Characterization and application potential of two newly isolated phages targeting the prevalent multidrug resistant Salmonella serovars in China.

The escalating global threat of multidrug resistant (MDR) Salmonella, a foodborne pathogen with animal-derived foods serving as the primary transmission vehicle, underscores the urgent need for effective lytic phages for biocontrol. From 142 environmental and farm samples in Shandong Province, we isolated 103 phages active against MDR S. Enteritidis and S. Typhimurium, which were the most prevalent Salmonella serovars in China. Two Siphoviridae phages vB-SenS-S1 and vB-SenS-SEC2 were selected for further study. With optimal multiplicities of infection (MOIs) of 10-2 (vB-SenS-S1) and 10-5 (vB-SenS-SEC2), both phages exhibited a 20 min latent period, yielding burst sizes of 52 and 37 PFU/cell, respectively. They also demonstrated stability across a range of temperatures (50-60 °C), pH levels (5-11), and after 1 h of UV exposure. Genomic analysis identified vB-SenS-S1 (43,002 bp, 47.04% GC) and vB-SenS-SEC2 (42,948 bp, 47.65% GC) as novel double-stranded DNA phages. Functional annotation confirmed the presence of genes essential for structural assembly, host lysis, and DNA replication/metabolism, and also verified the absence of resistance, virulence, and lysogeny-associated genes. Both phages vB-SenS-S1 and vB-SenS-SEC2 exhibited synergy with colistin and tetracycline. The synergy with colistin was particularly potent, leading to complete bacterial eradication in vitro. The in vivo therapeutic efficacy was further validated in both Galleria mellonella larvae and murine models of MDR Salmonella infection. Combination therapy with vB-SenS-SEC2 and colistin not only dramatically increased survival but also achieved a significant reduction in bacterial burden across multiple visceral organs of infected mice. Moreover, vB-SenS-S1 (108 PFU/mL) completely inhibited MDR Salmonella on chicken meat at 4 °C and -20 °C when initial contamination was ≤103 CFU/mL. This study not only expands the diversity of Salmonella phages but also highlights their potential as biocontrol agents in both clinical veterinary use and food decontamination, thereby enhancing food quality and safety at both the meat production source and the terminal product.

Of the numerous zoonoses part of which are reportable, according to the Federal Communicable Diseases Act, the food hygienist attributes topicality at present to salmonellosis, brucellosis, toxoplasmosis, taeniasis (T. saginata), trichinellosis, and sarcosporidiosis. In salmonellosis, combating is directed to breeding and keeping of Salmonella-free flocks, poultry offering favourable conditions for this purpose. Moreover, one tries to decontaminate feeds through certain pelleting machines with the aid of friction heat or by admixing propionic acid. In brucellosis, the possibility of infection by tourism, but also that through ingestion of imported dairy products (like soft cheese) play a certain role. Instruction of tourists and advising foreign labour from endemic areas are essential contributions to combating. Another important infection source of toxoplasmosis is infected raw pig meat. As the agent does not survive freezing temperatures under certain conditions, the decontamination procedure is particularly suitable. In pregnancy advising, the role of raw pig meat for development of congenital toxoplasmosis should be clearly explained. In trichinellosis, apart from the traditional examination by the trichinoscope, the modified digestion procedure using pepsin as a digestive fluid was successful. Moreover, it is suitable for detecting weakly infected swine. The importance of systematic examinations of all domestic and wild pigs became evident only recently in 1977, when in Bavaria an epidemic of trichinellosis occurred following the ingestion of undetected Trichinella containing wild pig meat. According to most recent knowledge on the cycle of development of sarcosporidia, the studies showed that the species Sarc. bovihominis and suihominis were pathogens for human beings. Quite apart from necessary supplementary examinations, the freezing procedure would here too, warrant an effective protection for the raw meat consuming population.

Factors in the spread of salmonellas in meatworks with special reference to contamination of knives.

Immersion of knives momentarily in hot water (82 degrees C) was ineffective in destroying salmonellas on knives used in a meatworks to carry out the bung dropping operation. Laboratory experiments confirmed that knives covered with meat products required 10 or more seconds to be effectively decontaminated at this temperature. Examination of knives used for slaughtering and for dressing beef carcases showed that knives coming into contact with hides had higher counts for salmonella and a higher percentage positive than knives used for other cutting operations. Knives used for cutting the skin of the forelegs and hindlegs had the highest counts.

Chernobyl fallout: internal doses to the Norwegian population and the effect of dietary advice.

Dietary studies and whole-body measurements were used to estimate the intake of radiocesium and the radiation dose received by different groups of people in Norway after the Chernobyl accident. Freshwater fish, milk, and reindeer meat were the major sources for radiocesium intake. Dietary advice, together with agricultural decontamination measures, resulted in a considerable reduction in the exposure level of the population. A majority (40-80%) of the specially selected groups (farmers-hunters and Sami reindeer herdsman) changed its diet significantly after the accident. Without dietary changes, specifically a reduction in the consumption of freshwater fish and reindeer meat, the Sami group would have had a 400-700% higher radiocesium intake, and the farmers-hunters' intake would have been up to 50% higher than what they actually had experienced.

[Decontamination of hot beef carcases with organic acids (author's transl)].

The decontamination effect of spraying hot beef carcases with solutions of lactic acid and solutions of the commercial preparation of pH liquid was studied. Spraying a 0.5 per cent lactic acid solution only resulted in a significant reduction of aerobic, gram-negative and gram-positive bacterial counts on severely contaminated areas. Treatment with a 0.75 per cent solution of lactic acid caused a significant decrease of almost all bacterial counts, whereas a 1 per cent lactic solution produced a significant reduction of all bacterial counts at the sites sampled. The maximum reduction was 2 log units per sq.cm. and varied with the level of contamination. A 5 per cent pH Liquid solution was ineffective and a 20 per cent pH Liquid solution had an effect similar to that of a 0.5 per cent lactic acid solution. The reduced bacterial counts persisted during three days of storage in the chilling room. When the above concentrations are applied, this will not cause any permanent changes of colour of the treated carcases. These changes do occur when a 10 per cent solution of lactic acid is used. This treatment can only be used to spray severely contaminated superficial layers which are to be removed. Recommendations are made for the possible use of organic acids in the slaughter line. The legal aspects are discussed.

Destruction of bacteria on fresh meat by hot water.

Strains of different bacterial genera; Escherichia coli (7), Salmonella serotypes (7), Enteropathogenic E. coli O 157 (2), Aeromonas hydrophila (4), Yersinia enterocolytica (1), Pseudomonas fragi (5), and Listeria monocytogenes (5), inoculated onto the surface tissues of pieces of fresh beef were found to be susceptible to the lethal action of hot water. On average, more than 3 log10 (99.9%) of these organisms were destroyed when water at 80 degrees C was applied for 10 or 20 s. Therefore, a hot water decontamination cabinet would be effective for destroying in situ any viable cells of these bacteria which might be present on the surface tissues of fresh beef carcasses.

Management of land and livestock contaminated with polychlorinated biphenyls.

Investigations were conducted on several small neighbouring beef cattle and sheep farms that were found to be contaminated with polychlorinated biphenyls (PCBs). The residues detected contained about 62% w/w chlorine and soil concentrations ranged from 0.35 to 1.9 mg/kg. Beef cattle and sheep grazing the contaminated land had PCB concentrations in their fat of 0.3 to 1.7 mg/kg and 0.19 to 0.45 mg/kg, respectively. The concentration of PCBs in the caudal fat of cattle was 0.3 to 2 times the concentration of PCBs in the land they were grazing and was positively related to stocking rates. PCBs were present in milk fat from cattle at about the same concentration as in caudal fat, and lactation appeared to contribute to decontamination. Wool grease from sheep contained about 1 to 2 times the concentration of PCBs in subcutaneous fat. Calves of contaminated cows also became contaminated. The half-life for decontamination of PCBs in a group of 8 young female beef cattle was calculated at 13 months. PCBs were detected in pasture samples at concentrations ranging from less than 0.01 mg/kg to 0.12 mg/kg. However, PCBs were not detected in any sample of hay cut from these paddocks. In 4 sheep fed a sole ration of this hay, traces of PCBs were detected in their fat following 89 days of feeding. Grazing livestock for meat production on land contaminated with PCBs is not recommended because livestock readily acquire residues, and PCBs persist in soil and livestock for long periods. Grazing sheep for wool production on land contaminated with PCBs may be an option.
