[Relationship between means of administration, concentration in the blood, organ distribution and excretion of prespidin-14C from the body].

Blood content, distribution among organs and tissues and passage from the organism of prospidine-C14 in rats, following its introduction per os and application to the skin in the form of an ointment, were studied. With these modes of introduction it is shown that prospidine and/or products of its biotransformation are capable of relatively quickly penerating through the wall of the gastro-intestinal tract and skin. However, specific radioactivity in the organs in such cases is scores of time lower than with intravenous administration. In case of the peroral introduction of the compound 72.3 per cent of the amount of the compound and/or of its metabolites administered are eliminated via the gastro-intestinal tract in two days.

[The effect of 11-deoxycortiocosterone acetate on the antitumor activity of prospidin].

Male rats with sarcoma 45 being intact with respect to the adrenal gland and thyroid were subjected to bilateral adrenalectomy or thyroidectomy. The experiments have indicated that 11-desoxycorticosterone acetate (DOCA) injected intramuscularly in the dosage of 30-45 and 80-90 mg/Kg daily during 6-8 days (terms of observation-14-16 days since the moment of tumor transplantation) lessens the antiblastic effect of prospidine (injected intraperitoneally in the same dosage and regimen), especially in an impaired mineral metabolism, caused by adrenalectomy. The growth of sarcoma 45 was found to be retarded in bilateral adrenalectomy or thyroidectomy in rats. Epinephrectomized animals showed the entire absence of this effect in DOCA injection. In thyroidectomized and adrenalectomized animals the antiblastic effect of prospidine was more pronounced than in intact (with respect to the thyroid and adrenal gland) rats.

Adrenalectomy

[Prospidin-C14 blood level, distribution in the organs and tissues and excretion from the body of rats].

Experiments were conducted on rats with sarcoma-45 and intact ones to study the prospidin-C14 content in the blood, distribution of the drug among the organs and tissues and its elimination from the organism. A single intravenous introduction of prospidin-C14 was found to be followed by its quick (in 2 hours) disappearance frob the blood. During the first 15 minutes the distribution of the drug over the organs and tissues was of a non-uniform nature and different from the distribution of other known antineoplastic agents, with relatively high concentrations of prospidin-C14 being demonstrable in the kidneys, lungs, the skin, intestine, bones, pancreas and low ones--in the liver, spleen and lymph nodes. The elimination of the drug from the organs and tissues is non-uniform. Prospidin-C14 and labeled products of its transformation are quickly (during the first 24 hours after administration) passed from the organism, chiefly with urine.

[An electron microscopic study of the behavioral characteristics of human lung tumor cells cultured on soft agar].

The ultrastructure of cells from seven human lung cancers and from the colonies formed by these cells in soft agar was investigated. Tumor cells developed to display the morphofunctional potentials of the initial tumors. Cultured cells of squamous-cell carcinomas contained numerous tonofilaments, those of adenocarcinomas developed microvilli on their apical surfaces and intracellular lumens. On the other hand, cells of squamous-cell carcinomas showed features specific of adenoma epithelium, i.e. well developed microvilli and intracellular lumens. Besides, cells of adenocarcinoma often contained large quantities of tonofilaments considered to be characteristic of epidermoid epithelium. The results obtained suggest a possibility of metaplastic transformation of the lung epithelium.

Adenocarcinoma

[Use of cytostatics as anti-cicatricial drugs after anti-glaucoma surgery in children, adolescents and young adults].

Anti-cicatricial cytostatic therapy with 5-fluorouracil and prospidin was carried out after antiglaucoma surgery in 78 eyes of 67 patients aged 6 to 42 (2/3 of the patients were aged from 21 to 40). Half (34) of the patients suffered from open-angle (pigmented in 3 cases) and 6 from closed-angle glaucoma, the rest patients presented with common hydrophthalmia, the Frank-Kamenetsky, Sturdge-Weber, Riger's syndromes, etc. The cytostatics were injected subconjunctivally. The results of 67 surgeries were followed up for 6 months to 3 years. Stable normalization of the ophthalmic tone and stabilization of the process were achieved in 60 eyes (89.2%). The authors emphasize that cytostatics may be recommended for children and young subjects after surgery involving fistulization, on condition of thorough biomicroscopic monitoring in the postoperative period, because of a possible disadaptation of the edges of the mucosal incision and corneal diseases.

Adolescent

[Results of radiation and chemoradiation therapy in esophageal cancer].

Eight therapeutic variants were worked out for 602 patients with esophageal cancer: 61 patients entered in the control group. The latter received routine radiation therapy. The results of therapy of 496 patients were studied. The most promising method was chemoradiotherapy using multifractionation of a daily dose of irradiation, the 4-year survival rate of 20% being achieved. For combination with radiation therapy the following schemes of chemotherapeutic agents were used bleomycin-methotrexate-prospidine, bleomycin-adriamycin-methotrexate, dactinomycin-methotrexate-prospidine, methotrexate-prospidine. Intensive detoxication therapy with parenteral feeding is recommended to decrease the frequency of complications and to improve therapy tolerance.

Antineoplastic Combined Chemotherapy Protocols

[Treatment of maxillary cancer].

The experience of the Moscow Research Institute of Oncology named after P. A. Herzen with the treatment of 368 primary patients with malignant tumors of the upper jaw, and the analysis of the literature data allowed a formulation of basic tendencies in therapy of this affection. These include a wide use of the combination therapy with preoperative radiation treatment; an elaboration of some extended variants of surgical interventions; an improvement of radiation technics, use of drug therapy, mainly as regional chemotherapy associated with radiotherapy. Further elaboration of the early diagnostics is also of importance. Patients in whom malignant tumors of the upper jaw were detected should be treated in specialized oncological departments, specially equipped and having well-trained medical staff.
