Effect of the interferon inducer tilorone in inbred CBA mice.

Tilorone hydrochloride (200 mg/kg) was administered intragastrically to inbred CBA mice. After 5, 18 and 48 hr the number of circulating leucocytes and peritoneal cells as well as the migration of unstimulated peritoneal cells, the blood corticosteroid level and interferon production were investigated. In spite of the considerable decrease of the number of mononuclear cells in the blood and polynuclear ones in the peritoneal exudate, the drug induced production of circulating interferon and stimulated its synthesis by peritoneal cells. The blood corticosteroid level and the mast cell count in the peritoneal cavity were significantly elevated, but the migration of peritoneal cells in antigen-free medium decreased.

[Further study of the hormonal regulation of natural immunity to weak transplantation antigens in inbred mice].

Blood plasma corticosterone level and the extent of immunological reaction to sex-antigen of skin transplant in inbred mice CBA, C57Bl/6, F1 (CBA X 57Bl/6), C3H, AKR was compared. Mice CBA had a high blood plasma corticosterone level and no immunological reaction to sex-antigen. A lower blood corticosterone level and the presence of reaction to sex-antigen was characteristic of mice C57Bl/6, AKR, C3H, F1(CBA X C57Bl/6). The number of endogenous colonies in the spleen of mice CBA was less than in mice C57Bl/6. Mice CBA had a narrow cortical thymus layer in comparison with the wide thymus cortex of mice AKR and C57Bl/6. Adrenalectomy of mice CBA caused a sharp widening of the thymus cortex.

[Body constitution of growing inbred animals under hypo-, normo- and hyperkinetic conditions].

The work was carried out in inbred mice C57BL/6, CBA, CC57Br/M strains, interstrain hybrids of the first generation FF1 and in rats of August and Wistar strains. Only one-month-old male animals, which were kept for two months under the conditions of hypo-, normo- and hyperkinesia were used in the study. Statistically significant relation between underloading, overloading and habitus changes was demonstrated. It was stated that habitus changes under the conditions of hypo-, normo- and hyperkinesia were genetically determined. Hereditary predisposition of the reactionary norm and the phenomenon of heterosis manifest themselves under different motor regimens.

Effects of B4 Coxsackievirus infection on cell-mediated immunity, natural cytotoxicity and interleukin-1 production by spleen cells, in mice.

By using CBA inbred mice as responders, the influence of in vivo B4 Coxsackievirus infection (in two separate experimental procedures) on anti-virus cell-mediated immunity (CMI), natural killer (NK) cell activity and lipopolysaccharide (LPS) induced interleukin-1 (IL-1) production in spleen cells were studied. Both experimental infection schedules were able to induce a "positive" CMI response. NK cell activity was also significantly stimulated, especially by the administration of high virus amounts/inoculum, with frequent injections. On the contrary, LPS-dependent IL-1 synthesis increased particularly following application of the other experimental schedule (low virus amounts, few injections). Some aspects concerning CMI, NK and IL-1 relationships, during several enteroviral diseases, are discussed.

Different response of maturing oocytes from two inbred strains of mice to sperm penetration.

Oocytes from two inbred strains of mice, CBA and KE, were inseminated at prometaphase of the first meiotic division. Pronuclei were formed in 21-36% of inseminated oocytes from the CBA strain. In the KE strain the formation of pronuclei occurred in 2-5% of oocytes and was at the same level as in the non-inseminated control. These results show that in oocytes of the CBA strain maturity of the cytoplasm is acquired earlier than in those of the KE or other so far studied strains of mice. This fact is discussed in the context of different maturation rates of oocytes from different strains. In oocytes which remained non-activated after penetration, transformation of sperm heads into separate chromosomes was observed. An interstrain difference in efficiency of this process was also found.

Genetic similarities and differences between sublines of certain inbred strains of mice held within Australia.

Inbred mice of the strains C57B1/6, CBA, C3H, Balb/c and DBA/2 from a total of 17 different sources within Australia were compared, utilizing 12 different blood proteins and a morphometric analysis of a series of lower jaws. Evidence for genetic differentiation between sublines was found in only two cases. Evidence for the recent contamination of an inbred strain with another stock was found in one case.

Effect of T-lymphocytes on normal haemopoiesis: studies in congenitally athymic nude mice.

The germ-free nude mouse represents a most useful animal for investigating the effects of a congenital deficiency of T-lymphocytes on various physiological processes, including haemopoiesis. Nude mice (nu/nu) of the CBA strain, nonmutant inbred CBA mice, and inbred C3H mice were reared in germ-free isolators and used to compare (1) the haematological parameters of nu/nu mice and CBA mice at different ages and (2) the labelling pattern of circulating leucocytes at various times after a single intraperitoneal injection of 3H-thymidine into 3-month-old nu/nu and C3H mice. The data suggest that the deficiency of T-lymphocytes in nu/nu mice may lead to a disturbance of haemopoiesis in 2 to 6-month animals which is characterised by a mild macrocytosis and a very marked reduction in the proportion of labelled leucocytes which can be seen in the blood after an injection of 3H-thymidine. Despite these perturbations, unstressed nu/nu mice were able to maintain adequate numbers of blood cells (other than lymphocytes) in their circulation. Nine-month-old nu/nu mice did not show a macrocytosis and the disappearance of the marcrocytosis at this age was associated with an increase both in the blood lymphocyte count and in the mass of lymphoid tissue in the spleen and mesenteric lymph nodes.

[Morphological and functional interrelationships of the thymus and adrenals in inbred mice].

The authors carried out morphological examination of the thymus, adrenal glands and the spleen in CBA, C57BL/6, AKR mice and studied the immunological reactivity of mice to sex antigen of the skin transplant with the determination of the blood plasma corticosterone level. In comparison with C57BL/6 and AKR mice, CBA mice displayed a narrow cortical layer in the thymus, wide thymus-dependent zones in the spleen, a high corticosterone level, and the absence of any reaction to the sex antigen of the skin transplant. Adrenalectomy in CBA mice led to a sharp dilatation of the thymus cortex, the appearance of numerous lymph follicles in the spleen and development of an immunological reaction to sex antigen of the skin transplant. Atrophy of the cortical layer of the adrenal glands witha sharp reduction of subanophilic lipids was noted in CBA B-mice.

Genetics of resistance to infection with Salmonella typhimurium in mice.

Eight strains of inbred mice fell into two sharply defined groups. Four strains (CBA, A/JAX, C3H/He, and DBA/2) were resistant (LD50, greater than 10(5)) to Salmonella typhimurium C5 given subcutaneously. The other four strains (Balb/c, C57BL, B10.D2 [new line], and DBA/1) were susceptible (LD50, less than 10). No intermediate resistance was seen. Examination of the F1, F2, and parental backcross generations bred from matings of CBA and Balb/c mice showed that resistance behaved as a simple Mendelian dominant. Resistance was not linked to H-2 genes, and no useful marker has yet been found. However, as previously demonstrated in the parent strains, resistance in the hybrids was related to the ability to produce a good delayed-type hypersensitivity reaction to an extract of S. typhimurium.

F antigen. III. Immunological and biological properties.

Relationship of the isoforms of F antigens from A2G and CBA inbred mouse strains have been worked out in detail in respect to their immunogenicity. Mice from several inbred strains have been shown to possess the ability of forming anti-F upon stimulation with xenogeneic F antigens from certain species. An immunoelectrophoretic method for sensitive quantitation of the antigen is described. During human ontogenesis the antigen appears very early. It could never be detected in serum except in cases of acute liver disease. A small fragment set free under denaturing conditions showed the ability of inhibiting precipitation of the complete antigen by antibody. The F protein seems to be a vehicle for this small fragment possessing haptenic properties and probably being responsible for the universal cross-reactivity.

Frequencies of mitogen-reactive B cells in the mouse. I. Distribution in different lymphoid organs from different inbred strains of mice at different ages.

Frequencies of mitogen-reactive B cells have been determined in vitro under culture conditions which allow every growth-inducible B cell to grow and mature into a clone of Ig-secreting PFC. The frequencies of LPS-reactive B cells in the spleen of 6- to 8-wk old mice were between 1 in 3 and 1 in 10 splenic B cells from the following inbred strains of mice: C3H/Tif; BALB/c; BALB/c nu/nu; C57BL/6J; DBA/2J; C57BL/6J x DBA/(2J)F(1); and CBA and A/J. Very similar frequencies are found for lipoprotein-reactive B cells in BALB/c, BALB/c nu/nu, C3H/Tif, and C3H/HeJ mice. No LPS-reactive cells but normal frequencies of lipoprotein-reactive cells were found in C3H/HeJ mice, genetically nonreactive to LPS. SJL mice had significantly lower frequencies of LPS- and of lipoprotein-reactive B cells (1 in approximately 30 B cells). The number of LPS- and of lipoprotein-reactive B cells in spleen was dependent upon the age of the mouse. Newborn spleen contained approximately 10 percent of the number of reactive cells found at 6- to 8-wk of age. From there the frequencies declined again to drop below 5 percent of the maximal number at ages beyond 11 mo. LPS-reactive B cells yielding IgM- and IgG-PFC responses could be found in mesenteric lymph nodes, bone marrow, thymus, thoracic duct, and peripheral blood of 6- to 8-wk old mice. Their frequencies were one in three to five lymph node cells, 1 in 50 to 100 bone marrow cells, one in 10(5) thymus cells, and 1 in 20 to 40 thoracic duct or peripheral blood cells.

The expression of IV6 beta[Gal beta 1-4(Fuc alpha 1-3)GlcNAc]-Gb5Cer in mouse kidney is controlled by the Gsl-5 gene through regulation of UDP-GlcNAc:Gb5Cer beta 1-6N-acetylglucosaminyltransferase activity.

We reported the polymorphic expression of GL-Y (IV6 beta[Gal beta 1-4(Fuc alpha 1-3)GlcNAc]-Gb5Cer in kidneys of inbred strains of mice in previous papers [J. Biochem. 101, 553-562 and 563-568 (1987)]. DBA/2 mice express a large amount of GL-X (Gb5Cer), but not GL-Y, in their kidneys, because of a defect on a single autosomal gene (Gsl-5). This suggested that DBA/2 mice lack the ability to transfer GlcNAc onto the C-6 position of GalNAc of Gb5Cer or GL-X. In this study, we characterized UDP-GlcNAc:GL-X beta 1-6N-acetylglucosaminyltransferase (beta 1-6GlcNAc transferase) in the microsomal fraction of mouse kidney. Maximum activity was detected with an incubation mixture containing sodium cacodylate buffer (pH 6.4), 0.1% Zwittergent 3-16 and 1 mM EDTA. Divalent cations were not required. The apparent Km values for UDP-GlcNAc and GL-X were 0.42 and 0.12 mM, respectively. The product of the enzymatic reaction was identified as IV6 beta GlcNAc-Gb5Cer by means of 1H-NMR spectroscopy and permethylation analyses. Then, we measured the beta 1-6GlcNAc transferase activity in the microsomal fractions of kidneys of inbred strains of mice and progeny obtained on mating. WHT/Ht, C57BL/10, BALB/c, and C3H/He mice, which express GL-Y in their kidneys, exhibited detectable amounts of activity, whereas CBA and DBA/2 mice, which do not express GL-Y, did not exhibit detectable activity.(ABSTRACT TRUNCATED AT 250 WORDS)

Physical characteristics of mouse sperm nuclei.

The nuclei of epididymal sperm, isolated from C57BL/6J and CBA/J inbred mice by their resistance to trypsin digestion, retain the shape differences of the intact sperm head. Various physical characteristics of these nuclei were measured and compared. The measurement of the projected dimensions of nuclei showed that the CBA nuclei are 13.5% longer than C57BL/6 nuclei (8.64 +/- 0.02 mum compared with 7.61 +/- 0.02 mum), 0.8% narrower (3.51 +/- 0.01 vs. 3.54 +/-0.01 mum) with 6.8% more area (22.34 +/- 0.10 vs. 20.91 +/- 0.09 mum2). However, the volumes of the nuclei as based on reconstructing calibrated electronmicrographs of serial sections of the nuclei indicated that CBA are about 7% smaller than C57BL/6 nuclei (3.72 +/- 0.08 vs. 4.01 +/- 0.03 mum3). The buoyant density of the CBA nuclei is 1.435 +/- 0.002 g/cm3 compared with 1.433 +/- 0.002 g/cm3 for the C57BL/6 nuclei as determined on linear CsCl and Renografin-76 density gradients and confirmed by a technique utilizing physiological tonicities. Therefore, the average mass of the CBA nuclei is less than that of the C57BL/6 nuclei (5.34 +/- 0.12 vs. 5.75 +/- 0.05 pg). The sedimentation velocities at unit gravity of nuclei from 11 inbred strains differ over a range of more than 6% with CBA nuclei sedimenting about 2.0% more slowly than C57BL/6 nuclei. We show that for these nuclei the sedimentation velocity can be related to their buoyant density, volume and a sedimentation shape factor. Within the errors of our measurements of these various characteristics, it was found that C57BL/6 and CBA nuclei have similar sedimentation shape factors. Therefore, the difference in sedimentation velocity between these nuclei appears to be primarily a result of differences in volume. The possible applications of these techniques to the physical separation of sperm are evaluated in the discussion.

Genetic differences in BCG-induced resistance to Schistosoma mansoni are not controlled by genes within the major histocompatibility complex of the mouse.

Induction of nonspecific resistance to Schistosoma mansoni infection after the i.v. injection of viable BCG was investigated in outbred mice and a panel of inbred and H-2 congenic strains. Significant protection was induced in CF1, A/J, C57BL/6, C57BL/10, DBA/2, C57BR, and SJL mice. BALB/c mice were not protected whereas CBA and C3H mice expressed intermediate degrees of protection. Expression of the protective phenomenon is not controlled by genes within the MHC as shown by the marked differences in response between BALB/c and DBA/2 (H-2d) as well as between C57BR and C3H (H-2k) mice. H-2 congenic strains with C57BL/10 background (B10.A and B10.D2) were high responders. BALB.B10 mice carrying the high responder (B10) MHC on the nonresponder (BALB/c) background were not protected. The degree of splenic hypertrophy did not correlate with the expression of nonspecific resistance. These results demonstrate that, in addition to controlling specific immune responses, genetic differences influence the nonspecific protective phenomena related to BCG administration as well.

Inheritance of antibody specificity. II. Anti-(4-hydroxy-5-bromo-3-nitrophenyl) acetyl in the mouse.

Mice of 17 inbred strains produced anti-(4-hydroxy-5-bromo-3-nitrophenyl)-acetyl (NBrP) of three different fine specificity types. Anti-NBrP antibodies of all allotype b mice (five strains tested) had a high relative affinity for (4-hydroxy-3.5-dinitrophenyl) acetyl (NNP) but low for (4-hydroxy-5-cloro-3-nitrophenyl) acetyl (NCP). Another category was characterized by high relative affinity for NCP but low for NNP. This category included most of the tested strains. The third category (CBA and C3H strains) had an intermediate fine specificity. Associated with fine specificity characteristics were anti-NBrP titers, mice of allotype b had lower titers than the other mice. Studies of congenic, recombinant inbred, F1 and backcross mice showed that both fine specificity and the magnitude of the anti-NBrP response of tbalb/C MICE WERE CONTROLLED BY AN ALLOTYPE-LINKED GENE. This gene was dominant over the C57BL/6 ALLELE. Lack of recombinant mice in the backcross generatioterns on the other suggest close linkage between the two genes.

Various epithelial and non-epithelial tumors spontaneously occurring in long-lived mice of A/St, CBA, C57BL/6 and their hybrid mice.

Various kinds of epithelial and non-epithelial tumors, and tumor-like lesions spontaneously developed in long-lived mice of strains A/St, C57BL/6 and CBA, and of (C57BL/6 X CBA) F1 hybrids, all of which had been fed in the 2nd Department of Pathology, Gifu University School of Medicine. Some tumors were successfully transplanted into the same strain of mice. An interesting tumor line showing phagocytic activity and growing only in the liver even when inoculated subcutaneously, which was obtained from a reticulum cell neoplasm, Type A by Dunn, originated in the liver of a (C57BL/6 X CBA) F1 hybrid male mouse.

Stereoselective enhancement of nociception by opioids in different strains of mice.

The effects of Mr2096, Mr2097, diprenorphine, (-)bremazocine and Mr2266 on jumping latencies were evaluated in Swiss, CXBK, C57BL, CXBH and CBA strains of mice. Mr2096 and Mr2097 respectively produced analgesia and hyperalgesia in these strains of mice. Thus their effects on nociception were mediated by stereoselective opioid receptors. Diprenorphine also produced hyperalgesia which might be mediated by both mu and kappa receptors. Kappa receptors also appeared to mediate hyperalgesia at least in Swiss and CBA strains, as both (-)bremazocine and Mr2266 shortened the jumping latencies.
