Development of high intensity narrow-band lamps and studies of the irradiation effect on human skin. Irradiation with high intensity lamps.

A compact light source has been developed. It consists of a high-pressure mercury lamp, a shutter, and water-cooled filters for selected wavelength regions. A mixing device permits simultaneous irradiation from two lamps of different wavelengths. The spectral distribution of the light for seven filter combinations and the construction of a power meter are also described. The application of the lamps to clinical investigations is illustrated by determination of minimal erythemal and blister doses, as well as pigmentation, for various groups of patients. The light intensities available are high enough to make pain threshold measurements possible in the UV and visible regions.

Toxicity and mutagenicity of radiation from fluorescent lamps and a sunlamp in L5178y mouse lymphoma cells.

Unfiltered broad spectrum radiation emitted by black light, cool white, and black light blue fluorescent lamps and a sunlamp, is both toxic and mutagenic to L5178Y mouse lymphoma cells when the cells are irradiated in phosphate-buffered saline. The increase in mutant frequency seen after exposure of the cells is linear throughout the range of exposures tested. The linear increase in mutagenesis is observed even at exposure levels which do not cause significant toxicity. To facilitate comparison of the differing rates of mutagenesis derived from exposure-response curves obtained for each light source, we have defined a parameter, joule-equivalent mutagenesis (jem), equal to mutants per 10(5) survivors per joule per square meter. Jem values are calculated using the integrated irradiance of each lamp. Based on jem values, the relative mutagenicity of the various lamps tested (compared with a germicidal ultraviolet lamp) is 3 x 10(-3) for the sunlamp, 1 x 10(-4) for the black light and cool white lamps, and 3 x 10(-5) for the black light blue lamp. The toxic and mutagenic effects of the lamps are in reasonable agreement with their relative spectral output from 290 to 330 nm.

Cell Line

Saliva-based RT-LAMP assays support heat shock protein 70 as a promising transcript marker for estrus identification in buffaloes.

Buffaloes do not exhibit overt estrus signs particularly during summer, leading to a significant economic loss to farmers. Previous studies have identified several candidate transcripts (HSP70, TIMP1, TLR4 and HSD17B1), abundant in buffalo saliva during estrus stage. However, there is no widely applicable technology for estrus detection targeting these transcripts. Therefore, the present study aimed to develop reverse transcription loop mediated isothermal amplification (RT-LAMP) assays for these candidate transcripts using buffalo saliva. Saliva samples were collected from 10 cyclic buffaloes and RT-LAMP assays were optimized for salivary RNA as well as direct saliva. Among the four candidate transcripts, HSP70 showed a statistically significant colour change (p-value = 0.0191) at the estrus stage compared to the diestrus stage. This abundance of HSP70 was also supported in large simulated population datasets (10,000 animals) generated using R. Further, the RT-LAMP assays were tested using direct saliva without RNA isolation, and the colour change in the samples during estrus suggested the feasibility of estrus identification using direct saliva, overcoming the tedious step of RNA isolation. The detection of HSP70 using either direct saliva or salivary RNA indicated its potential as a marker for estrus identification. Similarly, TLR4 appeared to be another potential biomarker for RT-LAMP reaction using direct saliva, but it needs further validation in both RNA and direct saliva samples. Overall, the proof-of-concept on RT-LAMP assays optimized for salivary transcripts in the present study would be useful for estrus identification in tropical production systems following further validation on a larger sample size.

[Experience with a new slit lamp concept (author's transl)].

Since the development of the first slit lamp by A. Gullstrand in 1911 this instrument is an indispensible part of our diagnostic repertoire. Nevertheless it was not until almost 40 years later that the slit lamp could be able to be used for the examination of the fundus as a result of the development of the Goldmann 3-mirror contact glass. Because of the increasing importance of antiretinal detachment prophylaxis the slit lamp examination also of the rear end of the eye its becoming more and more important because of its good stereoscopy, the excellent illumination and the high magnification. A newly developed slit-lamp apparatus fulfils these special requisites which have firstly been demanded recently. This apparatus is easy and comfortable to use, which makes easier above all the contact glass examination. Technical factors, possibilities of application and experience with this slit-lamp are discussed.

Ophthalmoscopes

[Narrow-band UVB phototherapy (Philips TL01 lamps) in psoriasis].

The authors report the results of an open study conducted on 53 patients with psoriasis treated by narrow-band UVB phototherapy, using Philips' TL01 lamp. With a simple procedure which did not require MED determination this treatment gave satisfactory results in 92 p. 100 of the cases, with mild burns in only 9 p. 100. The morphological type of psoriasis (patchy, guttate or nummular) had no influence on the therapeutic result, but the degree of infiltration of the lesions and their location on the lower limbs proved to be a factor of relative resistance. In most patients the results were obtained in 20 sessions with a mean cumulative dose of 20.19 +/- 2.7 J/cm2. Some patients had an additional treatment of 6 sessions. The results obtained with Philips' TL01 lamp were as satisfactory as those obtained with the conventional UVB lamps, but the TL01 lamp seemed to be easier to handle and better tolerated, which gives them some advantages over the latter.

[Activity of serum leucine aminopeptidase lamp in normal and pathologic pregnancies].

The activity of the enzyme leucine aminopeptidase (LAmP) in serum was examined in 305 pregnant women (174 women with normal pregnancy, and 131 women with disturbed pregnancy). The activity of this enzyme increases during the normal development of pregnancy. In disturbed pregnancies, the LAmP activity is lower as compared to normal pregnancies. A peculiar case: toxemia of pregnancy, where the activity of the enzyme is considerably increased. This fact may be compared with the increase of LAmP activity in cases of liver damage. The LAmP activity test in pregnancy is reliable and can be performed practically in every laboratory, thus deserving its place among the many tests meant to assess the normal function of the foeto-placental unit. This test may also help in the diagnostic of some specific pregnancies such as twin pregnancy, hydatidiform mole and extra-uterine pregnancy.

Slit-lamp beamsplitter for two stereoscopic views.

A double binocular beamsplitter, compatible with Zeiss optics, splits the light from each optical train of a slip-lamp microscope to provide an observer the exact stereoscopic view of the examiner. It has advantages in teaching slit-lamp examination techniques. Further, in surgery at the slit lamp, it allows the observer to function as an assistant and the surgeon to have both hands free.

An evaluation of ultraviolet radiation personnel hazards from selected 400-watt high intensity discharge lamps.

A study of selected mercury and multi-vapor lamps, commonly used for working environment illumination, was underta ken to evaluate the scope of the ultraviolet personnel hazards one may expect under various operating conditions of the lamps. Spectral irradiance measurements were made for operating conditions with and without reflector-type luminaires, glass covers on luminaries and outer glass bulbs on the lamps.

An accessory fill-in flash for the Nikon photo-slit lamp.

High-quality slit lamp photographs require a balance between slit beam exposure and exposure of surrounding ocular structures. Proper exposure of surrounding ocular structure is difficult to obtain with the Nikon photo-slit lamp. The purpose of this research was to design an accessory fill-in flash for this slit lamp, which would provide proper exposure of over all ocular structures. In addition, this paper describes the procedures used to obtain proper exposure values and provides an exposure table for this accessory flash unit.

Localization of malanin pigmentation in the skin with Wood's lamp.

Examination of the skin or hair with Wood's lamp has long been used to aid in the clinical diagnosis of some cutaneous disorders. A new observation reported here illustrates that Wood's light can be used to determine the depth of melanin pigmentation in the skin: contrast in epidermal pigmentation is increased while contrast in dermal pigmentation is decreased under Wood's lamp illumination compared to ambient visible light. The principles underlying this phenomenon are discussed. Use of the Wood's lamp to localize abnormal melanin pigmentation in the skin can be a guide to clinical diagnosis of hypermelanosis.

[Reading by fluorescent lamp light].

In dioptrics courses it has hitherto been taught that eyes of near-sighted patients who work in fluorescent light should be slightly under-corrected, since the near point is approximated in bluish light as a result of the chromatic aberration of the eye, in contrast to its location in white light or even the reddish light of incandescent lamps. The theory further states that this recommendation does not apply if close-range refraction occurs under fluorescent light. However, this is seldom the case. In our own investigations this approximation of the near point, under uniform illumination by fluorescent strip lights running across the ceiling, did not occur, at least not in "white" light (No. 33). On the contrary, under an incandescent lamp which was only half as powerful the near point was regularly closer to the eye than under white fluorescent light. The recommendation for under-correction of pulpit spectacles computed in incandescent light can therefore no longer be upheld.--While many patients complain of difficulties in diffuse incandescent light, these may be due to several other illumination factors which have not yet been fully investigated.--Some fundamental differences between diffuse fluorescent lighting and incandescent lamp lighting where there is a fall-off in luminosity are described.

Development of narrow-band lamps for testing and clinical use.

A compact narrow band lamp for high intensity irradiation has been developed. It consists of a high-pressure mercury lamp, a shutter and water cooled filters for spectral isolation of the light. A mixing device permits simultaneous irradiation with two different wavelenghts by using two lamps. The spectral distribution of the light within the bands is given for seven filter combinations. The construction and calibration of a power meter is also described.

Studies on retinal blood flow. I. Estimation of human retinal blood flow by slit-lamp fluorophotometry.

This article describes a new direct method for estimation of human retinal blood flow by slit-lamp fluorophotometry. The apparatus consists fundamentally of a modified slit lamp with an eye piece containing two fiberoptic probes connected to a double photometric detection system. Both optic fibers are focused on a retinal arterial segment, and the passage of a fluorescein bolus is automatically registered, giving, after correction, the mean transit time. Fluorescein angiography allows the determination of the volume of the arterial segment under study. Values for superior temporal retinal blood flow in healthy individuals show a mean value of 4.2 +/- 0.5 microliter/min. Studies in anemic patients show an increase of approximately 52% in the retinal blood flow. The potential causes of error associated with this technique, apart from the slight increase in intraocular pressure induced by the contact lens used, do not appear to be significant.

Slit-lamp attachment for examination of donor corneas in McCarey-Kaufman medium.

An attachment permits slit-lamp examination of excised donor cornea in McCarey-Kaufman medium. The bottle that contains the excised cornea is placed in a holder and examined with the slit lamp by means of a mirror placed at a 45 degree angle. This mirror reflects the slit beam up through the bottom of the bottle. This allows accurate examination of the donor cornea in a stable manner that was not previously possible.

Comparison of photo slit lamp and fundus camera photography of the optic disc.

The photo slit lamp has characteristics that may be important in optic disc photography for measuring glaucomatous cupping. This finding led to the question of whether disc photographs with the slit lamp showed less detail than fundus camera photographs. Tests were conducted on in-air resolution and of each instrument's ability to record fine vessel detail in normal human optic discs. The results showed that the instruments performed comparably in each test.

Fluorescein Angiography

On slit-lamp microscopy.

Theoretically the contact lens with a refractive power of -64 dioptres is superior to the concave pre-set lens (Hruby lens) or the convex lens (Bayadi). In practice the results are as follows: With the convex lens the examination of the vitreous is unsatisfactory. The reversed image with the convex lens will become familiar with some experience as one will also get used to the mirror images of the three-mirror contact lens. However, the abnormal curvature of the image is really a nuisance. On the other hand the large field of view is an advantage. The convex lens is only practically useful when it can be firmly attached to the slit-lamp microscope, as otherwise the new adjustment necessary for searching the fundus is too difficult. Attachment to the microscope is also of advantage with the Hruby lens. The concave contact lens of 64 dioptres is best suited for detailed examination of vitreous and fundus and for measurements. When no value is placed on a minute examination and a quick simple orientation suffices, the concave pre-set lens (Hruby lens) has the advantage that the lens does not have to be placed on the eye. If it is centred to the middle axis of the microscope, it facilitates especially quick observation. There will be cases where the pre-set lens will be the only answer, especially shortly after an operation or with very sensitive patients. Furthermore, the Hruby lens is preferable to the Bayadi lens because the vitreous can be better examined. The Hruby lens is also advantageous for examination of the region in the middle, lateral periphery of the fundus, 30 degrees - 60 degrees, and is superior in this area to the simple concave contact lens. The Bayadi lens seems to us only indicated for fundus examination of extremely high myopes. There it definitely offers advantages over and above the concave lens. For the most peripheral fundus, expecially below and above, the three-mirror lens with its modification is so far the best method. Especially for examination of the vitreous good brightness of the slit image is required for stereoscopic examination with as large an angle as possible between microscope and illumination. The lateral parts of the most peripheral fundus cannot be examined with the vertical slit in connection with the three-mirror lens. However, this is possible with the horizontal and tilted position of the slit and intermediate positions with an oblique slit. The slit must form an angle with the microscope in order to examine vitreous and fundus in optical section. With the indentation contact lenses ciliary processes and pars plana are now accessible to slit-lamp microscopy.

First report of tomato spotted wilt virus (Orthotospovirus tomatomaculae) and phytoplasma in China aster and development of duplex PCR, LAMP, and qPCR assays for rapid detection.

UNLABELLED: China aster (Callistephus chinensis) is an economically important ornamental crop widely cultivated for cut flowers and landscaping. During field surveys conducted in three districts of Karnataka, India, China aster plants exhibiting chlorotic and necrotic ring spots, leaf deformation, and witches' broom symptoms were collected and analyzed to determine the causal agents. Mechanical inoculation of symptomatic leaf sap onto cowpea (Vigna unguiculata cv. C-152) produced characteristic chlorotic and necrotic ring spots on newly emerging leaves indicating the presence of an infectious viral agent. Serological assay by DAC-ELISA followed by RT-PCR confirmed the presence of tomato spotted wilt virus (TSWV, Orthotospovirus tomatomaculae) in symptomatic plants. Similarly the plants exhibiting witches' broom symptoms tested positive for phytoplasma infection using universal and Nested primers PCR assays targeting the 16S rRNA gene. Sequence analysis of TSWV CP gene revealed more than 97% nucleotide identity with TSWV isolates reported from India and other countries. Based on these results, one representative isolate was selected for complete genome sequencing. The complete sequences of the L, M, S RNA segements were amplified cloned, and sequenced showing more than 97% nucleotide identity with global TSWV isolates available in database. Sequence analysis of 16S rRNA gene of the phytoplasma associated with witches' broom symptoms was identified as 'Candidatus Phytoplasma australasiaticum' belonging to the 16SrII-D subgroup, sharing 99.2% nucleotide identity with previously reported isolates. Phylogenetic analysis further supported the placement of both the TSWV and phytoplasma isolates within their respective taxonomic groups. To facilitate rapid and sensitive diagnosis, quantitative PCR (qPCR) and RT-LAMP assays were developed for TSWV detection. In addition a duplex PCR assay was optimized for simultaneous detection of TSWV and phytoplasma from infected China aster plants in a single reaction. This study represents the first reports of the complete genome characterization of TSWV and phytoplasma infection in China aster in India along with the development of sensitive qPCR, RT-LAMP, and duplex PCR assays for rapid detection of these pathogens providing valuable tools for disease diagnosis, epidemiological studies.
