[Epidemiology of dysentery in Dagestan].

Analysis of material on bacterial dysentery and other acute intestinal infections morbidity in the Dagestan ASSR for a period of 15 years showed high morbidity level and its variations, with reduction and elevation in individual years, and the leading role played by Shigella flexneri among the causative agents of dysentery. The greatest incidence of dysentery was revealed among children aged under one year and between 1 and 2 years. A high bacterial dysentery incidence was recorded in Dagestan throughout the whole year. However analysis of the seasonal dysentery curve showed the beginning of elevation in July, reaching the maximum in August, and lasting four months with a decline beginning in October. The persisting activity of the water route of dysentery transmission in the Dagestan ASSR requires particular attention to the organization of good-quality water-supply to the population of the republic.

[Comparison of the nematode fauna of winter wheat and wild grain in the foothill and lowland zones of Dagestan].

The nematode fauna of winter wheat and wild cereals in the foothill zone of Dagestan is richer than that in the lowland zone. From 117 species of nematodes recorded from the rhizosphere and organs of examined plants 105 species are reported from foothills and only 78 species from the lowland zone. In the foothill zone the average number of nematodes in 10g of root soil and organs of wheat amounts to 619 specimens, in wild cereals--to 347 specimens, in lowland zone--to 310 and 236 specimens respectively. This difference is caused by soil--climatic conditions.

[Seasonal problems of bacterial dysentery in the Dagestan ASSR].

During the period of 1959-1973 bacterial dysentery showed high morbidity rate in Daghestan in all seasons, this tendency existing both in urban and rural areas. The peculiarity of the monthly distribution of dysentery cases, observed all over the repulic, was the shift of the morbidity peak from August to September-October, and in some years even to November; this could be clearly observed from the analysis of the material as a whole and from the analysis of dysentery morbidity rates in rural and urban areas separately. This rise in dysentery morbidity rate in August--September might be due to the reassembling of children at various institutions after their mass return from summer resorts in the country, as well as to thhe influx of new children to crèches and kindergartens. Moreover, as the period of August--September is the bathing and fruit-harvesting season in Daghestan, a rise in morbidity rate in these months was obviously caused by the unsatisfactory level of sanitation and hygiene due to an acute deficiency of water.

[Effect of ethnic affiliation and the high altitude factor on the tubular bones of the hand in children from Dagestan (x-ray osteologic study)].

Dimentions of tubular bones were compared concerning ethnic and climatic-geographical peculiarities of populations (under the conditions of ethnic homogenity of the groups). The data obtained demonstrated that Avarian people living at the altitude of 2,000 m differ from Avarian people living at the altitude of 1,000--1,100 m and from Kumyk people living at the foothills (500--600 m) in faster growth of the tubular bones of the hand and in their larger dimentions. Activation in bone growth of the Avarian people living high in the mountains results in "discoordination" of increment values of separate dimentions and in decrease of the correlation level.

Development of a PCR-based technique for genotyping UGT1A1 gene and distribution of rs3064744 alleles in the Russian population.

BACKGROUND: Accurate determination of tandem thymine-adenine (TA) repeat numbers in the UGT1A1 promoter region (rs3064744) is essential for diagnosing Gilbert's syndrome and personalizing therapy with toxic agents like irinotecan and atazanavir. However, traditional polymerase chain reaction (PCR) assays face severe limitations due to the AT-rich sequence and overlapping melting temperatures (Tm) of the highly homologous 7TA and 8TA alleles. In this context, melting curve analysis (MCA) employing fluorophore-quencher systems has emerged as a promising alternative. The purpose of this study was to develop a novel genotyping approach combining optimized aPCR-MCA analysis with an automated classifier to overcome the limitations posed by the differentiation of highly homologous alleles and to demonstrate its practical application, providing the distribution of rs3064744 genotypes across four regional cohorts of the Russian population. METHODS: A specialized Dual Head 1D-convolutional neural network (1D-CNN) ensemble with Test-Time Augmentation (TTA) was developed. The model was trained and internally validated on 1,620 engineered plasmid samples, and independently evaluated on an external clinical test set of 440 unique patient genomic DNA specimens. Real-time PCR was performed on CFX96 and DTprime platforms. Additionally, population-wide screening was conducted on 997 archival clinical samples from Moscow, Sakha (Yakutia), Dagestan, and Rostov regions. RESULTS: While 5TA and 6TA alleles were easily separated, absolute Tm distributions of 7TA and 8TA alleles overlapped significantly, and non-uniform Tm shifts of 0.8 °C-1.4 °C occurred across platforms. Conventional absolute Tm thresholding was therefore inadequate. By assessing relative morphological curve divergence against co-amplified 7TA/7TA and 7TA/8TA reference anchors, the 1D-CNN ensemble neutralized instrument noise. It achieved 100% accuracy on internal validation and 100% concordance (440/440) with clinical reference pyrosequencing. Population screening revealed that Dagestan, Yakutia, and Rostov cohorts closely align with the European population. Rare 5TA and 8TA alleles were detected at low frequencies in Yakutia and Moscow. CONCLUSION: Combining LNA-modified aPCR-MCA with a comparative 1D-CNN model successfully circumvents thermodynamic limitations and eliminates human operator bias. This integrated system offers an accessible, high-throughput, and clinically valid solution for routine UGT1A1 pharmacogenetic testing.

[New data on the ambiguous nature of the effects of exogamy on human growth processes].

Growth peculiarities were followed in children of pre-school and school age (Ukrainians from Dnepropetrovsk Region, Kumyks, and Avartses from Dagestan, Tadjiks, Usbeks and Kirghizes from Middle Asia) with a special reference to exogamy estimated both by the presence or absence of relationship between their parents and by the distance between their birthplaces denoted as 0, I and II degrees of exogamy (DE). In every ethnic groups investigated with transition from 0 to DE to 1 DE increase in growth activity was noted. Transition from 1 DE to 11 DE in Ukrainians, Kirghizes and Usbeks was accompanied by some decrease in growth activity, while in Kumyks and Avartses it remains at the same level. The differences noted could be explained by various demographic conditions in population of the regions studied. As the studies demonstrated in Ukrainian children, activation in growth processes in the population with 1 DE is followed by the appearance of some peculiarities in dermatoglyphics and odontoglyphics which were absent in children with 0 DE and 11 DE. The data obtained demonstrate a certain role of genetic factors in developmental acceleration displaying at a moderate degree of exogamy.

[Role of the aqueous factor in the epidemiology of dysentery].

Analysis of the dynamics and territorial distribution of bacterial dysentery morbidity during 1959-1973 in the Dagestan ASSR demonstrated that the epidemically unfavourable conditions were largely connected with the use of water in open water bodies for domestic purposes and swimming as well as a wide network of irrigation canals. Persistence of the water factor in the dysentery epidemiology was connected with the ever increasing contamination of water in open water bodies and neglect of sanitary-hygienic requirements of the water supply to population of pertinent and sanitary laws.
