Naphtho[2,1-b][1,5] and [1,2-f][1,4]oxazocines as selective NK1 antagonists.

Previously we reported on the synthesis and properties of a series of highly potent piperidinyl 2-subsituted-3-cyano-1-naphthamide NK1 antagonists that includes 3 and 4. Here we report our efforts to alleviate a troublesome atropisomeric property of those derivatives by introduction of a tethering bridge that, in addition, could be used to lock the resulting cyclic derivatives in a purported NK1 pharmacophore conformation. Using 3 as a starting point, the naphtho[2,1-b][1,5]oxazocine, 17, was found to contain the optimal ring tether size (8) for retaining NK1 activity, was more NK1 versus NK2 selective, and reduced the number of atropisomers from four to two. Cyclic derivatives 29 and 32, which exist as essentially single atropisomers in the purported pharmacophore conformation, were prepared in the closely related naphtho[1,2-f][1,4]oxazocine series as part of an effort to use mono methyl substitution of the tethering bridge as a conformation stabilizing factor. Both 29 and 32 were found to be less active as NK1 antagonists than the non-methylated parent 28 possibly due to methyl group destabilization of receptor interaction. We discuss the above findings in the context of a previously proposed NK1 pharmacophore model and present a further refinement of that model.

2-substituted-4-aryl-6,7,8,9-tetrahydro-5H-pyrimido[4,5-b][1,5]oxazocin-5-one as a structurally new NK1 antagonist.

The structurally novel pyrimido[4,5-b][1,5]oxazocine derivative 3, a hybrid compound of pyrido[4,3-b]- and [2,3-b]-1,5-oxazocine (1 and 2, respectively), was designed and synthesized. We examined the atropisomeric property and the NK1 antagonist activity of 3. Compound 3 was found to possess both a feature of 1, free rotation about the biaryl bond, and a feature of 2, potent in vivo activity.

Design and synthesis of novel 9-substituted-7-aryl-3,4,5,6-tetrahydro-2H-pyrido[4,3-b]- and [2,3-b]-1,5-oxazocin-6-ones as NK(1) antagonists.

Novel 9-substituted-7-aryl-3,4,5,6-tetrahydro-2H-pyrido[4,3-b]- and [2,3-b]-1,5-oxazocin-6-ones were designed and prepared as part of a search for NK1 antagonists. Structure-activity relationship studies indicated that the conformational restriction resulting from the incorporation of an oxazocine ring and the presence of a terminal heteroatom on the cyclic amino group at the C-9 position play important roles in NK1, receptor recognition.

Design, synthesis, and evaluation of novel 2-substituted-4-aryl-6,7,8,9-tetrahydro-5H-pyrimido[4,5-b][1,5]oxazocin-5-ones as NK1 antagonists.

A series of novel bicyclic pyrimidine derivatives was prepared as part of a search for NK1 antagonist aimed at the treatment of urinary incontinence. Among them, 3g, a pyrimido[4,5-b][1,5]oxazocine derivative, bearing a 4-acetylpiperazinyl group and a 2-methylphenyl group, was shown to have potent NK1 antagonist activity with a K(B) value of 0.105 nM and markedly increased the effective bladder capacity of guinea pigs (59.4% at 0.3 mg/kg iv and 62.8% at 3 mg/kg id). Furthermore, the effect of 3g on bladder function appeared to differ from that of tolterodine, another classical anti-pollakiuria agent, as determined by the distention-induced rhythmic bladder contraction assay using a urethane-anesthetized guinea pig model. Compound 3g is expected to be a promising agent for the treatment of urinary incontinence.

Substituted 5H-dibenz[b,g]-1,4-oxazocines and related amino acids with antiinflammatory activity.

During an investigation of the antiinflammatory properties of a number of tetracyclic derivatives of 6,8-dichlorodibenz[b,f]oxepin-10(11H)-one, the ring-expanded 1,3-dichloro-5H-dibenz[b,g]-1,4-oxazocine (9) was prepared and found to be considerable pharmacological interest. It was subsequently found that the corresponding ring-opened amino acid 66, a close analogue of the antiinflammatory agent fenclofenac, also possessed significant antiinflammatory activity, superior both to the dibenzoxazocine and to fenclofenac. These findings prompted extensive synthetic programs in both areas, and a number of derivatives in the amino acid series showed potencies considerably in excess of the standard compound. These phenylacetic acids, however, were significantly more ulcerogenic than fenclofenac whereas the corresponding dibenzoxazocines showed few signs of ulcerogenicity at doses up to 1 g/kg.

Investigations of anthelminthic activity and synthesis of new derivatives of perhydroazepine, perhydro-1,4-diazepine, perhydro-1,4-oxazepine and perhydro-1,5-oxazocine.

Anthelminthic activity of new esters of 3,4,5-trimethoxybenzoic acid containing rests of the title heterocyclic amines has been determined. 3-(Perhydroazepinyl)-propyl ester 6 acts on enchytraeids in vitro 56-fold stronger than piperazine adipate, 2-(perhydroazepinyl)-ethyl ester 5 is the most effective in a therapy of mice infested with nematodes.

Identification of a novel fluorescent adduct formed in the reaction of malonaldehyde with adenosine.

Malonaldehyde was reacted with adenosine in aqueous solution at acidic conditions and the reaction mixtures were analysed by HPLC. Four major product peaks were observed in the chromatogram recorded by the UV detector at 320 nm. Two of the peaks could be deduced to the previously characterised malonaldehyde-adenosine reaction product 9-(beta-D-ribofuranosyl)-6-(5,7-diformyl-2H-3,6-dihydro-2,6-methano-1,3-oxazocin-3-yl)purine (M3A) and to the ribose analogue of the 2'-deoxy adduct 9-(2'-deoxy-beta-D-ribofuranosyl)-6-(3,5-diformyl-4-methyl-1,4-dihydro-1-pyridyl)purine (M2AA-dA). The two other peaks were due to previously uncharacterised adducts. Upon isolation of these adduct peaks it was found that the peaks are interconverted to each other, one adduct peak dominating over the other one. On the basis of data recorded by UV, fluorescence and 1H NMR spectroscopy, and mass spectrometry, the structure of the major adduct could be determined as 9-(beta-D-ribofuranosyl)-6-(3,5-diformyl-4-etanal-1,4-dihydro-1-pyridyl)purine (3M-A). The adduct is most likely formed by reaction of adenosine with a malonaldehyde condensation product consisting of three units of malonaldehyde. The highest yield of 3M-A was obtained in the reaction performed at pH 4.6 and 80 degrees C for 75 h. The minor form of the interconverting peaks could not be characterised on the basis of the spectral data. However, it is concluded that the minor peak most likely represents the hydrated form of 3M-A. Since these adducts are formed only in trace amounts at neutral pH and 37 degrees C and the formation requires high amounts of malonaldehyde, it is likely that the adducts are not formed in DNA in vivo and thus not contribute to the malonaldehyde genotoxicity.

Palladium-mediated intramolecular aryl amination on furanose derivatives: an expedient approach to the synthesis of chiral benzoxazocine derivatives and tricyclic nucleosides.

Pd-catalyzed intramolecular arylamination on sugar derivatives has been accomplished by using bulky biaryl phosphine ligands. An application of this methodology on a variety of D-glucose-derived substrates, 2a-f, led to the synthesis of highly functionalized cis-fused tricyclic oxazocines, 3a-e. The products could subsequently be transformed to the optically active benzoxazocine derivative 4 and tricyclic nucleoside 6. This is the first example of the synthesis of eight-membered rings via intramolecular cycloamination of furanose derivatives, which provides a very useful method for the catalytic synthesis of medium-ring heterocycles.

Investigation of the adducts formed by reaction of malondialdehyde with adenosine.

Malondialdehyde (MDA) forms oligomeric adducts with DNA bases. It has been proposed that the 2:1 MDA-guanosine (M2G) and 3:1 MDA-adenosine (M3A) adducts result from sequential addition of MDA molecules to the nucleoside base. Reaction of 1:1 MDA-guanosine (M1G) and 1:1 MDA-adenosine (M1A) adducts with MDA does not produce the multimeric adducts. This suggests they arise by reaction of the nucleoside bases with oligomers of MDA. If so, the proposed structure for M3A is inconsistent with the addition of an MDA trimer to adenosine. Therefore, we investigated the structure of this molecule. Two-dimensional double quantum filtered COSY and hetero-COSY NMR experiments were performed, and a series of insensitive nuclei assignment by polarization transfer (INAPT) NMR spectra were also recorded. The results of these experiments revealed the presence of a propano group and two alpha,beta-unsaturated aldehydes. The UV spectrum of M3A displayed a maximum at 326 nm, similar to that of N6-[3-oxo-1(E)-propenyl]adenosine (M1A). The adducts were reduced with sodium borohydride for comparison of the UV and NMR spectra. On the basis of our results, a new structure for M3A is proposed which is tentatively named 6(5*,7*-diformyl-2*H-3*,6*-dihydro-2*,6*-methano-1*,3*-oxazocin -3*-yl)-9-beta-D-ribofuranosylpurine.
