Tissue culture studies. VI. The effect of medium constituents on nucleic acids and uptake of p32.

The effect of horse serum alone, and of embryo extract alone, was compared with that of "complete medium" on the content and synthesis of ribo- and desoxyribonucleic acids and uptake of tracer P(32) by chick heart cultures in vitro. The factors mentioned are influenced by embryo extract in a manner similar to the effect in complete medium. Horse serum produced little synthesis of nucleic acids or uptake of tracer, giving only slightly more effect than Tyrode's solution alone. Cutting the tissue into smaller pieces caused considerably greater synthetic effects, and retarded necrosis of the implant.

[Simultaneous measurement of 32P and a 33P double-labelled sample by liquid scintillation counter and its application to phosphorus compounds isolated by paper chromatography].

Simultaneous determination of 32P and 33P radioactivities was carried out with the liquid scintillation counter (Aloca LSC 670). For the quenching correction, the channel ratio method was used, and the proper gain and window level were set to get the maximum performance number or Klein-Eisler value. Using this method the radioactivities of the 32P and 33P double labelled compounds which were separated with paper chromatography were determined. New index, RSA (relative specific activity, i.e. 32P/33P) was proved to be effective as well as specific activity for the studies of the transport and metabolism of phosphorus compounds in the plant roots.

Synthesis of [32P]dolichyl phosphate, utilizing a general procedure for [32P]phosphorus oxychloride preparation.

Equilibration of the phosphorus in radioactive phosphoric acid with the phosphorus in phosphorus oxychloride occurs if these compounds are refluxed together for approximately 24 h. This observation led us to develop a method for the preparation of radioactive phosphorus oxychloride on a small scale with high specific radioactivity. The labeled phosphorus oxychloride may be utilized directly in a one-pot reaction for the preparation of labeled phosphate esters or for the synthesis of more selective phosphorylating agents such as cyanoethyl phosphate. Since the method is very simple and capable of yielding highly labeled radioactive phosphate esters on a small scale, it is applicable to a number of different problems. The preparation of 32P-labeled dolichyl phosphate is described utilizing this procedure.
