In vitro culture and in vitro maturation of mouse preantral follicles with recombinant gonadotropins.

OBJECTIVE: To develop an effective method for in vitro maturation of preantral follicles isolated from mice ovarian tissue. DESIGN: Isolated preantral follicles were randomly allocated to designed experimental groups for study. SETTING: University-based research lab. PATIENT(S): Healthy, normal mice. INTERVENTION(S): Superovulation with pregnant mare serum gonadotropin and hCG. MAIN OUTCOME MEASURE(S): Morphological changes and E(2) production were assessed. RESULT(S): To obtain competent oocytes, preantral follicles must be cultured with medium containing insulin and recombinant gonadotropins (i.e., recombinant FSH and recombinant LH), with a change of medium daily. A high initial recombinant LH or recombinant FSH facilitates E(2) secretion, enhances granulosa cell outgrowth, and has earlier antral formation. However, prolonged culture in high-recombinant LH or recombinant FSH triggers early differentiation and luteinization of granulosa cells, which results in low metaphase II oocyte and blastocyst formation. CONCLUSION(S): We have developed a culture system that allows the successful maturation of preantral follicles in vitro. The matured follicles are a physiologically functional unit that not only secrete E(2) but also generate competent oocytes. In a special condition, 90% of the cultured follicles survived, 53.5% of them produced MII oocytes, and 50% of the derived MII oocytes were fertilized and reached the blastocyst stage after culture in vitro.

Dexamethasone effects on group B streptococcal infection in newborn rats.

BACKGROUND: We previously published that human neutrophil-mediated bacterial killing of group B Streptococcus (GBS) in vitro was dependent on the timing and concentration of dexamethasone exposure. HYPOTHESIS: Dexamethasone treatment would affect neutrophil mediated killing of GBS in an animal model. METHODS: Wistar rat pups were randomly allocated to receive placebo or dexamethasone before, early or late after GBS infection. Suckling rats were infected with 104 or 105 colony-forming units of GBS or nothing. Pups were followed for survival, quantitative bacteremia, growth and neutrophil-mediated bacterial killing. Neutrophils for bacterial killing were obtained via cardiac puncture before infection. Statistics included chi square for survival, Mann-Whitney U test for bacteremia, analysis of variance for growth and paired Student's t test for bacterial killing analyses. RESULTS: Dexamethasone treatment before invasive GBS infection decreases quantitative bacteremia, improves survival and improves neonatal neutrophil-mediated bacterial killing in suckling rats, whereas dexamethasone treatment after infection increases bacteremia and decreases survival. Regardless of timing of dexamethasone treatment, before or after invasive GBS infection, growth was significantly impaired in all suckling rats receiving dexamethasone compared with controls. CONCLUSION: Treatment with dexamethasone before invasive GBS infection improves survival and decreases bacteremia in suckling rats; this appears in part to be mediated by improved neonatal neutrophil-mediated bacterial killing. We speculate that this improvement in outcome may be a result of improved number or function of neutrophil cell surface receptors.

Effects of Taiwanese yam (Dioscorea japonica Thunb var. pseudojaponica Yamamoto) on upper gut function and lipid metabolism in Balb/c mice.

OBJECTIVE: This study investigated the effects of a Taiwanese yam, Dioscorea japonica Thunb var. pseudojaponica Yamamoto, on upper gut function and lipid metabolism in adult Balb/c mice. METHODS: Mice were randomly allocated to consume the control, 25%, or 50% yam diet in which yam in an uncooked lyophilized form was incorporated into the diet for 21 d. RESULTS: Growth rates were similar among groups, even though the apparent protein absorption rate was decreased by the 50% yam diet. Both yam diets decreased gastric villous width but did not significantly modulate other morphologic and proliferative indices. Brush-border leucine aminopeptidase activities in the small intestine were increased approximately 30% by the 25% and 50% yam diets, respectively. In contrast, sucrase activity was decreased 40% by the 25% yam diet and 50% by the 50% yam diet. The 50% yam diet consistently improved the cholesterol profile in the plasma and liver, whereas the 25% yam diet reduced only the level of low-density lipoprotein cholesterol in plasma. Changes in blood lipid levels were associated with reduced fat absorption. CONCLUSION: A 25% uncooked yam diet may benefit upper gut function and prevent hypercholesterolemia in humans, but the 50% yam diet negatively affected protein absorption.

Group comparison of serum ethinyl estradiol, SHBG and CBG levels in 83 women using two low-dose combination oral contraceptives for three months.

Serum ethinyl estradiol (EE2), sex hormone-binding globulin (SHBG) and corticosteroid-binding globulin (CBG) concentrations were studied in healthy young women randomly allocated to one of two low-dose combination oral contraceptives containing 30 micrograms EE2 and either 75 micrograms gestodene (F) or 150 micrograms desogestrel (M) per unit. There was either no (formerly non-pill users) or one (pill users) wash-out cycle before the study started with a pill-free pretreatment cycle in which the hormone status and basal SHBG and CBG levels were measured. Treatment was for three months. During treatment cycles 1 and 3, there were three test days each. Seven serum samples were obtained up to four hours and one sample 24 hours after intake of the first, tenth and the last (21st) pill. Additional samples were taken prior to morning ingestion of pills 5 and 15. For each individual and each test day, a representative serum pool has been constructed for SHBG and CBG analysis. EE2 concentrations were analyzed in all individual samples by means of a specific and sensitive RIA using anti-EE2-6 beta-CMO-BSA antiserum. Area under the curves (AUC) up to 4 and 24 hours, Cmax and tmax were evaluated and compared between the two treatment groups (n = 40 for F, n = 43 for M). SHBG and CBG concentrations were measured using commercially available immunoassay kits. Groups were large enough to detect a difference in group means of 75% of one standard deviation (alpha = 0.05, 1-beta = 0.9) of target variables, which is equivalent to 28 pg EE2/ml for Cmax, 69 pg.h.ml-1 for AUCEE2 0-4h, 257 pg.h.ml-1 for AUCEE2 0-24h, 39 nmol/l SHBG and 13.4 micrograms CBG/ml. Results clearly demonstrate that there were no differences between the two treatment groups in any of the target variables at any of the six test days distributed over a three-month period. Mean SHBG and CBG pretreatment levels of about 70 nmol/l and 37 micrograms/ml, respectively, increased to about 210 nmol/l and 88 micrograms/ml during the first treatment cycle and to about 230 nmol/l and 93 micrograms/ml during the third treatment cycle. Whereas the time of maximum EE2 serum levels did not differ significantly between test days, Cmax, AUCEE2 0-4h and AUCEE2 0-24h values increased by 30-35% or 40-50%, respectively, when test days 10 and 21 were compared to test day 1. Similar results were found for the third treatment cycle.(ABSTRACT TRUNCATED AT 400 WORDS)

Short-term progestin treatments prevent estrous induction by a GnRH agonist implant in anestrous bitches.

The objective of this study was to test the efficacy and safety of a short-term progestin treatment administered at two different times to prevent estrous induction in response to the administration of an implant releasing the GnRH agonist, deslorelin acetate (DA), in anestrous bitches. Interestrous intervals (IEI) observed prior to and post DA were compared. Forty-two anestrous bitches, with previous IEI history, were randomly allocated to one of the following treatments: PL: placebo sc (n = 12); MA: megestrol acetate 2mg/kg po for 8 days (n = 4); DA: 10mg sc (n = 8); MA&DA-1: MA beginning the day before DA (n = 8); and MA&DA-4: MA beginning 4 days before DA (n = 10). The dose of MA was identical for each treatment. All bitches were examined daily for 1 month and then every 3 months until the next spontaneous post-treatment estrous cycle. Post-GnRH estrous response occurred in 0, 0, 100, 50, and 10% of the PL, MA, DA, MA&DA-1, MA&DA-4, groups, respectively (<0.01). There was an interaction between the treatment and period for the duration of the IEI (< 0.01). Changes in IEI were different among treatments (p<0.01); the three DA-treated groups (147.5% +/- 10.3, 161.3% +/- 14.1, 148.6% +/- 19.2) differed from both the MA (12.9% +/- 17.6) and PL (8.1% +/- 7.8), but not among themselves. It is concluded that an 8 days megestrol protocol and DA on Day 4 was better than DA on Day 1 to prevent estrous response in anestrous bitches and that both protocols significantly increased the IEI.

Uptake of 5-fluorouracil (5-FU) in peritoneal metastases in relation to the route of drug administration and tumour debulking surgery. an autoradiographic study in the rat.

Patients with peritoneal metastases from colorectal cancer have a poor prognosis. Aggressive treatment by debulking surgery and intraperitoneal (i.p.) chemotherapy has been suggested as an alternative therapy. However, the drug penetrance into the tumour in relation to the administration route and surgical reduction of the tumour is not well known. We compared locoregional administration with intravenous (i.v.) injection. Thirty-four in-bred rats with peritoneal metastases were randomly allocated into eight groups and injected with 14C-labelled 5-fluorouracil (5-FU) either through the i.v. or i.p. route, with or without a preceding tumour debulking, and were sacrificed after 2 or 8 h. Tumour radioactivity was visualised by autoradiography and quantified by a computer-based image analysis. After 8 h, 19 debulked and i.p.-injected tumours had a higher drug uptake, 63.2+/-28 (mean+/-standard deviation (SD)) kBq/g than 62 native i.p.-injected tumours (32.8+/-14) or 22 debulked and i.v.-injected tumours (18.5+/-18, P=0.002). After 8 h, 9 small tumours ( /=median 571 pixels), 16 debulked and i.p.-injected tumours had a higher radioactivity (drug uptake) (150.7+/-63) at 2 h than 49 i.p.-injected native tumours (48.5+/-59) or 11 reduced and i.v.-injected tumours (19.9+/-13, P=0.03). At 8 h, 10 debulked and i.p.-injected tumours had a higher drug uptake (50.3+/-24) than 33 native and i.p.-injected (30.8+/-10) or 14 debulked and i.v.-injected tumours (16.0+/-19, P=0.001). These results indicate that a debulking procedure and locoregional treatment of peritoneal metastases is associated with an increased level of 5-FU in the tumours.

A novel 32P deployable balloon system inhibits formation of neointima in porcine coronary arteries after balloon-overstretching injury.

BACKGROUND: Intracoronary radiation has shown the potential to reduce formation of neointima in porcine models of restenosis. OBJECTIVE: To investigate the feasibility, safety, and efficacy of a new 'deployable-balloon' device with radioisotope 32P integrated into the balloon material. METHODS: Ten swine were subjected to balloon-overstretching injury in 20 coronary arteries and were randomly allocated to receive a radiation dose of 0, 15, or 20Gy prescribed to 1 mm from the surface of the radioactive balloon material. The animals were killed 4 weeks after the procedure. Their coronaries were perfusion fixed and stained. Vessel parameters (area of intima and length of fracture) and area of thrombus area were analyzed by computer-aided histomorphometry. RESULTS: Radiation treatment with the new 32P balloon device was performed without complications. The lengts of fracture for the three groups were similar (NS). Formation of neointima after balloon injury was less in members of the radiated groups than it was in controls (area of intima/length of fracture was 0.70 +/-0.12 mm for controls, 0.08+/-0.13mm with 15 Gy, and 0.07+/-0.17 mm with 20 Gy; P < 0.001). Vessels treated with 20 Gy had a greater total area of thrombus (0.00+/-0.00 versus 0.51+/-0.98 mm2, P< 0.01). CONCLUSIONS: Intracoronary radiation therapy using a new 32P deployable-balloon system is feasible and safe. A radiation dose of 15 Gy was sufficient to inhibit neointimal response in the porcine coronary-balloon-injury model.

Evaluation of sulbactam plus ampicillin for treatment of experimentally induced Klebsiella pneumoniae lung infection in foals.

Efficacy of sulbactam, a beta-lactamase inhibitor, in combination with ampicillin, was evaluated for treatment of experimentally induced pneumonia caused by beta-lactam-resistant Klebsiella pneumoniae. Infection was experimentally induced in 18 healthy weanling foals that were randomly allocated to 3 treatment groups: sulbactam plus ampicillin (S/A, 3.3 and 6.6 mg/kg of body weight, respectively), ampicillin (6.6 mg/kg), or vehicle only. Foals were treated daily for 7 days; the observer was unaware of treatment status. Compared with ampicillin and vehicle, treatment with S/A resulted in a statistically significant (P less than 0.05) decrease in severity of pneumonia, with regard to bronchoalveolar lavage cytologic findings (decreased total cell and neutrophil numbers, and increased lymphocyte numbers) and extent of macroscopic lesions in lung tissue of the noninoculated regions. Marked trends toward improvement of S/A-treated foals were observed for quantitative results of bacteriologic culture of bronchoalveolar lavage fluid samples (P less than 0.07), macroscopic pathologic features of the whole lung (P less than 0.1), and histopathologic variables (P less than 0.07), compared with ampicillin- and vehicle-treated foals. Treatment effects were not observed for radiographic, hematologic, and blood gas abnormalities that resulted from infection. In conclusion, the combination of sulbactam plus ampicillin was found to have synergistic effects in vivo, to reduce the extent and severity of experimentally induced gram-negative lung infection in foals.

Inhaled and intravenous corticosteroids both attenuate chlorine gas-induced lung injury in pigs.

BACKGROUND: The accidental release of chlorine gas is a constant threat in urban areas. The purpose of this randomized, blinded, controlled experiment was to examine the effects of post-injury administration of inhaled or intravenous corticosteroid in chlorine gas-injured pigs followed for 23 h. METHODS: Anaesthetized, ventilated pigs (n = 24) in the prone position were exposed to chlorine gas (400 parts per million in air) (1160 mg/m3) for 15 min, then randomly allocated to receive inhaled budesonide (BUD) and intravenous placebo, intravenous betamethasone (BETA) and inhaled placebo or inhaled and intravenous placebo. Haemodynamics, gas exchange and lung mechanics were evaluated for 23 h after exposure to chlorine gas. RESULTS: Airway and pulmonary artery pressures increased and arterial oxygenation fell sharply (from 13.5 +/- 0.8 to 6.7 +/- 0.9 kPa, P < 0.001) after chlorine gas exposure. These immediate changes were followed by a gradual improvement over 5-7 h to a stable level of dysfunction for the rest of the experiment in placebo animals. Arterial oxygen tension, pulmonary vascular resistance and airway pressure recovered faster and more completely in the budesonide and betamethasone groups than in the placebo group (P < 0.01). Lung wet weight to dry weight ratios were greater in the placebo group than in the budesonide and betamethasone groups (6.34 +/- 0.59 vs. 5.56 +/- 0.38 and 5.53 +/- 0.54, respectively, P < 0.05). There was a trend towards lower histological injury scores compared with placebo in animals that received budesonide (P = 0.05) or betamethasone (P = 0.07). CONCLUSION: Treatment of chlorine gas lung injury with nebulized budesonide or intravenous betamethasone had similar positive effects on recovery of lung function.

Effect of intraruminal propionic acid infusion on metabolism of mesenteric- and portal-drained viscera in growing steers fed a forage diet: I. Volatile fatty acids, glucose, and lactate.

This experiment investigated the effect of intraruminal infusion of propionic acid on ruminal VFA metabolism and the absorption of nutrients by the mesenteric- and portal-drained viscera of seven Friesian steers, average BW 127 kg, fed a dried grass-pellet diet. Each received by random allocation 0 (control), .5, or 1.0 mol of propionic acid/d for 7 d. Ruminal acetate and propionate irreversible loss rates and carbon exchange between VFA and CO2 were measured during continuous intraruminal infusions of 2-14C-acetic acid and 2-14C-propionic acid. Ruminal acetate irreversible loss rate was not affected by propionic acid infusion (overall mean 8.09, error mean square [EMS] 2.68 mol/d), whereas propionate irreversible loss increased incrementally with PA supply (3.22 vs 4.16, EMS .61 mol/d, for control and 1.0 mol of propionic acid/d, respectively, P = .09). Glucose irreversible loss rate was increased at the highest level of PA infusion (2.84, 2.83, and 3.22, EMS .06 mol/d, for control, .5, and 1.0 mol of propionic acid/d, respectively; P = .02 for control vs .5 + 1.0), although the proportion of glucose irreversible loss derived from propionate remained constant (.6). Net absorption into venous blood showed that propionate was extensively metabolized in the rumen wall and that the tissues of the small intestine utilized acetate. Utilization of glucose was reduced in portal tissues as a result of intraruminal infusion, and the data were used to derive a model of glucose and lactate interrelationships in gut tissues.

Effect of copper supplementation on the evidence of developmental orthopaedic disease in pasture-fed New Zealand Thoroughbreds.

The effect of copper status on the evidence of bone and cartilage lesions was investigated in 21 Thoroughbred foals. The foals and their dams were grazed on pasture containing 4.4-8.6 mg Cu/kg dry matter (DM). Four treatment groups were created by randomly allocating mares and their foals to either copper supplemented (0.5 mg Cu/kg liveweight (LW)/day), or control (pasture only) groups. This experimental design allowed the effect of copper supplementation of mare and foal to be examined independently. Parameters of bone and cartilage development were assessed in the foals both in vivo, and at post mortem at approximately age 150 days. Mare copper supplementation significantly (P<0.01) decreased radiographic indices of physitis in the distal third metatarsal bone of the foals at 150 days, and the prevalence of articular cartilage lesions (P<0.05). Articular cartilage lesions were minor in all foals, with no evidence of clinical developmental orthopaedic disease (DOD) in vivo, with the exception of minor radiographic changes assessed at post mortem. Copper supplementation of the foal had no effect on any of the bone and cartilage parameters. Copper supplementation of the mares did not abolish DOD in the growing foals, emphasising the probable multifactorial nature of this condition. However, mare supplementation may be a useful treatment regime on a farm where the incidence and severity of DOD are of concern.

Effect of intraruminal propionic acid infusion on metabolism of mesenteric- and portal-drained viscera in growing steers fed a forage diet: II. Ammonia, urea, amino acids, and peptides.

This experiment investigated the effect of intraruminal infusion of propionic acid on the net flux of nitrogenous compounds across the mesenteric- (MDV) and portal-(PDV) drained viscera of seven Friesian steers, average BW 127 kg (SEM 4.6), fed a grass-pellet diet. Each received by random allocation 0 (control), .5, or 1.0 mol of propionic acid/d for 7 d. Blood flow in mesenteric and portal veins was determined by downstream dilution of p-aminohippuric acid in order to determine net appearance rates across the gastrointestinal tissues. Net urea and ammonia flux was unaffected by propionic acid supply. Circulating plasma free amino acid concentrations were increased (P < .05) by propionic acid infusion (2,235, 2,428, and 2,427, error mean square [EMS] 44,370 microM, for control, .5, and 1.0 mol of propionic acid/d, respectively). Net amino acid flux rates were increased at the highest rate of propionic acid infusion across MDV and PDV (4.66, 3.69, and 6.11, EMS 2.98 mol/d for MDV [P < .05] and 2.98, 2.45, and 3.73, EMS 1.69 mol/d for PDV [P < .10] for control, .5, and 1.0 mol of propionic acid/d respectively). Positive venous-arterio concentration differences for peptide-bound amino acids (PBAA) across the MDV and PDV indicated net appearance across the gastrointestinal tissues, but this was not affected by propionic acid infusion. The data show that amino acid flux across postruminal tissues can be influenced by ruminal propionic acid supply and that this does not affect PBAA appearance.

Perivascular axillary block IV: blockade following 40, 50 or 60 ml of mepivacaine 1% with adrenaline.

Perivascular axillary blockade was performed on 90 patients with the aid of a catheter technique. The patients were randomly allocated to receive either 40, 50 or 60 ml of 1% mepivacaine with adrenaline 1:200,000. Blood concentrations of mepivacaine were measured up to 90 min after injection in seven, eight and ten of the patients from the three groups. Sensory and motor blockade was evaluated 20, 30 and 40 min after injection. All groups showed the same temporal development of the blockade, i.e. improval of the blockade during the period from 20 to 40 min after injection, but no difference was found in the sensory or motor blockade between the three groups. However, a further analysis of the incomplete blockades showed a better quality of the sensory blockade in the groups given 50 and 60 ml than in the group given 40 ml. None of the 90 patients showed any signs of systemic toxic reactions. The mean peak values of blood concentrations were 0.5-1.0 microgram/ml higher in the groups given 50 ml and 60 ml than in the group given 40 ml. On the basis of the present and two previous investigations on the dose response in perivascular axillary blockade, a dose of 50 ml 1% mepivacaine with adrenaline or another equivalent drug with vasoconstrictor is recommended.

[Ischemic preconditioning inhibits ischemia/reperfusion induced myocardial apoptosis in rabbits in vivo].

OBJECTIVE: To investigate the effects of ischemic preconditioning on myocardial apoptosis induced by ischemia/reperfusion. METHODS: Twenty four rabbits were randomly allocated to three groups(n = 8), pseudo-operation group(group P), ischemia/reperfusion group(group IR), and ischemic preconditioning group (group IP). Group IR and group IP were subjected to three hours of left anterior descending coronary artery occlusion followed by three hours of reperfusion. Ischemic preconditioning was achieved by three 5-minute cycles of ischemia, each followed by 5 minutes of reperfusion. Infarct size and area at risk were defined by dual staining with triphenyltetrazolium chloride and Evans blue dye. DNA laddering in the border zone myocardium of ischemic area at risk was revealed with agarose gel electrophoresis, and apoptosis index(AI) was obtained with flow cytometry. RESULTS: Infarct size, expressed as a percentage of the area at risk(IS:AR), was(60.8 +/- 10.8)% in group IR. Ischemic preconditioning reduced myocardial infarct size significantly to (33.1 +/- 4.9)% (P < 0.05). AI was (13.83 +/- 3.98)% in group IR, and were reduced significantly to (5.85 +/- 1.59)% in group IP(P < 0.01). DNA laddering indicative of fragmented DNA was clearly demonstrated in myocardial specimens sampled from the lateral border zones of the ischemic area at risk in group IR but were attenuated in group IP. CONCLUSION: It suggests that ischemic preconditioning inhibits the rabbit myocardial apoptosis induced by ischemia/reperfusion in vivo.

A comparison of intermittent vs. continuous and of adriamycin vs. methotrexate 5-drug chemotherapy for advanced breast cancer. A Cancer and Leukemia Group B study.

The therapeutic effectiveness of intermittent vs. continuous combination chemotherapy and of the substitution of adriamycin for methotrexate in a 5-drug regimen was evaluated in women with metastatic breast carcinoma. Patients were randomly allocated to receive continuous therapy with cyclophosphamide, methotrexate, 5-fluorouracil, vincristine, prednisone ( CMFVP -C, 86 patients), intermittent CMFVP ( CMFVP -I, 109 patients), or intermittent CAFVP (107 patients). The CR + PR rate with CAFVP (71%) was superior to CMFVP -C (50%, p = 0.003) and to CMFVP -I (50%, p = 0.002). The remission duration with CAFVP (14 months, median) was superior to CMFVP -I (7 months) (p less than 0.01), and tended to be superior to CMFVP -C (9 months) (p = 0.07). There was a survival advantage of CAFVP (19 months, median) over CMFVP -I (13 months) (p = 0.01), but not over CMFVP -C (16 months) (p = 0.24). Among CR + PR patients, the survival with CAFVP (29 months, median) was superior (p = 0.02) to both CMFVP -I (18 months) and CMFVP -C (21 months). The CMFVP -C regimen was associated with the highest incidence of leukopenia and neurologic toxicity, but the lowest incidence of GI toxicity. The results indicate that the CAFVP regimen is well tolerated and is superior to the CMFVP regimens.

The diversity of Escherichia coli serotypes and biotypes in cattle faeces.

AIM: To study the diversity of commensal Escherichia coli populations shed in faeces of cattle fed on different diets. METHODS AND RESULTS: Thirty Brahman-cross steers were initially fed a high grain (80%) diet and then randomly allocated into three dietary treatment groups, fed 80% grain, roughage, or roughage + 50% molasses. Up to eight different E. coli isolates were selected from primary isolation plates of faecal samples from each animal. Fifty-two distinct serotypes, including nine different VTEC strains, were identified from a total of 474 E. coli isolates. Cattle fed a roughage + molasses diet had greater serotype diversity (30 serotypes identified) than cattle fed roughage or grain (21 and 17 serotypes identified respectively). Cluster analysis showed that serotypes isolated from cattle fed roughage and roughage + molasses diets were more closely associated than serotypes isolated from cattle fed grain. Resistance to one or more of 11 antimicrobial agents was detected among isolates from 20 different serotypes. Whilst only 2.3% of E. coli isolates produced enterohaemolysin, 25% were found to produce alpha-haemolysin. CONCLUSIONS: Diverse non-VTEC populations of E. coli serotypes are shed in cattle faeces and diet may affect population diversity. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides new information on the serotype diversity and phenotypic traits of predominant E. coli populations in cattle faeces, which could be sources of environmental contamination.

Effects of caponization and different exogenous androgen on the bone characteristics of male chickens.

The effects of caponization and androgen implantation on the bone characteristics of male chickens were evaluated. Healthy Single Comb White Leghorn cockerels were caponized or sham operated (sham) at 12 wk old. Sixteen birds from each group were selected for a 14-wk experiment in trial 1. Sixteen birds from the sham group and 64 from the caponized group (randomly allocated into 4 treatments) were implanted with 10.4 +/- 0.4 mg (1.62-mm i.d., 3.6-mm o.d.) of cholesterol, testosterone (TES), 5alpha-dihydrotestosterone (5alpha-DHT), or 19-nortestosterone (19-NorT) and were assigned to trial 2 for a 14-wk experiment. The results from trial 1 showed that caponization increased BW (P < 0.05) and decreased tibia stress, ash content, and P content with higher blood P concentration (P < 0.05) as compared with the sham group. In trial 2, the cholesterol implantation group showed the lowest tibia breaking strength, bending moment, stress, and ash content (P < 0.05). The 19-NorT implantation group showed decreased (P < 0.05) blood Ca and P concentration but increased tibia ash and P content, reaching the same level as the sham group (P > 0.05). The adverse effects of caponization on bone characteristics could be improved using androgen implantation. Among the implantation groups, the 19-NorT implantation group showed the best improvement in tibia breaking strength and bending moment, followed by the TES and 5alpha-DHT groups. The TES group showed the best improvement in tibia stress, followed by the 19-NorT and 5alpha-DHT groups.

The effects of active immunization against gnRH on testicular development, feedlot performance, and carcass characteristics of beef bulls.

The objective was to determine the effects of a recombinant fusion protein anti-GnRH vaccine on testicular development, feedlot performance, and carcass quality of beef bulls. Crossbred beef bulls (n = 58, average weight 306 kg, 9 mo of age), were randomly allocated to two groups and received either an anti-GnRH vaccine (GnRH) or placebo (Control) by intramuscular injection on d 0, 56, and 112. There were group effects (P < 0.01; as a percentage of Control) on testicular weight (53%), daily sperm production (40%), and epididymal sperm reserves (16%). There were group x time interactions (P < 0.0001) for scrotal circumference and serum testosterone concentrations; at slaughter, bulls in the GnRH group had a smaller (P < 0.05) scrotal circumference (28.3 vs 33.9 cm) and lower (P < 0.05) serum testosterone concentrations (2.2 vs 8.6 ng/mL) than those in the Control group. Average daily gain, feed intake, and feed efficiency were not different between treatments during the backgrounding phase (d 0 to 84). During the finishing phase (d 98 to 182), ADG was greater (P < 0.05) for bulls in the Control group (1.69 vs 1.42 kg/d), as was carcass weight (6.9%; P < 0.01). However, GnRH bulls had numerically better feed efficiency (6.12 vs 7.08 kg DMI/kg gain; P < 0.23) and shear force values for ribeye that were 16% lower (P < 0.14) than Control bulls, warranting further investigation. Vaccinating bulls against GnRH suppressed testicular function, with growth and carcass characteristics similar to that expected with steers.
