Aberrations of chromosome 8 in mixed salivary gland tumors--cytogenetic findings on seven cases.

Cytogenetic findings on seven mixed salivary gland tumors are reported herein. The involvement of chromosome #8 in clonal chromosome aberrations in five of the seven tumors was particularly noteworthy. Four tumors had translocations involving chromosome #8 and one or two other chromosomes (#3, #7, #9, #13). The fifth showed a deletion of parts of the long arm of chromosome #8. In an attempt to define the critical segment on chromosome #8, we have identified the part between 8q11 and 8q13 as the critical region involved in all rearrangements. Thus far, our results confirm the results of the Swedish group, though the percentage of cases having #8 abnormalities is somewhat higher in our small series. The relationship between the two groups of cases, those with and those without chromosome abnormalities, will be discussed.

Immunocytochemistry of acinic cell carcinomas and mixed tumors of salivary glands.

Antisera of several secretory products of the salivary gland were used to investigate the histogenesis of acinic cell tumors and mixed salivary gland tumors for comparison. Amylase, lactoferrin, secretory piece, and proline-rich protein (PRP) immunoreactivity was detected in the majority of acinic cell tumors; staining was focal, except for PRP, which was diffuse. Lysozyme immunoreactivity was rare. There was discordance for immunoreactivity with several antisera in identifiable tumor lobules of half of the neoplasms. An antikeratin serum outlined microcystic and follicular areas but rarely solid foci. These findings support the contention that acinic cell tumors derive from a tubular type stem cell. Lactoferrin and secretory piece immunoreactivity was not common in mixed tumors and was confined to scattered ductal cells and luminal contents. Rare small foci of amylase and PRP immunoreactivity were found in two mixed tumors only.

Atypical features in salivary gland mixed tumors: their relationship to malignant transformation.

Although criteria for distinction between the benign and malignant elements in carcinoma ex mixed tumor have been adequately described, there have not been any attempts to identify clinical or histologic features in benign mixed tumors that indicate increased risk of malignant change. For this reason, 65 mixed tumors of the major and minor salivary glands that exhibited atypical histologic features were examined in an attempt to analyze which, if any, of these features might indicate a greater likelihood of malignant transformation. The atypical features evaluated were hypercellularity, capsule violation, hyalinization, necrosis, and cellular anaplasia. The mitotic rate was also analyzed. The age of the patient, and the site, size, and prediagnostic duration of the tumor were recorded and, together with the histologic findings, were correlated with follow-up information. Nine (13.8%) of the 65 tumors underwent malignant transformation. Five of these patients died of the tumor, two others had distant metastases and were alive with the disease, and two were free of disease. Benign mixed tumors that showed prominent zones of hyalinization or at least moderate mitotic activity were more likely to develop carcinoma than those that did not. chi 2 analysis indicated that only hyalinization was significant (P < 0.05), but, with Fisher's exact test (two-tailed), this and all of the other features evaluated revealed a P value greater than 0.05. The other atypical features failed to correlate with malignant change. Clinical findings at the initial diagnosis that indicated a greater likelihood of malignant transformation were occurrence in the submandibular gland, older patient age, and large tumor size.

[Cancer en cuirasse, induced by a malignant mixed salivary gland tumor].

A case of cancer en cuirasse is reported, deriving from a metastasizing malignant mixed tumor of the lacrimal gland. Based on histological criteria, the primary benigne prognosis of mixed tumors of lacrimal glands is discussed. According to sensitivity testing to cytostatic agents treatment with 375 mg of bleomycin achieved a recurrence-free interval of eight months. Pathogenesis of secondary effects are discussed (so-drug-induced called bleomycin-lung).

Polyclonal chromosomal evolution in a benign mixed salivary gland tumor.

Banding analyses of a human benign pleomorphic adenoma in the parotid gland revealed a polyclonal pattern where structural rearrangements predominated. These deviations were different from the anomalies previously observed in 100 mixed tumors. The reason found for the differences in all likelihood was x-ray treatment of tuberculous lymphadenitis in the neck during childhood. Implications regarding origin and development of pleomorphic adenomas are discussed.
