Kinetics of lysosomal storage of indigestible matter.

In lysosomal storage diseases and in accumulation of lipofusion in the lysosomes there is a gradual eroding of the lysosomal system due to overloading the lysosomes by molecules which cannot be digested or expelled. The kinetics of this accumulation is examined for tissue cultures in terms of the cell growth rate, lysosomal production rate, and of generation of the indigestible element.

Resonance Raman microprobe spectroscopy of rhodopsin mutants: effect of substitutions in the third transmembrane helix.

A microprobe system has been developed that can record Raman spectra from as little as 2 microL of solution containing only micrograms of biological pigments. The apparatus consists of a liquid nitrogen (l-N2)-cooled cold stage, an epi-illumination microscope, and a substractive-dispersion, double spectrograph coupled to a l-N2-cooled CCD detector. Experiments were performed on native bovine rhodopsin, rhodopsin expressed in COS cells, and four rhodopsin mutants: Glu134 replaced by Gln (E134Q), Glu122 replaced by Gln (E122Q), and Glu113 replaced by Gln (E113Q) or Ala (E113A). Resonance Raman spectra of photostationary steady-state mixtures of 11-cis-rhodopsin, 9-cis-isorhodopsin, and all-trans-bathorhodopsin at 77 K were recorded. The Raman spectra of E134Q and the wild-type are the same, indicating that Glu134 is not located near the chromophore. Substitution at Glu122 also does not affect the C = NH stretching vibration of the chromophore. The fingerprint and Schiff base regions of the Raman spectra of the 380-nm, pH 7 forms of E113Q and E113A are characteristic of unprotonated retinal Schiff bases. The C = NH modes of the approximately 500-nm, pH 5 forms of E113Q and E113A in H2O (D2O) are found at 1648 (1629) and 1645 (1630) cm-1, respectively. These frequencies indicate that the protonated Schiff base interacts more weakly with its protein counterion in the Glu113 mutants than it does in the native pigment. Furthermore, perturbations of the unique bathorhodopsin hydrogen out-of-plane (HOOP) vibrations in E113Q and E113A indicate that the strength of the protein perturbation near C12 is weakened compared to that in native bathorhodopsin.(ABSTRACT TRUNCATED AT 250 WORDS)

[Pigment formation and other biological properties of staphylococci].

A comparison was made of the pigment-forming capacity and the enzymatic activity, growth rate, virulence and sensitivity to antibiotics and egg lysozyme in 397 pigment, 203 pigment-free strains of staphylococci and 24 pigment-free Staph aureus mutants. Pigment formation did not always correlate with the general biological activity of the cultures, the rate of their growth and the sensitivity to antibiotics and lysozyme. Pigment-free staphylococcus mutants retained the principal properties of the initial strains (including the virulence and sensitivity to antibiotics and egg lysozyme) more frequently. It is supposed that the marker determining the pigment formation in staphylococci was not connected with the markers determining the most significant pathogenicity properties and also antibiotic resistance.

[Pigment dispersion in a family with albinism (author's transl)].

A report is given on a family wherein two males showed albinism; of the female members, who all had been light blonde in childhood, two were carriers (one of them with pigment changes in the iris and the fundus), a third had no ocular changes, while the fourth showed the typical phenomenon of bilateral idiopathic pigment dispersion without glaucoma. The occurrence of pigment dispersion in a family with albinism has not yet been described. It is regarded as accidental combination.

[Forum: dermopigmentation or medical tattooing. Brief notes on technical aspects of dermopigmentation].

Tattooing is a very old procedure widespread throughout the world. Recently dermatologists and plastic surgeons have applied this procedure for medical dermography. Incarcerating a pigment in the depth of the dermis has caused no problems for professionals and aestheticiens performing tattoos. Medical ethics oblige us to specify the nature of the pigment used. The instrument used for injecting the pigment should be well adapted for safety, technically, ergonomically and for asepsis.

Dermopigmentation is now part of the therapeutic armamentarium of plastic surgeons. In good indications and when applied according to a few simple rules, every surgeon can obtained reliable and reproducible results with medical dermography. This therapeutic procedure is performed on an outpatient basis, usually in several sessions; subsequent revisions are always possible provided certain basic errors are avoided. The authors describe each step of this technique.
