Minocycline-related cutaneous hyperpigmentation as demonstrated by light microscopy, electron microscopy and X-ray energy spectroscopy.

A 70-year-old patient with a chronic cutaneous ulcer treated by minocycline hydrochloride developed hyperpigmentation of the forearms. Biopsy material was studied by light microscopy, electron microscopy and X-ray energy spectroscopy. Granular gold-brown pigment was found in dermal histiocytes and eccrine myoepithelial cells, which gave positive reaction with Prussian blue and Fontana-Masson stains. Electron microscopy revealed intracytoplasmic granules of dark, homogeneous material and small fine particles. X-ray energy spectroscopy showed iron and other elements in smaller amounts. The different types of minocycline-related hyperpigmentation and the possible pathomechanism are discussed with special regard to the importance of the diagnostic methods.

Calcium deposition in human pituitary adenomas studied by histology, electron microscopy, electron diffraction and X-ray spectrometry.

The incidence and morphogenesis of calcification were investigated by histology, electron microscopy, electron diffraction and X-ray spectrometry in 312 surgically removed human pituitary adenomas. By light microscopy the incidence of calcification was 9%. The calcium deposits were evident in several adenoma types, but were most frequent and extensive in prolactin-producing adenomas. Histologically calcium deposits represented amorphous precipitates and psammoma bodies staining with hematoxylin, the von Kossa technique and alizarin red. By electron microscopy, calcified deposits were noted in the cytoplasm of adenoma cells and extracellular space. Electron diffraction and X-ray spectrometry of intracellular precipitates as well as psammoma bodies showed the presence of phosphate, calcium and organically bound sulphur with a ratio of 3.5:5:2, indicating that they contained calcium hydroxyapatite. Present findings provide evidence that mitochondria with microcrystal deposition of calcium represent the primary sites of calcification.

Light microscopy, electron microscopy, and X-ray diffraction analysis of glycerinated collagen fibers.

Light microscopy, transmission electron microscopy (freeze-fracture replicas and thin sections), and X-ray diffraction techniques were used to investigate the structure of rat tail tendon collagen fibers subjected to one of the following treatments: water, phosphate buffer, glutaraldehyde, glutaraldehyde followed by glycerol, glycerol, and glycerol followed by phosphate buffer. As seen by light microscopy, only treatment with glycerol induces a remarkable swelling of the native (untreated) collagen specimens. Replicas and thin sections show that this swelling is due to an expansion of the interfibrillar space, and to a dissociation of the collagen fibrils into microfibrils. X-Ray diffraction analysis reveals great disorder in the glycerol-swollen collagen fibers. However, this does not appreciably involve the microfibrillar and molecular structure. Light and electron microscopy as well as X-ray diffraction techniques show that the collagen fiber swelling induced by glycerol is an almost completely reversible process.

Structure and blood supply of intrinsic lymph nodes in the wall of the rabbit urinary bladder--studies with light microscopy, electron microscopy, and vascular corrosion casting.

The urinary bladder is especially subject to infection by virtue of its direct connection to the external urethral opening, and it is natural to anticipate the presence of a well-developed immunological mechanism to respond to this potential threat. The present study describes small, very highly vascular lymph nodes located in the wall of the rabbit bladder, which may be involved in a local response to foreign antigens. The vasculature and structure of these lymph nodes was described using a combination of vascular corrosion casting, ink injection, and light and electron microscopy. The distal abdominal aorta was cannulated, and after clearing the bladder vasculature with buffered saline, one of the following procedures was used: 1) the bladder was perfuse-fixed in preparation for light and electron microscopy; 2) the bladder vasculature was filled with India ink for vessel tracing; or 3) vascular corrosion casts of the vasculature were prepared by infusing resin comprised of a mixture of Mercox, methyl methacrylate monomer, and catalyst. The resulting casts were cleaned with KOH, formic acid, and water in preparation for scanning electron microscopy. Vascular casts and India ink injections revealed the presence of a number of isolated capillary tufts consisting of clusters of one to five "glomeruli," closely associated with the major vesicular vessels along the lateral walls of the bladder, and supplied by tertiary branches of these vessels. Light and electron microscopy showed that the capillary tufts represented the blood supply to small, ovoid lymph nodes located near the serosal surface of the bladder wall and usually restricted to the basal half of the bladder. These nodes were encapsulated and exhibited subcapsular sinuses, numerous small blood vessels, a limited number of high endothelial cells, and, occasionally, nerves and a follicular substructure. The nodes contained abundant lymphocytes, stellate stromal cells, macrophages, and eosinophils, but lacked the obvious cortical and medullary organization and germinal centers often seen in larger lymph nodes. Vascular corrosion casts, vascular ink injections, and microscopic examination confirmed the presence of small, highly vascular lymph nodes closely associated with the main vesicular vessels along the lateral walls of the rabbit bladder. A follicular substructure of the nodes appears to correspond with the "glomerular" capillary arrangement within the nodes as seen with corrosion casts. The rich blood supply may be indicative of the high metabolic demand of lymphatic tissue, and may be altered in response to the level of activity of the node. The close association between the lymphatic tissue and the rich blood supply to the nodes may allow a rapid mobilization of lymphocytes during a local immune response to foreign agents.

Endothelialization of a new Dacron graft in an experimental model: light microscopy, electron microscopy and immunocytochemistry.

Two types of synthetic vascular grafts, Dacron Triaxial and Dacron Gelseal Triaxial, were implanted into both the common carotids of sheep. The animals were sacrificed 1, 2, 8, and 16 weeks after surgery. Multiple specimens, obtained from grafts and anastomoses, were studied by light microscopy, transmission and scanning electron microscopy. A parallel immunocytochemical analysis was performed on some specimens. Dacron Triaxial grafts failed to develop a complete neointimal coverage. Myofibroblasts and fibroblasts were the dominant cells in such synthetic graft. Moreover, focal areas of stripping, platelet deposition, and thrombosis were observed at 8 and 16 weeks. In contrast, a stable endothelial coverage developed on the Gelseal Triaxial grafts after 16 weeks.

Study of gastrointestinal stromal tumors by light microscopy, electron microscopy and immunohistochemistry.

OBJECTIVE: To investigate the morphological features, immunohistochemical speciality of the gastrointestinal stromal tumors (GISTs), and its histogenesis as well. METHODS: The morphologic characteristics of GISTs were studied in 65 cases using light microscopy and 17 cases by electron microscopy. The expression of c-kit (CD117), CD34, vimentin, SMA, actin, desmin, S-100 and MBP were detected in all the cases with EnVision trade mark staining. RESULTS: Among 65 cases of GISTs, 46 were spindle cell type, 6 epithelioid cell type and 13 mixture type, equivalent to 85.5% (65 of 76) of all of the mesenchymal tumors of gastrointestinal tract admitted in the same period. The epithelioid cell type tumors composed of mainly the epithelioid cells, predominantly short spindle, oval or round in pattern, with an overall eosinophilic cytoplasm by hematoxylin-eosin stain. Focal cytoplasmic vacuolization was often seen. Sometimes signet-ring like cells and cells with a clear cytoplasm were seen in the epithelioid stromal tumor. The tumor cells arranged in interlacing fascicles forming whorls or sometimes cell clusters. Electronic microscopy showed the presence of interdigitating cell processes, in some areas, synapse-like structure and numerous desmosome-like junctions as well as a few gap junctions and small round neurosecretory granules. There were also abundant intermediate filaments and thin filaments of actin-type with longitudinal condensations (dense bodies). All of the 65 stromal tumors were strongly positive for vimentin (100%), 61 out of 65 tumors positive for CD117 (c-kit) (93.8%) and 51 out of 65 positive for CD34 (78.5%). Some cases also expressed SMA, actin, S-100 and MBP. CONCLUSIONS: GISTs were the most frequent mesenchymal tumor seen in the gastrointestinal tract. Under light microscope, the morphology of stromal tumors looks sometimes like a leiomyoma and Schwannoma. The application of immunohistochemical markers (particularly CD117 and CD34) and ultrastructural study are considered necessary for the differential diagnosis. GISTs may originate from the pluripotential precursor cells like the interstitial cells of Cajal.

Calcification in a pineal tumour studied by transmission electron microscopy, electron diffraction and x-ray microanalysis.

The calcification in a totally calcified pineal tumour was studied. Transmission electron microscopy indicated that the tumour was a pinealoma. The type of calcification was investigated by bright-field and dark-field transmission electron microscopy in addition to electron diffraction and electron-induced x-ray fluorescence. The calcified material consisted predominantly of amorphous calcium phosphate. The type of calcification differs from the normal calcification present in pineal acervuli which consists of crystalline hydroxypatite.

The significance of histologic analysis of skin lesions in porphyria cutanea tarda. Light microscopy, electron microscopy, immunohistochemical and immunofluorescence analysis.

Porphyria cutanea tarda (PCT) is one of several entities in the differential diagnosis of scleroderma. We report a 62-year-old man with PCT diagnosed since two decades. Clinical data and the results of light microscopy, immunohistochemistry, immunofluorescence and electron microscopy of skin biopsies are presented. The biopsy revealed sclerosis of the dermis mainly due to increased collagens I and III, and accumulation of collagen IV, which had caused the vessel wall to thicken. Immunofluorescence for detection of immune reactants was negative. It was concluded that the histomorphology of PCT of long duration may be similar to that of scleroderma. Nevertheless, in addition to clinical findings, detailed skin biopsy studies including contemporary techniques can contribute to the differentiation of these diseases.

Earliest enamel deposits of the rat incisor examined by electron microscopy, electron diffraction, and electron probe microanalysis.

In order to describe initial events in enamel mineralization and to help characterize inorganic-organic interactions in this tissue, the earliest rod and interrod enamel in mandibular incisors from normal young adult (100 gm) rats, perfused with 100% ethylene glycol, has been studied by transmission electron microscopy, selected area electron diffraction, and high-spatial-resolution electron probe microanalysis. Diffraction and probe data were correlated precisely from the same extracellular regions of the tissue. Sites were examined progressively as a function of location a) from the most recently deposited enamel adjacent to ameloblasts toward the dentin-enamel junction and b) from the apical portion of the tooth longitudinally toward its incisal end. Electron diffraction patterns consistent with that of a poorly crystalline hydroxyapatite were generated at all locations. Diffraction characteristics changed only slightly toward that of more crystalline hydroxyapatite at different locations. Earliest apical enamel generated molar Ca/P ratios in a range of 0.99-1.46 (average 1.24 +/- 0.15). Molar Ca/P ratios of the first enamel interrod elements increased from approximately 1.24 at ameloblast-enamel boundaries to approximately 1.40 at the dentin-enamel junction, small changes corresponding to those observed in electron diffraction characteristics.

Definitive support by transmission electron microscopy, electron diffraction, and electron density maps for the formation of a BCC lattice from poly[N-[3,4,5-tris(n-dodecan-1-yloxy)benzoyl]ethyleneimine].

Transmission electron microscopy (TEM), electron diffraction (ED), and electron density maps (EDM) experiments were carried out on a poly[N-[3,4,5-tris(n-dodecan-1-yloxy)benzoyl]ethyleneimine] [poly[(3,4,5)12G1-Oxz]] with a degree of polymerization (DP) of 20. All experiments confirmed the thermotropic body-centered cubic (BCC) Im3m lattice suggested previously by X-ray diffraction (XRD) experiments. The unit cell parameter determined by ED at 23 degrees C is a = 42.4 A, in good agreement with XRD results which show a = 42.6 A after quenching from 70 degrees C. EDM of the XRD results confirm that the supramolecular minidendrimer obtained from poly[(3,4,5)12G1-Oxz] adopts a spherical "inverse micellar-like" structure, with the polyethyleneimine backbone and the aromatic groups microsegregated and concentrated in the corners and in the center of the cubic unit cell. A space-filling continuum is realized by the n-alkyl groups that radiate out of the aromatic core of the spherical dendrimer. This manuscript is only the second example of complete structural analysis of a lattice generated from supramolecular objects and complements the previous example reported from our laboratory on the Pm3n lattice.

Histiocytoid hemangioma with features of angiolymphoid hyperplasia and Kaposi's sarcoma. A study by light microscopy, electron microscopy, and immunologic techniques.

We examined by light and electron microscopy 99 vascular lesions removed on 17 occasions over a 2-year period from a 55-year-old black man. The lesions all showed histologic features of a vascular neoplasm composed of enlarged "histiocytoid" endothelial cells and overlapping features of angiolymphoid hyperplasia with eosinophilia and Kaposi's sarcoma. An apparently unique feature was early loss of melanin from the overlying epidermis and a mononuclear inflammatory-cell infiltrate. The patient also had an abnormal immunologic state very similar to that seen in homosexuals with Kaposi's sarcoma.

Chondroitin sulfate facilitates an acrosome reaction in bovine spermatozoa as evidenced by light microscopy, electron microscopy and in vitro fertilization.

Bovine epididymal spermatozoa were incubated for 22 h in a modified Tyrode's medium. The percentages of sperm exhibiting an acrosome reaction were determined morphologically after fixing and staining specimens. The addition of chondroitin sulfate A (CS-A) significantly increased the incidence of acrosome reaction. When observed by electron microscopy, acrosome-reacted sperm had undergone vesiculation of the plasma and outer acrosomal membranes. Sperm incubated in the presence of CS-A demonstrated a significantly higher incidence of vesiculation when compared to the controls. Additionally, rates of in vitro fertilization of bovine oocytes were significantly elevated when sperm and ova were exposed to CS-A. These results suggest that glycosaminoglycans in the female reproductive tract may be responsible for some of the biochemical changes associated with fertilization, and a light microscope procedure can be used to assess occurrence of the acrosome reaction.

Demonstration of Chlamydia trachomatis in inguinal lymphadenitis of lymphogranuloma venereum: a light microscopy, electron microscopy and polymerase chain reaction study.

Intravacuolar organisms in vacuolated macrophages were associated with areas of necrosis and suppuration in 12 patients with suppurative inguinal lymphadenitis. The intravacuolar organisms measured 0.2 to 2.0 micrometers in diameter, stained Gram negative with the Brown-Hopp's tissue Gram stain, faintly blue with hematoxylin and eosin stain, and black with the Warthin-Starry silver impregnation stain. The organisms lined vacuolar membranes and/or clumped in centers of vacuoles. Electron microscopy revealed elementary and reticulate bodies and intermediate forms characteristic of the genus Chlamydia. Cultures of three lymph nodes in McCoy cells grew Chlamydia trachomatis, lymphogranuloma venereum (LGV) serovars. Polymerase chain reaction using primers for chlamydial 16S ribosomal DNA confirmed the organisms as Chlamydia in lymph nodes from nine patients. Recognition of chlamydial organisms by light microscopy in tissue sections of lymph nodes allows a definitive diagnosis of lymphogranuloma venereum.

Localization of gallium in tumor cells. Electron microscopy, electron probe microanalysis and analytical ion microscopy.

The incorporation of gallium in tumor cells in vitro and in solid tumors was studied with electron microscopy and two microanalytical techniques. It was thus possible (1) to directly prove that gallium is concentrated in lysosomes of tumor cells; (2) to demonstrate that in these lysosomes, gallium is precipitated with phosphorus; and (3) to propose a mechanism of gallium concentration in tumor cells; in this mechanism, the accumulation of gallium as phosphate salts in lysosomes is directly related to the acid phosphatase activity of these organelles.
