Detection of human herpesvirus 6 and human herpesvirus 7 in the submandibular gland, parotid gland, and lip salivary gland by PCR.

In order to define the major sites of persistence of human herpesvirus 6 (HHV-6) and HHV-7, PCR with DNAs from more than 100 specimens of 3 different salivary glands was performed. HHV-6 DNA was detected in 52 (88.1%) of 59 submandibular gland, 17 (63.0%) of 27 parotid gland, and 9 (52.9%) of 17 lip salivary gland specimens. On the other hand, HHV-7 DNA was detected in 59 (100%) of 59 submandibular gland, 23 (85.2%) of 27 parotid gland, and 10 (58.8%) of 17 lip salivary gland specimens. These findings demonstrate that salivary glands are a site of persistent infection of both HHV-6 and HHV-7 and that among the three types of salivary gland examined, the submandibular gland is the primary one in which these herpesviruses, especially HHV-7, persist.

Immunohistochemical localization and mRNA detection of Rab3D and/or Rab3B in rat von Ebner's glands, parotid gland, pancreas, and liver.

In examining the secretory mechanism of exocrine glands, we focused on the small GTP-binding proteins, Rab3D and Rab3B, which function in the final steps of exocytosis in non-neuronal tissues. These proteins were observed in von Ebner's glands by 32P-GTP overlay. mRNA isolated from von Ebner's glands, the pancreas, parotid glands, and liver was subjected to reverse transcription PCR and probed with primers and nested primers for Rab3D and Rab3B. Rab3D was found in all three exocrine glands and the liver, while Rab3B was found in the liver. Utilizing immunofluorescence histochemistry, Rab3D was localized to hepatocytes of the liver and to secretory granules of the exocrine glands, and Rab3B to liver and pancreatic islets. Isoproterenol evoked decreases in alpha-amylase- and Rab3D-labelled parotid secretory granules, and pilocarpine stimulated decreases in secretory granules labelled for lingual lipase and Rab3D from von Ebner's glands, and amylase and Rab3D from pancreas. Neither secretagogue affected Rab3B in pancreatic islets. These observed parallel decreases in response to beta-adrenergic (parotid) or cholinergic (von Ebner's and pancreas) secretagogues indicate that the function of Rab3D in exocytosis in these exocrine organs is similar and that the type of secretagogue does not determine the function.

Vascular neoplasms of the parotid gland. Parotid vascular tumors.

Vascular neoplasms of the parotid gland are common in early childhood, particularly in females. We reviewed the clinical, histologic, and treatment details of 10 cases of hemangiomas and one case of lymphangioma that involved the parotid gland. Histologically, the cellularity and increased division figures in these lesions should not be interpreted as a sign of a malignant condition. A watchful expectancy for spontaneous regression and preservation of the facial nerve at surgery are advocated.

[Peptidergic innervation of human salivary glands (parotid gland and submandibular gland)].

Immunocytochemistry and a radioimmunoassay were used to investigate the existence and distributions of various regulatory peptide immunoreactivities (ir) in human submandibular and parotid glands. Numerous nerve fibers containing vasoactive intestinal polypeptide (VIP) and peptide histidine methionine (PHM), or neuropeptide tyrosine (NPY) and C-flanking peptide of NPY (CPON)-ir were found in close proximity to acini, ducts and blood vessels. Only a few calcitonin gene-related peptide (CGRP)- and substance P (SP)-ir nerve fibers could be demonstrated and were mainly localized around blood vessels and ducts. Galanin and the recently discovered peptides helospectin and pituitary adenylate cyclase activating peptide were unable to be detected in the salivary glands studied. Preliminary quantitative investigations of four human submandibular glands using radioimmunoassay showed that VIP-ir had the highest concentration, followed by NPY-ir and CGRP-ir; SP-concentrations were below the detection limit. The possible physiological significance of these peptides for salivary secretion is discussed.

The short-term effects of a single injection of isoproterenol on proliferation in the submandibular gland, parotid gland and oesophagus in vivo.

Mitotic and labelling indices were studied in the submandibular, parotid and oesophageal cells of male mice within the first 6 hr (but particularly within the 1st hr) of a single injection of isoproterenol or saline, using the metaphase arrest agent (vincristine) which was previously tested for efficacy in submandibular gland. There was a significant increase in the metaphase index of the salivary glands over control values 5, 15, 30, 45 and 60 min after isoproterenol. In contrast, there were no significant changes in the metaphase index of basal cells of the oesophagus. There was no significant change in the labelling index in isoproterenol-treated mice in comparison with saline-injected control animals. Possible explanations for the rapid mitotic response in murine salivary glands are considered; a rapid efflux from G2 into mitosis is thought to be the most likely.

Expression of CD44 standard and variant isoforms in parotid gland and parotid gland tumours.

In this study, we investigated the distribution of the standard form of the CD44 (CD44s) cell adhesion molecule and of its v3 and v6 isoforms in samples of foetal and adult parotid gland tissue, in comparison with samples of parotid gland adenomas and carcinoma ex pleomorphic adenoma. Foetal parotid gland showed CD44s and CD44v3 expression in the peripheral small primordial ducts and acini, while CD44v6 was only focally expressed. Adult parotid gland tissue showed a similar distribution of CD44s and variants, with a predominant expression in acinar structures and a weaker expression at duct level. In parotid gland adenomas, a diffuse and intense expression of CD44s and variants 3 and 6 was observed only in pleomorphic adenomas, while expression of CD44s was prevalent in Warthin's tumour, myoepithelioma and oncocytoma. The malignant areas of carcinoma ex pleomorphic adenoma showed a markedly decreased expression of CD44v3 and CD44v6 in comparison with the adjacent pleomorphic adenoma component. In conclusion, the prevalent expression of CD44s and variants in pleomorphic adenoma in comparison with other adenomas may be related to the abundant extracellular matrix production present in these tumours, while loss of CD44v3 and CD44v6 associated with the onset of carcinoma ex pleomorphic adenoma could promote stromal invasion, eventually contributing to the development of distant metastases.

An unusual reason of parotid gland enlargement; parotid gland tuberculosis.

Although pulmonary tuberculosis is common in Turkey, parotid gland tuberculosis is rarely seen. A 39 years old female presented with left side swelling on her face. The diagnosis was made excisional biopsy. Histologic examination of the operative specimen revealed necrotising granuloma concordant with tuberculosis.

Infantile hemangioma of the accessory parotid gland.

Parotid gland tumors, especially those of the accessory parotid gland, are rare in infancy. Although infantile hemangiomas have been frequently reported as a cause of parotid gland tumors, their association with the accessory parotid gland has not been reported. This article describes the presentation of a hemangioma of the accessory parotid gland in an infant. Review of the literature and treatment methods are discussed.

Effects of alloxan diabetes and insulin in vivo on rat parotid gland.

Parotid gland growth and secretory enzyme levels were studied in male Sprague-Dawley rats following the induction of alloxan diabetes. Diabetes resulted in a retardation of parotid gland, as well as body growth, and in a reduction of parotid gland DNA, RNA, and total protein compared with control rats. Morphologically, parotid glands of diabetic animals were characterized by an intracellular accumulation of lipid within acinar and intercalated ductal cells. Parotid amylase was reduced 40% in diabetic rats compared with control rats. In contrast, peroxidase levels increased by 54%, and DNase was unaffected. Insulin treatment of diabetic rats led to a restoration of gland and body growth. Parotid gland DNA, RNA, total protein, and secretory enzyme levels returned to control values within 7 days. Thus, insulin in vivo may play a major role in the regulation of parotid gland growth and function.

Apparent diffusion coefficient mapping of the normal parotid gland and parotid involvement in patients with systemic connective tissue disorders.

PURPOSE: We hypothesized that a difference in restricted diffusion would exist in patients with connective tissue disorders (CTD) as compared with those without CTD. Our purpose was to determine whether the apparent diffusion coefficient (ADC) measurement could be used to identify parotid abnormalities in patients with CTD. METHODS: One neuroradiologist, who was unaware of patient histories, retrospectively measured the ADC values for the parotid glands in 121 patients who underwent clinically indicated brain MR imaging in which the parotid glands were sufficiently depicted. Regions of interest were obtained from both the left and right parotid glands. After the medical records were reviewed and exclusion criteria were used, 90 non-CTD and seven CTD patients (systemic lupus erythematosus = 5; discoid lupus erythematosus = 1; Sjögren syndrome = 1) remained. The two groups were then compared. Statistical analysis consisted of Wilcoxon sign rank and Mann-Whitney tests. RESULTS: The combined mean ADC for both parotid glands in 90 healthy patients was 0.50 +/- 0.28 x 10(-3) mm(2)/s (95% CI, 0.44 x 10(-3), 0.56 x 10(-3)). The combined mean ADC for both parotid glands in the seven CTD patients was 0.96 +/- 0.24 x 10(-3) mm(2)/s (95% CI, 0.79 x 10(-3), 1.14 x 10(-3)). The mean ADC for the CTD patients' parotid glands was significantly higher than that of the non-CTD patients (P =.0001), which suggests there is less restricted diffusion in parotid glands affected by CTD when compared with normal parotid glands. CONCLUSION: These results suggest that ADCs may be used to detect parotid abnormalities in patients with CTD that are not identified by standard imaging. Although preliminary, the results indicate a potential role for ADC mapping in detection of subclinical parotid disease.

Proliferative behavior of differentiating cells in the developing rat parotid gland.

Parotid glands of litters of rats at age intervals from 20 days in utero to 100 days were assayed for DNA content and examined by light- and electron-microscopy. The age differences in total DNA and DNA concentration indicated that there was a rapid rate of proliferation of parenchymal cells until 25 postnatal days, after which the rate declined rapidly, and that there was a rapid increase in cell size between 18 and 25 days. These findings were substantiated by histologic observations, such as the presence of numerous mitotic figures until 25 days of age, and the rapid maturation of the acinar cells between 18 and 25 days. These data suggest that the acinar cells of the rat parotid gland comprise an expanding cell population. Light- and electron-microscopic observations consistently indicated that cells with mitotic figures were about as well differentiated as other parenchymal cells at all stages of gland development, including mature acinar cells observed at ages 23 and 25 days. These observations support the view that the division of cells in advanced stages of differentiation may be important in the growth of certain organs and tissues.

Differentiation of myoepithelial cells in the developing rat parotid gland.

Parotid glands of rats were prepared for light and electron microscopy and for the histochemical demonstration of myofibrils and alkaline phosphatase (AkPase) activity. Through 18 days in utero, the epithelial cells of the developing gland remain relatively undifferentiated. At 20 days in utero, a few cells in the outer layer of the terminal buds and adjacent segments of ducts acquire a cilium, the initial indication that they are differentiating into myoepithelial cells (MEC). Up until the time of birth, the only additional characteristics of MEC that the outer cells develop are to flatten against the underlying cells, begin to send out processes, and produce a few dilated cisternae of rough endoplasmic reticulum. Myofibrils and AkPase activity are first detected at the light microscopic level at five days after birth, around both the developing acini and intercalated ducts. Progressive increases in AkPase activity and in the size and number of myofibrils continue until the acini and intercalated ducts are invested with well-differentiated MEC at 15 days. Subsequently, as the acini undergo maturation during the weaning period (18-25 days), the MEC cease to surround the acini and assume the adult pattern of investing only the intercalated ducts. The pattern of MEC differentiation in the parotid gland differs from those in the sublingual and submandibular glands of the rat in several important respects. They begin to differentiate last, yet mature almost as early as do the MEC of the sublingual gland; they begin to differentiate prior to, rather than simultaneously with, the secretory cells; and their distribution changes as the acinar cells become mature.

Analysis of the immunoactivator sites of parotid protein isolated from bovine parotid glands.

Parotid protein (parotin) was isolated from bovine parotid glands. To analyze the active site of parotid protein, the parotid subunit (PS) was digested by trypsin and then fractionated on a Sephadex G-25 column. Fraction A induced mainly polyclonal antibody responses and interleukin 1 (IL-1)-like activities. FrAA-1, consisting of 58 amino acids (LYILYFFQSDNEDKEKVVRQEEGEE-RITALLMNGSALKQEEWWEKEDTDDTAIVLLK) was isolated from fraction A by chromatography on QAE-Sephadex A-25 columns. FrAA-1 was found to possess IL-1-like activity. There was only 29% homology between FrAA-1 and four IL-1 molecules. When 10- and 20-residue peptides based on the amino acid sequence of FrAA-1 were synthesized, P-10.2 (TDDTAIVLLK) alone exerted an IL-1-like effect on C3H/HeJ thymocytes, whereas P-20.1 (SDNEDKEKVVRQEEGEERIT) alone elicted polyclonal IgM and IgG antibody production in human lymphocytes. These results suggest that the active sites for polyclonal B-cell activator (PBA) and IL-1-like activity have different locations in FrAA-1.

Ultrastructure of the ferret parotid gland.

Parotid glands of two male and two female adult ferrets were examined using electron microscopy. The acinar cells contained an abundance of rough endoplasmic reticulum, an extensive Golgi complex, and electron-dense granules with a denser spot in each. The acinar lumen and the intercellular canaliculi were lined by a large number of prominent microvilli. The intercalated ducts were long and in their proximal part the cells contained small electron-dense granules, whereas the distal portions were lined by agranular cells. The majority of cells in the intralobular striated ducts contained a large number of granules. These granules were of varying shapes and were moderately electron-dense. Each granule had a close fitting membranous envelope. The agranular cells had a prominent Golgi zone, free ribosomes and a few lysosomes. Both the granular and agranular duct cells had an abundance of mitochondria towards the basal regions of the cells. The excretory ducts resembled the intralobular striated ducts. No sexual dimorphism was observed in the ferret parotid.

A cytochemical study of the effect of cholinergic and beta-adrenergic stimulation on calcium fluxes of rat parotid gland.

Parotid gland tissue from carbachol-treated and isoproterenol-treated rats was studied cytochemically by pyroantimonate precipitation and x-ray microanalysis in an effort to identify any intracellular calcium reservoirs that might be available for use in a stimulus-secretion coupling mechanism, and to determine any differences that might exist in terms of calcium utilization due to the cholinergic or beta-adrenergic receptor mechanisms. Stimulation by either secretagogue results in a reduction of mitochondrial and plasma membrane calcium, but the reduction in mitochondrial calcium deposits of carbachol-treated animals is only one-half that of the beta-adrenergic-treated animals. This could possibly suggest a greater dependency on mitochondrial calcium for beta-adrenergic stimulated animals.

The co-localisation of substance P and VIP in cholinergic-type terminals of the rat parotid gland.

Parotid glands from the rat were examined for substance P (SP) and vasoactive intestinal polypeptide (VIP)-like immunoreactivity with the peroxidase-antiperoxidase (PAP) and immunogold methods. The majority of nerve terminals associated with the acinar secretory cells contained numerous small agranular vesicles measuring 40-60 nm in diameter and a few larger vesicles which had an electron-dense core and measured 90-120 nm in diameter. Electron-dense peroxidase reaction product indicative of SP and/or VIP-like immunoreactivity was found within the larger dense-cored vesicles and attached to the outer membrane of the small agranular vesicles. Some nerve terminals associated with the acinar cells contained no reaction product irrespective of whether sections were incubated for SP or VIP. With the immunogold method gold particles indicative of SP and/or VIP-like immunoreactivity were found associated with the larger dense-cored vesicles with very little gold labelling over the small agranular vesicles. When ultrathin sections were incubated for both SP and VIP-like immunoreactivity all of the labelled terminals examined contained gold particles indicative of the presence of both peptides. In several terminals individual dense-cored vesicles contained gold particles of different sizes which indicates co-existence of SP and VIP within the same vesicle. Several nerve terminals associated with the acinar cells contained no gold labelling of their synaptic vesicles. Occasionally nerve terminals were found around blood vessels that were positive for SP-like immunoreactivity and VIP-like immunoreactivity but none was found, using the immunogold method, that contained both peptides. Very few nerve terminals were found associated with ducts and none contained reaction product or gold particles indicative of SP or VIP-like immunoreactivity. The ultrastructural features of the nerve terminals containing SP and/or VIP-like immunoreactivity could not be distinguished from those that have been described as representing cholinergic terminals. The fact that the postganglionic parasympathetic secretomotor neurons contain, in addition to acetylcholine, two neuropeptides and the possible functional implications thereof are discussed.

Lymphangioma of the parotid gland.

Parotid gland tumors in infancy and childhood are rarely seen and infrequently documented. Among them, hemangioendothelioma and mixed tumors are the most common. Parotid lymphangioma is very rare. This report presents a case of parotid lymphangioma and discusses the clinical, gross, and histological features, differential diagnosis, and treatment.
