Effects of neonatal and late unilateral enucleation on optokinetic responses and optic nerve projections in the rabbit.

Rabbits were unilaterally enucleated at the age of 0 or 21 days or at adult age. After survival times of 6-21 months optokinetic nystagmus (OKN) was measured and retinofugal connections were traced with anterograde transport of horseradish peroxidase or 3H leucine, injected into the eye. Non-enucleated animals served as controls. The asymmetry of monocular OKN in normal rabbits, characterized by a strong preference for pursuit of motion in the nasal (anterior) direction, was only slightly alleviated after enucleation. Responses to stimulation in the nasal direction were unchanged; responses to stimulation in the temporal direction showed modest improvements especially after enucleation at adult age and to a smaller degree after enucleation at 0 or 21 days. Redistribution of retinofugal fibers from the eye remaining after enucleation was very limited. Contralateral connections, including those to the lateral geniculate nucleus, showed a normal distribution. Of the ipsilateral connections, those to the lateral geniculate nucleus were normal in extent and density, while those to the superior colliculus were enhanced, in agreement with previous workers (Chow et al. 1973, 1981). Changes in ipsilateral pretectal projections were extremely small; particularly no connections to the nucleus of the optic tract were developed in any of the normal or enucleated animals. Of the accessory optic nuclei, the medial terminal nucleus received a very small ipsilateral projection in normal rabbits, which was markedly enhanced after enucleation especially at 0 and 21 days, but even at adult age. It is concluded that functional and anatomical plasticity of OKN circuits in the rabbit is very limited from the time of birth.

Upper extremity function in spina bifida.

Poor upper extremity function is often recorded in meningomyelocele patients. Only 2 of the 25 patients we assessed, 5 to 19 years old, showed normal upper extremity function in the clinical neurological examination and a timed hand function test simulating daily activities. Slow performance with unsystematic variability was typical. Poor hand function correlated strongly with hydrocephalus. A trend towards better performance with increasing age may indicate that the difficulties are overcome in some patients. While patients without hydrocephalus showed a near-normal distribution in the seven subtests, patients with hydrocephalus needed more time than normal children. Patients with shunt-treated hydrocephalus did not cope as well as patients without a shunt. Mean age in the three groups differed and may partly explain the differences.

Multivariate analysis of trace metal levels in tannery effluents in relation to soil and water: a case study from Peshawar, Pakistan.

Tannery effluents and relevant ground water and soil samples collected from various tanning industries of Peshawar were analyzed for Na, Ca, K, Mg, Fe, Mn, Cr, Co, Cd, Ni, Pb and Zn by the AAS method. The metal concentration data for the three media are reported in terms of basic statistical parameters, metal-to-metal correlations and linear regression analyses. Metal distributions in the three media were quite divergent and showed non-normal distributions with high standard deviation and skewness parameters. Sodium exhibited the highest mean levels of 1,277mg/L, 881mg/L and 12,912mg/kg in the effluent, ground water and soil samples, respectively. Among other metals, Cr concentrations were 410mg/L, 0.145mg/L, 100mg/kg and Ca, 278mg/L, 64.8mg/L, and 2,285mg/kg in the effluent, ground water and soil samples, respectively. Some significant correlations were observed between effluent and soils in terms of Na, Cr, Ni, Co and Pb. The ground water-soil interrelationship suggested that Na levels in the soil and ground water were significantly correlated with each other (r=0.486, P<0.01). Similarly, Cr in the soil is strongly correlated with Ca in ground water (r=0.486, P<0.01). These results were duly supported by the linear regression analysis of data. The source identification studies conducted using Principal Component Analysis (PCA) and Cluster Analysis (CA) evidenced that ground water and soil were being contaminated by the toxic metals emanating from the tannery effluents.

The distribution of the tail moments in single cell gel electrophoresis (comet assay) obeys a chi-square (chi2) not a gaussian distribution.

The parameter tail moment in single cell gel electrophoresis (comet assay) is calculated as the product of the two values: the percentage of DNA in the comet tail and the tail length in microm. Experiments were performed with cultured mammalian cells: B-Lymphoblasts, epithelial cells of a kidney tissue and a plate-epithelial cell line of a human carcinoma. They were irradiated in suspension with UV A at lambda = 343 nm, generated by an excimer laser-pumped dye laser. DNA migration was assessed and analysed. It is demonstrated that the distribution of the tail moments can be fitted by a chi2 (chi-square) distribution, whereas the factors of the product tail moment tend to be normally distributed. From this result, consequences for the statistical evaluation of the results can arise, especially for the computation of the confidence limits and for the valuation of the parameter tail moment from other comet assay experiments.

High order wave aberration of eyes.

This paper is concerned with the means and distribution of the wavefront variances and their root mean squares (RMS) of normal eyes, as measured by various techniques and computed for various pupil sizes. Using data from a subjective crossed cylinder aberroscope [Howland and Howland J. Opt. Soc. Am. A 67(1977)1508] it was found that the logarithms of the wavefront variances are approximately normally distributed. A comparison of data from this subjective study with that of six other studies using a variety of techniques of wavefront measurement showed that the relationship between RMS wavefront deviations of high order aberrations (third order polynomial terms and above) obeyed the relationship: log (RMS) (microm) = -1.918 +/- 0.048 SE + 3.023 +/- 0.111 x log (pupil radius) (mm), with R2 = 0.971.

Relaxation training as complementary therapy for mild hypertension control and the implications of evidence-based medicine.

Relaxation therapy for the treatment of hypertension presented a number of challenges, in terms of conducting and using research as well as gaining the wider acceptance of this complementary therapy in common practice in Hong Kong. Two issues were central to the current implementation of complementary therapies: the evidence-based practice movement and the management of risk during the implementation of new therapies. A major concern was how to maximize the chance of inclusion of research on complementary therapies in evidence-based practice systematic reviews. This was thought to be achievable by making the data amenable to meta-analysis when small samples may be unavoidable. The empirical work examined the effects of three relaxation therapies for the reduction of high blood pressure in nine Chinese subjects. Subjects were randomly assigned to three groups: (a) progressive muscle relaxation, (b) stretch release relaxation and (c) cognitive imagery relaxation. Systolic, diastolic blood pressure and heart rate were assessed in a baseline session, the 8th post-treatment session, and a 30-days follow-up session. Datawere analyzed using ANOVA and Paired sample t-test. One-Sample Kolmogorov-Smirnov Tests for the normal distribution were performed among the three groups. The results verified that the normality assumption for the data had not been violated. Results revealed that in the context of the study all relaxation therapies can reduce blood pressure in Chinese subjects, but stretch release relaxation and progressive muscle relaxation therapies appeared to be more effective in lowering blood pressure compared to cognitive imagery relaxation. These data are in a form available for meta-analysis at a later date. The study also shows a degree of clinical significance and validation with respect to a Hong Kong population. It demonstrates ways in which the inclusion of research on complementary therapies in systematic reviews can be maximized.

Alterations of haemostatic and fibrinolytic markers in adult patients with growth hormone deficiency and with acromegaly.

Alterations of coagulation and fibrinolytic systems might contribute to the increased cardiovascular and cerebrovascular mortality observed in patients with both chronic growth hormone (GH) excess (acromegaly) and deficiency (GHD). However, contrasting results have been so far reported. To assess the importance of GH in modulating haemostatic system, several haemostatic variables in patients with GHD and acromegaly were measured. Twenty-four adult patients with GHD (8 childhood- and 16 adult-onset; age: 41+/-12 years, insulin like growth factor-I, IGF-I: 6.7+/-4 nmol/L), 10 non-diabetic acromegalic patients (age: 39+/-15 years; IGF-I: 109+/-37 nmol/L) and 64 healthy volunteers age- and sex-matched with cases were studied. The plasma levels of tissue-type plasminogen activator antigen (t-PA), prothrombin fragment 1+2 (F1+2) and thrombin-antithrombin complex (TAT) were measured by ELISA. Plasminogen activator inhibitor type I (PAI-1) was measured by an immunoactivity assay and fibrinogen by von Clauss method. GH levels were measured by IFMA and IGF-I by RIA. GHD patients had higher PAI-1 (12.7+/-16.7 vs 4.8+/-5.3 U/ml, p<0.01), fibrinogen (363+/-104 vs 291+/-71 mg/dL, p< 0.05) and TAT levels (6.8+/-9 vs 3.6+/-2.8 ng/ml, p<0.05) than controls. Taking the 95th pecentile of the normal distribution in the control group as the cut-off point for normal plasma levels of the haemostatic variables, high PAI levels were found in 25% of patients with GHD (P<0.01), while high fibrinogen and TAT levels were observed in 21% (P<0.05). The alterations were mostly present in patients with adult-onset GHD, with the exception of hyperfibrinogenaemia which was equally present in adult- and childhood-onset patients. Acromegalic patients had higher mean fibrinogen levels than controls (398+/-111 vs 291+/-71 mg/dL, p< 0.05), 40% having hyperfibrinogenaemia (P<0.01, vs controls). They also had t-PA levels lower than controls and GHD. No correlations between hormonal and haemostatic variables were found. Body mass index and waist to hip ratio correlated positively with PAI-1 levels in GHD patients only. In conclusion, this study shows that several abnormalities of coagulation variables (increased PAI-1. fibrinogen and TAT levels) are present in patients with GHD, while only hyperfibrinogenaemia is found in patients with acromegaly. These changes do not appear to be directly related to IGF-I levels or to the degree of GH deficiency/excess. However, these abnormalities may be an additional trigger for the development of coronary heart disease and thromboembolic complications mostly in patients with GHD.

[Insulin autoantibodies and islet cell antibodies in recently appearing diabetes mellitus type I. Association with age of manifestation and HLA phenotype].

Insulin autoantibody (IAA) and islet cell antibody (ICA) titres were measured in 108 newly diagnosed type I diabetics (49 male, 59 female, mean age 20 [1-38] years) and 103 non-diabetic controls (41 male, 62 female, mean age 23 [16-46] years). IAA titres in the controls were normally distributed, with a mean of 5 +/- 11 nU/ml. The upper limit of normal was established as 49 nU/ml (mean + 4 standard deviations). Raised IAA and ICA titres were present in 45% and 44% of type I diabetics, respectively, with 59% positive for either IAA or ICA or both. IAA were markedly age-dependent, being positive in 70% (26 out of 37) of diabetics under the age of 15 years, and in 32% (23 out of 71) at the age of 15 years or more (P = 0.0004). There was a less marked difference for ICA titres (positive in 62% of patients less than 15 years, and in 35% of those of 15 years of older; P less than 0.01). IAA were significantly more common in HLA DR4 positive patients than in HLA DR4 negative patients (56% vs 11%; Pc less than 0.00015). With regard to age a significant association between IAA and HLA DR phenotype was present only in homozygous (Pc less than 0.03) and heterozygous (P less than 0.0003) patients aged 15 years or older. By contrast, ICA was not significantly correlated with HLA phenotype. These data suggest a genetic predisposition for the development of IAA.

A robust confidence interval for location for symmetric, long-tailed distributions.

A procedure called the wave-interval is presented for obtaining a 95% confidence interval for the center (mean, median) of a symmetric distribution that is not only highly efficient when the data have a Normal distribution but also performs well when some or all of the data come from a long-tailed distribution such as the Cauchy. Use of the wave-interval greatly reduces the risk of asserting much less than one's data will support. The only table required is the usual t-table. The wave-interval procedure is definitely recommended for samples of ten or more, and appears satisfactory for samples of nine or eight.

Making best use of available residue data for acute intake assessment.

Risk assessment for pesticide residues in food has previously concentrated on chronic intake, probably with the assumption that if the chronic intake is acceptable, acute intake will also be acceptable. Data generation is expensive so we should extract, where possible, information useful for acute intake estimation from supervised residue trials. Residues in individual units of fruit or vegetables are more variable than the residues in composite samples and the distribution in individual units is probably best approximated by a log-normal distribution, but further evidence is needed. Theory provides a method for calculating the variability of replicate composite samples and applying it to individual units. The expected residue in the 95th or 99th percentile single fruits may then be calculated. An example for folpet residues in apples showed that the 95th and 99th percentile apples would contain 1.8 x and 2.3 x the residue in the composite sample, respectively. For abamectin in apples the 95th and 99th percentile factors were 6.7 and 15, respectively. The 95th and 99th percentile factors for azinphos-methyl in apples, based on single apple analysis, were 1.7 and 2.1, respectively.

Short-term variability of pulse rate and blood pressure in cardiac surgery patients.

The magnitude and mathematical nature of short-term variations of pulse rate and mean arterial blood pressure has been studied in 27 post-operative cardiac surgical patients over a continuous period of 200 min in each case. Similar variability was observed in all patients. Short-term variations were composed predominantly of a series of rhythmic changes ranging from one synchronous with respiration to others between 2 and 5 min in cycle length. There was consistent variance of the beat-by-beat values for both variables about a continuously updating 5 min mean. The average standard deviation was 3.75 beats/min for pulse rate and 0.48 kPa (3.64 mm Hg) for mean arterial blood pressure. For both variables the distribution about the 5-min mean did not differ significantly from a normal distribution in 50 out of the 54 records. These findings have implications for the reproducibility of current methods of estimating mean pulse rate and blood pressure, and the change necessary before two estimates may be regarded as significantly different. The results are applicable both to the interpretation of Ward Charts by medical staff and to automated monitoring systems.

Pilot study of indoor radon in Greek workplaces.

Radon and gamma dose rate measurements have been performed in 561 workplaces in 19 prefectures of Greece. The distribution of radon concentration can be well described by a log-normal distribution. Most of the radon concentrations are between 50 and 200 Bq m(-3) with an arithmetic mean of 123 Bq m(-3). The maximum measured value of radon gas concentration is 695 Bq m(-3). About 10% of the workplaces exceed 200 Bq m(-3). Only a small fraction ( approximately 1%) of workplaces exceed the European Commission action level (400 Bq m(-3)). Despite the relative small fraction of workplaces which exceed the value of 400 Bq m(-3), it is clear from the results of the present work that for certain prefectures, further and more extensive research is needed.

Indium-111 labeled leukocyte imaging following hepatic artery embolization.

The use of In-111 labeled leukocytes for abscess localization is becoming well established. The first report of In-111 imaging following hepatic embolization is presented. A 45-year-old man with adenocarcinoma of the colon and metastatic liver disease was treated for intractable pain using particulate embolization of the hepatic artery. In-111 leukocyte imaging was performed to rule out abscess formation. The distribution of the labeled leukocytes demonstrated hepatic uptake commensurate with Tc-99m sulfur colloid (SC) images. Areas of embolization did not accumulate tracer. Pathologic examination at autopsy correlated with the distribution of the labeled leukocytes. Thus, therapeutic embolization did not alter the normal distribution of this tracer in functional hepatic tissue.

Logarithmic distribution of amplitudes of compound muscle action potentials evoked by transcranial magnetic stimulation.

Motor evoked potentials were recorded from the first dorsal interosseous muscle on the dominant side evoked by transcranial magnetic stimulation in 20 healthy volunteers. Amplitude data revealed skewness of distribution and variance heterogeneity. Natural logarithmic transformation of amplitude data results in normal distribution and ensures variance homogeneity. There was a negative first-order autocorrelation between consecutive recordings, which indicates that a stimulus-free interval of 5 s in consecutive stimulations is insufficient to ensure independent events. In a variability study, no systematic changes in latency and amplitude during day-to-day examinations or in consecutive examinations were found. There was a significant change in latency data (p = 0.02) during a gradual increase of constant elements in the stimulation procedure, whereas the amplitude data were unchanged. Estimation of random contributors to the total variability in the magnetic stimulation technique was performed. Intersubject differences were the major contributor to the variability of latency data but not to variability of logarithmic amplitude data. The variance estimates of different examiners' contribution to the variability was 0.02 for latency data and 0 for amplitude data. This indicates that staff members familiar with the principles in magnetic stimulation and following a thorough verbal and written instruction in the standardized examination procedure can perform the stimulation procedure without increasing the data variability.

Lattice Boltzmann simulation of fluid flow in fracture networks with rough, self-affine surfaces.

Using the lattice Boltzmann method, we study fluid flow in a two-dimensional (2D) model of fracture network of rock. Each fracture in a square network is represented by a 2D channel with rough, self-affine internal surfaces. Various parameters of the model, such as the connectivity and the apertures of the fractures, the roughness profile of their surface, as well as the Reynolds number for flow of the fluid, are systematically varied in order to assess their effect on the effective permeability of the fracture network. The distribution of the fractures' apertures is approximated well by a log-normal distribution, which is consistent with experimental data. Due to the roughness of the fractures' surfaces, and the finite size of the networks that can be used in the simulations, the fracture network is anisotropic. The anisotropy increases as the connectivity of the network decreases and approaches the percolation threshold. The effective permeability K of the network follows the power law K approximately (beta), where is the average aperture of the fractures in the network and the exponent beta may depend on the roughness exponent. A crossover from linear to nonlinear flow regime is obtained at a Reynolds number Re approximately O(1), but the precise numerical value of the crossover Re depends on the roughness of the fractures' surfaces.

Observations of coarsening of air voids in a polymer-highly-soluble crystalline matrix during dissolution.

A combination of magnetic resonance imaging and x-ray microcomputed tomography has been used to visualize the development of the internal micro-structure within compressed tablets made from a combination of insoluble particles (Eudragit, a polymer) and soluble particles (diltiazem hydrochloride, a drug), during dissolution in water. Air voids in the tablet are seen to coarsen. The size distribution of the air voids is well fitted by a log-normal distribution with a mean size that grows linearly with time. There is evidence for both diffusion of voids and sudden collapse of individual voids, presumably as they coalesce. The behavior of the voids is studied and compared with models of coarsening; the implications for tablet dissolution are considered.

Pushing the boundaries of molecular replacement with maximum likelihood.

The molecular-replacement method works well with good models and simple unit cells, but often fails with more difficult problems. Experience with likelihood in other areas of crystallography suggests that it would improve performance significantly. For molecular replacement, the form of the required likelihood function depends on whether there is ambiguity in the relative phases of the contributions from symmetry-related molecules (e.g. rotation versus translation searches). Likelihood functions used in structure refinement are appropriate only for translation (or six-dimensional) searches, where the correct translation will place all of the atoms in the model approximately correctly. A new likelihood function that allows for unknown relative phases is suitable for rotation searches. It is shown that correlations between sequence identity and coordinate error can be used to calibrate parameters for model quality in the likelihood functions. Multiple models of a molecule can be combined in a statistically valid way by setting up the joint probability distribution of the true and model structure factors as a multivariate complex normal distribution, from which the conditional distribution of the true structure factor given the models can be derived. Tests in a new molecular-replacement program, Beast, show that the likelihood-based targets are more sensitive and more accurate than previous targets. The new multiple-model likelihood function has a dramatic impact on success.

Haematologic profile of natural populations: red cell parameters.

Age-sex specific reference values and frequency distribution of erythrocyte parameters were derived from a representative sample of a natural population consisting of 1094 men and 1284 women between 10 and 96 years of age. Some differences have been found between previously reported reference values and those presented here, the importance of which lies not so much in their magnitude as in the fact that the latter are based upon a probability sample of a natural population unselected as to disease and are presented as percentile distributions by age and sex rather than as the more frequently used normal distribution. These results are intended for use as reference data both for clinical and for population estimates of haematological parameters.
