Comparison of volatile constituents of Persicaria odorata(Lour.) Soják (Polygonum odoratum Lour.) and Persicaria hydropiper L. Spach (Polygonum hydropiper L.).

Polygonum odoratum Lour. has been reclassified as Persicaria odorata (Lour.) Soják [Wilson, K. L. Polygonum sensu lato (Polygonaceae) in Australia. Telopea 1988, 3, 177-182]; other synonyms currently used are Vietnamese mint or Vietnamese coriander and, in Malaysia, Daun Laksa or Laksa plant. The aerial parts of Laksa plant are highly aromatic, and they contain many organic compounds such as (Z)-3-hexenal, (Z)-3-hexenol, decanal, undecanal, and dodecanal that are typical for green, citrus, orange peel, and coriander odors. In addition to these aldehydes, 3-sulfanyl-hexanal and 3-sulfanyl-hexan-1-ol were discovered for the first time in this herb. The fresh leaves are pungent when they are chewed, although the active compound has never been identified. The pungency of Persicaria hydropiper (L.) Spach (formerly Polygonum hydropiper L., synonym water pepper) is produced by polygodial, a 1,4-dialdehyde derived from drimane terpenoids. We also identified polygodial as the active pungent compound in P. odorata (Lour.) Soják.

Antioxidant activity of extract from Polygonum aviculare L.

Free radicals induce numerous diseases by lipid peroxidation, protein peroxidation, and DNA damage. It has been reported that numerous plant extracts have antioxidant activities to scavenge free radicals. Whether Polygonum aviculare L. (Polygonaceae) has antioxidant activity is unknown. In this study, dried Polygonum aviculare L. was extracted by ethanol, and the extract was lyophilized. The antioxidant activities of extract powder were examined by free radical scavenging assays, superoxide radical scavenging assays, lipid peroxidation assays and hydroxyl radical-induced DNA strand scission assays. The results show that the IC50 value of Polygonum aviculare L. extract is 50 microg/ml in free radical scavenging assays, 0.8 microg/ml in superoxide radical scavenging assays, and 15 microg/ml in lipid peroxidation assays, respectively. Furthermore, Polygonum aviculare L. extract has DNA protective effect in hydroxyl radical-induced DNA strand scission assays. The total phenolics and flavonoid content of extract is 677.4 +/- 62.7 microg/g and 112.7 +/- 13 microg/g. The results indicate that Polygonum aviculare L. extract clearly has antioxidant effects.

Astragalus mongholicus and Polygonum multiflorum's protective function against cyclophosphamide inhibitory effect on thymus.

The protective effects of two Chinese herbs, astragalus mongholicus, polygonum multiflorum and astragalus mongholicus-polygonum multiflorum in combination against thymus injury induced by cyclophosphamide were evaluated by transmission electron microscopy, image analysis, DNA gel electrophoresis as well as flow cytometry. Results showed that mice pretreated with cyclophosphamide had degenerated thymus with less normal thymocytes; when those mice were treated with the herbs, thymus morphology improved. The apoptosis analysis showed the thymus treated with the herbs had fewer apoptotic thymocytes than the thymus pretreated with cyclophosphamide only. In conclusion, astragalus mongholicus and polygonum multiflorum have protective effects on the thymus against cyclophosphamide-induced injury. Their protective effects partly attribute to reduced apoptosis. Astragalus mongholicus-polygonum multiflorum in combination has better effects than either of the two herbs.

[Effect of crystal no. 4 of polygonum cuspidatum on the viability of island flaps with venous stasis: an experimental study].

The effects of crystal No. 4 of Polygonum cuspidatum on microcirculatory changes of island flaps in rats following resumption of blood flow after venous occlusion of six hours were observed with a special TV set with magnification of 500X. The pathological events and survival rate of flaps were recorded as well. The results showed that the survival rate, amount of open capillaries, amount of blood flow, and velocity of blood flow of flaps treated by crystal No. 4 of polygonum cuspidatum were much higher than those of untreated control flaps. The white thrombosis and endothelial damage of dermal vessels were alleviated by administration of crystal No. 4 of polygonum cuspsidatum. The experiment also showed that there were no differences between flaps treated with crystal No. 4 of polygonum cuspidatum preoperatively and flaps treated with it postoperatively.

Influence of ethyl acetate extract and quercetin-3-methyl ether from Polygonum amphibium on activation lymphocytes from peripheral blood of healthy donor in vitro.

The influence of an ethyl acetate extract from Polygonum amphibium L. and quercetin-3-methyl ether isolated from them were examined on the human immune system. The investigations were made on peripheral blood mononuclear cells from healthy donors. The cells were stimulated by plant extract and quercetin-3-methyl ether in 24 h culture and analysed inflow cytometry. The following mice monoclonal antibody anti human activate markers were used: anti HLA-DR PE, anti CD25 FITC, anti CD69 FITC and anti CD71 FITC. The level of Interleukine-2 in blood serum and in culture supernatants was measured by ELISA method. Ethyl acetate extract from Polygonum amphibium caused the rise in the number of CD25 and HLA-DR positive lymphocytes and increased the expression of CD25 and CD71 antigens. The level of IL-2 was increasing for the duration of the culture, independently from the presence of stimulators. Besides quercetin-3-methyl ether, from herb of Polygonum amphibium L., trans-taxifolin, quercetin and kaempferol were isolated The structure of the isolated compounds was determined by spectroscopic (UV, (1)H NMR, (13)C NMR and CI-MS) methods.

Shade tolerance plasticity in response to neutral vs green shade cues in Polygonum species of contrasting ecological breadth.

Here we examined species differences in perception and response to two distinct types of shade cue, reduced photosynthetically active radiation (PAR) with and without reduced red : far red ratio (R : FR), in Polygonum persicaria and Polygonum hydropiper, two closely related annuals of contrasting ecological breadth. We compared plasticity data for light-gathering traits from glasshouse experiments at equivalently reduced PAR under neutral shade (R : FR 1.03) and green shade (R : FR 0.702). Species shared the ability to distinguish between the two types of shade, as shown by the ability of each to respond differently to neutral vs green shade for one or more traits. However, the species' responses to these cues differed significantly. Polygonum persicaria expressed stronger shade-tolerance responses (increased leaf allocation and leaf area ratio) to reduced PAR alone than to green shade. By contrast, P. hydropiper expressed slightly less plasticity for these traits in neutral than in green shade. The pronounced plastic response of P. persicaria to neutral shade may contribute to the range of habitats this widespread species can occupy, which includes neutral-shade environments such as urban settings.

[Preliminary survey on medicinal plants of Polygonum in Gansu Province].

OBJECTIVE: To gain a clear idea on the resources of medicinal plants of Polygonum in Gansu. METHODS: Field investigation, specimen collection, taxonomic study, scientific names study and literature retrieval were carried out. RESULTS: The distribution, ecological environment and medicinal parts of 23 species and 7 varieties of Polygonum from Gansu have been clarified. CONCLUSION: The investigation may prove to be useful for the utility and further study of Polygonum in Gansu province.

[Effect on gastrointestinal functions of Polygonum paleaceum].

OBJECTIVE: To study the effect on gastrointestinal functions of Polygonum paleaceum Wall. METHODS: Defecation reflex, gastric emptying, motility of the gastroinestinal tract of mice and isolated intestinal smooth muscle of rabbits were used. RESULTS: Polygonum paleaceum Wall could inhibit defecation reflex and depress the motility of the gastrointestinal tract, delay gastric emptying in mice. It also had analgesia action, but didn't affect rabbit intestinal contraction spontaneously or induced by acetylcholine (Ach) in vitro. CONCLUSION: It suggested that Polygonum paleaceum Wall possessed the action on gastrointestinal functions.

Analysis of estrogenic compounds in Polygonum cuspidatum by bioassay and high performance liquid chromatography.

The estrogenic activity of traditional Chinese herb-Polygonum cuspidatum Sieb. et Zucc. was investigated by a recombinant yeast screening (YES) assay. Anthraquinones are the main components in the plant, of which emodin is the most abundant one. The ethyl acetate fraction of the ethanol extract of Polygonum cuspidatum was separated on a silica gel TLC plate and seven sub-fractions were collected. The results of bioassay demonstrated that Hzs1 and Hzs6 showed higher estrogenic activities than that of others and the potency of these two compounds were approximately 10(-4) g/L and 10(-3) g/L, respectively. HPLC analysis was performed to determine the activities and the active components. Combining the results of HPLC analysis and estrogenic activity test by YES led to the conclusion that an unknown bioactive compound might exist in the extraction of Polygonum cuspidatum.

The investigations into the interferon-like activity of Polygonum L. genus.

The interferon inducing abilities of eleven ethanolic extracts obtained from nine taxons of Polygonum L. genus, were tested in the cell culture of monkey kidney. The extracts from the herb of Polygonum amphibium L. and the rhizome and fruit of Polygonum bistorta L. induced a substance showing an interferon-like activity. Biological activity studies showed that the protective titre (the highest dilution which protected cells by 50% against virus infection) of the interferon-like materials was 1:10-1:15.

[Effects of meteorological factors and mineral elements on the content of resveratrol in Polygonum cuspidatum rootstalk].

By determining monthly the content of resveratrol and eight mineral elements (Ca, Cu, Fe, K, Mg, Na, Zn, Mn) in the rootstalk of Polygonum cuspidatum, by considering the dynamic meteorological factors and by analyzing the principal components, we are investigating the dynamic relationship laws among mineral elements, meteorological factors and the content of resveratrol in Polygonum cuspidatum rootstalk in varied seasons. The results indicated that the correlation coefficient between Cu and Fe; between Mg and K; between Mg and Zn; between K and Zn; between Ca and average sunlight hours per month are 0.812, 0.871, 0.793, 0.729, 0.602. Their correlation is markedly in the positive 0.01 levels. The correlation coefficient between Cu and average air temperature in a month; between Mn and average air temperature in a month; between Fe and K are -0.738, -0.712, -0.766. Their correlation is markedly in negative 0.01 levels. The correlation between Cu and the content of resveratrol is markedly in negative 0.05 levels. Of the five principal factors affecting the content of resveratrol, two were found to be principal: "mineral element factors of promoting enzyme reacts" and "the meteorological factors of high temperature and strong shines". The percentage results of these two principal are 33.539% and 33.358%, making a total of 68.897%. The two factors above are also the main reasons for the variation of Polygonum cuspidatum resveratrol. A regression equation was set up through correlation and regression analysis between the changing content of resveratrol and the principal component.

[Studies on active substance of anticancer effect in Polygonum cuspidatum].

OBJECTIVE: To supply the scientific basis of research and development of the medicinal value of Polygonum cuspidatum. METHODS: One composition was isolated from the roots of Polygonum cuspidatum by cytotoxicity based fractionation and identified by HPLC-MS, UV scanning and IR. The inhibition and morphology of L-02, Hep G2, SHZ-888, MCF-7, MCF-7/ADM cells growth caused by this composition was determined by MTT assay and HE dyeing. RESULTS: This composition was identified as trans-and cis-resveratrol. It could specifically inhibit proliferation of many cancer cells but not human normal liver cell. We investigated the cytotoxicity of resveratrol to adriamycin-resistant MCF-7 cell in virtro. CONCLUSION: Resveratrol is a new anticancer composition which is less toxicity and higher efficiency in Polygonum cuspidatum.

[Pharmacological studies of Polygonum capitatum Buch-Ham. ex D. Don].

The pharmacological effect of aqueous extract of Polygonum capitatum has been studied. The experimental results show that Polygonum capitatum markedly decreases WBC and RBC in urine of pyelonephritis mode in rats, the death rate of Escherichia coli infected mice, and the temperature of feverish rabbits. It has also been found that after oral administration of Polygonum capitatum the animal urine markedly inhibits the growth of Escherichia coli, but diuretic action of the herb has not been observed.

An unusual homoisoflavanone and a structurally-related dihydrochalcone from Polygonum ferrugineum (Polygonaceae).

The homoisoflavanone 5,7-dihydroxy-6-methoxy-3-(9-hydroxy-phenylmethyl)-chroman-4-one (1) and its structurally related 2',4',6'-trihydroxy-3'-methoxy-alpha-hydroxymethyl-beta-hydroxy-dihydrochalcone (2) along with the known pashanone (3), flavokawin B (4) and cardamonin or alpinetin chalcone (5) pinostrobin (6) and 5,8-dimethoxy-7-hydroxychroman-4-one (7) were isolated from dry leaves of Polygonum ferrugineum (Polygonaceae). To our knowledge, this is the first report of the isolation of a homoisoflavanone from the Polygonum genus and the Polygonaceae family, and could be an important chemotaxonomic finding. In addition, the pattern of substitution of this homoisoflavanone is different from others previously reported.

Quantitative analysis of indigo and indigo precursors in leaves of Isatis spp. and Polygonum tinctorium.

Analysis of extracts from two woad species (Isatis tinctoria and Isatis indigotica) and Polygonum tinctorium revealed that only one indigo precursor (indican) was present in Polygonum, but two precursors were found in Isatis spp. This was done using high performance liquid chromatography (HPLC), coupled to an evaporative light scattering detector (ELSD). In Isatis spp., the indigo precursors indican and a fraction representing isatan B were identified. The proportion of indican and isatan B was different between the two Isatis spp. tested. For the first time, it was possible to quantify the precursors in woad plant species, and the results were found to be in good agreement with those made from total indigo quantification using two different spectrophotometric methods or a derivatization technique.

In vivo antioxidant action of a lignan-enriched extract of Schisandra fruit and an anthraquinone-containing extract of Polygonum root in comparison with schisandrin B and emodin.

The in vivo antioxidant action of a lignan-enriched extract of the fruit of Schisandra chinensis (FS) and an anthraquinone-containing extract of the root of Polygonum multiflorum (PME) was compared with their respective active constituents schisandrin B (Sch B) and emodin by examining their effect on hepatic mitochondrial glutathione antioxidant status in control and carbon tetrachloride (CCl 4 )-intoxicated mice. FS and PME pretreatments produced a dose-dependent protection against CCl 4 hepatotoxicity, with the effect of FS being more potent. Pretreatment with Sch B, emodin or alpha-tocopherol (alpha-Toc) also protected against CCl 4 hepatotoxicity, with the effect of Sch B being more potent. The extent of hepatoprotection afforded by FS/Sch B and PME/emodin pretreatment against CCl 4 toxicity was found to correlate well with the degree of enhancement in hepatic mitochondrial glutathione antioxidant status, as evidenced by increases in reduced glutathione level and activities of glutathione reductase, glutathione peroxidase as well as glutathione S-transferases, in both control and CCl 4 -intoxicated mice. alpha-Toc, which did not enhance mitochondrial glutathione antioxidant status, seemed to be less potent in protecting against CCl 4 hepatotoxicity. The ensemble of results indicates that FS/PME produced a more potent in vivo antioxidant action than alpha-Toc by virtue of their ability to enhance hepatic mitochondrial glutathione antioxidant status and that the differential potency of FS and PME can be attributed to the difference in in vivo antioxidant potential between Sch B and emodin. Long-term administration of Polygonum multiflorum Thunb. reduces cerebral ischemia-induced infarct volume in gerbils.

Focal cerebral ischemia was produced by an occlusion of the middle cerebral artery for 1, 3 and 24 hours in gerbils. Infarct volumes were determined by 2,3,5-triphenyltetrazolium chloride (TTC) transcardiac perfusion 24 hours after cerebral ischemia. Significant and consistent infarct sizes were produced in gerbils subjected to 24-hour occlusion of the middle cerebral arterey when compared to the 1 and 3-hour occlusion group. Longterm pretreatment of the 50% ethanol extract of Polygonum multiflorum Thunb. for 2 weeks significantly reduced the infarct volume by 50% as compared to that of the 24-hour occlusion group. The results revealed that long term pretreatment of Polygonum multiflorum Thunb. may protect the brain against focal cerebral ischemia.
