[Lipoxygenase inhibitors. IV. Synthesis and cyclization reactions of open-chain N1-aryl-substituted amidrazones].

alpha-Carbonyl carboxylic acid arylhydrazonochlorides obtained by Japp-Klingemann reaction are the starting substances for the synthesis of alpha-carbonyl carboxylic acid arylhydrazonoamides, -esters and -thioesters. The inhibiting activity of these compounds against 15- and 5-lipoxygenase is described. Reactions of derivatives of amidrazones with formaldehyde give triazole, triazoline and unexpected benzotriazepine derivatives.

Kinetic studies on the peroxyoxalate chemiluminescence reaction: determination of the cyclization rate constant.

Although more currently utilized as analytical tool because of its high sensitivity and good reproducibility, the mechanism of the peroxyoxalate system, a chemiluminescence reaction with quantum yields only comparable to bioluminescence systems, has been extensively studied. The light emission mechanism can be divided in the pathway before chemiexcitation, which contains the rate-limiting steps, and the fast and kinetically non-observable chemiexcitation step. In this work, we obtain information on the mechanism of the slow pathways, attribute values to several rate constants prior to chemiexcitation and suggest a mechanistic scheme that could help optimization of conditions when the peroxyoxalate reaction is used as analytical tool.

Cyclization of pyrrhocoricin retains structural elements crucial for the antimicrobial activity of the native peptide.

Pyrrhocoricin is a naturally occurring antimicrobial peptide from the European fire bug Pyrrhocoris apterus. It has submicromolar activity against a range of Gram-negative bacterial strains and has created recent interest as a lead for the development of novel antibiotic compounds. In this study, we have used NMR spectroscopy to determine the solution structures of pyrrhocoricin and a synthetic macrocyclic derivative that has improved in vivo pharmaceutical properties. Native pyrrhocoricin is largely disordered in solution, but there is evidence of a subpopulation with ordered turn regions over residues 2-5, 4-7, and 16-19. The macrocyclic derivative incorporates a nine amino acid linker joining the N- and C-termini, which does not adversely affect the antimicrobial potency but leads to a broader spectrum of activity. The NMR data suggest that the turn conformations in the cyclic derivative are similar to those in the native form, thus implicating them in the biological function.

The formation of cyclized silyl derivatives of beta-hydroxyamines and their analyses by means of gas chromatography mass spectrometry.

A generally applicable silylation method for beta-hydroxylated primary, secondary, tertiary and even quaternary amines is presented. These aminoalcohols form 6-membered heterocycles under the influence of a reagent mixture consisting of 1,3-bis-(chloromethyl)-1,1,3,3-tetramethyldisilazane and chloromethyldimethyl-chlorosilane. Examples of analogue ring closures with a gamma-hydroxyamine and an alpha-amino acid are also given. The formation of the derivatives and their properties, are discussed, mainly from the viewpoint mass spectrometry.

Efficient synthesis of 4-(3'-furanyl)butenolide derivatives via PdII-catalyzed oxidative heterodimeric cyclization reaction of 2,3-allenoic acids and 1,2-allenyl ketones.

The oxidative cyclization/dimerization reaction between two classes of allenes with different functionalities was reported to provide an efficient route to polysubstituted 4-(3'-furanyl)-2(5H)-furanones, which are not readily available from the known methods. The highly optically active butenolides could be easily formed from the optically active 2,3-allenoic acids, which was obtained conveniently through chiral resolution with optically active amines, that is, cinchonidine or alpha-methyl benzylamine. A mechanistic study showed that the reaction proceeded via a matched double oxypalladation-reductive elimination process. The Pd(II) species may be regenerated via the subsequent cyclometallation of two equivalents of 1,2-allenyl ketones with Pd(0) and protonlysis of Pd enolates formed with the in situ generated HCl.

Pd-Cu bimetallic catalyzed domino cyclization of alpha-allenols followed by a coupling reaction: new sequence leading to functionalized spirolactams.

A novel regioselective metal-catalyzed spirocyclization of alpha-allenols-cross coupling (Heck, Sonogashira, and Suzuki) reaction sequence, leading to potentially bioactive spirocyclic lactam derivatives has been developed. Precursors for the tandem spirocyclization-coupling reaction, alpha-allenols 2 a-d were obtained starting from alpha-oxolactams 1 a-c via indium-mediated Barbier-type carbonyl-allenylation reaction in aqueous media by using our previously described methodologies.
