Characteristics of two cell lines (TE-1 and TE-2) derived from human squamous cell carcinoma of the esophagus.

Two epithelial cell lines (TE-1 and TE-2) have been established from a well or poorly differentiated human squamous cell carcinoma of the esophagus. TE-1 has been subcultured 120 times during 2 years and 10 months, and TE-2, 50 times for almost 2 years. Cultured cells grew as isolated and piled-up colonies of epithelial cells. The average doubling time of the TE-1 cell line was 60 hr and that of TE-2, 72 hr. Distinctive marker chromosomes and a male karyotype were present in TE-1, but no marker chromosomes were seen in TE-2. Scanning electron microscopic examinations of both TE-1 and TE-2 confirmed the presence of desmosomes and interdigitated microvilli. Transmission electron micrographs of TE-1 showed the presence of abundant cell organelles, and a few organelles were found in the scanty cytoplasm of TE-2. There was a marked difference in the cell organelles between TE-1 and TE-2. Heterotransplantation of the cultured TE-1 and TE-2 cells produced tumors, the histological appearance of which was similar to that of the original ones. The carcinoembryonic antigen level of the medium in the confluent culture of TE-2 was 270 ng/10(6) cells. In the cytoplasm of TE-1 cells the number of paracrystals, which were produced by treatment with vinblastine sulfate, increased by the addition of cholera toxin to the medium.

Energy dependence of the neutron sensitivity of C--CO2, Mg--Ar and TE--TE ionisation chambers.

The neutron sensitivity relative to 60Co of commercially available C--CO2, Mg--Ar and TE--TE ionisation chambers was measured as a function of energy from 1 to 44 MeV. The sensitivity function was obtained by the method of Kuchnir, Vyborny and Skaggs from differences in measurements made at two angles in mixed fields having an isotropic gamma-ray component. Such fields were produced by bombardment of a thick beryllium target with 16 and 28 MeV deuterons, 44 MeV 3He-ions and 35 and 46 MeV protons. The results show that the relative neutron sensitivity of the C--CO2 and Mg--Ar chambers increases continuously with energy, whereas that of the TE--TE chamber is relatively constant.

Unusual manifestation of tuberculosis: TE fistula.

Fistulas between the tracheobronchial tree and the esophagus (TE fistula) caused by tuberculosis are rare; usually they are associated with readily apparent pulmonary and/or mediastinal infection, and require surgical management. The patient we describe presented with a TE fistula as the only manifestation of active tuberculosis. This case represents the first report of successful nonsurgical treatment of tuberculous TE fistulas.

[Increase functions of the type dw/dt = kwm(te--t)q and their integrals (author's transl)].

In consequent continuation of the investigations into the increase functions dW/dt = kWm(En--Wn), dW/dt = kWm(E--W)n and dW/dt = ktp (E--W)n the type dW/dt = kWm(tE--t)q with q greater than 0 is added. As in the last paper, 2 cases of integration have to be distinguished, namely for m = 1 and 0 less than m less than 1 respectively. In both cases adultness W = E is reached after a finite time tE. A quite different aspect results for the turning point of the growth function with the ordinate covering values between E/e and E for m = 1 and from 0 up to a maximum and down to E/e when 0 less than m less than 1. As in former papers graphs give an illustration of the variability of the growth curves and the increase functions. The already used example is taken up and shows a fargoing neighbourhood of the growth functions resulting from dW/dt = kWm (tE--t)q and dW/dt = ktp (E--W)n, thus hinting that quite different increase equations can lead to similar results.

Characterisation of Trichuris incognita n sp in Côte d'Ivoire: a morphological, genomic, and genome-wide association with drug sensitivity study.

BACKGROUND: Trichuriasis is a neglected tropical disease that affects up to 500 million individuals and can cause considerable morbidity. For decades, trichuriasis was thought to be caused by one species of whipworm, Trichuris trichiura. The aim of this study was to investigate the origin of differences in response rates to the best available anthelmintic treatment for trichuriasis-a combination of albendazole and ivermectin-in Côte d'Ivoire by analysing the parasite population. METHODS: In this morphological, genomic, and genome-wide association study (GWAS) with drug sensitivity we used long-read and short-read sequencing approaches and assembled a high-quality reference genome of Trichuris incognita n sp isolated in a primary interventional study conducted in the Lagunes district of Côte d'Ivoire. Children aged 6-12 years were screened between July 14, 2022, and July 31, 2022; children positive for T trichiura on duplicate Kato-Katz smears and with infection intensity of 200 eggs per gram or more were eligible and treated first with albendazole (400 mg) and ivermectin (200 μg/kg) then with oxantel pamoate (20 mg/kg). We constructed a species tree of the Trichuris genus using 12 434 orthologous groups. We sequenced individual worms, which were used to confirm the phylogenetic placement and investigate patterns of adaptation through comparative genomic analyses. Finally, we conducted a GWAS to compare albendazole-ivermectin sensitive worms to drug non-sensitive worms. FINDINGS: 670 children were screened, of whom 243 were enrolled and from whom 271 worms were isolated after the first treatment and 827 worms after the second treatment. Sufficient DNA was recovered from 747 worms of which 721 were suitable for further bioinformatic analysis; of these, 179 were albendazole-ivermectin sensitive worms and 542 were drug non-sensitive worms. We present and characterise a new, human-infecting Trichuris species named T incognita n sp, which is morphologically indistinguishable from T trichiura, but forms a distinct phylogenetic clade, closer to Trichuris suis than to the canonical human-infective T trichiura. Comparative genomic analysis of genes suspected to confer resistance to either albendazole or ivermectin in helminths revealed a high number of β-tubulin orthologs, present in the whole population of T incognita n sp, compared with the canonical T trichiura species, but these genes were not associated with a resistant phenotype. The GWAS did not provide conclusive evidence of adaptation to drug pressure within the same species. INTERPRETATION: Our results demonstrate that trichuriasis can be caused by multiple whipworm species, and that differences in response rates might result from species responding differently to drug treatment, rather than from the intraspecies establishment of resistance. This discovery, coupled with the high tolerability of T incognita n sp to albendazole-ivermectin, marks a substantial shift in how we understand and approach whipworm infections. Near-Whole-Genome Sequencing of Peste Des Petits Ruminants Virus Lineage IV From the Savannah District, Northern Côte d'Ivoire in 2023.

Peste des petits ruminants (PPR) is a highly contagious viral disease affecting sheep and goats, causing substantial economic losses in endemic countries. In the Savannah district of Côte d'Ivoire, knowledge of the genetic diversity and molecular epidemiology of the PPR virus (PPRV) remains limited. This study investigated the genetic diversity and phylogenetic relationships of PPRV circulating in this region using whole-genome sequencing (WGS). A cross-sectional survey was conducted between September and December 2023. Nasal swabs collected from sheep and goats were screened for PPRV ribonucleic acid (RNA) using real-time reverse transcription polymerase chain reaction (RT-qPCR). Samples with low quantification cycle (Cq) values of less than 35 and successful multiplex PCR amplification profiles were selected for sequencing using the Oxford Nanopore MinION platform. Near-complete consensus genomes were generated through reference-based assembly and analysed alongside representative strains from all recognised PPRV lineages. Of the 355 samples analysed, 25 (7.0%) tested positive for PPRV RNA, with positive detections in all three surveyed regions (Poro, Tchologo and Bagoué). The four samples with the lowest Cq values, originating from all three administrative regions, were successfully sequenced, generating genomes that covered 82.0%-86.2% of the reference genome at a depth of ≥ 10 ×. The missing regions were mainly located at the 5' and 3' genomic termini, as well as in limited internal regions associated with amplicon dropout. Phylogenetic analysis revealed that all four sequences belonged to lineage IV and exhibited high nucleotide similarity (98.1%-99.9%). The Ivorian strains clustered with recent lineage IV viruses from West, North and Central Africa, whereas historical Ivorian lineages I and II formed distinct clades. These findings confirm the predominance of lineage IV in northern Côte d'Ivoire and provide baseline genomic data to support molecular epidemiological surveillance in the region.

Isolation and partial purification of plasma membrane-associated antigens from human osteosarcoma (TE-85) cells in tissue culture.

The plasma membrane of a cloned line of TE-85 (human osteosarcoma) cells subcultured for the last 4 years was isolated. The isolation was by hypotonic swelling, cell homogenization, and discontinuous sucrose gradient ultracentrifugation for 16 hr. Tumor-specific water-soluble antigens were identified by limited papain digestion of the isolated plasma membrane. Fractionation by diethylamino-ethyl anion-exchange column chromatography yielded antigenic fractions that inhibited the reaction of immune serum (from an unrelated patient with osteosarcoma) with the plasma membrane of TE-85 cells in tissue culture by indirect immunofluorescence test. Microimmunodiffusion confirmed the specificity of the isolated antigen against the sera of other patients with osteosarcoma. The definition of the antigen fraction may permit evaluation of antigen-antibody interaction in tumor immunity.

Whole genome of Lasiodiplodia theobromae, Lasiodiplodia euphorbiaceicola, and Lasiodiplodia brasiliensis isolates associated with Mango Dieback and stem-end rot in Côte d'Ivoire.

Mango productivity is threatened by dieback and fruit rot from Lasiodiplodia spp. (Botryosphaeriaceae fungal family). We report high-quality genomes of Lasiodiplodia theobromae, Lasiodiplodia euphorbiaceicola, and Lasiodiplodia brasiliensis from Côte d'Ivoire, enhancing genomic resources for comparative and pathogenicity studies of these fungal plant pathogens.

Interplay between the role of DNA methylation in regulating gene expression and TE-silencing in a reptilian methylome.

DNA methylation is a major component of eukaryotic genomes with an important role in the defence against transposable elements, to transcriptionally silence their activity and prevent transposition. DNA methylation also plays a major role in the regulation of gene expression. This dual role can come into conflict, where DNA methylation in gene regulatory regions becomes perturbed due to transposable element transposition, leading to disruption of gene expression. Here, we describe how this conflict is reflected in DNA methylation patterns in the sand lizard genome where there is recent transposable element activity. Using long-read sequencing technology we show that CpG islands in gene transcriptional start sites are typically hypomethylated and associated with higher gene expression. Outside transcriptional start sites, a majority of CpG islands overlapped transposable elements and were associated with hypermethylation, consistent with a host-defence role in suppressing transposition activity. We identify 605 instances where transcriptional start sites were associated with transposable elements (4.3% of all genes). These instances were far rarer in conjunction with a CpG island, when methylation signatures would be in conflict. Transposable elements were found to be closer to and at higher density the more hypermethylated a transcriptional start site was, suggesting strong selection against selfish genetic elements transposing into hypomethylated transcriptional start sites.
