Immunofluorescence studies with a specific antiserum to the microfibrillar protein of elastic fibres. Location in elastic and non-elastic connective tissues.

The microfibrillar protein (MFP) of foetal bovine ligamentum nuchae elastic fibres was prepared and used to produce a monospecific antiserum. Indirect immunofluorescence studies employing the specific antiserum demonstrated its selectivity for MFP associated with elastic fibres. The antiserum was demonstrated to react with vascular elastic tissue, perivascular connective tissue and reticular basement membranes in a variety of tissues. This evidence suggests that the MFP (or an immunologically related protein) is not confined to elastic fibres but is widely distributed in connective tissue being associated with both elastin and collagen fibres.

[Elastic activator appliances: Bimler's elastic appliance and Klammt's elastic activator].

The action of two elastic activators (Bimler and Klammt) is studied in twenty Class II/1 malocclusions, with a mean age of 10.2 years and a treatment time of 2.7 years. Intercanine and intermolar widths, and "anterior arch height" (Korkhaus) were measured on models at the beginning and at the end of treatment. On lateral X-Ray films following parameters (Ricketts) were measured: facial axis, facial depth, convexity, lower facial height, and corpus axis. Conclusions are: both appliances have a good transverse response, Bimler's Gebissformer increase vertical growth, and Klammt's elastic activator decrease convexity by a more anterior mandibular positioning.

Enzymatic destruction of the elastic lamella at the mouth of cerebral berry aneurysm? An ultrastructural study with special regard to the elastic tissue.

Destruction of the elastic tissue is probably an acquired lesion and is decisive for the appearance of cerebral arterial aneurysms at the sites of congenital media defects. The elastic component in the mouths of aneurysms has therefore been studied by electron microscopy, using two new staining methods, i.e. ruthenium-red staining and prolonged osmium-tetroxide treatment. The hypertrophic, duplicated, elastic lamellae showed a disintegration of their luminal portions not earlier described. In close connection with the disintegrated portions, extracellular lysosome-like granules were observed. It is hypothesized that discharged leucocyte grannules containing elastase help to destroy the elastic lamellae.

Iron gallein elastic method---a substitute for Verhoeff's elastic tissue stain.

A method for staining elastic fibers in formalin fixed, paraffin embedded sections is described. After deparaffinizing and dehydration, sections are stained for 30 minutes in a solution prepared by mixing equal parts of 1% gallein dissolved in ethylene glycol and absolute alcohol (1:4), and 1.16% aqueous ferric chloride in 1% hydrochloric acid. The sections are washed in water and then differentiated in 2% ferric chloride for 2 minutes. After washing in water, the sections are counterstained with a variant of Van Gieson's picric acid-acid fuchsin for 1 minute. The results are similar to Verhoeff's elastic stain with elastic fibers staining black. An advantage to this staining procedure is that visually controlled differentiation is not necessary.

Pre-elastic (oxytalan) fibres in the developing elastic cartilage of the external ear of the rat.

During chondrogenesis in the external ear of the rat oxytalan fibres precede the appearance of mature elastic fibres by 6-7 days. The spatial distribution of oxytalan fibres in fetal and neonatal pre-cartilage corresponds to that of the elastic fibres in mature cartilage. These findings support the hypothesis that oxytalan fibres in the pre-cartilage of the external ear of the rat are pre-elastic in nature.

Age Factors

Measurement of particle sizes by elastic and quasi-elastic light scattering.

A method of measuring an average particle radius in a highly polydisperse dispersion using the wavelength dependence of turbidity is described. For particles which are no larger than 0.3 of the wavelength of light used, a polynomial representation of the scattering cross-section can be used. For larger particles, more extensive numerical calculations are required. The use of the method is illustrated by determining the average particle radius of casein micelles by both elastic and quasi-elastic light scattering techniques. A polydisperse homogeneous sphere model is found to be a reasonably accurate representation of casein micelles. Several modifications of the model which would improve the agreement between the two techniques are mentioned briefly.

Casein micelle size from elastic and quasi-elastic light scattering measurements.

The average molecular weight, particle radius and size distribution of particles in skim milk from eight cows in mid-lactation have been measured by means of elastic and quasi-elastic light scattering techniques. The properties of sub-micellar casein particles in the milk of each cow were also studied. Particular attention has been given to the effects of particle size heterogeneity in the interpretation of results. The weight average molecular weight of the particles from different cows varied from 2.6-10(8) to 15-10(8) and the corresponding average particle radius varied between 90 and 130 nm. An unusual feature of these particles is their high water content, which was found to vary from 2.4 to 6.4 ml/g with a positive correlation between average particle density and average particle mass. Variations in particle water content can be most readily understood in terms of a gel-like casein micelle.

[Prophylaxis of thrombosis, varices, endema-indications for ready-made elasticated stockings? Comparative pressure measurements of ready-made elasticated stockings of Compression Classes I-IV (author's transl)].

Comparative pressure measurements of ready-made elasticated stockings of compression classes I-IV were carried out on 28 patients. Considerable differences in pressure sometimes occur, stockings with a high compression cannot be put on without help. Not all of them can be boiled, the properties sometimes alter after washing. Elasticated stockings are not indicated in lymphedemas etc. Only stockings of compression class I are suitable for the prophylaxis of thrombosis in bedridden patients.

The use of tannic acid-glutaraldehyde in the study of elastic and elastic-related fibers.

Fixation with tannic acid-glutaraldehyde permits distinction of oxytalan, elaunin and elastic fibers in the electron microscope. The results obtained using tannic acid at concentrations of 1.0%, 0.5% and 0.25% in 3% glutaraldehyde were compared. The 0.25% concentration is recommended for studying fine details of connective fibrils and for regular staining of elastin.
