The role of action reflexes in the damping of mechanical oscillations.

Reflex responses measured during voluntary contraction of the muscle being stretched are known to differ markedly from reflex responses elicited from passive muscle. The term 'action tonic stretch reflex' or 'action TSR' has been used previously to describe a reflex response to continuous stretch, separated from voluntary activity by means of a cross-correlational and spectrographic analysis. In this paper it is proposed that the action TSR play a functional role during voluntary movement by damping the transient oscillations associated with the natural resonant frequencies of the limbs. It is suggested that oscillations excite an action TSR force response with phase lead ahead of muscle stretch. This force response can be resolved into two force components, one of which has a 90 degree phase lead ahead of muscle stretch and behaves like a viscous friction reaction force causing damping of oscillations. Three experiments which support this proposition are described. (1) Analog computer model stimulation studies of a muscle supporting a mass-spring load reveal that damping only occurs when the force response has a phase lead ahead of muscle stretch. When the force response has a phage lag behind muscle stretch, the system is unstable and the amplitude of oscillation increases with time. (2) It is demonstrated that when a mass-spring load is supported as rigidly as possible by the human arm, reaction forces from the arm damp and mass-spring oscillations more rapidly than when the mass-spring is rigidly supported. Electromyogram (EMG) recordings reveal that mass-spring oscillations excite action TSR responses with phase lead ahead of muscle stretch. (3) Recordings of elbow angle and biceps EMG during rapid forearm flexion or extension movements followed by sudden stops reveal critically damped oscillations in the elbow angle signal (i.e. no more than one or two small overshoots), which are accompanied by EMG action TSR responses with phase lead ahead of muscle stretch.

Distortion of non-invasive cardiac pulse curves. A capillary-damped pick-up and a calibration unit forapex cardiograms and other pulse curves.

A calibration unit and a capillary-damped pick-up, of funnel type, intended for apex cardiograms and other non-invasive cardiac pulse curves are presented. The physical properties of a recording system, suitable for non-invasive cardiac diagnostic techniques, are reviewed. In 4 identical recording units consisting of a funnel with and without capillary damping, connected via air transmission to a transducer, the overamplification in the underdamped systems varied between 2-4 and 4-3, and in the capillary-damped systems between 1-1 and 1-2. The capillary damping gave an approximately linear relation between phase shift and frequency in the high frequency range with a calculated delay of about 4ms. The low frequency time constant in the systems varied between 1-9 and 4-6 s, depending on the amplifier setting used, which for sine curves of frequency 0-5 and 2 Hz means an apparent prematurity for their maxima and minima varying between 53 and 1 ms, this being larger with lower frequency time constants and lower frequency curves.

Studies with a new antifolate 2,4-diamino-5-adamantyl-6-methylpyrimidine (DAMP): tissue distribution and disposition of 2,4-diamino-5-adamantyl-6-methylpyrimidine and its metabolite.

The pharmacokinetics of a potentially useful antitumor agent, 2,4-diamino-5-adamantyl-6-methylpyrimidine (DAMP) has been studied in rat using 14C-labeled drug. It is reported that DAMP is metabolized rapidly by liver microsomes to an unidentified compound. The metabolite seems to be excreted by liver more rapidly than the unchanged drug, and within 24 hours all radioactivity is eliminated, 80% through kidney and 20% through the bile. Both DAMP, and its metabolite accumulate readily in pancreas and kidney. In contrast, in the brain mostly unchanged DAMP could be detected. The accumulation of the drug in tissues other than liver is much greater after i.v. than after i.p. administration.

Damped oscillations in continuous culture of Lactobacillus plantarum.

Lactobacillus plantarum exhibited long-period damped oscillations when grown aerobically with glucose as the rate-limiting substrate. Hydrogen peroxide accumulated in the cultures and its concentration also exhibited damped oscillations. It is suggested that inhibition of growth by hydrogen peroxide is involved in the production of the damped oscillations of microbiol population density.

Integrated analysis of plasma metabolomics and proteomics reveals the biological characteristics of damp-heat and stasis-toxin syndrome in colorectal cancer.

OBJECTIVE: To investigate the biological attributes of core syndromes in colorectal cancer, namely, the damp-heat and stasis-toxin syndrome (SRYD). METHODS: Between October 2021 and October 2022, a cohort comprising 40 patients with colorectal cancer (CRC) diagnosed with damp-heat and stasis-toxin syndrome (SRYD group), 40 patients with CRC without this syndrome (non-SRYD group), and 40 healthy controls (Normal group) was recruited at Jiangsu Province Hospital of Chinese Medicine. Untargeted metabolomics analysis was conducted on plasma samples from all 120 participants, while differential protein analysis using four-dimensional data-independent acquisition proteomics was performed on 20 randomly selected samples per group. A combined analysis of proteomics and metabolomics data followed, and the identified potential diagnostic biomarkers were subsequently used to train and validate multiple machine learning models. RESULTS: Proteomic analysis revealed 130 differential proteins in the colorectal cancer with damp-heat and stasis-toxin syndrome (CRC-SRYD) group, enriched in pathways including complement and coagulation cascades, as well as nuclear factor kappa-B (NF-κB) signaling. Metabolomic analysis identified 584 differential metabolites within the same group, showing enrichment in pathways such as primary bile acid biosynthesis, central carbon metabolism in cancer, and glucagon signaling. Integrated pathway analysis indicated heightened activity of the NF-κB signaling pathway in the CRC-SRYD group. A biomarker panel, comprising 6 proteins and 9 metabolites selected through the ReliefF algorithm, was used to construct a diagnostic model with random forest, achieving an accuracy of 93.33%, sensitivity of 80.00%, and specificity of 100%. CONCLUSION: This study systematically elucidates plasma metabolomic and proteomic alterations in patients with CRC, establishing a robust diagnostic model for CRC syndrome (CRC-SRYD). Further investigation is warranted to clarify the underlying molecular mechanisms and biological foundations.

[Ecological study of Pyroglyphides mites in house dust in the Grenoble area. A qualitative approach and incidence of various parameters: seasons, altitude, temperature and relative dampness in their multiplication].

This study concerns the analysis of 70 samples of dust mites taken periodically from 13 sample areas or mattresses at different altitudes. It shows the presence of Pyroglyphid mites in all the mattresses studied. Four species were found: Dermatophagoides pteronyssinus, Dermatophagoides farinae, Euroglyphus maynei and Dermatophagoides evansi. From a quantitative point of view, the statistical analysis of the results obtained allows the following conclusions to be drawn: -- All the Pyroglyphids (adults and young) undergo seasonal fluctuations at low, middle and high altitude. In the plains and at middle altitude these mites have a seasonal maximum and minimum. At high altitude only the seasonal maximum can be determined. -- The number of Pyroglyphids found is much higher in the plaine than at middle altitude and at middle altitude than at high altitude. The influence of altitude can be demonstrated with certitude. -- The conditions of temperature and relative dampness most favourable to the multiplication of mites are found in the plaine. At middle and high altitude these conditions are only met in summer. In flat country the statistical analysis allows us to show the existance of a correlation between the rate of dampness and the number of Pyroglyphids, and more specially, between the dampness rate and Dermatophagoides farinae, a predominant species at this sampling area. At middle and high altitude such a correlation cannot be shown clearly. At the three altitudes considered, no correlation can be established between the temperature and the number of Pyroglyphid mites found.

Regulation of IgE antibody production by serum molecules. V. Evidence that coincidental sensitization and imbalance in the normal damping mechanism results in "allergic breakthrough".

Experiments presented in this paper were designed to test a new concept concerning the possible pathogenesis of the allergic phenotype. This concept, termed "allergic breakthrough" considers that one of the avenues toward the allergic phenotype involves coincidental sensitization combined with an imbalance in the normal damping mechanism that serves to limit IgE antibody production. The three predictions of this concept that can be tested experimentally are: 1) manipulations that are effective in heightening or re-establishing the damping mechanism should manifest persistence insofar as IgE antibody synthesis to the relevant allergen is concerned; 2) once allergic breakthrough has occurred, the height of production of IgE antibodies specific for the sensitizing agent should remain elevated at levels characteristic of the allergic phenotype, even after the threshold of damping activity has returned to a normal level; and 3) allergic breakthrough should display specificity in that breakthrough would occur in response to subsequent exposure to the specific antigen to which coincidental sensitization initially occurred, but not for other unrelated antigens. The studies presented herein confirm each one of these predictions, thereby providing substantial support for the validity of this concept as one possible distinguishing feature between individuals manifesting the nonallergic and allergic phenotypes, respectively.

Electrocardiographic and serum enzymic alterations associated with cardiac alterations induced in dogs by single transthoracic damped sinusoidal defibrillator shocks of various strengths.

The safety margin between the strength of shock needed to defibrillate the ventricle and shocks which produce cardiac damage has not previously been reported. This study quantitates the shock intensity required to produce ECG alterations, serum alpha-1LDH and MB CPK isoenzyme elevation and myocardial damage using single transchest damped sinusoidal defibrillator shocks. Shocks of 1 to 20 amperes per Kg. of body weight were applied. Fifty-six dog weighing 2.4 to 15 kilograms were shocked with defibrillator pulses via 10 centimeter diameter electrodes applied to the thorax. Electrocardiograms were taken to be analyzed for arrhythmias, S-T segment changes, and T wave changes. Serum enzyme levels were determined in 25 dogs. Macroscopic and histopathologic studies were conducted on the hearts. Transchest single damped sine wave shocks did not produce microscopically detectable cardiac damage until at least a threefold current overdose was applied. No macroscopic morphologic alterations were observed until at least a sixfold current overdose was applied and no deaths occurred until a twelvefold or greater current overdose was delivered. Incidence and severity of ECG changes, increase in serum enzyme activity, incidence and severity of cardiac damage, and incidence of mortality all correlated positively with shock strength. However, these four adverse effects did not correlate well with each other. Transient ECG changes were very frequent following shock application regardless of the morphologic damage produced, and hence the transient changes have little value as indicators or predictors of damage. Persistent ECG changes were predictive of morphologic changes but were not sensitive enough to detect damage in mildly injured hearts. Likewise, elevated serum cardiac isoenzyme activity was a reliable but insensitive indicator of damage.

[Damping of oscillations in arterial pressure in the internal carotid arteries].

In dogs, recording of fluctuations of the inlet and outlet pressure in the internal carotid artery showed a decrease of the amplitude of fluctuations along the artery, the damping index (Id) being about 60-90%. The Id was almost unchanged when the skull was either intact or largely opened. The amplitude of blood pressure fluctuations in the circle of Willis was always decreased after occlusion of the vertebral arteries, being evidently unrelated to superposition of the pulse waves that reach the circle of Willis through the internal carotid and the vertebral arteries. Therefore the decrease of the amplitude of pulsation was dependent on the damping of the pulsatile waves along the internal carotid arteries. The rise of the tone of the latter increased the Id, and vice cersa. The Id was almost independent of the size of amplitude and of the frequency of the fluctuations of the systemic arterial pressure, but decreased when the latter rised.

The damped pendular rotation test in central vestibular disorders.

Nystagmic rhythm in the damped pendular rotation test (DPRT) was analysed electronystagmographically both in healthy subjects (control) and patients with peripheral and central vestibular lesions. The patients were tested in a semi-dark room with eyes open and, in addition, a serial subtraction task was performed to maintain the state of alertness throughout the rotation. In the controls 49 years of age and under dysrhythmia was found in 11%, while in persons 50 years of age and over the abnormal rate was 29%. In patients with acoustic tumors, who had undergone surgery in which the translabyrinthine and middle fossa approach had been utilized, dysrhthmia was present post-operatively in only a few cases. In patients 49 years of age and under, operated on through the suboccipital approach, dysrhythmia was found post-operatively, or was at least more remarkable than pre-operatively in most cases. Nystagmic rhythm was, on the other hand, regular with cerebellar degenerative process or atrophy. Pre-operative dysrhythmia in patients with cerebellar and cerebral tumors was in some cases post-operatively converted to regular rhythmic nystagmus in pendular stimulations. Therefore, in cerebellar and cerebral tumors, dysrhythmia was thought to be due to secondary effects in brainstem, as these tumors can cause intracranial hypertension and circulatory disturbances in surrounding brain tissues. In Menière's disease and sudden deafness, dysrhythmia and salvos in DPRT were found in the age groups, 49 years of age and under and 50 years and over, as frequently as in the healthy and control group. In one case with Menière's disease which showed dysrhythmia, labyrinthectomy was done. This surgical manipulation of the labyrinth did not eliminate dysrhythmia, though a temporary improvement was obtained. This result indicates that the dysrhythmia may be of central origin. These findings also suggest that nystagmic rhythm in DPRT with mental arithmetic is significant in assessing brainstem dysfunctions, when the age and state of alertness of the patient are taken into consideration, or when comparing the pre- and post-operative findings in intracranial lesions.

A data analysis microcomputer package (DAMP) for biomedical signals.

The advent of cheap, powerful microcomputer systems makes the analysis of data via sophisticated techniques available to the personnel who are non-specialists in computing systems. The DAMP package described here is intended for use on personal computers and has therefore been written in BASIC for portability. The analysis techniques are powerful, comprising algorithms to perform sample-data generation, plotting displays, digital data filtering, auto-correlation functions, fast Fourier transforms and autoregressive modelling. The last technique contains a number of options including the display of z-plane plots, frequency response of the model, residual plotting and auto-correlation of the residuals. Illustrative results are shown from psychological mood data and rat locomotor activity. The package is designed both to instruct a user in the techniques of spectral analysis, and also to provide a range of methods for investigating time and frequency behaviour of biomedical data.

Increased acetylation of H3K14 in the genomic regions that encode trained immunity enzymes in lysophosphatidylcholine-activated human aortic endothelial cells - Novel qualification markers for chronic disease risk factors and conditional DAMPs.

To test our hypothesis that proatherogenic lysophosphatidylcholine (LPC) upregulates trained immunity pathways (TIPs) in human aortic endothelial cells (HAECs), we conducted an intensive analyses on our RNA-Seq data and histone 3 lysine 14 acetylation (H3K14ac)-CHIP-Seq data, both performed on HAEC treated with LPC. Our analysis revealed that: 1) LPC induces upregulation of three TIPs including glycolysis enzymes (GE), mevalonate enzymes (ME), and acetyl-CoA generating enzymes (ACE); 2) LPC induces upregulation of 29% of 31 histone acetyltransferases, three of which acetylate H3K14; 3) LPC induces H3K14 acetylation (H3K14ac) in the genomic DNA that encodes LPC-induced TIP genes (79%) in comparison to that of in LPC-induced effector genes (43%) including ICAM-1; 4) TIP pathways are significantly different from that of EC activation effectors including adhesion molecule ICAM-1; 5) reactive oxygen species generating enzyme NOX2 deficiency decreases, but antioxidant transcription factor Nrf2 deficiency increases, the expressions of a few TIP genes and EC activation effector genes; and 6) LPC induced TIP genes(81%) favor inter-chromosomal long-range interactions (CLRI, trans-chromatin interaction) while LPC induced effector genes (65%) favor intra-chromosomal CLRIs (cis-chromatin interaction). Our findings demonstrated that proatherogenic lipids upregulate TIPs in HAECs, which are a new category of qualification markers for chronic disease risk factors and conditional DAMPs and potential mechanisms for acute inflammation transition to chronic ones. These novel insights may lead to identifications of new cardiovascular risk factors in upregulating TIPs in cardiovascular cells and novel therapeutic targets for the treatment of metabolic cardiovascular diseases, inflammation, and cancers. The undamped and damped series elastic components of a vascular smooth muscle.

Small arterial resistance vessels (internal diameter about 175 micrometer) have been mounted on a myograph that enabled their wall tension, T, and internal circumference, L, to be measured and controlled with a time resolution of about 4 ms. Maximally activated vessels were subjected to isometric releases (step changes in L) and isotonic releases (step changes in T) of varying extents and at two different temperatures (27 degree C and 37 degree C). The recovery from an isometric release was monotonic and did not include the two phases seen in skeletal muscles. The isotonic release response did, however, contain a velocity transient lasting about 150 ms: the velocity immediately after the release was about six times the steady shortening velocity. The form of both the isometric and isotonic release responses and their dependence on the extent of release can be explained in terms of a modified Hill model in which the "series elastic component" (SEC) is replaced by the series combination of an undamped-SED (that is, an undamped elastic element) and a damped-SEC (a Voigt element). Although the initial response to both types of release was independent of temperature, all stages of subsequent responses were temperature dependent, with Q10's in the range 1.5 - 2.0. The results suggest that the responses to isotonic and isometric releases may in part be due to active processes.

Ultrastructure of the turkey's basilar papilla and the damping elements in the scala media.

The ultrastructure of the turkey's inner ear was investigated by use of scanning and transmission electron microscopes. It was shown that hair cells of the basilar papilla are separated from each other by microvilli of the supporting cells. Organized compartments in the lower surface of the tectorial membrane of the turkey are arranged in polarized fashion as are the hair bundles. Kinocilia were not identified in the hair bundles examined in this study. Well-organized filamentous elements were observed in the cutucular plates. The continuation of these filamentous elements into the stereocilia indicates that the theory of adjustable resistor in generating cochlear potential may be closely associated with these elements. Membrane bound homogeneous material was found in the scala media between the tegmentum vasculosum and the tectorial membrane. The presence of this homogeneous damping material is believed to be essential in offsetting the sensitivity and enhancing the time resolution ability of the relatively short avian cochlea.

[Damping method of traction].

Damping of the system of skeletal extension with steel coil springs, ball bearing blocks and capron fishing-line was shown by the authors to be a necessity, since the routinely used "rigid" systems proved to be less effective. The authors have elaborated a new method of elimination of angular deviations of fragments and dislocations in width.

Cardiac damage produced by transchest damped sine wave shocks.

High-energy transchest damped wave sine defibrillation shocks have been shown to produce cardiac damage when applied in rapid sequence. However, there are no reports as to whether single, threshold-intensity shocks produce damage. In this study, nonfibrillating dogs were subjected either to a single, threshold-intensity (1 A/kg) shock, or to a series of 6 shocks with high intensity (3 to 4.8 A/kg). Electrodes of 8 cm diameter were used to apply the shock through the chest wall. Dogs receiving shocks of adequate (but not excessive) strength to defibrillate showed no cardiac damage, although they exhibited transient ventricular arrhythmias after the shock was applied. All dogs receiving the higher intensity, multiple shocks showed gross and microscopic evidence of cardiac damage. Ventricular lesions were observed in both right and left ventricular free walls and were sometimes transmural in extent. ECG analysis of the records from the dogs receiving multiple, high-intensity shocks showed second and third degree A-V block, ventricular ectopic beats, ventricular tachycardia, S-T segment changes, and T-wave inversion. Although multiple, high-energy, high-current defibrillation shocks produce permanent cardiac damage in dogs, threshold shocks do not produce morphologic changes.

Evaluation of the operating internal resistance, inductance, and capacitance of intact damped sine wave defibrillators.

A method is developed for determing actual values of circuit elements in a damped sine wave (Lown waveform) defibrillator solely from measurements of the output, using two or more power resistors and a storage oscilloscope. If a defibrillator containing capacitance C, inductance L, and internal resistance RI, is discharged into increasing 5- to 100-ohm resistive loads R, it is shown for underdamped output waveforms that â = RI/2L + R/2L and ĉ = CRI + CR, where â = pi[t2tan(pit1/t2)], ĉ = 2â/[â2 + (pi/t2)2], t1 = time from onset to peak, and t2 = time from onset to first zero crossing of the output waveform on the oscilloscope trace. Linear plots of â vs R are constructed for seven defibrillators, and values of RI and L computed as intercept/slope and 1/(2 slope) respectively. C is given by the slope of a linear plot of ĉ vs R. Delivered energy is accurately predicted as stored energy X R/(RI + R).
