[Extraction and purification of slow myosin from the masseter muscle of Ovis aries].

Ovis aries masseter is a typical muscle consisting of slow-twitch fibres. The muscle homogeneity is well evidenced histochemically by the myosin ATPase reaction. The extraction and purification of myosin from sheep masseter is reported. After its centrifugation and dialysis followed by ion-exchange chromatography, the protein analysed on SDS polyacrylamide gels showed the usual pattern of slow myosin. The light-chain LC2 was present as a double band, a fact which has already been reported and which could be ascribed to SH group oxidation. From the present data sheep masseter can be considered a good source of slow myosin, and we have used it satisfactorily in the preparation of specific antibodies.

[Participation of structural gene markers and anonymous DNA sequences in genetic differentiation in the species Ovis aries L. and Ovis nivicola borealis].

Genetic variability and genetic differentiation in two Ovis species--domesticated (Askanian sheep, Sokilska and Kulunda sheep) and wild (bighorn sheep) were analyzed using different types of molecular-genetic markers--genetic-biochemical (30 loci) and DNA (ISSR-PCR) ones. High level of genetic variability was revealed. The average heterozygosity of biochemical markers loci was in range of 0.073-0.188. Markers involved into process of genofond divergence of two closely related species were revealed.

A chromosomal level genome assembly of Nguni Sheep, Ovis aries.

Nguni sheep (Ovis aries) are indigenous to the Southern Africa region and common within the smallholder and poor resources farming systems. They are well adapted to different agroecological regions. However, limited genomic resources such as high-quality reference genomes have hindered our understanding of its adaptation and establishment of an effective breeding program. To address this, we assembled a chromosomal-level genome of Nguni sheep using a combination of PacBio HiFi reads and Omni-C reads. The genome size was estimated to be 2.9 Gb with a contig/scaffold N50 74 Mb and 99.6 Mb and a genome completeness of 96.1%, as estimated by the Benchmarking Universal Single-Copy Orthologs (BUSCO) program. The final genome encompassed a total of 25,926 protein-coding genes. The findings of this study provide a valuable genomic resource for understanding the adaptability of the Nguni sheep and the establishment of effective breeding programs.

Effect of porcine zonae pellucidae immunisation on ovarian follicular development and endocrine function in domestic ewes (Ovis aries).

Domestic ewes (Ovis aries) were immunised with porcine zonae pellucidae (pZP) or pZP conjugated to keyhole limpet haemocyanin (KLH) in adjuvant(s) to examine the feasibility of the species to serve as a model for further development of pZP-based vaccines in ungulates. Two immunisation groups were employed, with a third group receiving only adjuvant (n = 5 per group). Early in the study, oestrous activity was monitored by the use of a vasectomised ram fitted with a marking harness. Eventually, ewes were exposed to an intact ram for breeding. In addition, weekly serum and every-other-day faecal samples were collected to measure pZP antibodies and progesterone metabolite concentrations respectively. At the conclusion of the study, fecundity was established, and ovarian tissue was examined. Ewes immunised against pZP : KLH with adjuvant produced minimal antibody absorbance levels, displayed normal oestrous cycles, became pregnant upon introduction of the intact ram and exhibited normal ovarian histopathology. Ewes immunised against pZP with adjuvant produced high antibody absorbance levels, were acyclic following primary immunisation and were infertile. Examination of the ovarian tissue revealed atrophic changes that included: (1) the absence of growing follicles; (2) significant reduction in the number of primordial follicles; and (3) the presence of abnormal granulosa cell clusters lacking oocytes. Antisera displayed immunoreactivity to the major components of pZP, and immunohistochemical labelling of ovarian tissue showed specificity to the ZP. These data are the first generated in an ungulate species showing deleterious effects of pZP immunisation on folliculogenesis and oestrous cyclicity.

A diagnostic assay discriminating between two major Ovis aries mitochondrial DNA haplogroups.

Two major Ovis aries mitochondrial DNA (mtDNA) haplogroups have been described in independent studies. HinfI RFLP data of mitochondrial genomes from a large sample set (n = 239) indicated an ancient mutation which differentiates between the two mtDNA types. A completely determined sheep mtDNA sequence was used to assign this mutation to the COI gene and to develop a PCR based assay discriminating between the two phylogenetic branches. The haplogroup specificity of the mutation was further investigated in 26 randomly selected individuals. The animals were unequivocally assigned to their respective groups on the basis of the developed test and their complete control region sequences. The assay provides a rapid and economic means of discriminating between both major domestic sheep mtDNAs.

Procedure for successful interspecific embryo transfer from mouflon (Ovis gmelini musimon) to Spanish Merino sheep (Ovis aries).

Embryos from five anesthetized mouflons (Ovis gmelini musimon), superovulated with FSH-o (Ovagen) were transferred into preselected Spanish Merino sheep (Ovis aries). Myorelaxation was complete in four of five donor mouflons. The status of the uterus of potential recipients was evaluated by transrectal ultrasonography, and those ewes with fluid in the uterine horn were rejected. The corpus luteum in each ewe was assessed ultrasonographically the day before surgery. Plasma progesterone levels and the quality of the corpora lutea were the criteria for selection of recipients. Ten embryos were transferred to the five selected Spanish Merino recipients, resulting in four pregnancies and seven live-born lambs, including three sets of twins. This study shows that determination of plasma progesterone levels combined with ultrasonographic assessment of the corpus luteum provides information useful for screening of potential recipients.

Erythrocyte enzymes in Ovis aries. Effect of cell age.

1. Activities of 20 red cell enzymes were compared in 8 normal female Ovis Aries, in a serially bled ewe followed by 243 days, and in young and old red cell populations separated by density gradient centrifugation. 2. These comparisons indicate that the erythrocyte enzymes which show significant changes with cell age are glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, hexokinase, phosphoglucose isomerase, lactate dehydrogenase, glutathione reductase, enolase and phosphoglycerate kinase. 3. The usefulness of these data in interpretation of enzyme activities from fetal Ovis Aries is discussed.

Interspecific transfer of IVM IVF-derived red sheep (Ovis orientalis gmelini ) embryos to domestic sheep (Ovis aries ).

Two embryo production methodologies were investigated to generate Red sheep embryos for use in an interspecific embryo transfer program. In Experiment 1, 4 multiparous female Red sheep (Ovis orientalis gmelini ) were implanted with CIDR type G devices for 11 d. Forty-eight hours prior to CIDR removal, a total of 22.5 mg bid of FSH-P was administered over a 3-d period. Laparoscopic embryo collection was performed 5 d post breeding, and embryos were transferred to domestic recipient ewes (Ovis aries and Ovis orientalis musimon ). In Experiment 2, 7 nulliparous female Red sheep were implanted with CIDR devices and injected with 200 IU of PMSG and 25 mg of FSH-P on the 8th day of implant insertion. At 60 to 70 h post PMSG/FSH-P treatment, follicular oocytes were aspirated laparoscopically. The recovered oocytes were matured in M199 (with fetal calf serum, FSH, LH, penicillin and streptomycin) at 39 degrees C in a humidified atmosphere containing 5% CO(2). At 24 h oocytes were fertilized with frozen-thawed semen at a concentration of 1.6 x 10(6) sperm/ml. The ova/embryos were placed in CR2 or BOEC culture medium at 20-22 h post IVF. Following 3 to 4 d in culture, embryos were transferred laparoscopically to the uterine horn of synchronized recipients. In Experiment 1, 4 embryos and 6 UFO were collected from 2 embryo donors, respectively. Two embryos were transferred with the aid of a laparoscope to each of 2 Rambouillet recipients, one of which gave birth to a healthy Red sheep lamb at 158 d of gestation. In Experiment2, a total of 62 oocytes was collected from 7 oocyte donors; 16 developed to the 16- to 32-cell stage and were transferred to 8 recipients. Three of these IVM-IVF embryos were transferred laparoscopically to 2 Mouflon recipients, resulting in no pregnancies. Thirteen IVM-IVF embryos were transferred to 6 Rambouillet recipients. Each of these gave birth to a single healthy Red sheep lamb. Gestation lengths of the 3 IVM-IVF lambs ranged from 152 to 162 d. This research demonstrates that when using compatible species IVM-IVF technology in conjunction with interspecific ET can lead to the production of live offspring and can be used to propagate exotic ovine species.

Epidemiology of parasitic protozoan infections in Soay sheep (Ovis aries L.) on St Kilda.

The feral Soay sheep (Ovis aries L.) population on Hirta, St Kilda, is host to a diverse component parasite community, but previous parasitological studies of the population have only focussed on the metazoan species. This paper reports the first epidemiological study of the protozoan species comprising Cryptosporidium parvum, Giardia duodenalis and 11 species of Eimeria in Soay sheep across 3 years of varying host population density. Prevalence and intensity of almost all species of protozoa significantly decreased with host age, with the exception of E. granulosa, which increased in prevalence with host age. The prevalence of C. parvum appeared to vary positively with host population density but that of G. duodenalis did not vary significantly with density. Most species of Eimeria showed a distinct lag in infection level following the host population crash of 2002, taking up to 2 years to decrease. Mixed Eimeria species intensity and diversity were highest in 2002, a year of low host density. Parasite diversity decreased with host age and was higher in males. There were 5 positive pair-wise associations between protozoa species in terms of prevalence. The results of this study highlight the potential for protozoal infection to shape the evolution of parasite resistance in wild host populations harbouring diverse parasite species.

A low rate of replacement substitutions in two major Ovis aries mitochondrial genomes.

Two mitochondrial DNA molecules which represent major Ovis aries mtDNA haplogroups were cloned and comparatively sequenced to assess the degree of intraspecific variation. A total of 9623 bp that correspond to 58% of both mitochondrial genomes were determined. The control region, the Cytb, ND2, ND3, ND4L, COIII and 12 tRNA genes, including the origin of L-strand replication, were completely characterized. Partial sequence information was obtained from the 12S and 16S rRNA and an additional six protein coding and six tRNA genes. The control regions of the two mtDNAs showed a nucleotide divergence of 4.34% while coding regions differed by 0.44%. The number of sheep coding region substitutions was similar to values observed in intraspecific comparisons of mitochondrial DNAs that represent remote points in genealogical trees of mice and humans. However, replacement substitutions were only observed at approximately 30% of the rate in mice and approximately 20% of the rate in humans. Nucleotide substitutions with a potential for phenotypic effects were found in the 12S and 16S rRNA and in the ND1 and COIII genes.

Eimeria ahsata Honess from domestic sheep (ovis aries) in Italy.

Examination for coccidia of 20 domestic sheep (Ovis aries) from a farm in Rome Province revealed the following 5 species: Eimeria ahsata Honess, E. intricata Spiegl, E. ovina Levine & Ivens, E. ovinoidalis McDougald, and E. parva Kotlán, Mócsy & Vajda. Of these species, only E. ahsata was not previously reported in Italy. For this reason, a description of this coccidium and a statistical analysis of size differences between E. ahsata and E. ovina are given.

[Vascularization and glial structures of the area postrema in sheep (Ovis aries) (author's transl)].

The area postrema of the sheep (Ovis aries) reveals the typical position and structure as seen in the domestic animals. The fetal development of the organ concerning especially the vascularization does not give any reference to a phylogenetic connection with the area postrema of birds. The glial border of the organ ist strikingly marked by the funiculus separans, which seems to be lacking in other domestic animals.

Association of nucleolus organizer chromosomes in domestic sheep (Ovis aries) shown by silver staining.

The nucleolus organizer regions of domestic sheep (Ovis aries), as shown by silver staining, are located terminally on chromosomes 1, 2, 3, 4, and 25. Significant differences between individuals in the number of Ag-NORs per cell were found. The frequency of involvement of individual chromosome pairs in nucleolar organization was found to be a characteristic of individual animals. Association frequencies of individual chromosomes were accounted for by their frequency of participation in nucleolar organization. No evidence for nonrandom association of chromosome pairs was found.

[Peculiarities of the innervation of paranasal cavity mucosa in Ovis aries].

The innervation of the mucous membranes in the paranasal cavities of "ovis aries" appears homogeneous as regards the various nasal sinuses. It is represented by numerous nervous bundles which are in the deep and middle parts of the mucous membranes, and have sensory terminals and fibres assigned to the vessels and glandular adenomeres. In the subepithelial stratum can be noted a superficial plexus from which come off the fibres assigned to the epithelial elements of the mucous membranes.

Identification of nucleolus organizer chromosomes in sheep (Ovis aries L.) by sequential GBG/Ag-NOR and RBG/Ag-NOR techniques.

Seven Gentile di Puglia sheep (Ovis aries L., 2n = 54) reared in southern Italy were cytogenetically investigated to reveal which chromosomes carry nucleolus organizer regions (NORs). The use of sequential GBG/Ag-NOR and RBG/Ag-NOR techniques in prometaphase preparations revealed NOR-localization at the telomeres of chromosomes 1p, 2q, 3q, 6 and 25 on the basis of the standard R-banded karyotype. This procedure also permitted the characterization of G- and R-banding patterns of acrocentrics 6 and 25 which are often confused with the closest pairs in sheep and related species karyotypes. Indirect comparisons among nucleolus organizer chromosomes (NOCs) of sheep, goat, cattle and river buffalo, when using a common R-banding nomenclature, revealed that (a) sheep and goat have the same five homologous NOCs, (b) sheep and cattle have three homologous NOCs (c) sheep and river buffalo have two NOCs in common, and (d) the four species have only one homologous NOC in common.

[Detection of hydrodynamically operative tissue in the substantia spongiosa of the femoral head in Bos taurus and Ovis aries].

The authors show the presence of CC-Tensulae in Bos taurus and Ovis aries. They define CC-Tensulae as tense membranes in openings between the trabeculae of spongy bone. These structures were interpreted in man as a hydrodynamic subsystem which serves the flow and the impulses of mechanical forces. The authors point especially to the similarity that exists between their findings and what has been seen in man.
