Estrous behavior and the estrus-to-ovulation interval in Nelore cattle (Bos indicus) with natural estrus or estrus induced with prostaglandin F2 alpha or norgestomet and estradiol valerate.

Estrous behavior and the estrus-to-ovulation interval are essential for estimating the best time to artificially inseminate cattle. Because these parameters are not well characterized in the Nelore breed (Bos indicus), the main purpose of the this study was to determine the estrus-to-ovulation interval in Nelore heifers and cows with natural estrus or with estrus induced by treatments with PGF2 alpha or norgestomet and estradiol valerate (NEV). The cows and heifers were observed continuously (24 h a day) to determine the onset of estrus and to study estrous behavior in the cows. Ten hours after the start of estrus the ovaries were scanned every 2 h by ultrasonography to monitor the dominant follicle until ovulation. Blood samples were collected periodically to determine progesterone levels by RIA. Administration of PGF2 alpha (2 injections, 11 days apart) did not induce estrus in most Nelore females in spite of the presence of functional CL, indicated by progesterone concentrations above 6.0 ng/ml in 25 of 28 animals. Treatment with NEV induced high sexual receptivity in cows (10/11), but only 66% ovulated. Cows with natural or induced estrus exhibited behavioral estrus of 10.9 +/- 1.4 h, and ovulation occurred 26.6 +/- 0.44 h (n = 26) after the onset of estrus. In most of the cows (53.8%) estrus began at night (between 1801 and 600 h), and 34.6% it started and finished during the night. It is concluded that in Nelore females ovulation occurs approximately 26 h after the onset of estrus. Additionally, estrous behavior is shorter than in European breeds, and there is a high incidence of estrus at night, which makes it difficult to detect and, consequently, impairs Al in Nelore cattle. The observation that a high percentage of Nelore females with an active CL did not respond to usual dosages of PGF2 alpha warrants further investigation.

A note on the effect of continuous ram presence on estrus onset, estrus duration and ovulation time in estrus synchronized ewes.

During fall season, 18 multiparous Corriedale ewes were divided into two equal groups for the continuous (CON) and intermittent (INT) presence of a ram. Estrus was synchronized with fluorgestone acetate intravaginal sponges that were left 14 days, plus an injection of 200&mgr;g of a prostaglandin F-2alpha analog at sponge removal. Estrus was detected three times a day (at 6 a.m., 2 p.m. and 10 p.m.) by using rams with harnessess and markers. Ovulation time was determined by laparoscopy, starting 24h after estrus detection. Estrus onset was (mean+/-S.E.M.) 32.9+/-1.6 and 45.3+/-4.4h for the CON and INT groups, respectively (P<0.01). Estrus duration was 31.1+/-0.9 and 30.2+/-1.2h, for the same groups, respectively (P>0.05). Ovulation time and the interval from sponge removal to ovulation (ISRO) for the CON and INT groups was 29.0+/-1.5, 62.0+/-2.0, 26.7+/-1.3 and 72.0+/-4.2h, respectively. Ovulation time was not different (P>0.05), but ISRO was shown to be different between treatments (P<0.05). It is concluded that the continuous presence of a ram after sponge removal hastens estrus onset and reduces the interval between sponge removal and ovulation, without modifying estrus duration and time between estrus onset and ovulation.

Chemical properties of bovine cervical mucus during normal estrus and estrus induced by progesterone and/or PGF2alpha.

Ninety-two Friesian cows were used to determine the chemical properties of cervical mucus during normal estrus and estrus induced by progesterone (P4)-releasing intravaginal devices (PRID) and/or prostaglandin F2alpha. The animals were assigned to 4 groups (no treatment, a PRID for 12 days plus injection of 1000 IU PMSG at the removal of PRID, a double i.m. injection of PGF2alpha 11 days apart, or PRID for 7 days plus an im injection of PGF2alpha 24 h before the removal of PRID). A number of cows with normal estrus exhibited three consecutive estrous cycles after delivery. Cows that had not shown estrus for 3 months after delivery had their ovaries palpated twice at 10-day intervals, to determine their ovarian activity. Then PRID and/or PGF2alpha was administered in cows that had a palpable corpus luteum in one of the two palpations (cyclic cows). A double artificial insemination (AI) was performed to the cows of the three induced-estrus groups, while the cows with normal estrus received only one AI. Cervical mucus samples were collected from all cows 5 to 30 min before the first AI. Additionally, samples of cervical mucus were collected from 20 cows during their first estrus after the induced one. The results are summarized as follows: 1) The biochemical properties of cervical mucus in the first three estrus periods after delivery were similar. 2) These properties were similar both in normal estrus after delivery and in the first estrus after an induced one. 3) Glucose and fructose concentrations for normal estrus were similar to those for induced estrus groups. 4) Total protein and cholesterol concentrations were significantly lower (P < 0.001) in normal than in induced estrus, while no difference was found among the induced estrus groups. 5) Pregnancy rates of the cows did not differ significantly among the normal and the induced-estrus groups. 6) The percentages of cows in the induced-estrus groups that produced cervical mucus with total protein and cholesterol concentrations similar to those for the normal estrus groups was very low.

The effect of administering a dopamine agonist (Cabergoline) on follicular and luteal development during pro-estrus and estrus in the female greyhound.

To study the effect of the dopamine agonist Cabergoline, on ovarian activity in the female dog during pro-estrus and estrus, 6 greyhounds were treated with 5 microg/kg per os of Cabergoline for 20 days beginning on the first day of pro-estrus; 6 animals were left untreated (controls). Ovarian morphology was determined by ultrasound examination once a day during pro-estrus and twice a day during estrus. Follicles were divided into three classes on the basis of their diameter: F1 (<3mm), F2 (3-6mm) and F3 (>6mm). The presence and diameters of post-ovulatory follicles (F-POST) and corpora lutea (CL) were also recorded. Blood samples were taken once a day during pro-estrus and twice a day during estrus. The plasma was assayed for LH, prolactin and progesterone by radioimmunoassay. There were no differences between Cabergoline-treated and control dogs in the duration of pro-estrus or estrus. There was a progressive increase in follicle diameter from the start of pro-estrus when follicles were mainly class F1 to the day of estrus when follicles were mainly class F3. Three days after the start of estrus, the first F-POST follicles were detected. This pattern of development continued and on day 5 the first CLs were detected. By day 9, only CLs were detected. The peak of pre-ovulatory LH was within 3 days of estrus and ovulation was detected in all animals within 3 days of the LH peak. There were no differences in LH concentrations between groups. Plasma prolactin levels varied between animals and were reduced in treated dogs, however, this was not statistically significant (P=0.12). Plasma progesterone levels were below 1.0 ng/ml before the LH surge and thereafter gradually increased. There were no differences in plasma progesterone concentrations between treated and control dogs. In conclusion these results show that the administration of the dopamine agonist Cabergoline during pro-estrus and estrus did not affect the duration of pro-estrus or estrus or the pattern of follicular and luteal development in female dogs.

Methods to reduce or eliminate detection of estrus in a melengestrol acetate-PGF2alpha protocol for synchronization of estrus in beef heifers.

Three experiments were conducted to evaluate methods to decrease or eliminate the detection of estrus inherent to a melengestrol acetate (MGA)-PGF2alpha (PGF) protocol for synchronization of estrus in heifers. In each experiment, all heifers received 0.5 mg of MGA x animal(-1) x d(-1) for 14 d (d -32 to -19) and PGF (25 mg, i.m.; d 0, 0 h) 19 d after the last feeding of MGA (MGA-PGF protocol). In Exp. 1, heifers (n = 709) were assigned to each of the following protocols: 1) the MGA-PGF protocol with AI 6 to 12 h after detection of estrus (estrus AI; MGA-PGF); 2) MGA-PGF plus 100 microg, i.m. of GnRH on d -7 (1x GnRH) and estrus AI; or 3) MGA-PGF, GnRH on d -7, and GnRH (100 microg, i.m.) at 48 h after PGF, coincident with insemination (2x GnRH-TB48). In Exp. 2, heifers (n = 559) received the MGA-PGF protocol and were inseminated by either estrus AI or fixed-time AI (TAI) at 60 h, coincident with an injection of GnRH (GnRH-TB60). In Exp. 3, all heifers (n = 460) received the MGA-PGF protocol and were inseminated by estrus AI when detected up to 73 h. Heifers not observed in estrus by 73 h received TAI between 76 and 80 h. Half the heifers inseminated by TAI received no further treatment (TB80), and the remaining half was injected with GnRH at insemination (GnRH-TB80). Variance associated with the interval to estrus and the proportion in estrus from d 0 to 5 was similar for 1x GnRH and MGA-PGF treatments in Exp. 1. Pregnancy rate (d 0 to 5) did not differ for the MGA-PGF and 1x GnRH treatments (62.5 and 60.4%, respectively), and both were greater (P < 0.05) than TAI pregnancy rate in the 2x GnRH-TB48 treatment (42.3%). In Exp. 2, the peak estrous response occurred 60 h after PGF. Pregnancy rate during the synchrony period was greater (P < 0.05) for the MGA-PGF (255/401; 63.6%) than the GnRH-TB60 (74/158; 46.6%) treatment. In Exp. 3, 75.7% of heifers (348/460) were detected in estrus by 73 h and were inseminated, with a conception rate of 74.4%. Pregnancy rates after TAI did not differ between TB80 and GnRH-TB80 (14/56 = 25% and 19/ 56 = 33.9%, respectively). Total pregnancy rate was 63.5% for heifers inseminated after detected estrus and by TAI. Collectively, these data indicate that the exclusive use of TAI for heifers treated with the MGA-PGF protocol resulted in lower pregnancy rates than when AI was performed after detection of estrus. However, estrus AI for 3 d and TAI at the end of d 3 could result in pregnancy rates similar to those achieved after a 5-d period of detecting estrus.

Synchronization of estrus and fertility in zebu beef heifers treated with three estrus synchronization protocols.

The effects on estrus and fertility of 3 estrus synchronization protocols were studied in Brahman beef heifers. In Treatment 1 (PGF protocol; n=234), heifers received 7.5 mg, i.m. prostianol on Day 0 and were inseminated after observed estrus until Day 5. Treatment 2 (10-d NOR protocol; n = 220) consisted of norgestomet (NOR; 3 mg, s.c. implant and 3 mg, i.m.) and estradiol valerate (5 mg, i.m.) treatment on Day -10, NOR implant removal and 400 IU, i.m. PMSG on Day 0, and AI after observed estrus through to Day 5. Treatment 3 (14-d NOR+PGF protocol; n = 168) constituted a NOR implant (3 mg, sc) on Day -14, NOR implant removal on Day 0, PGF on Day 16, and AI after observed estrus through to Day 21. All heifers were examined for return to estrus at the next cycle and inseminated after observed estrus. The heifers were then exposed to bulls for at least 21 d. During the period of estrus observation (5 d) after treatment, those heifers treated with the PGF protocol had a lower (P<0.01) rate of estrual response (58%) than heifers treated with the 10-d NOR (87%) or 14-d NOR+PGF (88%) protocol. Heifers treated with the 10-d NOR protocol displayed estrus earlier and had a closer synchrony of estrus than heifers treated with either the PGF or the 14-d NOR+PGF protocol. Heifers treated with the 14-d NOR+PGF protocol had higher (P<0.05) conception and calving rates (51 and 46%) to AI at the induced estrus than heifers treated with the PGF (45 and 27%) or the 10-d NOR (38 and 33%) protocol. Calving rate to 2 rounds of AI was greater (P<0.05) for heifers treated with the 14-d NOR-PGF (50%) protocol than heifers treated with the 10-d NOR (38%) but not the PGF (43%) protocol. Breeding season calving rates were similar among the 3 protocols. The results show that the 14-d NOR+PGF estrus synchronization protocol induced a high incidence of estrus with comparatively high fertility in Brahman heifers.

Estrus synchronization with melengestrol acetate in cyclic ewes. Insemination with fresh or frozen semen during the first or second estrus post treatment.

A total of 540 cyclic ewes were randomly allocated to 1 of 6 groups according to synchronization or not with melengestrol acetate (MGA), insemination with fresh or frozen semen, and insemination during the first or second estrus post treatment. The MGA was effective in synchronizing estrus, since the percentage of ewes showing estrus during the first 6 days after treatment was significantly higher (P<0.05) for treated (79.5%) than for nontreated ewes (33.5%); 74% of the treated ewes showed estrus during a 72-h period. Progesterone concentrations in plasma samples obtained at random from 34 treated ewes on Day 8 post estrus revealed that 94% of them ovulated and formed a functional CL. Synchronization was maintained during the second estrus post treatment, since 71.9% of the ewes showed the second estrus during a 72-h period. Treatment with 0.22 mg of MGA/head/d for 14 d had a detrimental effect on fertility when insemination was carried out during the first estrus post treatment. Delaying insemination until the second estrus post treatment caused a less marked reduction in conception rates. Thus, MGA can be a useful alternative for estrus synchronization of a large number of ewes. Artificial insemination can be delayed until the second estrus post treatment, improving fertility without loosing the advantages of estrus synchronization.

Estrus synchronization efficiency of PGF2alpha injection in Shorthorn-Hereford and crossbred Charolais cattle not having exhibited estrus at 4 or 7 days prior to treatment.

The objective of this study was to compare the estrus synchronization efficiency of PGF2alpha in cattle (n = 470) not having shown estrus for 4 (D4 treatment) or 7 d (D7 treatment) after onset of the breeding season. The physiological status of crossbred Charolais cows was studied, and their reproductive performance was compared to that of Shorthorn-Hereford cows. The percentage of cows in estrus during the 7 d prior to PGF2alpha treatment was superior (P < 0.01) to that observed during the 4 d preceding PGF2alpha. The daily rates of estrus were similar during the 2 periods. For both the Shorthorn-Hereford (87.8 vs 74.7%; P < 0.03) and crossbred Charolais (87.8 vs 66.3%; P < 0.005) females, the estrus synchronization rate during the 5 d post PGF2alpha was higher in treatment D7 than in the D4 treatment. Therefore, for both Shorthorn-Hereford (92.8 vs 81.4%) and crossbred Charolais (93.1 vs 75.0%) the D7 program permitted insemination in 12 d (before and after PGF2alpha) of a higher percentage (P < 0.01) of females than the D4 program did in 9 d. The intervals PGF2alpha to estrus and their variances were similar for both treatments. The conception rate of Shorthorn-Hereford was the same whether they were inseminated after spontaneous (73.4%) or PGF2alpha-induced estrus (D4 treatment: 78.5%; D7 treatment: 72.2%). However, crossbred Charolais cows inseminated after a PGF2alpha-induced estrus in the D4 treatment had a fertility rate (50.9%) slightly inferior (P < 0.07) to that of cows inseminated after spontaneous estrus (70.1%) or after PGF2alpha-induced estrus in the D7 treatment (67.4%). The reproductive performances of multiparous Shorthorn-Hereford and crossbred Charolais were similar in treatment D7; however, in the D4 treatment, the conception rate of Shorthorn-Hereford was higher than that of crossbred Charolais (81.0 vs 42.9%; P < 0.002). Primiparous crossbred Charolais in the D4 treatment had a slightly lower (P < 0.10) synchronization rate (48%) than nulliparous (71%) and multiparous crossbred Charolais (78%). In contrast, the reproductive performances of nulliparous, primiparous and multiparous crossbred Charolais were similar in the D7 treatment. These results indicate that the efficiency of PGF2alpha to synchronize estrus is greater when the estrus detection period increases from 4 to 7 d before PGF2alpha. Only in the D7 treatment was the fertility rate of crossbred Charolais similar to that of Shorthorn-Hereford.

Assessment of estrus detection by visual observation and electronic detection methods and characterization of factors associated with estrus and pregnancy in beef heifers.

One hundred and sixty-four beef heifers representing Angus, Brahman and their crosses were subjected to estrus synchronization treatment following evaluation of weight, body condition score and reproductive tract. Heifers were assigned to 1 of 2 methods of estrus detection, either visual observation for signs of standing estrus or a rump-mounted pressure-sensitive detection device. All heifers were artificially inseminated during a 25d insemination period and then bred by a bull. The effectiveness of estrus detection and timely insemination were evaluated by the type of detection method, breed and breeding event resulting in a pregnancy. Although there was not a significant difference in first service conception for method of detection, at the end of a 25-d insemination period, 60.5% of the visually observed heifers were pregnant, while only 45.8% of the heifers detected by the mount detection device were pregnant (P = 0.05). The reduced 25-d conception rate in the pressure-sensitive detection group suggested that insemination of detected heifers may not have been optimal for pregnancy. The mean time to estrus after implant removal and the time of insemination were evaluated. Heifers pregnant at the first service had a shorter time to estrus (32.11 +/- 1.6 h, P=0.13), a longer mean interval from the start of estrus to insemination (12.10 +/- 1.2 h, P = 0.16) and a positive mean interval between the end of standing estrus and the time of insemination (3.17 +/-1.7 h, P=0.19) than heifers not conceiving at the synchronized estrus (38.5 +/- 2.1, 7.42 +/- 1.9 h and -2.04 +/- 2.1 h, respectively). Breed differences were observed in estrus durations (Angus 8.52 +/- 1.2 h, Brahman 6.65 +/- 1.2 h, crossbred 11.90 +/- 1.2 h; P = 0.03), number of mounts (19 +/- 3.6, 25 +/- 5.4, 37 +/- 5.5, respectively; P=0.02) and gestation length (281 +/- 1.2, 291 +/- 1.8, 286 +/- 1.1 d, respectively; P = 0.001).

Synchronization of estrus in suckled beef cows for detected estrus and artificial insemination and timed artificial insemination using gonadotropin-releasing hormone, prostaglandin F2alpha, and progesterone.

We determined whether a fixed-time AI (TAI) protocol could yield pregnancy rates similar to a protocol requiring detection of estrus, or estrous detection plus TAI, and whether adding a controlled internal device release (CIDR) to GnRH-based protocols would enhance fertility. Estrus was synchronized in 2,598 suckled beef cows at 14 locations, and AI was preceded by 1 of 5 treatments: 1) a CIDR for 7 d with 25 mg of PG F(2alpha) (PGF) at CIDR removal, followed by detection of estrus and AI during the 84 h after PGF; cows not detected in estrus by 84 h received 100 mug of GnRH and TAI at 84 h (control; n = 506); 2) GnRH administration, followed in 7 d with PGF, followed in 60 h by a second injection of GnRH and TAI (CO-Synch; n = 548); 3) CO-Synch plus a CIDR during the 7 d between the first injection of GnRH and PGF (CO-Synch + CIDR; n = 539); 4) GnRH administration, followed in 7 d with PGF, followed by detection of estrus and AI during the 84 h after PGF; cows not detected in estrus by 84 h received GnRH and TAI at 84 h (Select Synch & TAI; n = 507); and 5) Select Synch & TAI plus a CIDR during the 7 d between the first injection of GnRH and PGF (Select Synch + CIDR & TAI; n = 498). Blood samples were collected (d -17 and -7, relative to PGF) to determine estrous cycle status. For the control, Select Synch & TAI, and Select Synch + CIDR & TAI treatments, a minimum of twice daily observations for estrus began on d 0 and continued for at least 72 h. Inseminations were performed using the AM/PM rule. Pregnancy was diagnosed by transrectal ultrasonography. Percentage of cows cycling at the initiation of treatments was 66%. Pregnancy rates (proportion of cows pregnant to AI of all cows synchronized during the synchronization period) among locations across treatments ranged from 37% to 67%. Pregnancy rates were greater (P < 0.05) for the Select Synch + CIDR & TAI (58%), CO-Synch + CIDR (54%), Select Synch & TAI (53%), or control (53%) treatments than the CO-Synch (44%) treatment. Among the 3 protocols in which estrus was detected, conception rates (proportion of cows that became pregnant to AI of those exhibiting estrus during the synchronization period) were greater (P < 0.05) for Select Synch & TAI (70%; 217 of 309) and Select Synch + CIDR & TAI (67%; 230 of 345) cows than for control cows (61%; 197 of 325). We conclude that the CO-Synch + CIDR protocol yielded similar pregnancy rates to estrous detection protocols and is a reliable TAI protocol that eliminates detection of estrus when inseminating beef cows.

Influence of equine chorionic gonadotropin on weaning-to-estrus interval and estrus duration in early-weaned, primiparous, female swine.

The weaning-to-estrus interval (WEI) influences the total nonproductive days (NPD) accumulated by the breeding herd and affects herd productivity. Short lactation lengths (LL) are commonly followed by prolonged WEI, which are also associated with short estrus duration (ED). Equine chorionic gonadotropin treatment is a tool that has been used to reduce WEI, especially for low-parity females. The objectives for this study were to evaluate the effect of LL on the association between WEI and ED and to estimate the effects of postweaning eCG administration on WEI and ED for early-weaned females. Two treatments (TREAT) consisting of 750 IU of eCG (n = 96) or control (n = 77) were applied 1 d after weaning to first-parity, weaned females. The study was conducted on a commercial farm having a target LL of 18 d. Estrus detection was conducted three times daily, and estrus duration was determined as the interval between the first and the last positive response to back pressure. Analyses of variance were conducted to estimate the effects of LL and TREAT on WEI and the effects of TREAT and WEI on estrus duration. Mean LL was 17.9+/-1.7 d, mean WEI was 106.6+/-29.2 h, and mean estrus duration was 55.9+/-15.5 h. Even though the frequency of short WEI tended to increase with longer LL, mean WEI was shortest for females weaned after 18 d and longest for those weaned after 20 d (P<.05). The WEI for females receiving eCG (98.7+/-2.7 h) was shorter (P = .0001) than that for control females (121.5+/-3.3 h). The WEI was also affected by a LL x TREAT interaction (P = .0014), indicating that the interval was longer (P<.05) for control females weaned after 17 and 20+ d than for other females. The LL and TREAT did not affect estrus duration (P = .20 and P = .157, respectively). However, estrus duration was reduced as the WEI increased (P = .0001), and it was also influenced by a WEI x TREAT interaction (P = .024). A linear regression model estimated that the association between WEI and estrus duration was stronger in the eCG group than in the control group (R2 = .51 and .15, respectively; both P<.001). In conclusion, the use of eCG postweaning was associated with more precise prediction of estrus duration as a function of the WEI and allows optimization of breeding management in early-weaned, primiparous females.

Estrus behavior and fecal steroid profiles in the Asiatic lion (Panthera leo persica) during natural and gonadotropin-induced estrus.

In this paper the behavior of the Asiatic lion was studied during natural and gonadotropin-induced estrus in relation to fecal estradiol and progesterone concentration. The average length of estrus was 5.4 days and no significant difference was observed between natural and gonadotropin-induced estrus. Vocalization and rolling were the major estrus behavioral activities of Asiatic lions and the frequency of these activities were similar in both natural and gonadotropin-induced estrus and treatment with exogenous gonadotropin did not alter estrus behavioral activities. A significant positive correlation was observed between fecal estradiol and frequency of estrus behavior during natural and gonadotropin-induced estrus. Following gonadotropin treatment estrus could be induced in 69% of animals and these induced animals ovulated following hCG treatment. This study reports for the first time the successful use of the non-invasive fecal steroid assay for monitoring the induction of estrus and ovulation in the Asiatic lion.

Detection of estrus by visual observation and radiotelemetry in peripubertal, estrus-synchronized beef heifers.

The effectiveness of two estrus-detection methods (visual observation and radiotelemetric, pressure-sensitive, rump-mounted devices) was compared in peripubertal, crossbred yearling beef heifers. Heifers (n = 50) were fitted with a pressure-sensitive device affixed to their rumps to which a battery-operated radio transmitter was connected. Activation of the sensor sent a radiotelemetric signal to a microcomputer via a fixed radio antenna. Heifer identification, date, time, and duration of standing events were recorded. Estrus was synchronized by feeding melengestrol acetate (MGA) for 14 d and then injecting PGF2 alpha 17 d after the last daily dose of MGA. Following the injection of PGF2 alpha, heifers were observed visually for signs of estrus at 0730 and 1630 (45 min each). Estrus was detected in 41 of 49 heifers (one lost the device) by visual observation and(or) the radiotelemetric device. Number of standing events during estrus, determined by the radiotelemetric device, averaged 50.1 +/- 6.4 per heifer, with the duration of estrus ranging from 2.6 to 26.2 h (average = 14 +/- .8 h). Visual observation failed to detect 11 of 41 heifers (37%) that were detected by the radiotelemetric device. Heifers with fewer standing events (19.3 vs 60.5; P < .001) and estrus of shorter duration (8.4 vs 15.6 h; P < .001) were those not identified by visual observation. Based on intervals between the first detected standing event of estrus and time of insemination, a wide window of fertility seems to exist in some heifers. Radiotelemetric devices were useful in identifying a greater (P < .05) proportion (100%) of heifers in estrus (increased efficiency) compared with visual observation (73%). However, accuracy of both methods was similar.

Incidence of premature estrus in lactating dairy cows and conception rates to standing estrus or fixed-time inseminations after synchronization using GnRH and PGF(2alpha).

Fixed-time AI (TAI) after GnRH-PGF(2alpha)-GnRH treatment is a method to achieve pregnancies in dairy herds without estrous detection. However, cows that fail to respond to the initial GnRH may have compromised TAI conception rates due to asynchronous ovarian response. This study documented the percentage of GnRH-treated Holstein cows (n=345) in two herds that displayed estrus at an inopportune time for optimum TAI conception rate (< or =48h post-PGF(2alpha); premature estrus (PE)) and compared conception rates of two TAI protocols in cows that did not display PE. At biweekly herd health exams, cows diagnosed as not pregnant to a previous AI and cows >80 days postpartum with no AI were treated with 100 microg GnRH (day -7) and 25mg PGF(2alpha) (day 0). Cows detected in PE by twice-daily visual observation from day -7 to day 2 were bred by AI 8-12h later. Cows not detected in PE were randomly assigned by parity, body condition score, and postpartum interval to receive either: (1) 100microg GnRH at 48h after PGF(2alpha) and TAI 16 to 18h later (Ovsynch); or (2) TAI at 72h post-PGF(2alpha) and a concurrent 100 microg GnRH injection to those cows not detected in estrus between 48 and 72h post-PGF(2alpha) (modified Ovsynch (MOV)). All hormone injections were im. Twenty percent (68/345) of the cows were detected in estrus before 48 after PGF(2alpha), of which 5% (17/345) were detected in estrus before PGF(2alpha) (< or =day 0). Herd influenced the percentage of cows in the PE group (herd A versus herd B; 25% versus 14%; P<0.05). Conception rates were not affected by treatment (PE versus Ovsynch versus MOV; 32% (21/65) versus 30% (37/125) versus 32% (47/145); P>0.10). However, within MOV-treated cows, conception rates were greater (P<0.05) in cows detected in estrus (46% (23/50)) compared with cows not detected in estrus (25% (24/95)). In conclusion, 20% of GnRH-treated cows displayed PE and necessitates estrous detection during this period if maximal pregnancy rates are to be achieved. Although additional estrous detection is required compared to Ovsynch, reduced cow handling and hormone usage, efficient use of expensive semen through greater conception rates in cows detected in estrus, and comparable TAI conception rates, suggests the MOV protocol may be a cost effective alternative to Ovsynch in many dairy herd reproductive management programs.

Characteristics of estrus before and after first insemination and fertility of heifers after synchronized estrus using GnRH, PGF2alpha, and progesterone.

Our objectives were to determine fertility of heifers after synchronization of estrus using PGF2alpha, preceded by progesterone (P4), GnRH, or both, and to examine the variability of estrual characteristics in heifers before first and second AI. Dairy (n = 247) and beef (n = 193) heifers were assigned randomly to each of three treatments: 1) 50 microg of GnRH (injected i.m.) administered on d -7 followed by 25 mg of PGF2alpha (i.m.) on d -1 (GnRH + PGF; modified Select Synch protocol); 2) placement of an intravaginal progesterone (P4)-releasing insert on d -7, PGF2alpha on d -1, and insert removal on d 0 (P4+PGF); and 3) 50 microg of GnRH plus a P4 insert on d -7, followed by 25 mg of PGF2alpha on d -1, and insert removal on d 0 (P4+GnRH+PGF). Characteristics of estrus were examined before first AI and before the next eligible AI (18 to 26 d later), including duration of estrus, number of standing events, and total and individual duration of standing events. In addition, all heifers were checked visually at least twice daily for estrus. Blood samples were collected on d -7, -1, and 0 for determination of P4, and pregnancy status was diagnosed by ultrasonography 27 to 34 d after AI. Rates of detected estrus were less (P < 0.05) in dairy than in beef heifers, and greater (P < 0.05) in heifers treated with P4. Pattern of conception and pregnancy rates among treatments differed between beef and dairy heifers (treatment x group interaction; P < 0.05). In dairy heifers, conception and pregnancy rates were greatest with P4+PGF, followed by P4+GnRH+PGF and GnRH+PGF, respectively. The opposite was observed among treatments in beef heifers. Administration of P4 without the preceding injection of GnRH produced the lowest pregnancy rates in beefheifers. Ofthe quantified sexual behavioral characteristics during the synchronized estrus, the number of standing events and total duration of standing events were greater (P < 0.01) than those observed during the next eligible estrus before second AI, whereas duration of estrus was unaffected.

Timing of insemination for dairy cows identified in estrus by a radiotelemetric estrus detection system.

The optimal time of artificial insemination (AI) was determined from data for 2661 AI in 17 herds utilizing a radiotelemetric system for estrus detection that has the potential for continuous 24-h surveillance to monitor behavioral events associated with estrus. The system consisted of pressure-sensitive radio frequency transmitters affixed over the sacrum region of cows. The activation of the sensor sent a radiotelemetric signal to a microcomputer via a fixed antenna. Cow identification, date, time, and duration of each standing event were recorded in the software program provided with the system. Each farm selected a 3-h interval to AI for cows that were identified in estrus during the previous 24 h. Pregnancy status was determined from data for return to estrus and palpation of the uterus 35 to 75 d following AI. Standing events during estrus averaged (+/- SD) 8.5 +/- 6.6 per cow, and the number of events per estrus across herds averaged from 6.2 +/- 5.1 to 12.8 +/- 9.9 per cow. The duration of estrus ranged from 5.1 +/- 3.8 to 10.6 +/- 6.8 h across herds; the mean was 7.1 +/- 5.4 h. The interval from the first standing event to AI affected the probability of pregnancy; the highest conception rates for AI occurred between 4 and 12 h after the onset of standing activity. The probability of pregnancy was higher for cows > 100 d in milk, exhibiting > 2 standing events during estrus, and inseminated during March, April or May.

Comparative studies on the effectiveness of Sil-estrus implants, Veramix sheep sponges and prostaglandin F2alpha in synchronizing estrus in West African dwarf sheep.

Sil-estrus implants, Veramix sponges and PGF2alpha were successfully used to synchronize estrus in normocyclic West African dwarf sheep. The intervals from end of treatment to onset of observable estrus in the three groups of treatment were 42.33+/-1.99, 77.67+/-15.20 and 41.62+/-2.23 h respectively, for Sil-estrus, Veramix sponges and PGF2alpha (P>0.05). Of all the treated sheep, 100% of those treated with Sil-estrus and PGF2alpha and 66.67% of the sheep treated with Veramix sponges were in estrus within 48 h post treatment. Neither the duration of standing estrus nor the succeeding estrus cycle length was significantly affected by the treatment. These results indicate that progestogens and prostaglandin F2alpha can be usefully employed in the management of reproduction of West African dwarf sheep.

Comparison of timed insemination with insemination at estrus following synchronization of estrus with a MGA-prostaglandin system in beef heifers.

Three trials utilizing 231 beef heifers were conducted in 1993 to determine if a timed insemination would result in similar synchronized pregnancy rates as insemination by estrus following synchronization of estrus using the 14-d MGA-prostaglandin system. All heifers were fed 0.5 mg MGA/h/d fof 14 d and given a 25 mg injection of PGF(2)alpha im 17 d after the final day of MGA feeding. Heifers in Group 1 (timed AI treatment) were inseminated at 72 h after the prostaglandin injection independent of whether or not they were observed in estrus. Heifers in Group 2 (AI by estrus) were inseminated 12 to 18 h after the onset of estrus. Since the trial was a significant source of variation for synchronized pregnancy rate, the effect of treatment on pregnancy rate was analyzed for each trial. Synchronized pregnancy rates in Trials 2 and 3 were similar in both treatment groups; 37 vs 35% and 61 vs 58% for the timed AI vs AI by estrus (Groups 1 and 2) in Trials 2 and 3, respectively. In both of these trials the degree of estrous synchrony was high. In Trial 1, the synchronized pregnancy rate in heifers that were time-inseminated was significantly lower than that of heifers that were inseminated by estrus (29 vs 57%). The lower synchronized pregnancy rate of Group 1 (timed AI) heifers in Trial 1 appeared to be due to the low degree of estrous synchrony in this trial. Our results indicate that using timed insemination with the 14-d MGA-prostaglandin system will give similar synchronized pregnancy rates as inseminating by estrus in groups of beef heifers where the degree of synchrony is high. However, in heifers where the degree of estrous synchrony is low, a timed insemination reduces synchronized pregnancy rates.
