[Factors influencing pathogenicity of Collectotrichum gloeosporioides f.sp. veronicae to Veronica persica].

The effects of the characters of the fungus, the growing period of plants and environmental factors on the pathogenicity of Colletotrichum gloeosporioides f.sp. veronicae infecting Veronica persica were studied in pot experiments. The results showed that 1.08 x 10(8) spores.ml-1 suspension could kill the seedlings with two cotyledons within one week and cause old plants serious disease. 1 x 10(8) spores.ml-1 was optimal concentration and 1 or 2 weeks old was the best age of the spores for killing birdseye speedwell. The optimal temperature was 15-25 degrees C for the disease development. More than two days' dew exposure was required to kill the weed. However, the disease was also developed better when corn or soybean flour was added or the darkness period during dew exposure was prolonged.

Insights into phylogenetic relationships of Veronica species (Plantaginaceae) based on comparative chloroplast genomics.

INTRODUCTION: Veronica L. is one of the most species-rich genera in Plantaginaceae and several species have medicinal, horticultural, or ecological value. METHODS: In this study, the complete chloroplast genomes of three Veronica species were assembled and annotated using Illumina sequencing data. RESULTS: The plastomes exhibited a typical quadripartite structures, with total lengths of 150,202 bp for Veronica biloba L., 151,159 bp for Veronica ciliata Fisch. and 151,098 bp for Veronica vandellioides Maxim. Each genome contained 130-132 unique genes, including 86-87 protein-coding genes, 36-37 tRNA genes, and 8 rRNA genes. Comparative analyses of 24 Veronica plastomes indicated that the IR/SC junctions were largely conserved, although slight boundary shifts occurred around rps19, ndhF, and ycf1. Forward, palindromic, complement, and reverse repeats were detected, and A/T mononucleotide repeats were the dominant SSR type. Nucleotide diversity analysis identified rpl32-trnL, trnK-rps16, rpl32, ycf1, ndhF, accD, matK, and rpoB as highly variable regions. Phylogenetic analyses recovered Veronica as a well-supported monophyletic lineage and clarified the plastid positions of the three newly sequenced species. Divergence time estimation suggested that the estimation suggested of Veronica was around 14.9 Ma, with V. biloba, V. ciliata and V. vandellioides diverging approximately 3.9 Ma, 0.6 Ma, and 6.9 Ma, respectively. DISCUSSION: Because the analyses were based on plastid genomes, the inferred topology should be interpreted as chloroplast phylogenetic evidence rather than a complete species-history reconstruction. These results provide plastome resources and molecular evidence for taxonomy, species identification, and future evolutionary studies of Veronica.

Iridoid glucosides of Paederota lutea and the relationships between Paederota and Veronica.

In a chemical investigation of the water soluble compounds in Paederota lutea eight known iridoids were isolated together with a new one with a 8,9-double bond, namely paederotoside (10-O-benzoyl-6'-O-alpha-arabino(1-->6)-beta-glucopyranosyl arborescosidic acid) and the 6-hydroxy-flavone glucoside 4'-O-methylscutellarein 7-O-beta-glucopyranoside. The known iridoid glucosides were 8-epiloganic acid, gardoside, aucubin, catalpol and the 6-O-esters of catalpol: veronicoside, catalposide, amphicoside and verproside. The compounds isolated show that Paederota has a glycoside content almost identical to that of a general Veronica species, and this is in good agreement with the results from recent investigations of the phylogeny of Veronica and its closest relatives, where Paederota is placed as a sister-group next to Veronica. In an analysis of the iridoid glucosides from some of these relatives, it is shown that Veronica, Paederota, Picrorhiza and Veronicastrum are all characterized by containing 6-O-esters of catalpol. Some less closely related taxa namely: Lagotis, Wulfenia, Plantago, Aragoa and Globularia instead contain 10-O-esters of catalpol or aucubin.

Phylogeny of Veronica--a combination of molecular and chemical evidence.

A molecular phylogenetic tree of the genus Veronica, based on sequences of the ITS region of nuclear ribosomal DNA, provided a framework to test the use of iridoid chemical data in the classification of the genus. The distribution of ten iridoid glucosides, determined in 27 species of Veronica, supported the molecular results. The nine clades of the molecular tree appeared to correlate with a specific pattern of catalpol esters and chromosome base numbers. Some relationships within Veronica, previously suggested on the basis of morphological and karyological characters, were supported by our data. Neither the molecular, nor the chemical features were in favour of the classical circumscription and classification of the genus.

Systematic significance of seed morphology in veronica (plantaginaceae): a phylogenetic perspective.

BACKGROUND AND AIMS: A new infrageneric rearrangement for Veronica has been proposed based on the most recent evidence from DNA sequence data, morphological evidence, and biogeographical considerations. Looking for morphological synapomorphies for each monophyletic subgenus has been problematic, due to difficulties arising from widespread homoplasy (mainly parallel evolution). In an attempt to overcome these difficulties, previously underexplored morphological characters are starting to be studied in more depth. METHODS: A molecular phylogenetic hypothesis was used based on sequences of ITS (nuclear ribosomal DNA) and plastid trnL-F regions, as a framework to test the use of seed coat ultrastructure (studied under scanning electron microscope) in the systematics of the genus. A sample of 132 taxa representing ten of the 13 subgenera in Veronica, excluding the species of the southern hemisphere Hebe complex and the exclusively North American subgenus Synthyris, was studied. KEY RESULTS AND CONCLUSIONS: The results demonstrate that, in many cases, the ultrastucture of the testa can be employed to assess relationships of taxa within the genus, and the character provides additional support for molecular trees. Further characters relevant for the classification of Veronica, i.e. base chromosome number, iridoid chemical data, life cycle, inflorescence position, have been taken into consideration in a discussion where an attempt is made to highlight the best traits to characterize each subgenus investigated.

Anti-inflammatory and cytotoxic activities of five Veronica species.

Biological activities of five Veronica species (Scrophulariaceae), V. cymbalaria, V. hederifolia, V. pectinata var. glandulosa, V. persica and V. polita were studied for their anti-inflammatory and cytotoxic activities. Their methanol extracts showed both the inhibitory activity of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated macrophages and cytotoxic activity against KB epidermoid carcinoma and B16 melanoma. When the methanol extracts were fractionated between water and chloroform, water fractions significantly inhibited NO production without any cytotoxicity, while chloroform fractions showed cytotoxicity dose-dependently. When the radical scavenging activity was determined using 2,2-diphenyl-1-picryl-hydrazyl (DPPH), water fractions of the five Veronica species scavenged free radicals effectively, suggesting that the inhibitory effect of this species on NO production was due to their radical scavenging activity. On the other hand, chloroform fractions of Veronica species except for V. cymbalaria showed similar cytotoxic activity against KB and B16 melanoma cells.

Bioassay-guided isolation of iridoid glucosides with antinociceptive and anti-inflammatory activities from Veronica anagallis-aquatica L.

Extracts obtained from the herbs of various Veronica species are used as folk remedy worldwide for the treatment of various inflammatory ailments including rheumatism. In vivo anti-inflammatory and antinociceptive activities of Veronica anagallis-aquatica L. aerial parts were investigated. Methanolic extract of the plant was shown to possess significant inhibitory activity against carrageenan-induced hind paw edema model and of p-benzoquinone-induced writhings in mice. Through bioassay-guided fractionation and isolation procedures eight compounds, aquaticoside A (1), aquaticoside B (2), aquaticoside C (3), veronicoside (4), catalposide (5), verproside (6), verminoside (7) and martynoside (8) were isolated and their structures were elucidated by spectral techniques. Catapol derivative iridoid glucosides, verproside (6) and catalposide (5), were found to possess potent antinociceptive and anti-inflammatory activities, per os without inducing any apparent acute toxicity as well as gastric damage. Results of the present study supported the utilization of the plant in Turkish folk medicine.

Acylated flavone glycosides from Veronica.

A survey of the flavonoid glycosides of selected taxa in the genus Veronica yielded two new acylated 5,6,7,3',4'-pentahydroxyflavone (6-hydroxyluteolin) glycosides and two unusual allose-containing acylated 5,7,8,4'-tetrahydroxyflavone (isoscutellarein) glycosides. The new compounds were isolated from V. liwanensis and V. longifolia and identified using NMR spectroscopy as 6-hydroxyluteolin 4'-methyl ether 7-O-alpha-rhamnopyranosyl(1"'-->2")[6"-O-acetyl-beta-glucopyranoside] and 6-hydroxyluteolin 7-O-(6"-O-(E)-caffeoyl)-beta-glucopyranoside, respectively. Isoscutellarein 7-O-(6"'-O-acetyl)-beta-allopyranosyl(1"'-->2")-beta-glucopyranoside was obtained from both V. intercedens and V. orientalis and its 4'-methyl ether from V. orientalis only. Complete 1H and 13C NMR spectral assignments are presented for both isoscutellarein glycosides. Two iridoid glucosides new to the genus Veronica (melittoside and globularifolin) were also isolated from V. intercedens.

Acylated flavonoids and phenol glycosides from Veronica thymoides subsp. pseudocinerea.

A new acylated flavone glucoside, 3'-hydroxyscutellarein 7-O-(6''-O-protocatechuoyl)-beta-glucopyranoside (1), and a new phenol glucoside, 3,5-dihydroxyphenethyl alcohol 3-O-beta-glucopyranoside (6) were isolated from Veronica thymoides subsp. pseudocinerea together with seven known flavone, phenol and lignan glycosides; 3'-hydroxyscutellarein 7-O-(6''-O-trans-feruloyl)-beta-glucopyranoside (2), 3'-hydroxy, 6-O-methylscutellarein 7-O-beta-glucopyranoside (3), luteolin 7-O-beta-glucopyranoside (4), isoscutellarein 7-O-(6'''-O-acetyl)-beta-allopyranosyl (1''' --> 2'')-beta-glucopyranoside (5), 3,4-dihydroxyphenethyl alcohol 8-O-beta-glucopyranoside (7), benzyl alcohol 7-O-beta-xylopyranosyl (1" --> 2')-beta-glucopyranoside (8), and (+)-syringaresinol 4'-O-beta-glucopyranoside (9). Compounds 2, 3 and 7-9 were reported for the first time in the genus Veronica. The structures of the isolates were determined by means of spectroscopic (UV, IR, 1D and 2D NMR, HR ESI-MS) methods. Isolated compounds (1-7) exhibited potent radical scavenging activity against the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical.

External flavonoid aglycones from Veronica chamaedrys L. (Scrophulariaceae).

External flavone aglycones in the overground parts of Veronica chamaedrys L. (Scrophulariaceae) have been analysed by isocratic reversed-phase high performance liquid chromatography. Luteolin, apigenin, luteolin-3'-methyl ether and scuterallein-6,4'-dimethyl ether have been identified. The latter compound is a new flavonoid identified in Veronica genus.

Experimental anti-ulcer activity of Veronica officinalis L. extracts.

In indomethacin-induced ulcers in albino rats, the anti-ulcerogenic effects of some extracts prepared from Veronica officinalis L. were investigated. The extracts had a significant anti-ulcerogenic activity. In ulcer healing experiments performed in rats by administering reserpine, the extracts were found to enhance the regeneration of the gastric mucosa. These results seem to confirm the popular observations according to which the decoction from Veronica officinalis L. possessed useful properties in the treatment of gastric ulcers.

Three new (1-3) and four known iridoid glucosides (4-7) as well as a known phenylethanoid glycoside (8) were isolated from the aerial parts of Veronica anagallis-aquatica and their structures were determined as 6'-O-benzoyl-8-epiloganic acid named aquaticoside A (1), 6'-O-p-hydroxybenzoyl-8-epiloganic acid named aquaticoside B (2), 6'-O-benzoyl-gardoside named aquaticoside C (3), veronicoside (4), catalposide (5), verproside (6), verminoside (7) and martynoside (8) on the basis of 1D and 2D NMR spectral analysis.

Genome size variation and evolution in Veronica.

BACKGROUND AND AIMS: The amount of DNA per chromosome set is known to be a fairly constant characteristic of a species. Its interspecific variation is enormous, but the biological significance of this variation is little understood. Some of the characters believed to be correlated with DNA amount are alpine habitat, life history and breeding system. In the present study, the aim is to distinguish between direct causal connections and chance correlation of the amount of DNA in the genus Veronica. METHODS: Estimates of DNA amount were analysed for 42 members of Veroniceae in connection with results from a phylogenetic analysis of plastid trnL-F DNA sequences and tested correlations using standard statistical tests, phylogenetically independent contrasts and a model-based generalized least squares method to distinguish the phylogenetic effect on the results. KEY RESULTS: There appears to be a lower upper limit for DNA amount in annuals than in perennials. Most DNAC-values in Veroniceae are below the mean DNA C-value for annuals in angiosperms as a whole. However, the long-debated correlation of low genome size with annual life history is not significant (P = 0.12) using either standard statistical tests or independent contrasts, but it is significant with the generalized least squares method (P < 0.01). CONCLUSIONS: The correlation of annual life history and low genome size found in earlier studies could be due to the association of annual life history and selfing, which is significantly correlated with low genome size using any of the three tests applied. This correlation can be explained by models showing a reduction in transposable elements in selfers. A significant correlation of higher genome sizes with alpine habitats was also detected.

Comparative phylogeography of the Veronica alpina complex in Europe and North America.

The Veronica alpina complex comprises eight species of alpine habitats over a wide range of mountain systems in the Northern Hemisphere. The occurrence of sympatric species in the European and North American mountain systems allowed us not only to investigate the effect of the ice ages on intraspecific phylogeographical patterns and genetic diversity in different continents of the Northern Hemisphere, but also to compare these patterns in closely related species. Plastid DNA trnL-F sequences and AFLP (amplified fragment length polymorphism) fingerprints were used to infer the phylogenetic history of the group and phylogeographical patterns within species. Hybrid origin of tetraploid eastern North American V. wormskjoldii from western North American V. nutans (= V. wormskjoldii s.l.) and Eurasian V. alpina is suggested. A number of phylogeographical groups have been found both in V. alpina from Europe and in V. nutans from western North America. Phylogeographical substructuring in the Alps is inferred for V. alpina but not for V. bellidioides, which is moreover characterized by an overall very low genetic diversity. Western North American V. cusickii is much more genetically diverse than its sympatric relative, V. nutans, an effect that is likely due to differences in the breeding system. Populations of V. nutans are differentiated into three groups, those from the Cascades and from the southern and the northern Rocky Mountains. Genetic diversity seems to be higher in the North American V. nutans than in the morphologically and ecologically similar European V. alpina. A possible scenario to explain this pattern is suggested.

Iridoid glucosides from Veronica hederifolia.

A new iridoid glucoside, urphoside A, and six known iridoid glucosides, pikuroside, aucubin, veronicoside, catalposide, amphicoside, and verminoside, were isolated from Veronica hederifolia together with a known megastigmane glucoside, 3-hydroxy-5,6-epoxy-beta-ionol-9-O-beta-D-glucopyranoside, and a hexitol, dulcitol. The structures of the isolated compounds were established by the extensive 1D- and 2D-NMR spectroscopy.

New phenylethanoid glycosides from Veronica pectinata var. glandulosa and their free radical scavenging activities.

A known phenylethanoid glycoside, ehrenoside (1), was isolated together with three new phenylethanoid glycosides, verpectoside A (2), B (3) and C (4) from the aerial parts of Veronica pectinata var. glandulosa. On the basis of spectral analysis (UV, FAB-MS, 1H-, 13C- and 2D-NMR), compounds 2-4 were determined to be 2-(3,4-dihydroxyphenyl)ethyl-O-alpha-L-arabinopyranosyl-(1-->2)-[alpha-L-rhamnopyranosyl-(1-->3)]-(4-O-trans-feruloyl)-beta-D-glucopyranoside, 2-(3,4-dihydroxyphenyl)ethyl-O-beta-D-glucopyranosyl-(1-->2)-[alpha-L-rhamnopyranosyl-(1-->3)]-(4-O-trans-caffeoyl)-beta-D-glucopyranoside and 2-(3,4-dihydroxyphenyl)ethyl-O-beta-D-glucopyranosyl-(1-->2)-[alpha-L-rhamnopyranosyl-(1-->3)]-(4-O-trans-feruloyl)-beta-D-glucopyranoside, respectively. Isolated phenylethanoid glycosides exhibited potent radical scavenging activity against the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical.

Iridoid glucosides from Veronica pectinata var. glandulosa.

A new highly oxygenated iridoid glucoside, urphoside B (1) was isolated from the Veronica pectinata var. glandulosa together with seven known iridoid glucosides, aucubin, catalpol, veronicoside, catalposide, verproside, amphicoside and 6-O-veratroyl catalpol. The planar as well as the stereo structures of the isolated compounds were determined by means of extensive 1D- and 2D-NMR spectroscopy and confirmed by HR-Mass.

Phenylethanoid glycoside from Veronica undulata.

From a water extract of whole plants of Veronica undulata, a new phenylethanoid triglycoside and five known glycosides were isolated. The structure of the new compound was elucidated from chemical and spectroscopic evidence.
