Kinetics of excision repair of UV-induced DNA damage, measured using the comet assay.

The kinetics of UV- (254 nm) irradiation-induced DNA single-strand breaks (SSBs), generated during the excision repair of UV-induced DNA damage, in leukemic lymphocytes and in normal blood mononuclear cells (MNCs) were studied using the alkaline comet assay. The cells were isolated by density gradient centrifugation from peripheral blood of patients with chronic lymphocytic leukemia (CLL) and from healthy study subjects. The cytotoxicity of UV irradiation was determined in vitro in peripheral blood mononuclear lymphocytes from 36 CLL patients and from eight healthy donors using the incorporation of radioactive leucine in 4-day cultures. A remarkable difference in excision repair capability was observed between normal and leukemic lymphocytes. In contrast to normal lymphocytes, there was always a subpopulation of CLL cells that did not complete the repair of UV-induced DNA damage during the 24-h repair period. Furthermore, differences were also recorded between UV-sensitive and UV-resistant CLL cases. The differences in DNA migration between the maximum increase (59-77 microm) and that at 24 h after irradiation (21-66 microm) was statistically significant in two of three patients exhibiting UV-resistance. Correspondingly, only in one of three patients exhibiting UV-sensitivity was the difference in DNA migration statistically significant (maximum increase: 44-107 microm, vs. 24 h after: 42-100 microm). Our results confirm an abnormal pattern of the CLL cell response to UV irradiation. Furthermore, we identified defective processing of UV-induced DNA damage in CLL versus normal lymphocytes, particularly in UV-sensitive cases.

Stromal cell-derived factor-1 as a chemoattractant for follicular center lymphoma B cells.

BACKGROUND: Follicular center lymphoma displays widespread lymph node involvement at diagnosis. The chemoattractants that control the locomotion of follicular center lymphoma B cells have not been established. Stromal cell-derived factor-1 (SDF-1) is a CXC-class chemokine that enhances the migration of normal human B cells and is expressed in peripheral lymphoid tissues. Here we have investigated 1) whether SDF-1 stimulates the in vitro locomotion of follicular center lymphoma B cells and of their presumed normal counterparts (i. e., germinal center B cells) and 2) whether the same cells express SDF-1 transcripts. METHODS: B cells were purified by immunomagnetic bead manipulation. Messenger RNA was detected by reverse transcription-polymerase chain reaction. Migration was assessed by the filter and collagen invasion assays. All P values were two sided. RESULTS: Follicular center lymphoma B lymphocytes showed a statistically significant migratory response to 300 ng/mL SDF-1, both in the filter and in the collagen assays (P =.002 for each). Such response was mediated by the SDF-1 receptor, CXCR4. CD40 monoclonal antibody (MAb) and tonsillar germinal center B cells treated with CD40 MAb and recombinant interleukin 4, but not freshly isolated, migrated statistically significantly faster in the presence than in the absence of SDF-1 (P =.002 in both filter and collagen assays). Freshly isolated follicular center lymphoma and germinal center B cells expressed SDF-1 transcripts. CONCLUSIONS: This study shows that SDF-1 substantially enhances the migration of follicular center lymphoma B cells but not the migration of freshly purified germinal center B cells. This difference may be related to the extended survival of follicular center lymphoma versus germinal center B cells. SDF-1 produced in follicular center lymphoma lymph nodes may play a role in the local dissemination of tumor cells.

International migration and New Zealand labour markets.

"This paper seeks to assess the value of the overseas-born members of the labour force in ensuring a flexible labour supply in New Zealand since the beginning of the 1970s. Three main issues are considered: first, the role of the labour market in New Zealand's immigration policy; second, international migration trends and the labour market; and third, the evidence on migration and labour market segmentation in New Zealand." Data used are from official external migration statistics, quinquennial censuses, and recent research. The author notes that "in New Zealand immigration measures are currently being taken that emphasize that immigration continues to add to the flexibility of the labour market while uncontrolled emigration is a major cause of labour market instability." Population and health.

Population and health ecology as subsystems of the health system are of special interest for public health. The population as a financer and beneficiary of health care determines various needs and possibilities of their realization. In order to establish solid socio-medical diagnosis of population, it is the key parameter, which is being realized through population statistics considering the number and distribution of population according to biological, socio-economic and cultural characteristics, as well as through vital and migration statistics. Health ecology as a subsystem represents the interest of public health from the aspect of monitoring of ecology expositions and the state of other contents of ecology and their impact on health. The establishment of sustainable development of human environment represents a philosophy, which is based on inter-generation equality meaning that present generation should give the future generation an environment in which is possible to live healthy life and realise any other progress. Population and health ecology are subsystems within which is necessary to seek needs that have to be met with the aim of improvement of population in terms of health.

Possible approaches to improving the international comparability of immigration and emigration statistics: an illustration of the Canadian situation

"This article seeks to highlight the extent of the lack of standardization which exists in the field of international migration statistics and, using statistics for Canada as an example, to illustrate how significant progress towards resolving this problem could be made if sufficient numbers of countries made their immigration and emigration statistics gradually comply more fully with the United Nations recommendations."

[Genetic-demographic processes in the Muscovite population in the mid-1990s. Migration and emigration as a factor for changing the genetic diversity of the population].

On the basis of official migration statistics and marriage records, the genetic parameters of migration in the Moscow population over the past 120 years were determined. Although the number of officially registered migrants decreased, the coefficient of marriage migration in the mid-1990s was higher than in the mid-1980s (m = 0.4). The in-migrants considerably differed from Moscow-born residents with respect to their age at marriage, ethnicity, level of education, and speciality. The emigration of the Moscow residents to foreign countries was selective with respect to the educational level and adversely affected the labor, intellectual, and cultural resources of Moscow (the "braindrain"). In the long run, the observed ethnic differences between in-migrants and emigrants may considerably influence the ethnic composition and, hence, genetic diversity of the Moscow population.

Human postprandial gastric emptying of indigestible solids can occur unrelated to antral phase III.

According to animal experiments, postprandial gastric emptying of indigestible solids is mainly related to the antral phase III activity of the migrating motor complex. Gastric emptying of indigestible solids in humans has not been directly correlated to pressure recordings. The aim of the present study was to investigate the postprandial emptying pattern of indigestible solids in humans and its relation to fed and fasted antral motility. Ten healthy volunteers participated. After an overnight fast they had a standard breakfast. Two sizes of radiopaque markers (ROMs) were given with the test meal; ten cubes each of side measurement 1.5 mm and 3 mm, respectively. Emptying of the ROMs from the stomach was followed by fluoroscopy with simultaneous antral manometry. In six of the subjects, fasting antral manometry was performed on one day and on another day, the emptying of 7 mm cylindrical particles together with 3 mm cubes, in the absence of a gastric tube was recorded. All ROMs were emptied within 5 h (range 1.5-4.5 h). In all subjects, the smaller particles (1.5 mm) showed a slight, insignificant tendency to move from the stomach more rapidly than the larger (3 mm) particles. None of the subjects had an antral phase III before all ROMs were emptied from the stomach. Instead, the typical irregular postprandial pressure activity was present in all subjects until the emptying was completed. Furthermore, the highest postprandial motility index during the emptying study was far below the motility index during phase III, but comparable to the motility index during late phase II. Emptying of the 7 mm particles occurred significantly more slowly at 1.5-2.5 h, but otherwise was similar to the emptying of the smaller particles. There was no difference between emptying of the 3 mm cubes with or without the presence of the tube. Contrary to common opinion, gastric emptying of indigestible solids after a meal can occur unrelated to the antral phase III, at least up to a particle size of 3 mm and perhaps even 7 mm. These findings are of great importance for the evaluation of gastric emptying of indigestible solids, including the pharmacodynamics of orally administered drugs.

Immigration to Israel, January-June 1991.

"This publication presents data on the immigration to Israel in the first half of 1991. In the period under review the number of immigrants to Israel amounted to 110,600...compared to 57,600 immigrants in the first half of 1990 and about 141,900 immigrants in the second half of 1990." About 80 percent of immigrants were from the former Soviet Union, and 17 percent from Ethiopia.
