Two new cyclopentenone derivatives and a new cyclooctadienone derivative from Erigeron annuus (L.) PERS., Erigeron philadelphicus L., and Erigeron sumatrensis RETZ.

Two new cyclopentenone derivatives, erigerenones A (1) and B (2), and a new cyclooctadienone derivative, erigerenone C (3), were isolated from the aerial parts of Erigeron philadelphicus L. Compound 2 was also isolated from the aerial parts of Erigeron annuus (L.) PERS. and Erigeron sumatrensis RETZ. The structures of 1-3 were elucidated on the basis of their spectral data.

Five new sesquiterpenoids and a new diterpenoid from Erigeron annuus (L.) PERS., Erigeron philadelphicus L. and Erigeron sumatrensis RETZ.

The aerial parts of Erigeron annuus (L.) PERS., E. philadelphicus L. and E. sumatrensis RETZ. (Compositae) have been investigated chemically. A new sesquiterpenoid, 6beta,14-epoxyeudesm-4(15)-en-1beta-ol (1), and a new diterpenoid, philadelphinone (6), have been isolated from E. philadelphicus. Four new sesquiterpenoids, (7R*)-opposit-4(15)-ene-1beta,7-diol (2), 11-methoxyopposit-4(15)-en-1beta-ol (3), 15-methoxyisodauc-3-ene-1beta,5alpha-diol (4) and 10alpha-hydroxycadin-4-en-15-al (5), have been isolated from E. annuus. Compounds 2 and 4 were also isolated from E. sumatrensis. The structures of the new compounds were elucidated on the basis of their spectral data.

[Study on effects of Erigeron breviscapus extract on anticoagulation].

OBJECTIVE: To study the effect of Erigeron breviscapus extract on anticoagulation. METHOD: Coagulation time (CT), prothrombin time (PT), platelet factor III (PF3) and euglobulin lysis time (ELT) have been used to evaluate the anticoagulation activities of Erigeron breviscapus extract. RESULT: Erigeron breviscapus extract significantly delayed CT and PT, inhibited the activity of PF3, and decreased ELT. CONCLUSION: Erigeron breviscapus extract plays an role in anticoagulation by inhibiting PF3 and prothrombin V. It can also enhance the activity of fibrimolysis.

Diplospory and Parthenogenesis in Sexual x Agamospermous (Apomictic ) Erigeron (Asteraceae) Hybrids.

Segregation for asexual seed production was evaluated for 130 experimental F1 hybrids resulting from a cross between diploid (2n=18) sexual Erigeron strigosus and triploid (2n=27) agamospermous Erigeron annuus. Paternity of hybrids was documented using 13 RAPD markers. The distribution of F1 chromosome numbers is bimodal, centering on diploid and triploid ploidal levels but with underrepresentation of diploids. Diplosporous versus meiotic megagametophyte development was ascertained microscopically for >/=100 ovules per plant. Diplospory ranges from 0% to 100% among all progeny but is uniformly low (0%-3%) for 17 diploid hybrids. The inheritance of diplospory in Erigeron appears to be best explained by a one-locus-two-allele polysomic model with selection against gametes homozygous for diplospory. Parthenogenesis, estimated via seed counts, ranges from 0% to 60% and apparently is contingent upon diplospory, as seed production was absent or very low in predominantly meiotic hybrids. However, the absence of parthenogenesis in many highly diplosporous hybrids indicates that these two aspects of agamospermous development are not strictly associated. The segregation of both diplospory and parthenogenesis in this population will permit further genetic dissection of these traits with molecular marker-based analyses.

[The effects of Erigeron breviscapus preparation on the imaging and neuronal metabolites at the early time points after reperfusion following the ischemic cerebral injury in rats].

OBJECTIVE: To investigate the effects of Erigeron breviscapus preparation on the imaging and neuronal metabolites after reperfusion in the ischemic cerebral injury in rats. METHOD: Twenty-three male Sprague Dawley rats with an average body weight of (165 +/- 15) g (mean +/- S) were used, and were randomly divided into two groups: group A rats (n = 11) underwent an operation of ischemic brain injury, group B rats (n = 12) underwent the same operation and received the treatment of Erigeron breviscapus preparation (1.5 mg.kg-1 weight, i.p.). The right middle cerebral artery occlusion (MCAO) of rats in both groups was induced by 5/0 nylon suture for 2 hours. The reperfusion was conducted for four hours and six hours respectively following MCAO. T2 weighted-imaging (T2WI) and 1H-magnetic resonance spectroscopy (1H-MRS) were performed, to study the changes of the imaging and the neuronal metabolites N-acetyl aspartate (NAA), creatine/phosphocreatine (Cr/PCr), choline (Cho) and lactose (Lac) in cerebrum following cerebral ischemia. RESULT: The hyperintense signals in the right cerebrum in group B decreased compared with those in group A, the T2 values decreased, the level of NAA increased, the ratio of Cr/NAA and Cho/NAA decreased, and no lactose was observed. The brain surface vessels of rats in group B were in the state of dilation. CONCLUSION: Erigeron breviscapus preparation is beneficial to the reestablishment of the blood circulation in the ischemic brain, and to the improvement of the neuronal metabolism and survival.

Analgesic and antiinflammatory activities of Erigeron floribundus.

This study was intended to evaluate the analgesic and antiinflammatory activities of an aqueous extract of Erigeron floribundus (H.B. & K) or (syn": Conyza sumatrensis (Retz) E.K. Walker) (Asteraceae). Phytochemical analysis was carried out using standard methodologies. The analgesic investigations were carried out against two types of noxious stimuli, chemical (formalin-induced pain and acetic acid-induced writhing) and thermal (hotplate and tail immersion tests). The effects following aspirin and naloxone pretreatments were also studied. For the antiinflammatory activities, the carrageenan-induced oedema of the hindpaw of rats was used and the paw volume measured plethysmometrically from 0 to 24 h after injection. This was compared to a standard drug indomethacin (10 mg/kg). The results were subjected to statistical analysis. The plant had saponins, flavonoids, glycosides, alkaloids, oils, phenols and tannins and significantly increased the reaction time of hotplate and immersion tests. It decreased the writhings of acetic acid-induced abdominal contractions and lickings of formalin-induced pain. Aspirin had no effect on hotplate and tail immersion tests but showed an effect on writhing test. These results showed that the plant had both central and peripheral acting effects and this was confirmed by its effect on both phases of formalin-induced pain. The extract also significantly decreased the rat paw oedema volume at 50 mg/kg and above. In conclusion, Erigeron floribundus has central and peripheral analgesic properties as well as antiinflammatory activities.

Inheritance of apomeiosis (diplospory) in fleabanes (Erigeron, Asteraceae).

Unreduced egg formation (apomeiosis) in flowering plants is rare except when it is coupled with parthenogenesis to yield gametophytic apomixis via apospory or diplospory. Results from genetic mapping studies in diverse apomictic taxa suggest that apomeiosis and parthenogenesis are genetically linked, a finding that is compatible with the conventional rationale that apomeiosis is unlikely to evolve independently because of deleterious fitness consequences. An Erigeron annuus (apomictic) x E. strigosus (sexual) genetic mapping population, however, included a high proportion of plants that were highly apomeiotic (diplosporous) but nonapomictic; that is, they lacked autonomous seed production. To evaluate the function and inheritance of diplospory in Erigeron, a diplosporous triploid (2n=3x=27) seed parent was crossed with a sexual diploid (2n=2x=18) E. strigosus pollen parent to produce an F1 of 31 plants. Chromosome numbers and molecular markers (AFLPs) document the inheritance of the maternal genome through unreduced eggs resulting in recombinant but predominantly (77%) tetraploid F1s (2n=4x=36; 2n+n, B(III)). Quantitative evaluation shows continuous variation in the proportion of diplosporous (vs meiotic) ovules (41-89%) in tetraploid F1s despite the presumed equal genetic contribution from the diplosporous mother. These findings demonstrate the functional independence of diplospory and suggest that variation in the trait in F1s is likely due to segregating paternal modifiers. In addition, of six aneuploid (4x-1, 4x-2) F1s, three lack a subset of maternal AFLP markers. These plants likely arose from aberrant megagametogenesis resulting in the loss of maternal chromatin prior to fertilization.

[Protective effect of erigeron injections on hepatotoxicity induced by intestinal ischemia-reperfusion in mice].

The liver injury was induced by intestinal ischemia-reperfusion and the protective effect of erigeron injection (EI) was studied in mice. The results showed that serum alanine transaminase (ALT) and glutathione S-transferase (GST) activity increased while the root of superior mesenteric artery was blocked with non-injury artery clap for 20 minute and reperfusion for 1 hour. EI could protect the above changes induced by intestinal ischemia-reperfusion. EI could reduce the activities of serum ALT and GST, decrease the content of hepatic MDA and significantly increase the activity of SOD. The pathological changes of hepatocytes induced by intestinal ischemia-reperfusion were less in EI groups. These results suggested that EI could protect the liver of mice from injury induced by intestinal ischemia-reperfusion. One of the mechanisms of hepatoprotective action was related to the antioxidative function of erigeron.

[Isolation and structure of 6'-O-caffeylerigeroside from Erigeron multiradiatus].

The plant, Erigeron multiradiatus (Wall.) Benth(Family: Compositae), is a perennial herb distributed abundantly in the mountainous area of the southwestern part in China. It is used in folk medicine for the treatment of common cold, panting cough, rheumatic, enteritis and toothache. In this paper, we report a new glycoside named as 6'-O-caffeylerigeroside isolated from Erigeron Multiradiatus (Family: Compositae) together with four known compounds. The known compounds were identified as pyromeconic acid(I), scopoletin(II), 4-hydroxybenzoic acid(III) and vanillic acid(IV). The structure of 6'-O-caffeylerigeroside was elucidated as gamma-pyrone-3-O-[6"-(3',4'-dihydroxy-cinnamoyl)]-beta-D-glucoside(V) on the basis of chemical evidence and spectral data.

Effects of Erigeron injection on vascular endothelium in patients after intracoronary stenting.

OBJECTIVE: To observe the levels of serum C-reactive protein (CRP), endothelin-1 (ET-1), nitric oxide (NO), and superoxide dismutase (SOD) in patients after intracoronary stenting (ICS), and the effects of Erigeron Injection (EI) on them. METHODS: Seventy-two patients, who received ICS and had symptoms of chest stuffiness, palpitation and chest pain, were randomly divided into two groups, with 36 patients in the control group treated with Plavix alone for anti-platelet aggregation, and the other 36 patients in the treated group treated with Plavix and EI in combination. CRP, ET-1, NO and SOD were determined and compared before and 1, 2 and 3 weeks after treatment. RESULTS: As compared with those in the control group, improvement of symptoms in the treated group was significantly better, with the levels of CRP and ET-1 lower and levels of SOD and NO higher or approaching to normal ranges and significant difference was shown between the two groups (P < 0.01). CONCLUSION: EI could alleviate uncomfortable feelings such as chest stuffiness in patients after ICS, and improve the function of vascular endothelium.

Effects of Erigeron breviscapus ethanol extract on neuronal oxidative injury induced by superoxide radical.

Antioxidant activity of the ethanol extract of Erigeron breviscapus (EEEB) were studied by using neuron oxidative injury model induced by superoxide radical. EEEB at the dose of 10-140 mug/ml reduced significantly the lipid peroxidation levels and lactic dehydrogenase (LDH) release from neuron exposed to superoxide radical (generated by xanthine and xanthine oxidase). These results suggest that neuroprotective actions of EEEB may be due to its antioxidant or radical scavenging activity.

Fungitoxic metabolites from Erigeron apiculatus.

Four enyne derivatives (1-4) and quercitrin were isolated during a bioassay-guided chromatographic separation of a methanolic extract of Erigeron apiculatus. Matricarialactone (1) and lachnophyllumlactone (2) showed a high fungitoxic activity against Pyricularia oryzae. Matricaria acid methyl ester (3) and lachnophyllum acid methyl ester (4) were, instead, less active.

Two-step purification of scutellarin from Erigeron breviscapus (vant.) Hand. Mazz. by high-speed counter-current chromatography.

Scutellarin, a flavone glycoside, popularly applied for the treatment of cardiopathy, has been purified in two-step purification by high-speed counter-current chromatography (HSCCC) from Erigeron breviscapus (vant.) Hand. Mazz. (Deng-zhan-hua in Chinese), a well-known traditional Chinese medicinal plant for heart disease. Two solvent systems, n-hexane-ethyl acetate-methanol-acetic acid-water (1:6:1.5:1:4, v/v/v/v/v) and ethyl acetate-n-butanol-acetonitrile-0.1% HCl (5:2:5:10, v/v/v/v) were used for the two-step purification. The purity of the collected fraction of scutellarin was 95.6%. This study supplies a new alternative method for purification of scutellarin.

Mendelian segregation for two-factor apomixis in Erigeron annuus (Asteraceae).

The inheritance of asexual seed development (apomixis) in Erigeron annuus (Asteraceae) was evaluated in a triploid (2n=3x=27) population resulting from a cross between an apomictic tetraploid (2n=4x=36) pollen parent and a sexual diploid (2n=2x=18) seed parent. Diplospory (unreduced female gametophyte formation) and autonomous development (embryo and endosperm together) segregated independently in the population yielding four distinct phenotype classes: (1) apomictic plants combining diplospory and autonomous development, (2) diplosporous plants lacking autonomous development, (3) meiotic plants with autonomous (though abortive) development and (4) meiotic plants lacking autonomous development. Each class was represented by approximately one-quarter of the population (n=117), thus corresponding to a two-factor genetic model with no linkage (chi(2)=2.59, P=0.11). Observations demonstrate that autonomous embryo and endosperm development (jointly) may occur in either reduced or unreduced egg cells. The cosegregation of the traits is attributed to tight linkage or pleiotropy. The data are consistent with the hypothesis that autonomous development in E. annuus is regulated by a single fertilization factor, F, which initiates development of both the embryo and the endosperm in the absence of fertilization.

Gamma-pyranone derivatives and other constituents from Erigeron annuus.

Two new gamma-pyranone derivatives and a new triterpene, together with eight known compounds, were isolated from the aerial parts of Erigeron annuus. Their structures were elucidated by spectroscopic methods. Besides, the anti-tumor activities of the gamma-pyranone derivatives to human hepatoma (SMMC-7721), human embryo liver (L-02) and human leukemia (HL-60) cells were evaluated.

Molluscicidal and antifungal activity of Erigeron speciosus steam distillate.

The steam-distilled fraction of the aerial parts of Erigeron speciosus (Lindl) DC was tested for activity against strawberry plant pathogenic fungi Botrytis cinerea Pers ex Fr, Colletotrichum acutatum Simmonds, C fragariae Brooks, C gloeosporioides (Penz) Penz & Sacc, and the intermediate host snail Planobdella trivolvis that harbors the trematode, Bolbophorus confusus, that infests and causes severe infections in pond-raised catfish in the Mississippi Delta region of the USA. Bioautography on silica TLC plates demonstrated antifungal activity in the steam distillate. Preliminary bioassays of the steam distillate indicated the presence of phytochemicals toxic to P trivolvis. The bioactive compounds methyl 2Z, 8Z-deca-2,8-diene-4,6-diynoate and its 2E, 8E isomer were isolated by bioassay-guided fractionation and chromatographic techniques and identified by 1H NMR spectroscopy.

The effect of Erigeron breviscapus on proliferation of pulmonary artery smooth muscle cells in hypoxic porcines.

In order to study the effect of Erigeron Breviscapus (EB) on proliferation of pulmonary artery smooth muscle cells (PASMC) in hypoxic porcines, immunohistochemical and MTT methods were employed to measure the proliferation of PASMC. It was found that the proliferation of PASMC in porcines was obvious, and the expression of proliferating cell nuclear antigen (PCNA) was significantly high within 48 h after exposure to hypoxia. The EB could inhibit the proliferation and the expression of PCNA in PASMC under hypoxia, but it had no effect on the proliferation and expression of PCNA in PASMC under normal condition. The EB could inhibit the proliferation and the expression of PCNA in PASMC induced by phorbol 12-myristate 13-acetate (PMA), an agonist of PKC in normal and hypoxic conditions. It was concluded that the hypoxia could enhance the proliferation and expression of PCNA in PASMC. The EB can inhibit the proliferation and expression of PCNA in PASMC under hypoxia through PKC-signal way. The EB may be used in treating the pulmonary hypertension by inhibiting the proliferation of PASMC and the pulmonary vascular remodeling.
