[Developmental abnormalities of mineralization in populations with varying exposure to fluorine compounds].

The aim of the study was to determine the influence of environmental factors on developmental defects of dental enamel. The examination was carried out in a group of 238 schoolchildren (group I) aged 14 to 15 years, living since birth in a region with elevated levels of chemical compounds (fluorine, ammonia, sulphur) in the atmosphere. The control group (group II) consisted of 240 schoolchildren of the same age, living in regions with normal atmospheric concentrations of these compounds. The Epidemiological Index of Developmental Defects of Dental Enamel (DDE Index), Dean fluorosis index, and DMF index were used in the clinical examination. Additionally, a questionnaire was given to parents of the examined children. It was found that the number of children and teeth with developmental defects of dental enamel was greater in group I. Significant differences between boys and girls were noticed. The most common developmental defect of dental enamel in both groups was white/creamy opacity. The next common defect was hypoplasia. The incidence of diffuse white/creamy mottling and lines was higher in the study group than in controls. This was the basic finding which differentiated the two populations studied. Both groups differed as to the Dean index. Nevertheless, a comparison of Dean and DDE indices led to inconsistent conclusions. The distribution of developmental defects of dental enamel was similar in both groups. The defects were most often found on the buccal surfaces of maxillary premolars and maxillary central incisors. Children with developmental defects of dental enamel had a significantly higher incidence of caries and elevated mean DMF, as compared to children without any abnormalities of mineralization. Similar correlations were not found in the control group. The questionnaire revealed a positive correlation between the education level of parents, standards of living, and of abnormalities mineralization in both groups of children.

[Activity of acid phosphatase in saliva of industrial workers exposed to fluorine compounds].

Assuming that fluorine is a strong inhibitor of phosphatases, the activity of acid phosphatase in the saliva of 42 phosphorus fertilizers plant workers was studied. The workers were chronically exposed to fluorine compounds. Saliva had been collected for studies before the workers entered the productive plant and after the 1st and 2nd working hour. Enzyme activity was determined by Bessey and Lowry's method and was related to both the volume of the saliva and total protein level. The studies revealed a significant decrease in the activity of acid phosphatase after the 1st and 2nd working hour in those working in the division with a higher exposure to fluorine compounds. The results of these studies were confirmed by in vitro studies carried out additionally. Taking into account biological activity of acid phosphatase it seems that the observed phenomenon of inhibiting enzymatic activity may be connected with the frequency of parodontopathy in the studied group of workers. The results of these studies may be used for working out a biological test of workers' exposure.

[Oxygenating intra-alveolar lavage (OAL) with the use of some new fluorinated compounds].

Certain fluorinated carbonate compounds can carry O2 and CO2 in a physiological manner. This faculty was exploited in the elaboration of an experimental intra-alveolar oxygenating washing protocol employing such compounds as intrapulmonary oxygenating liquids conveyed by transtracheal flooding of the alveoli in 20 rabbits. AP, PaP, CVP and EP were monitored in accordance with a 35' protocol, during which the positive oxygenation and negative emptying phases of the washing process were repeated at an average 2 cycles/min. A respiratory exchange capable of keeping all the animals alive until the recommencement of spontaneous ventilation was noted, along with only slight changes in lung structure. Once an in-depth study is made of certain fundamental phenomena related to structure and lung function, it is felt that this washing method can be used clinically for the treatment of hypoxaemic situations in which normal alveocapillary exchange is impeded by extraneous material, as in desquamative interstitial pneumonia, pulmonary alveolar proteinosis, etc.

[Toxic effects of fluorine compounds on the the fetus and their effect on the course of pregnancy].

The effect of transfer fluorine compounds through placenta is a possibility their influence on fetus. There are known many examples of unprofitable activity fluorine compounds during pregnancy. There is mentioned among others increase percentage of preterm labours, stillbirths, congenital abnormalities and also deaths of newborns just after delivery. The mechanisms of toxic influence fluorine compounds on fetus have probably character of direct influence on embryo's cells or disturb the punction of placental tissue with following consequences. The real teratogenic fluorine compounds activity was not completely proved.

Fluorine compounds in anaesthesia (9). Examination of six aliphatic compounds and four ethers.

Six heavily fluorinated aliphatic compounds and four heavily fluorinated derivatives of diethyl ether have been examined as anaesthetics, using mice. Two compounds are felt to be worth further examination. These are 1,1,1,-trifluoro-2,2-dichloroethane (halothane with the bromine atom replaced by a second chlorine) and 1,1,2,2,4,5,5-heptafluoro-3-oxapentane (ordinary diethyl ether with seven hydrogen atoms replaced by fluorine).

[Effect of inorganic fluorine compounds on the functional state of the pituitary-testis system].

The radioimmunological method was applied to the study of blood testosterone, LH, and FSH content in 41 men suffering from fluorosis, aged from 33 to 45 years. Nineteen men who had no contact with fluorine compounds served as control. In comparison with healthy individuals testosterone content proved to be decreased and FSH content elevated in patients with fluorosis. Blood LH content was increased only in those patients with fluorosis who had long contact with fluorine compounds (over 15 years). The changes in the blood hormone concentration were connected with disturbances of the hormonal, and, possibly, of the germenative function of the testes. The hypophysis proved to suffer less, and the changes of its function were apparently secondary in character.

Fluorine compounds inhibit the conversion of active type-1 protein phosphatases into the ATPMg-dependent form.

1. The modulator protein slowly converts the glycogen-bound protein phosphatase from liver, as well as its catalytic subunit, into an inactive form that requires protein kinase FA and MgATP for reactivation. The inactivation process could be completely prevented by addition of either 0.3 mM-NaF or 0.3 mM-phenylmethanesulphonyl fluoride (PMSF). The effectiveness of the proteinase inhibitor was not due to production of free fluoride. With the catalytic subunit a half-maximal effect of either fluorine compound was obtained at 25-50 microM. 2. The inactivation process was instantaneously blocked by the addition of NaF or PMSF at any moment during the incubation of the catalytic subunit with modulator. This fluoride effect was reversible. It did not result from a decreased affinity of modulator for the catalytic subunit. The use of analogues of PMSF showed that the fluorine atom was essential, but structural aspects were also an important determinant. 3. The relative efficiency of fluorine compounds in preventing the inactivation of the catalytic subunit by modulator corresponded to their relative potency as inhibitors of the phosphorylase phosphatase activity, but the latter effect required at least 20-fold higher effector concentrations. Incubation of the catalytic subunit with 10 mM-PMSF or -NaF caused an irreversible inhibition of the enzyme. 4. It is possible to prepare stable complexes of catalytic subunit and modulator, either active or ATPMg-dependent. Both species displayed the same molecular size during gel filtration. The inactive complex could be reactivated by incubation with MgATP and protein kinase FA. NaF and PMSF increased the final extent of re-activation at limiting concentrations of the protein kinase.

Analytical techniques used for monitoring the biodegradation of fluorinated compounds in waste streams from pharmaceutical production.

During the assessment of the environmental impact of new pharmaceutical processes the selection and testing of suitable environmental treatment technologies is carried out. A large component of process waste stream treatment practice is aerobic biotreatment in wastewater treatment plants, as it is cost effective and generally more environmentally friendly than harsher chemical/physical treatments. Pharmaceutical syntheses use a range of halogenated compounds (either as reagents, solvents or intermediates) which pose particular challenges to microbial degradation. This is especially so for some fluorinated compounds due to the resilience to enzymatic cleavage of the C-F bond in some cases. The data presented here were obtained from a case study involving the monitoring of the biodegradation of 4-fluorocinnamic acid by means of a range of chromatographic techniques. These methods were used to monitor not only the disappearance of the compound but also the formation of degradation products in order to confirm mineralisation. In addition mass spectrometry was used to elucidate the metabolic pathway.

Toxic fluorine compounds. The use of the omega-fluorine atom in the study of the metabolism of fatty acids containing ethyl, n-propyl and isopropyl branches.

1. Some omega-fluoroalkanoic acids containing a branched ethyl, n-propyl or isopropyl group in the alpha- or beta-position have been prepared. 2. A study of the toxicity of these acids revealed that the ethyl group partially inhibited, and the n-propyl and isopropyl groups completely inhibited, the normal processes of oxidative degradation.

[The diagnosis and treatment of osteoarthrosis deformans in subjects in contact with fluorine compounds].

A comparative study is presented of 378 workers with osteoarthrosis deformans (OD) contacting with fluorine compounds and in 106 patients with primary OD. The diagnostic criteria in these two categories of patients coincided. But the pain syndrome in the joints in professional fluorosis (PF) was associated with ossalgias (85%), tenderness of the bones (50.1%) and in 100% by symmetric hyperostosis. OD in PF was accompanied by a more pronounced density, was not accompanied by formation of nodes, and secondary synovitis was 4 times frequent. In OD plus PF males prevailed (96.7 e), and were averagely 10 years younger. In OD+PF treatment diet, calcium agents, glutamic acid should be used that bind and excrete fluorine from the body.

[Lysozyme activity in the saliva of industrial workers exposed to fluorine compounds].

The authors performed investigations in Phosphoric Fertilizers Works in Gdańsk to find out whether or not the work environment contaminated by fluorine compounds affects the activity of lysozome--the protective enzyme of saliva. In some in vitro experiments lysozome activity in exposed workers' saliva was found to be significantly lowered, as opposed to that in unexposed workers. In vivo experiments did not indicate any effects of work conditions on the activity of this enzyme. The obtained results, in vivo and in vitro, do not demonstrate any clear effects of work environment in the Phosphoric Fertilizers Works in Gdańsk, upon lysozome activity in workers' saliva.
