Nuclear morphometry of MIB-1 positive and negative tumor cells in primary and metastatic malignant melanoma of the skin.

The purpose of this study was the evaluation of nuclear area, nuclear axis ratio, perimeter and roundness of nuclei of tumor cells with and without Ki-67 antigen expression (demonstrated immunohistochemically using MIB-1 antibody) in primary and metastatic malignant melanoma of the skin. The parameters were further analyzed with respect to their association with the depth of malignant invasion according to Clark [7] and tumor thickness according to Breslow [6]. 142 malignant melanomas (53 primary and 89 metastatic), were assessed employing a computerized image analyzer Quantimet 600S (Leica). The mean nuclear area of MIB-1 positive nuclei was significantly larger than that of the negative ones (p < 0.0001) both in primary and metastatic malignant melanoma. In comparison to the primary melanoma the nuclei of metastatic melanoma cells had a larger area and were more rounded, while the MIB-1 positive nuclei additionally showed a greater degree of polymorphism of their area and shape. With growing invasion thickness according to Breslow and increased Clark's level, the mean nuclear area of tumor cells increased, and their shape became more round. The MIB-1 positive tumor cell nuclei of primary melanomas with metastases were significantly out of round in comparison to primary melanomas without metastases. The results indicate an association between the area and shape of melanoma cell nuclei and the presence of metastases, and between the nuclear area of tumor cells and such factors related to poor prognosis as the depth of invasion and the tumor thickness.

Morphometry of the normal human ear: a cross-sectional study from adolescence to mid-adulthood.

The objective of this study was to supply information about: (1) normal gender-based dimensions of ears (linear distances and ratios, area); (2) left right symmetry; and (3) growth changes between adolescence and mid-adulthood. The three-dimensional co-ordinates of several soft-tissue landmarks on the ears and face were obtained by an electromagnetic digitizer in 40 male and 33 female adolescents aged 12-15 years. 73 young female and 89 young male adults aged 19-30 years, and 41 male and 38 female adults aged 31-56 years. From the landmarks, paired car width and length were calculated and averaged for age and gender, as well as the relevant ratios, ear areas and angles relative to the facial midline, and indices of left right symmetry. Comparisons were performed by factorial analysis of variance. All ear dimensions were significantly larger in men than in women (P < 0.005). A significant effect of age was found (P < 0.005), with larger values in older individuals. On average, the three-dimensional position of ears was symmetric, with symmetry coefficients ranging between 94 and 96%. Data collected in the present investigation could serve as a database for the quantitative description of human ear morphology during normal adolescent and adult growth.

Morphometry of the FRTL-5 cells after irradiation.

The morphological changes of in vitro irradiated FRTL-5 cells and their ability to grow in semi-solid medium were studied morphometrically. FRTL-5 cells were grown in medium with 4 different concentrations of TSH (0, 0.1, 1, 10 mU/ml). After irradiation with 0 Gy, 2 Gy and 4 Gy, the cells were seeded on glass cover-slips and in methocel. Fourteen days after irradiation, the morphometric analysis of FRTL-5 cells and their nuclei was performed. The results showed that irradiation and different doses of TSH have influence on FRTL-5 cell size, more on their nuclei than on the cells as a whole. Growing of FRTL-5 cells in the methocel indicates the possible transformation of these cells after long-culturing in the TSH medium and after irradiation.

[Radiologic morphometry of the hip in Beninese black children].

PURPOSE: The author aims to objectivize simple parameters of normal anatomy of the hip related to age and sex in children as a contribution to interpretation criterions in medical imagery. Materials and methods. He has examined antero-posterior radiographs of 155 normal hips (right: 81; left: 74) among 129 children aged from six months to 16 years (mean age=8.9 years). They were 86 boys and 43 girls (ratio M/F=2). RESULTS: The results are as follow:--the joint space of the hip (JS/H): cranial part (cranial joint space: CJS/H): 5.5 mm +/- 0.5; medial part (medial joint space: MJS/H): 7 mm+/-0.5; --the bony roofingless part of the femoral head (BR/FH): 1-6 years: 1-3 mm; 7-13 years: 4-5 mm; 14-16 years: 6 mm; --the neck shaft angle of the femur (NSA/F); 1-6 years: 150 degrees-140 degrees; 7-16 years: 135 degrees; --the horizontal and the vertical diameters of the femoral head (HD/FH; VD/FH); x being the age of the children, the HD/FH grows according to a linear regression function as Ydh = 2x + 13+/-2 and the VD/FH as Ych = 2x + 18+/-2; --the centre acetabular edge index (CEI): the CEI decreases from 1 to 16 years: medium: 2.75-1.77; maximum: 2.31-1.67; minimum: 3.62-1.91; --the centre-acetabular cartilage index (CCI)): the CCI grows from 1 to 16 years: medium: 0.57-0.70; maximum: 0.62-0.73; minimum: 0.5-0.68. No influence of the side of the hip nor the sex of the children is observed. CONCLUSION: This study brings out exhaustive and original data on the growth velocity of the hip skeleton in children and supplies with criterions of diagnosis according to age in childhood.

Morphometry error due to non-perpendicular section of microvessel characterized by two-dimensional isotropic arrangement.

Formulas for calculation of the section non-perpendicularity error in estimation of the perimeter and cross-sectional area of microvessel profiles are proposed for the two-dimensional isotropic approximation. A relationship was established between the bias and random error associated with non-perpendicularity corresponding to theta(min) for microvessel profiles meeting the condition theta(min) < or = B/C < or = 1, where B and C are the minimum and maximum axes of the microvessel profile, respectively.

[Prognostic value of optic morphometry in colorectal cancer].

UNLABELLED: In many types of cancer, certain morphometric characteristic of tumor cells correlate with patient survival. Our observations suggested that the survival of patients with colorectal carcinomas is negatively correlated with tumor-cell nucleus size. METHODS: We investigated relationships between postsurgery survival and nucleus morphometric in 90 patients who had undergone resection for a colorectal tumor. The nucleus-size variables considered were maximum diameter, minimum diameter, perimeter, area, and form factor (means for 100 nuclei from each patient were used in all cases). RESULTS: Our results confirmed that patients with large maximum nucleus diameter (where large = greater than the first quartile) have significantly worse survival than patients with smaller maximum nucleus diameter (mean survival, 28 vs. 43 months). Similar results were obtained for the other nucleus-size variables. Stepwise Cox regression analysis was then performed, with postsurgery survival time as the dependent variable and the following candidate independent variables: age, gender. Dukes class, degree of histologic differentiation, the various nucleus-size variables, and relative frequencies of different nucleus shapes (spherical, oval, cylindrical, fusiform, and irregular). The variables selected for the prognostic model were Dukes class, relative frequency of irregular nuclei, and maximum nucleus diameter. CONCLUSIONS: These findings indicated that nucleus size and shape are useful predictors of survival. Even in Dukes class is known, consideration of nucleus size and shape significantly improves prediction of survival.

Ultrastructural morphometry of the diffusion surface in a case of pulmonary asbestosis.

A combination of electron microscopy and stereologic principles has allowed an evaluation of the contribution of structures in the blood-air barrier to gas exchange across it. In the present study this method was applied to biopsy material from the lungs of a patient with asbestosis. Within the limits of the sampling method, the results of the study are interpreted to indicate that measurable diffusion abnormalities were principally due to changes other than an increase in membrane thickness.

Quantitative nuclear morphometry by image analysis for prediction of recurrence of ductal carcinoma in situ of the breast.

Clinical management of ductal carcinoma in situ (DCIS) remains a challenge because significant proportions of patients experience recurrence after conservative surgical treatment. Unfortunately, it is difficult to prospectively identify, using objective criteria, patients who are at high risk of recurrence and might benefit from additional treatment. We conducted a multi-institutional, collaborative case-control study to identify nuclear morphometric features that would be useful for identifying women with DCIS at the highest risk of recurrence. Tissue sections of archival breast tissue of 29 women with recurrent and 73 matched women with nonrecurrent DCIS were stained for DNA, and nuclei in the DCIS lesions were evaluated by image analysis. A clear correlation between mean fractal2_area (FA2) and nuclear grade was observed (P < 0.001), allowing an objective determination of nuclear grade. Several nuclear morphometric features, including mean and variance of variation of radius, mean area, mean and variance of frequency of high boundary harmonics (FQH), and variance in sphericity, were found to be useful in discriminating recurrent from nonrecurrent DCIS subjects. However, the nuclear features associated with recurrence differed between high- and low-grade lesions. For lesions with high FA2 (nuclear grade 3), mean variation of radius, mean FQH, and mean area alone yielded recurrence odds ratios of 4.55 [95% confidence interval (CI) 0.45-45.96], 3.86 (95% CI, 0.88-16.98), 2.90 (95% CI, 0.31-27.2), respectively. Using a summed feature model, high-FA2 lesions showing three poor prognostic features had an odds ratio of 15.63 (95% CI, 1.22-200), compared with those with zero or one poor prognostic feature. Lesions with low mean FA2 (nuclear grade 1 or 2) showing high variances in sphericity and FQH had an odds ratio of 7.71 (95% CI, 1.77-33.60). Addition of other features did not enhance the odds ratio or its significance. These results suggest that nuclear image analysis of DCIS lesions may provide an adjunctive tool to conventional pathological analysis, both for the objective assessment of nuclear grade and for the identification of features that predict patient outcome.

Morphometry in anatomy: image analysis on fine structure and histochemical reactions with special reference to radioautography.

Anatomy now has a step to transfer from qualitative science to quantitative science. The methods of anatomical investigation employ optical instruments. Anatomy initially described qualitative features of the structure of the body, but now it turns to transfer to new fields measuring the number, size, length, area and volume of not only, structure but also the histochemical components of the structure with quantitative analysis. In order to quantify histochemical reactions in various organs, light and electron microscopic radioautograms were prepared from the liver, pancreas, intestines and kidney which were labeled with 3H-thymidine, 3H-uridine, 3H-leucine, 3H-glucosamine and 3H-glycerol, fixed in glutaraldehyde and osmium tetroxide, embedded in Epoxy resin, sectioned either thick or thin, coated with either Konica NR-M2 or -H2 radioautographic emulsions, exposed, developed, stained for either light or electron microscopic radioautographs. The radioautographs were observed with Olympus Vanox AHB-LB or Nikon Optiphot-2 light microscopes or Hitachi H-700, JEOL JEM-4000EX electron microscopes at 200 and 400 kV. The radioautograms were analysed with various image and analysed such as PPA-250, MOP, Digigramer-G, IBAS-II, Quadra 900, Luzex 300 to count the number of silver grains and to measure the size and number of structures. Some electron microscopic radioautographs were analysed in analytical electron microscopes, combined with Hitachi H-700 Horiba EMAX-1800E, JEOL JEM-200CX Kevex 7000-77, and JEOL JEM-4000EX Tracor-Northern TN-5400 to quantify silver grain contents. The results demonstrated that correct data can be obtained by controlling both full-automatic and semi-automatic image analysers by operating them manually.

Quantitative morphometry--an easy and inexpensive method for comparative studies of antigen expression during development.

The morphometric studies due to the development of computerized techniques became a very suitable tool for quantitative analysis. In the present paper we describe an easy and relatively inexpensive method useful for comparative studies on changes of expression of examined molecules during development of tissue. We used with a success this method for studies on the expression of CD31 adhesion molecule during the fetal and newborn lung development. We found that expression of CD31 is regulated during the perinatal period. It might be related to the dynamic changes in developing vascular bed in rat lung.

The evaluation of murine pleural lavage fluid cellular composition in experimental hemorrhagic shock with special regard to mast cells morphometry.

In the course of a hemorrhagic shock, pathological changes occur, which result in intensifying the insufficiency of various vital organs. It can also lead to the development of the multiorgan dysfunction syndrome (MODS) that is the cause of high posthemorrhagic mortality. As a result of the ischemia in the lung there appear proinflammatory factors that mobilize and activate mast cells, inducing degranulation in them. The aim of the study was the analysis of cellular composition and cytomorphometric evaluation of mast cells present in the lavage fluid from the pleural cavity of rats in a sham operated group and in the group presenting hemorrhagic shock. The results revealed an increase of the total cell count in the lavage fluid from the pleural cavity. In the cytological smears a statistically significant accumulation of inflammatory cells was present, especially neutrophils. The increase in mast cells and eosinophils number was not statistically significant. There was not a change in the morphometric parameters of mast cells except the circularity index. A decline of the circularity index indirectly may suggest the degranulation of mast cells, which reflects an inflammatory process in the lungs.

The application of electron microscopic morphometry as helpful method in the diagnosis of focal segmental glomerulosclerosis (FSGS) early phase. I. Morphometric electron microscopic studies of renal glomeruli in cases of focal segmental glomerulosclerosis (FSGS), minimal change disease (MCD) and mesangioproliferative glomerulonephritis (GNMES) in children.

Electron microscopy morphometric studies were carried out on material obtained from children with minimal change disease (MCD), mesangioproliferative glomerulonephritis (GNMES) and focal segmental glomerulosclerosis (FSGS). The results indicated that an increase in the volume of the matrix in mesangioproliferative glomerulonephritis was due to an increase in the number of cells. The amount of produced matrix in mesangioproliferative glomerulonephritis was proportional to the number of cells in mesangium (so there is no overproduction of matrix). The ratios of mesangial matrix volume to cell volume and matrix volume to the volume of the entire mesangial region in MCD and GNMES were similar and differed significantly from the ratios found in FSGS. The present findings indicate that morphometric studies may be useful in the early diagnosis of glomerular sclerosis. This is particularly significant in cases where light microscopic examination fails to reveal changes indicative of glomerular sclerosis.

The application of electron microscopic morphometry as a helpful method in the diagnosis of focal segmental glomerulosclerosis (FSGS) early phase. II. Clinical usefulness of electron microscopic morphometric studies in cases of minimal change disease (MCD) and mesangioproliferative glomerulonephritis (GNMES) with suspicion of progression into focal segmental glomerulosclerosis (FSGS).

Clinical and morphological analysis (including morphometric studies of electron microscopic material) was made in 15 children with MCD and 15 children with GNMES. In both groups, an early phase of FSGS was suspected on the basis of electron microscopic studies. Moreover, analysis included the results obtained in 13 children with the diagnosis of FSGS and in whom repeated biopsies were performed. In most of them, MCD or GNMES was diagnosed from the first biopsy. In most children in whom electron microscopic studies revealed an increase in matrix area in some mesangial regions, thereby suggesting an early stage of glomerular sclerosis, the results of morphometric studies resembled or were identical to the results obtained from a control group with the established diagnosis of FSGS. These findings indicate that morphometric studies of electron microscopic material are significant. The results, when compared with the clinical data, confirm the usefulness of such a diagnostic procedure.

Comparative morphometry of fibers and capillaries in soleus following weightlessness (SL-3) and suspension.

This work is a continuation of efforts to assess responses of rat skeletal muscle to weightlessness and earthside laboratory experiments with unloading of hind limbs. The soleus is a slow twitch, load bearing (antigravity) muscle. Both exposure to weightlessness (W) and to the hypokinesia/hypodynamia of whole body suspension (WBS) results in soleus atrophy. Cross sectional areas of both slow and fast twitch fibers decrease during 7 days of W, and 7 or 14 days of WBS. Density and area changes tended to reverse to control levels during 7 days of recovery (R) following WBS. Capillary density was increased with 7 days of W, and 7 or 14 days of WBS. During 7 days of R the capillary density returned toward control levels. In summary, the reduction in fiber cross sectional areas and increase in fiber and capillary densities support the hypothesis that in both forms of disuse, i.e., W and WBS, there is a loss in soleus muscle cell mass and not in fiber numbers.

Echocardiographic morphometry of the right chambers of the heart in permanent cardiac pacing.

Permanent cardiac pacing is a method of choice in the treatment of specific arrhythmias and conduction disturbances. Clinical studies show that cardiac performance diminished at the site of impulse spreading. It determines local hypotrophy below the position of the pacing lead (early electric activation) with hypertrophic changes in the opposite lying myocardium (late electric activation). It seems that morphological changes, especially research by intravital methods, so relevant in permanent pacing to today's invasive cardiologist, are not understood in full. In connection with this we decided, on the basis on the echocardiographic examination, to evaluate in detail the morphology of the right ventricle and atrium in patients with permanent pacing. Research was carried out on a group of 124 patients (68 males, 56 females) from 40-93 years of age (avg. 68 +/- 14 yrs): 86 patients had implanted pacemakers or AICD (group I), the control group consisted of 38 patients with other cardiac diseases without any pacemaker devices (group II). We measured echocardiographically the following diameters: end-diastolic and systolic diameters of the right ventricle/atrium in short and long axis, diameter of the tricuspid orifice valve and calculated area of the tricuspid orifice based on a special formula. Regarding the morphometric parameters of the right ventricle and right atrium, we confirmed that all diameters of group I were overshooting in correlation to group II. Those differences, such as RVd-short and -long, RVs-long, RVinflow, RA-long and -short, TRId, were statistically significant. Regarding the area of the tricuspid orifice (TRIa), we did not observe any changes in the two examined groups. We concluded that patients with implanted devices have changes in the morphometric parameters of the right ventricle, atrium and orifice, but they do not depend on the duration of pacemaker implantation.
