banding patterns. 3) A dendrogram constructed from DNA fingerprint patterns reflected generally the genealogy of the sublines used. 4) DNA fingerprinting, therefore, seemed to be suitable for the genetic monitoring and assessment of genetic relationships among sublines of inbred mice having close relationships.

Variations in serum calcium between strains of inbred mice.

Serum calcium concentrations of C3H/Fg, C3H/HeJFg, AKR/Fg, A/Fg, and LCS/Fg mice were compared at 4 and 7 mo of age. In the C3H/Fg and A-strain mice, calcium levels did not differ significantly between the 2 strains nor between the 2 age groups. The C3H/HeJFg, LCS/Fg, and C57BL/Fg strains formed a distinct group with similar calcium levels, but at a significantly higher level than C3H/Fg and A/Fg group. Again there was no significant difference between the 4- and 7-mo groups. The AKR/Fg strain was distinct from both groups in that higher calcium levels characteristic of the second group (C3H/HeJFg, LCS/Fg, and C57BL/Fg, were seen at 4 mo of age, but lower calcium levels similar to those of the C3H/Fg and A/Fg strains were found at 7 mo of age.

[Escape-avoidance learning in two strains of inbred mice when two types of response can be alternatively effective].

Three experiments demonstrated that escape-avoidance learning is closely related to the response type dominantly occurred in mice of an inbred strain. Inescapable shock elicited locomotion (L-typed response) in C57BL/6 mice, and rearing or jumping (R-typed response) in C3H/He (Experiment 1). In the shuttle-box situation where both of these responses can be effective to terminate CS tone and electric shock, C57BL established L-typed avoidance mainly, while C3H tended to learn avoidance by R-typed response (Experiment 2). The two strains learned the escape in the shuttle-box by the same types of response as they showed in avoidance learning (Experiment 3). These findings lead us to conclude that different inbred strains of mice innately react to electric shock by their specific types of response. Strain differences in escape-avoidance learning could thus be attributed to the compatibility of the specific response type with requirement of the task situation.

Hyperprolinemia and prolinuria in a new inbred strain of mice, PRO-Re.

A hyperprolinemia was discovered, in a new inbred strain of mice, which was equivalent to about a sevenfold elevation above the concentration of proline in the blood of either of the original parental lines, or of 12 other inbred strains with diverse genetic constitution. In addition, mice of this PRO/Re strain exhibited a marked prolinuria, whereas the other 14 inbred strains had no proline detectable in their urine.
