Ascorbic acid and copper in linoleate oxidation. I. Measurement of oxidation by ultraviolet spectrophotometry and the thiobarbituric acid test.

The UV absorption method and the thiobarbituric acid (TBA) test for oxidation of an aqueous suspension of linoleate were compared. The absorption method depends on the formation of hydroperoxides having conjugated double bonds that absorb strongly at 233 nm. The absorption at 233 nm increased markedly during oxidation of linoleate catalyzed by either ascorbic acid or cupric ions. The concentration of ascorbic acid in the reaction mixture was also measured by UV absorption at 265 nm and pH 7.0. Color development in the TBA test also increased markedly with the extent of oxidation of linoleate. When ascorbic acid was the catalyst, UV absorption detected early stages of oxidation with greater sensitivity than the TBA test. The reverse was true when Cu(++) was the catalyst. In general, the relation between the two procedures will depend on whether copper is present when the TBA test is made.

No effect of influenza vaccination on theophylline pharmacokinetics as studied by ultraviolet spectrophotometry, HPLC, and EMIT assay methods.

The effect of influenza vaccination on steady-state pharmacokinetics of theophylline was studied in six healthy young adults by comparing pharmacokinetic parameters found on days 4 and 5 during a 5-day course of theophylline alone with those obtained after influenza vaccination on day 4 of a second study phase. Theophylline plasma concentrations were measured by means of high-pressure liquid chromatography (HPLC) analysis and, in part, with a manual ultraviolet spectrophotometric method and with an EMIT assay. On the fourth and fifth days of each of the two periods of drug administration, theophylline plasma concentration-time curves were evaluated, and the following pharmacokinetic parameters were compared: trough plasma concentration (cmin), peak plasma concentration (cmax), time to peak (tmax), and the area under the curve during a dosing interval (AUC). None of these pharmacokinetic parameters of theophylline before and after vaccination were found to be significantly different with any of the analytical methods.

[Rate of controlled-release urea pervasion through membrane determined by ultraviolet spectrophotometry].

Application of controlled-release nitrogenous fertilizers can improve the efficiency of fertilizers and reduce the environmental pollution. Controlled-release urea (coated urea) is one of the controlled-release nitrogenous fertilizers developed quickly in the recent years. The rate of controlled-release urea pervasion through membrane is the most important index of the capacity of controlled release. There is a maximum absorption at lambda=426 nm with complex in acidic solution, using p-dimethylaminozenzaldehyde as color reagent, and the absorbance exhibits a linear reponses to the urea concentration over the range of 7.5-210 microg x mL(-1). The method for determining the rate of controlled-release urea pervasion through membrane was realized through determining the content of urea in the liquor, the recovery efficiency of the method is 96.1%-103.9%.
