Nondestructive Larval Genotyping of Danio rerio for Mitochondrial and Nuclear DNA Genetics.

The rapid advancement of nuclear and mitochondrial genomic editing tools has created an urgent need for efficient, nonlethal larval genotyping methods in zebrafish (Danio rerio) research. This study optimizes and validates a nondestructive proteinase K digestion method for mitochondrial and nuclear DNA genotyping while characterizing its impact on larval survival and gene expression. Using optimized protocol parameters, we demonstrate successful amplification of different mitochondrial and nuclear genetic loci with consistently high sensitivity. Molecular validation through PCR, restriction fragment length polymorphism analysis, and Sanger sequencing confirmed the specificity and reliability of the extracted DNA. The method successfully detected C-to-T base edits in the mt-tl1 gene introduced using the FusX TALE Base editor system, demonstrating its applicability to gene editing studies. Both 48-well and optimized 96-well formats were used, enabling this approach to be deployed at scale. This optimized method enables researchers to correlate genotypes with phenotypes in longitudinal studies while maintaining specimen viability, particularly valuable for investigating early-onset mitochondrial diseases, and utilizes standard laboratory equipment and reagents, facilitating widespread adoption in zebrafish research while adhering to ethical principles in reducing animal mortality.

Magnetic resonance imaging-controlled laser-induced interstitial thermotherapy.

RATIONALE AND OBJECTIVES: Laser-induced interstitial thermotherapy (LITT) may become an attractive modality for minimally invasive tumor therapy. Magnetic resonance imaging (MRI) could be used to assist this procedure. METHODS: A T1-weighted turbo fast low-angle shot (FLASH) sequence for on-line monitoring of the laser-influenced region (liver, muscle) was investigated. Sequence parameters were optimized for maximal image contrast. Magnetic resonance imaging-controlled LITT was performed in vitro, in vivo (rabbits), and in 8 human investigations (6 patients). Special laser applicators were used to establish a uniform laser light distribution. RESULTS: With the MRI sequence used, the LITT region is visualized as a bright area outlined by a dark border. This dark border corresponds to an isotherm of 45 +/- 2 degrees C depending on the sequence parameters used. CONCLUSION: With the T1-weighted turbo-FLASH sequence, MRI can be used for on-line monitoring of interstitial laser-induced thermotherapy in moving organs.

Decolourization of aqueous dye solutions by a novel adsorbent: application of statistical designs and surface plots for the optimization and regression analysis.

Adsorption of Neolan Blue 2G (Acid Blue 158) and Basic Methylene Blue (Basic Blue 9) was investigated using a hybrid adsorbent that was prepared by pyrolysing a mixture of carbon and flyash at 1:1 ratio. A 2(4) full factorial central composite design with nine replicates at the center point and thus a total of 31 experiments were successfully employed for batch experimental design and analysis of the results. The combined effect of pH, temperature, particle size and time on the dye adsorption was studied. An empirical model was developed and validated applying ANOVA analysis incorporating interaction effects of all parameters and optimized using response surface methodology. The optimum pH, temperature, particle size and time were found to be 2.20, 27.85 degrees C, 0.0565 mm, 245 min, respectively, for Acid Blue 158 and those for Basic Blue 9 were 13.40, 28.45 degrees C, 0.0555 mm and 230 min, respectively. Complete removal (100%) was observed for both the dyes using the hybrid adsorbent.

Separation of (Z)- and (E)-isomers of thioxanthene and dibenz[b,e]oxepin derivatives with calixarenes and resorcinarenes as additives in nonaqueous capillary electrophoresis.

Five acidic calix[4]arenes with carboxylic or sulfonic groups at either the upper or lower rim of the cavity and one resorc[4]arene were investigated to separate three thioxanthenes (flupentixol, clopenthixol, chlorprothixene) and a dibenz[b,e]oxepin derivative (doxepin) with cis-/trans-isomerism by nonaqueous capillary electrophoresis (NACE). Partial filling of the capillary with the UV-absorbing selectors led to a low detection limit and an advantageous signal-to-noise ratio (S/N). A sufficient electrophoretic mobility of the calixarenes towards the anode was necessary to outweigh the oppositely directed electroosmotic flow (EOF). This depended from the functional groups, the dissociation and the hydrodynamic radius of the cyclophanes. In contrast, the resorcinarene was useable only by addition of sodium dodecyl sulfate (SDS) because only the complex of the two selectors had an anodic apparent electrophoretic mobility. p-Sulfonyl-calix[4]arene (ss-a1) was the most capable selector for all E/Z-isomers with maximal alpha-values ranging from 1.056 for doxepin to 1.224 for chlorprothixene. This was due to the sufficient migration in reversed direction to the EOF even at low pH* values of 3.0. Otherwise, electrostatic as well as hydrophobic interactions with the positively charged isomers seem to contribute to a superior recognition. Increasing the concentration up to 15 mM ss-a1 and using acidic media (pH* 5.0) led to high separation efficiency. Changing the organic solvent provides a powerful tool to improve selectivity with N,N-dimethylformamide-methanol (DMF-MeOH)-mixtures for thioxanthenes. Further electrophoretic parameters were optimized, such as the concentration of the electrolytes, the addition of SDS, the kind of electrolytes and the voltage. Distinct differences in selectivities were found between the derivatives with thioxanthene and dibenzo[b,e]oxepin ring system, respectively. Further, the different basic side chain was responsible for the different selectivity at higher pH* values. In contrast, the substitution at position 2 of the thioxanthenes played a secondary role. Based on the studies of single parameters a method for a simultaneous separation of the four pairs of isomers within 13 min was developed.

Model fitting and projection of the AIDS epidemic.

Previously it was possible to fit detailed models to incidence data (for example, of AIDS) only by trial and error and good judgment; the large number of parameters obstructed optimization of, for example, the (approximate) likelihood. Here, we analyze a model for the spread of AIDS in a homosexual population and identify a minimal set of primary components that dictate the dynamics of the Model: the initial growth rate theta, the basic reproductive ratio R0, and the heterogeneity coefficient S. It is then shown that it is sufficient to maximize the likelihood over these three primary components; further maximization over the remaining secondary parameters does not produce a significant improvement in the fit or affect the projection of the epidemic. This method also allows construction of confidence limits for the projected incidence curve, allowing us to quantify the uncertainties associated with such model fitting procedures. The method is tested on simulation data to analyze how the accuracy of estimates and projections changes as we gain more data.

An in vitro system for the enzymological analysis of avian hepatitis B virus replication and inhibition in core particles.

A detailed analysis of the hepatitis B virus (HBV) replication reaction is important both in understanding viral biology and in developing effective antiviral drugs. This can best be achieved by studying the viral reverse transcriptase (RT) in its natural context, encapsidated within viral core particles in a multiprotein complex, rather than as an isolated enzyme. In order to facilitate a precise enzymological analysis of the avian HBV-RT reaction and its inhibition within replicating cores, a scheme for the purification and analysis of intracellular core particles derived from infected liver tissue has been devised, optimized and evaluated. The purification scheme itself is simple and rapid, and results in preparations with a 25-fold increase in endogenous polymerase activity that persists for over 5 h under assay conditions. In order to assess the suitability of these preparations for mechanistic studies, a thorough evaluation of purity was undertaken, revealing predominantly pure viral protein and nucleic acid, free of contaminating cellular polymerases and phosphatase activities that potently degrade nucleotides and antiviral drugs. Parameters governing optimal polymerase activity have been determined, and an assay for DHBV-RT activity has been developed which offers the highest purity and specific polymerase activity currently available to study hepadnaviral replication and inhibition.

[Study on enrichment process of total flavonoids from Euphorbia himifusa with macroporous resin].

OBJECTIVE: To study the optimal technology parameters of the enrichment process for total flavonoids of Euphorbia himifusa with macroporous resin. METHODS: The enrichment process was studied by orthogonal experiment design using orthogonal form L9(3(4)) with the content of total flavonoids from Euphorbia himifusa as indexes, and three factors were chosen in this experiment, including the saturation of the extract sicca from Euphorbia himifusa, duration of absortion and the consistence of alcohol as eluent. RESULTS: The results showed that the optimal enrichment process was that 10 ml of the extract of Euphorbia himifusa (i.e. lml 75% alcohol contains 0.5 g the extract sicca from Herba Euphorbiae Humifusae) was absorbed 30 min with a column of macroporous resin(r 15 mm x H160 mm) and the total flavonoids was eluted from the macroporous resin with 95% alcohol. CONCLUSION: The elutive rate of total flavonoids was above 93% by means of the macroporous resin. So this process enriching the total flavonoids of Euphorbia himifusa is feasible.

Optimal javelin trajectories.

A companion paper has treated computer simulation of javelin flight using measured lift, drag and pitching moments. In the present paper we present, categorize and discuss the relative significance of various initial conditions in such a simulation. Since the differential equations describing flight are autonomous, the eventual javelin range and entry angle are unique functions of the initial conditions. A series of successively less constrained optimum solutions is defined, the last of which is the global optimum javelin trajectory. Sensitivities of these trajectories to perturbations from the optima and their implications for throwers are discussed. Finally, we investigate the effects of some design and environmental parameters on optimal initial conditions and trajectories.

Development of a sensitive, colorometric microarray assay for allergen-responsive human IgE.

With the existence of several thousand unique human allergens, a multiplex format, such as protein microarrays, is an attractive option for allergy screening. To determine the feasibility and sensitivity of using an enzyme-based, colorimetric protein microarray assay, three common allergens (mold, dust-mite, grass) were arrayed and added sera assayed for responsive human IgE. Normal, low positive, and negative control samples were assayed to determine optimal reaction parameters. Sensitivity of the assay (in international units, IU) was determined by constructing a standard curve using World Health Organization (WHO) standards. The system described here can reliably detect allergen-specific IgE below 0.35 IU, the current WHO standard cutoff. By taking advantage of the sensitivity of enzyme-linked immunosorbent assays (ELISAs) and the multiplex format of microarrays, we have achieved a high throughput system, capable of screening patients for allergen-susceptibility with optimal sensitivity.

Determination of the fluoroquinolone enrofloxacin in edible chicken muscle by supercritical fluid extraction and liquid chromatography with fluorescence detection.

A supercritical fluid extraction (SFE) method for the extraction of enrofloxacin from a chicken breast muscle was examined. A liquid chromatograph, equipped with a fluorescence detector, was used for the detection of enrofloxacin. Optimal extraction parameters, such as extraction time, supercritical fluid volume, modifier concentration, pressure, and temperature, were determined by examining SFE recoveries from control muscle samples spiked with enrofloxacin at different levels. In all of the experiments, high recovery values were observed, ranging from 101 to 104%. The extraction of enrofloxacin from real muscle samples was examined in chickens that were treated orally with enrofloxacin. Extraction was carried out by the SFE method after each oral treatment and under optimal extraction conditions at set intervals over time. The SFE, combined with liquid chromatographic analysis, showed that the concentration of enrofloxacin in the chicken muscles decreased continuously with time, giving a negligible concentration 72 h after the treatment. These results suggest that SFE is a useful approach for the extraction of enrofloxacin from chicken breast muscles.

Optimization of self-microemulsifying drug delivery systems (SMEDDS) using a D-optimal design and the desirability function.

D-optimal design and the desirability function were applied to optimize a self-microemulsifying drug delivery system (SMEDDS). The optimized key parameters were the following: 1) particle size of the dispersed emulsion, 2) solubility of the drug in the vehicle, and 3) the vehicle compatibility with the hard gelatin capsule. Three formulation variables, PEG200, a surfactant mixture, and an oil mixture, were included in the experimental design. The results of the mathematical analysis of the data demonstrated significant interactions among the formulation variables, and the desirability function was demonstrated to be a powerful tool to predict the optimal formulation for the explored system.

CO2 laser treatment of traumatic pulpal exposures in dogs.

BACKGROUND AND OBJECTIVE: Successful non-devitalizing treatment of localized pulpal lesions in mature teeth is not ensured using conventional endodontic techniques. The objective of this study was to evaluate CO2 laser surgical treatment of pulpal exposures in canine patients. STUDY DESIGN/MATERIALS AND METHODS: 17 permanent teeth with pulpal exposures of < or = 48 h duration received localized laser pulp surgery. Laser Parameters: pulse duration: 0.01 s, pulse interval: 1.0 s, spot size: 0.004 cm2, fluence: 276 J/cm2. Exposures were dressed with CaOH and Glass ionomer. Clinical and radiographic evaluations were performed by one blinded clinician 24 and 52 weeks after treatment. RESULTS: 15/17 laser-treated teeth assessed over > or = 1 year post-treatment remained clinically and radiographically healthy. CONCLUSION: These results demonstrate the feasibility of using the CO2 laser for localized pulp surgery. Further studies must optimize laser parameters and identify the range of clinical pathologies which can be treated using this modality.

Single molecule fluorescence burst detection of DNA fragments separated by capillary electrophoresis.

A method has been developed for detecting DNA separated by capillary gel electrophoresis (CGE) using single molecule photon burst counting. A confocal fluorescence microscope was used to observe the fluorescence bursts from single molecules of DNA multiply labeled with the thiazole orange derivative T06 as they passed through the approximately 2 micrometer diameter focused laser beam. Amplified photoelectron pulses from the photomultiplier are grouped into bins of 360-450 micros in duration, and the resulting histogram is stored in a computer for analysis. Solutions of M13 DNA were first flowed through the capillary at various concentrations, and the resulting data were used to optimize the parameters for digital filtering using a low-pass Fourier filter, selecting a discriminator level for peak detection, and applying a peak-calling algorithm. Statistical analyses showed that (i) the number of M13 molecules counted versus concentration was linear with slope = 1, (ii) the average burst duration was consistent with the expected transit time of a single molecule through the laser beam, and (iii) the number of detected molecules was consistent with single molecule detection. The optimized single molecule counting method was then applied to an electrophoretic separation of M13 DNA and to a separation of pBR 322 DNA from pRL 277 DNA. Clusters of discreet fluorescence bursts were observed at the expected appearance time of each DNA band. The autocorrelation function of these data indicated transit times that were consistent with the observed electrophoretic velocity. These separations were easily detected when only 50-100 molecules of DNA per band traveled through the detection region. This new detection technology should lead to the routine analysis of DNA in capillary columns with an on-column sensitivity of approximately 100 DNA molecules/band or better.

[Relation of parameters of frequent transesophageal electric stimulation of the atria during the treatment of atrial flutter and electrophysiologic characteristics of the myocardium].

After arresting paroxysms with frequent transesophageal cardiostimulation (TECS), 20 patients with coronary heart disease accompanied by episodes of Type 1 atrial flutter (AF) were studied by using a technique of intracardiac electrophysiological examination. The length of the atrial cycle in AF was found to correlate with that of successful TECS (r = 0.83). The correlation increases when a difference of atrial functional and effective refractory periods is subtracted from the length of the atrial cycle in tachycardia (r = 0.92). The equations of simple and multiple linear regression were derived, which defined the optimal frequency parameters of stimulation with a sufficient accuracy to stop Type I AF episodes. The efficacy of frequent TECS in arresting 324 spontaneous AF episodes was 92.9%.

Characterizing complex peptide mixtures using a multi-dimensional liquid chromatography-mass spectrometry system: Saccharomyces cerevisiae as a model system.

A rugged, reproducible, multi-dimensional LC-MS system was developed to identify and characterize proteins involved in protein-protein interactions and/or protein complexes. Our objective was to optimize chromatographic parameters for complex protein mixture analyses using automated peptide sequence recognition as an analytical end-point. The chromatographic system uses orthogonal separation mechanisms by employing strong cation exchange (SCX) in the first dimension and reversed phase (RP) in the second dimension. The system is fully automated and sufficiently robust to handle direct injections of protein digests. This system incorporates a streamlined post analysis results comparison, called DBParser, which permitted comprehensive evaluation of sample loading and chromatographic conditions to optimize the performance and reproducibility. Peptides obtained from trypsin digestion of a yeast soluble extract provided an open-ended model system containing a wide variety and dynamic range of components. Conditions are described that resulted in an average (n = 4) of 1489 unique peptide identifications, corresponding to 459 non-redundant protein sequence database records (SDRs) in the 20 microg soluble fraction digest.

A randomized study to evaluate the effect of a perioperative infusion of dopexamine on colonic mucosal ischemia after aortic surgery.

PURPOSE: Colonic ischemia after aortic surgery is associated with increased mortality and morbidity rates. This study was conducted as a single-center side arm to a multicenter, randomized, placebo-controlled study to evaluate the effect of dopexamine hydrochloride on its incidence. METHODS: Thirty patients, mean age 65.1 years (range, 46-84), undergoing elective infrarenal aortic surgery were entered. Preoperative hemodynamic and respiratory parameters were optimized. Patients were then randomly assigned to receive a perioperative infusion of dopexamine at 2 microg/kg per minute (n = 12) or 0.9% saline placebo (n = 18). All patients underwent colonoscopy and biopsy preoperatively and 1 week postoperatively. Specimens were assessed for evidence of mucosal ischemia, presence of mast cell tryptase, myeloperoxidase activity, and both the inducible and endothelial isoforms of nitric oxide synthase. RESULTS: There was no significant difference in perioperative fluid and blood requirements or hemodynamic and respiratory parameters between the two groups. However, there was significantly less evidence of mucosal ischemic changes in dopexamine-treated patients (n = 1) compared with placebo (n = 8) (P =.049). Furthermore, when preoperative biopsies were compared with those performed 1 week postoperatively, nine (50%) patients in the placebo group and two (16.7%) in the dopexamine group scored worse. Although there was no significant difference in inflammatory markers between the two groups, both mast cell tryptase and myeloperoxidase expression were increased in patients with histologic evidence of ischemia (P <.05). Furthermore, inducible nitric oxide synthase staining within the vascular (P =.001) and lamina propria (P <.05) components of the mucosa was also significantly greater. CONCLUSION: A perioperative dopexamine infusion affords significant histologic protection to colonic mucosa after aortic surgery.

Glass encapsulation of flavours with chemically modified starch blends.

Carrier compositions based on blends of various octenylsuccinic acid anhydride (OSAN)-modified starches were utilized in the glass encapsulation of flavours by means of melt extrusion and spray drying. Dextrinized, acid and enzymatically hydrolysed OSAN starches were used in various combinations. Processing parameters were optimized for desirable physical chemical and functional properties of the encapsulating compositions. One key property in the optimization of the extrusion process was setting rate of the exiting melt into a glass. Glassy states of identical spray-dried and extruded compositions were characterized by MDSC for glass transition temperature(s), heat capacity change and enthalpy relaxation. Flavour retention, surface flavour, moisture, particle density and flavour particle size distribution were also measured and compared for the extruded and spray-dried compositions. In addition, elastic recovery and caking were evaluated for extruded compositions. The glass transition parameters were correlated directly or indirectly with most of the physical properties of encapsulating compositions, especially flavour retention, surface flavour and caking. In extruded compositions, two phases were revealed by the presence of two glass transitions, whereas spray-dried compositions showed only one broad glass transition.

Effect of reaction parameters on synthesis of citronellyl methacrylate by lipase-catalyzed transesterification.

The methacrylate ester of citronellol was synthesized using various lipases as catalyst. The effect of different reaction parameters such as amount of lipase, solvent, temperature, and acylating agent on the conversion of citronellol to citronellyl methacrylate was studied. Methyl methacrylate, vinyl methacrylate, and 2,3-butanedione mono-oxime methacrylate were used as acylating agents. Porcine pancreatic lipase (PPL), Candida rugosa lipase (CRL), and Pseudomonas cepacia lipase (Amano-PS) were used as biocatalysts. Diisopropyl ether (DIPE) was found to be the most suitable solvent. The stereoselectivity of CRL in transesterification of (+/-)-citronellol was tested for the optimized reaction parameters.
