Biochemical and cytochemical effects of cytosine arabinoside (ARA C) and hydrocortisone on HeLa cells.

The biochemical and cytochemical effects of cytosine arabinoside (Ara C) and of hydrocortisone on HeLa cell in vitro have been studied. In cultures treated with Ara C, a relationship exists between the cytocidal effects of the drug and an increase in alkaline phosphatase. Although hydrocortisone had no influence on the proliferative rhythm, it induced an increase in alkaline phosphatase. Results obtained with the cytochemical technique were evaluated, particularly in relation to enzyme localization.

Cytochemical effect of a stable PGI2 analog iloprost on guinea-pig heart in calcium-free and calcium paradox conditions.

Morphological and cytochemical effects of stable prostacyclin analog, iloprost were studied on isolated and perfused guinea-pig hearts according to Langendorff in Ca-free and Ca paradox conditions. Samples from the papillary muscles and left ventricular wall-muscles were examined. For calcium cytochemistry lead acetate and potassium pyroantimonate, and for nickel cytochemistry dimethylglyoxime (DMG) techniques were used. Electron microscopic studies showed that during Ca-free perfusion (for 15 min) and the following Ca repletion (for 15 min) disruption of the sarcolemma, damages in the sarcotubular system and in the mitochondria and gradual separation of intercalated discs were observed. Treatment with iloprost (1 microM) prevented those alterations in Ca-free and Ca paradox conditions. Iloprost caused a change in the intracellular distribution of Ca2+, decreased the cytochemically-demonstrated calcium in the cytoplasma, and inhibited the formation of calcium phosphate granules in mitochondria. Iloprost did not modify the number of nickel-DMG complexes either in Ca-free or in Ca paradox conditions. This study indicates that iloprost prevents the morphological damages caused by Ca paradox. The study also suggests that iloprost has an effect on the intracellular handling of calcium.

Expulsion of Echinostoma trivolvis (Cort, 1914) Kanev, 1985 and retention of E. caproni Richard, 1964 (Trematoda: Echinostomatidae) in C3H mice: pathological, ultrastructural, and cytochemical effects on the host intestine.

C3H mice were infected with 30 metacercarial cysts of either echinostome to study the pathological, ultrastructural, and cytochemical effects of the infection on the mouse small intestine. In mice infected with Echinostoma caproni, the intestine showed villous atrophy with fused or eroded villi. The microvilli of the enterocytes were sparse and distorted and showed reduced alkaline phosphatase activity. The crypts of Lieberkuhn were hyperplastic and showed a marked reduction in goblet and Paneth cells. As compared with uninfected controls, there was a marked reduction in glucose-6-phosphatase activity in the enterocytes of the infected gut. Collagen fibers and the number of fibroblasts were increased under the epithelium. In mice infected with E. trivolvis, the tips of the intestinal villi were bent and blunted. The microvilli of the enterocytes were less tightly packed than those of uninfected controls. The mitochondria in the enterocytes were irregularly shaped, contained intracristal bodies, and showed increased cytochrome oxidase activity as compared with those of uninfected controls. The crypts were hyperplastic but showed an increase in the numbers of goblet and Paneth cells. The fibroblasts and collagen fibers showed abnormal development. The ultrastructural and cytochemical differences seen in this study reflect the uniqueness of the host-parasite relationship of each of these echinostome species in the gut of the C3H mouse.

Ultrastructural and cytochemical effects of trypan blue on TSH stimulation of thyroid follicular cells.

Ultrastructural and cytochemical techniques were used to study the effects of trypan blue on the response of mouse-thyroid cells to exogenous stimulation by thyroid stimulating hormone (TSH). The dye delayed the response to TSH resulting in decreased colloid-droplet formation in the apical region of the cells. The dye did not stop the shift of trimetaphosphatase activity from lysosomes to phagolysosomes. The duration of the TSH-induced response was shorter in the dye treated thyroids. Small vesicles, with trimetaphosphatase reaction product, were found near Golgi elements, phagolysosomes, and the plasma membrane facing rhe intercellular space of adjacent follicle cells. Their enzyme activity was not affected by exposure to the dye. These data indicate that the primary effect of trypan blue on the response of thyroid follicle cells to TSH stimulation was reduced endocytosis in the apical region resulting in fewer colloid droplets.

[Cytological and cytochemical effects of a liver extract from an adult rat on chick embryo hepatocytes and fibroblasts cultivated in vitro].

The effects of an extract from adult liver on chick embryo hepatocytes and fibroblasts cultivated in vitro are studied with cytological and quantitative cytochemical methods. This substance inhibits the multiplication of hepatocytes adn prevents these cells from entering into the S phase of the cycle. No effect is observed on the chick embryo fibroblasts. Not any morphological alterations are observed in the two cell types.

Cytochemical effects of in vitro dopamine treatment on the Na-KATPase activity in strial marginal cells.

Dopamine is often used clinically for the treatment of patients with shock. In a previous study, the Na-KATPase (K-NPPase) activity of strial marginal cells was inhibited after the repeated in vivo administration of dopamine hydrochloride. In the present study, the K-NPPase activity of strial marginal cells was determined using a cerium-based cytochemistry after an in vitro incubation with dopamine. The enzyme reaction product was found in untreated normal strial marginal cells, but it was almost completely undetectable after an in vitro treatment with 10 mM dopamine. This finding suggested that dopamine directly inhibited the Na-KATPase activity of strial marginal cells, and that these cells might have a dopamine receptor.

[Hematologic and cytochemical effects in specialists working with radiolocalization and radio navigation means].

Examination of workers servicing radio location, radio navigation and communication means proved electromagnetic radiofrequency waves to induce some peripheral blood changes: cytopenia, Hb decrease, lower RBC and WBC counts, increased RBC with basophilic granularity, WBC metabolism alteration (higher acid phosphatase and myeloperoxidase activity), disordered lymphocytes subunits (T-helpers, T-suppressors) ratio and T- and B-cells numbers.
