Altered B-lymphocyte membrane architecture indicated by ganglioside accessibility in C3H/HeJ mice.

We have analyzed both the total ganglioside composition and the surface accessibility of C3H/HeN B lymphocytes and C3H/HeJ B lymphocytes. Seventeen individual resorcinol-positive moieties were visualized by two-dimensional thin-layer chromatography of the purified gangliosides from both strains. Complete homology between strains was seen in the patterns of total gangliosides purified from the endotoxin-responsive and -hyporesponsive strains, with only minor differences in the relative concentrations of four gangliosides. In comparison, only 12 individual gangliosides were accessible to surface labeling following galactose oxidase treatment in these same strains, suggesting that some gangliosides are masked at the cell surface in both strains. However, labeling of the more polar components was greatly reduced in the endotoxin-hyporesponsive (C3H/HeJ) strain, suggesting that these gangliosides have decreased accessibility to galactose oxidase at the cell surface. Therefore, while the total ganglioside compositions of the two strains were nearly equivalent, there were dramatic differences in ganglioside surface accessibility. These findings indicate that an alteration in membrane structure that is associated with the endotoxin hyporesponsiveness observed in C3H/HeJ B lymphocytes exists.

OCT2 pre-positioning facilitates cell fate transition and chromatin architecture changes in humoral immunity.

During the germinal center (GC) reaction, B cells undergo profound transcriptional, epigenetic and genomic architectural changes. How such changes are established remains unknown. Mapping chromatin accessibility during the humoral immune response, we show that OCT2 was the dominant transcription factor linked to differential accessibility of GC regulatory elements. Silent chromatin regions destined to become GC-specific super-enhancers (SEs) contained pre-positioned OCT2-binding sites in naive B cells (NBs). These preloaded SE 'seeds' featured spatial clustering of regulatory elements enriched in OCT2 DNA-binding motifs that became heavily loaded with OCT2 and its GC-specific coactivator OCAB in GC B cells (GCBs). SEs with high abundance of pre-positioned OCT2 binding preferentially formed long-range chromatin contacts in GCs, to support expression of GC-specifying factors. Gain in accessibility and architectural interactivity of these regions were dependent on recruitment of OCAB. Pre-positioning key regulators at SEs may represent a broadly used strategy for facilitating rapid cell fate transitions.

Food store accessibility.

Although occupational therapists try to help their wheelchair-bound clients reenter the community or maintain independence, many facilities' noncompliance with accessibility standards makes such planning difficult. In the present study, a survey was used to investigate the wheelchair accessibility of 20 grocery and convenience stores. The results of the survey indicated that types of stores with greatest accessibility, in descending order, were (a) rural grocery stores, (b) urban grocery stores, (c) urban convenience stores, and (d) rural convenience stores. No overall differences were found between the urban and rural stores. The types of deficiencies found and their effects on clients' shopping are described. A follow-up study was conducted to see if the store managers, who were sent letters notifying them of their stores' deficiencies, would correct these problems. Six months after notification, the survey was repeated. Five of the 20 managers made changes in specific deficient areas, whereas the remaining 15 made no changes. The survey results and the fact that 25% of the stores made changes after receipt of a letter indicate that occupational therapists can be effective advocates for accessibility and thus provide a vital link to productive living for persons in wheelchairs.

[The handicapped and public transportation].

The article gives an overview of efforts made in various countries (the U.S.A., Canada, Sweden, and the Federal Republic of Germany) toward achieving greater accessibility of their public transportation systems. Various initiatives are shown to have been created in the 70ies on the part of public authorities and public transport providers, that have entailed the development of special vehicles, special information systems and architectural solutions. On analyzing the policies of both the transport providers and the competent public authorities it is, however, found that, although creation of an essentially barrier-free transportation network would be possible from the technical point of view, they refrain from subscribing to this objective. This has resulted in transportation of the disabled being oriented toward special services because these are far less costly than fundementally re-designing existing public transportation systems.
