[Effect of an excess of serotonin in the brain on consolidation of temporary connections].

As a result of intravenous administration of 100 mg/kg of 5-oxytryptophane (5-OTP) to mice, the serotonin content in the brain rises in ten minutes by 35 to 37 per cent, and in an hour, by 58 to 60 per cent, and declines in a day to its initial level. In case of elaboration of passive avoidance reflexes (PAR) an hour after 5-OTP injection, their preservation considerably decreases in a day's time, while 5-OTP administration immediately following PARs elaboration does not affect their preservation. However, 5-OTP injected after PARs elaboration to animals subjected to preliminary cooling, sharply deteriorates their preservation. These facts in conjunction with the previously obtained date attest that excess of serotonin in the brain has a negative effect on the earliest stages of consolidation. When the animals are cooled and the brain temperature is reduced, the consolidation processes are retarded; as a result, the temporary connections prove to be still vulnerable to the action of serotonin excess set up after PAR elaboration.

[Prolonged effect of 5-hydroxytryptophan on trace processes of neuronal activity].

In experiments on rabbits a study was made of the influence of serotonin's precursor, 5-oxytriptophane (5-OTP), on trace processes in spike activity of neurones in the visual and sensorimotor cortical areas. Rhythmic light was used as a stimulus with a frequency of 2 or 5 per second. Administration of 5-OTP produced prolonged action of the trace processes, significant for 2 per second frequency and less pronounced for 5 per second frequency of the flashes. This property was possessed both by the neurones which increased their activity frequency and those which reduced it. It is assumed that the prolonging effect of 5-OTP is the consequence of its modulating influence on the brain activity, which is manifested in a selective potentiation of the functioning of a neuronal system with a relatively lower frequency of spontaneous firing.

Qualitative evidence of service user experiences and perspectives on long-acting injectable buprenorphine for opioid treatment - a scoping review.

BACKGROUND: There is substantial literature on opioid treatment program (OTP) formulations and how they relate to the pharmacotherapy service user experience. As a newer formulation, less is known about service user experiences of long-acting injectable buprenorphine (LAIB). The aim of this scoping review is to map the qualitative evidence and gaps in the literature on service user experiences and perspectives of LAIB. METHODS: Our search strategy included Medline, Embase, PsycINFO, CINAHL, Scopus and Web Science, and citation chaining, from January 2016 to June 2025. Studies were included if reporting qualitative descriptions of LAIB service user experiences of treatment for opioid dependence, inclusive of qualitative, mixed methods (description of qualitative data only), case reports and English language. Articles were screened by two reviewers. A living experience first author led the analysis using inductive coding and thematic analysis, to produce a descriptive summary of synthesised findings alongside key study characteristics and quality appraisal, adhering to the Systematic reviews and Meta-Analysis for Scoping Reviews (PRISMA-ScR) checklist. RESULTS: After screening 838 titles/abstracts and reviewing 150 full texts, 40 studies met the eligibility criteria. All were conducted in high income countries, principally the US (n=12); Australia (n=10); and England and Wales (n=9). We identified five themes: Navigating LAIB treatment; Embodied and relational effects of LAIB; Impact and role of the service provider; Narratives of harm reduction and recovery; Stigma and criminalisation. LAIB was commonly experienced as increasing convenience, stability and freedom from daily supervised dosing, enabling improved work, travel, privacy and social participation. Reduced clinic/dosing contact often lessened enacted stigma and treatment burden. However, experiences were heterogenous. Some participants described injection-site discomfort, uncertainty about dose adequacy, reduced flexibility once injected, and ambivalence about LAIB effects. There was inconsistency in LAIB service user reports on service connection, isolation and psychosocial support. Treatment experiences were strongly shaped by provider practices. CONCLUSIONS: Findings underscore the need for integrated, flexible, harm-reduction oriented and person-centred LAIB treatment models that prioritise choice, autonomy and therapeutic relationships to maximise benefit for service users. However, evidence of LAIB service user experiences is concentrated in high-income countries, and the absence of perspectives from low- and middle-income country settings represents a substantial gap in the evidence base.

[Study of the monoaminergic mechanisms of the effect of haloperidol in experiments using cats].

Monoaminergic mechanisms of the haloperidol action on the systems "rewards" and "punishments" were studied in 22 cats with nichrome electrodes implanted into the hypothalamus. Haloperidol was introduced against the background of the dopaminomimetic amantadine, precursors of biogenic amines L-DOPA, tryptophan and 5-OTP. The inhibitory influence of haloperidol on the system "rewards" was found to be materialized largely at the expense of its serotonin-negative effect, while in the activating action of haloperidol on the system "punishment" its serotonin- and dopamino-negative effects partook.

[Cyclic changes in the excitability of the cerebral cortex in alert rats under conditions of direct interference with serotonin and noradrenaline metabolism].

Excitability recovery cycles in the visual cortex before and after administration of 5-OTP, d, 1-DOPA, PCPA and L-methyl-DOPA were studied on 23 alert rats with electrodes chronically implanted in the brain. The appearance of cyclicity under the influence of the drugs was established in the trace processes elicited by a photic stimulus. Differences between the periods of cyclic changes were revealed in conditions of activity predominance of the serotoninergic system (300-400 ms) and the noradrenergic system (200 ms). Their function in the short-term memory mechanisms is discussed.

[Characteristics of rat training on emotionally varied reinforcement under conditions of an oriented change in the level of brain serotonin and noradrenaline].

The effect of serotonin and noadrenaline precursors on the training of the animals on emotionally different reinforcement was studied. 5-OTP (10 mg/kg) facilitated the training of rats on the food reinforcement, but aggravated it on the pain reinforcement. D, L-DOPA (20 mg/kg) facilitated the training on the food reinforcement. The influence of precursors on the peculiarity of the serotonin and noradrenaline distribution in the brain structures of trained animals was also determined by the biological significance of the reinforcement used in the training.
