Chiral discrimination in lithium complexes of bis(5H-pyrroles) and bis(oxazolines).

The 1:1 and 2:1 complexes of chiral bis(5H-pyrroles) and bis(oxazolines) with the lithium cation have been studied by means of DFT methods (B3LYP/6-31G and B3LYP/6-311+G). The energetic, geometric, electronic, and orbital properties of the complexes have been analyzed. The chiral discrimination in the 2:1 complexes (homo vs heterochiral ones) indicate that in all the cases the heterochiral complexes are more stable than the homochiral ones, except for the tert-butyl derivatives. The chiral discrimination energies will be discussed on the basis of different parameters related to the lithium atom such as the N-Li distance, the orbital interaction between the lone pair of the nitrogen and an empty orbital of the lithium, and its atomic contribution to the total energy of the complexes.

Determination of absolute stereochemistry and an alternative synthesis of homopumiliotoxin 223G: identification on chiral GC columns with the natural alkaloid.

An alternative asymmetric synthesis of (+)-(lS,9aS)-homopumiliotoxin 223G (1) was accomplished via (1R,2R, 9aS)-1-(benzyloxy)-2-hydroxy-1-methyl-3[(E)-isobutylidene]++ +quinolizidi ne (4), which was synthesized according to the intramolecular nickel(II)/chromium(II)-mediated cyclization of the N-(iodoalkenyl)aldehyde 2. Compound 4 was converted to the acetate and subjected to reduction with lithium in ammonia, whereupon deprotection of the O-benzyl group and removal of the acetoxyl group occurred in a single operation to afford (+)-homopumiliotoxin 223G. The same sequence using (+/-)-4 was applied to the synthesis of racemic 223G. Gas chromatography of a sample of racemic 223G showed no separation into enantiomers on four different cyclodextrin-based chiral GC columns. We found, however, that the O-acetates of (+/-)-223G gave a nearly baseline separation on either a beta-cyclodextrin column or a permethylated beta-cyclodextrin column. The O-acetate of synthetic (+)-223G was identical on either of these two columns, with the first eluting O-acetate from acetylated (+/-)-223G and also with the acetylated 223G present in a frog skin extract, thus allowing us to confirm unambiguously the 1S,9aS absolute configurations of natural 223G.

Synthesis of highly enantioenriched all-carbon quaternary centers: conjugate additions of chiral organolithium nucleophiles to alpha,alpha-dinitrile beta,beta-disubstituted olefins.

[reaction: see text]. Highly enantioenriched quaternary centers are obtained by the reaction of chiral lithiated intermediates complexed to (-)-sparteine with tetrasubstituted, alpha,alpha-dinitrile activated olefins. Lithiated N-Boc-N-Aryl benzylamine furnishes products with drs from 78:22 to 95:5, with ers exceeding 94:6. Lithiated N-Boc-N-Aryl allylamine reactants provide enecarbamate products with drs from 55:45 to 99:1, with ers ranging from 87:13 to 97:3.

Diastereoselective additions of chiral vinylzinc reagents to alpha-chiral aldehydes.

[reaction: see text] Additions of vinylic zinc bromide reagents to alpha-chiral aldehydes (R(1) = CH(2)OTBS, R(2) = Me; R(1) = Me, R(2) = OTBS) in the presence of lithiated (+)- or (-)-N-methylephedrine proceed with predominant reagent control to afford anti or syn adducts stereoselectively, except when the aldehydes possess an alkoxy substituent at the alpha- or beta-positions (R(1) = Me, R(2) = OBn; R(1) = CH(2)OBn, R(2) = Me), in which case chelation-controlled adducts predominate.

Asymmetric total synthesis of sperabillins B and D via lithium amide conjugate addition.

Diastereoselective conjugate addition of homochiral lithium (R)-N-allyl-N-alpha-methylbenzylamide to methyl (2E,5E)-hepatadienoate, followed by protecting group manipulation and subsequent iodocyclocarbamation allows a concise route to the core fragment, methyl (3R,5R,6R)-3,6-diamino-5-hydroxyheptanoate, of sperabillins B and D. Differentiation between the C-3 and C-6 primary amino groups of this core amino acid was readily achieved by treatment with acetone, giving the 5,6-isopropylidene and C-3-imine protected diamine, with subsequent regioselective acylation of the C-6-nitrogen facilitating the total synthesis of sperabillin D in 10.8% overall yield, and the first asymmetric synthesis of sperabillin B in 5.8% overall yield.

Dearomatising rearrangements of lithiated thiophenecarboxamides.

Thiophene-3-carboxamides bearing allyl or benzyl substituents at nitrogen undergo dearomatising cyclisation on treatment with LDA. Rearrangements transform the dearomatised products into pyrrolinones, azepinones or partially saturated azepinothiophenes.

The stereocontrolled total synthesis of altohyrtin A/spongistatin 1: fragment couplings, completion of the synthesis, analogue generation and biological evaluation.

The antimitotic marine macrolide altohyrtin A/spongistatin 1 has been synthesised in a highly convergent and stereocontrolled manner, thus contributing to the replenishment of the largely exhausted material from the initial isolation work. Coupling of the AB- and CD-spiroacetal subunits by a stereoselective aldol reaction was achieved by using either a lithium (67 : 33 dr) or boron enolate (90 : 10 dr). A highly (Z)-selective Wittig coupling was used to unite the northern hemisphere aldehyde with the southern hemisphere phosphonium salt . Deprotection and subsequent regioselective macrolactonisation on a triol seco-acid completed the synthesis of altohyrtin A. Two structural analogues were also prepared and evaluated as growth inhibitory agents against a range of human tumour cell lines, including Taxol-resistant strains, alongside altohyrtin A and paclitaxel (Taxol), revealing that dehydration in the E-ring is tolerated and results in enhanced cytotoxicity (at the low picomolar level), whereas the presence of the full C44-C51 side-chain appears to be crucial for biological activity.

Imidazolin-2-iminato titanium complexes: synthesis, structure and use in ethylene polymerization catalysis.

The Staudinger reaction of the imidazolin-2-ylidenes, 1,3-di-tert-butylimidazolin-2-ylidene (1a), 1,3-diisopropylimidazolin-2-ylidene (1b), 1,3-diisopropyl-4,5-dimethylimidazolin-2-ylidene (1c), 1,3-bis(2,4,6-trimethylphenyl)imidazolin-2-ylidene (1d) and 1,3-bis(2,6-diisopropylphenylimidazolin-2-ylidene (1e), with trimethylsilyl azide furnishes the corresponding N-silylated 2-iminoimidazolines 2a-e, which react with [(eta-C5H5)TiCl3] to afford half-sandwich cyclopentadienyl titanium complexes of the type [CpTi(L)Cl2] (3) (L = imidazolin-2-iminato ligand). Similarly, the reactions of 1,3-di-tert-butyl-2-(trimethylsilylimino)imidazoline (2a) with [(eta-tBuC5H4)TiCl3] results in the formation of [(eta-tBuC5H4)Ti(L)Cl2] (4) (L = 1,3-di-tert-butylimidazolin-2-imide). Bis(1,3-di-tert-butylimidazolin-2-iminato)titanium dichloride (5) is obtained from the reaction of two eq. of 2a with TiCl4. Treatment of 5 with methyllithium results in the formation of the corresponding dimethyl complex [L2Ti(CH3)2] (6), whereas [CpTi(L)(CH3)2] (7) is similarly obtained from 3a. The molecular structures of 3a, 3b, 3c, 3e x C7H8, 4 and 7 are reported revealing linearly coordinated imidazolin-2-iminato ligands together with very short Ti-N bond distances. All dichloro complexes (3a-e, 4 and 5) can be activated with methylaluminoxane (MAO) to give active catalysts for ethylene homopolymerization. In most cases, moderate to high activities are observed together with the formation of high (HMWPE) or even ultra high molecular weight polyethylene (UHMWPE).

Influence of organic cations on basic amino-acid uptake by human placental villi.

Human placental chorionic villi were incubated for 30 min with [3H]lysine or [3H]arginine and the distribution ratios (intracellular:extracellular concentrations) were determined. The ratios remained unchanged when Na+ in Earle's buffered salt solution was replaced with Li+. When Na+ was replaced with choline there was a significant increase is distribution ratios (lysine 1.34 +/- 0.33 v. 3.99 +/- 0.15, arginine 1.95 +/- 0.37 v. 5.05 +/- 1.16). Leucine, a neutral amino acid with a Na(+)-independent transport system, was unaffected by choline substitution. The distribution ratio for alanine, which is Na(+)-dependent, was reduced (2.50 +/- 0.41 v. 1.45 +/- 0.20). Two other quarternary amines, acetyl-beta-methylcholine and tetraethylammonium chloride (TEA) caused similar increases in the distribution ratios of the basic amino acids. Hordenine, a tertiary amine, was less effective and there was little or no effect with ephedrine, a secondary amine. The choline effect was first observable at concentrations of 105 mM. With TEA, there was a progressive increase in distribution ratios beginning at 29 mM. Lysine efflux was measured after incubation of villi with lysine in Earle's buffer or choline buffer. Lysine was rapidly released to the fresh medium with 25% more retained in choline-exposed villi. The amines may cause alterations in the kinetics of basic amino-acid transporters or may modify other aspects of placental physiology permitting an increase retention of the basic amino acids.

Olfactory and gustatory stimuli in food-aversion learning of rats.

Two studies of food-aversion learning in the rat examined some crucial conditions for the potentiation of an odor component by a novel taste. Wistar rats were exposed twice to an odor-taste compound and then were treated either immediately or the next day with lithium chloride, a noxious substance. Immediately poisoned rats acquired an aversion to the odor component when it was paired with a familiar taste. Only in one experimental condition did some weak conditioning occur: when the new odor was presented together with a new taste. Thus, the classical phenomenon of overshadowing of the less salient odor component by the more salient taste was observed in all of our studies. This was true for situations in which the odor was presented near the drinking spout as well as for situations in which the odor was presented at some distance in front of the drinking source (Experiment 1). In addition, the weaker an odor component was, the more easily it was overshadowed by the taste (Experiment 2). All results fit well into classical rules of Pavlovian conditioning and fail to support the assumption that special mechanisms exist in food-related odor conditioning in rats.

Analyses of kinetic solvent isotope effects of a hammerhead ribozyme reaction in NH4+ and Li+ ions.

Hammerhead ribozymes have been considered to be divalent-metalloenzymes. However, this was recently questioned by the finding that the reaction can proceed without any divalent metal ions in the presence of high concentrations of monovalent ions such as NH4+/Li+ ions. Thus, one might think divalent metal ions are not involved in chemical step in the catalytic mechanism. To investigate the involvement of the monovalent ions, we analyzed the deuterium solvent isotope effects in the reactions. Our present analysis indicates a proton transfer(s) occurs only in the NH(4+)-mediated reaction, not in the Li(+)-mediated. Most simple interpretation is that NH4+ works as a general acid catalyst and Li+ as Lewis acid catalyst. This suggests hammerhead ribozymes can change catalyst upon their surrounding conditions.

Shark pit organs: response to chemicals.

Nerve fibers from pit organs and canal neuromasts are distinguished by the nature of their electrophysiological response to mechanical and chemical stimulation. Pit organs respond to touch but have a relatively high threshold compared with canal neuromasts. They respond readily to sodium and potassium chloride solutions, the rate of discharge increasing with the concentration of the solution. Order of effectiveness with 1 molar solutions of monovalent cations is as follows: potassium, rubidium > sodium, ammonium > cesium, lithium. Anions are ineffective. Divalent cations such as calcium and magnesium are inhibitory. Responses to acid, sugar, and quinine are either very slight or inhibitory.

Stimulatory effect of lithium ion on proline transport by whole cells of Escherichia coli.

The effect of monovalent cations on proline transport in whole cells of Escherichia coli K-12 has been examined. Lithium ion added to the uptake medium stimulated proline transport severalfold and K+ and Na+ were slightly effective, whereas Rb+, Cs+, and NH4+ were completely without effect. The stimulatory effect of Li+ on proline transport was not due to an increase in osmolarity of the uptake medium, and d 5 mM p-chloromercuribenzene sulfonic acid completely blocked this effect of Li+ without having any effect on the basal rate of proline transport. The Arrhenius plots for Li+-stimulated transport showed a clear transition point at 35 degrees C in addition to 20 degrees C which was also detectable in the basal transport. Lithium ion stimulated proline transport synergistically in the presence of glucose and succinate as a carbon source. The addition of 2.5 mM KCN or 0.5 mM arsenate did not inhibit this synergistic effect, although the presence of these inhibitors inhibited completely the stimulation of proline transport induced by the addition of carbon source. Carbonylcyanide m-chlorophenylhydrazone and 2,4-dinitrophenol blocked both the basal and Li+-stimulated proline transport. When membrane potential of E. coli cells was measured by the dibenzyldimethylammonium uptake method, the incubation of Li+ with the cells did not affect the preexisting membrane potential. These results suggest that Li+ stimulates proline transport by intact cells of E. coli in a manner somewhat affecting membrane component(s) different from the transport carrier of proline. It is uncertain whether the effect of Li+ is directly involved in the mechanisms of energy coupling of proline transport.
